1.Effect of proanthocyaindin on angiogenesis in rats with ischemic hindlimb

Objective To investigate the effect of proanthcyaindin on angiogenesis of rats with ischemic hindlimb.Methods Twelve male SD rats were randomized divided into control group (n =6) and proanthcyaindin group (PC group,n =6).Lower limb ischemia rat model was establish,rats in both group were administration by oral daily,and PC group was given proanthocyanidins 200 mg/(kg · d),while the control group received the same volume of saline.Forteen days after surgery,ischemic tissues of adductor were collected for several tests including the expression of CD31 in the ischemic muscle tissue and the number of new blood vessels by immunohistochemical staining,hypoxia inducible factor-1 protein expression by western blot,and vascular endothelial growth factor level test by enzyme-linked immunosorbent assay.Results The capillary count showed that the angiogenesis situation of PC group was (69.67 ±3.11)/HP,higher than that in control group((111.00 ± 3.11)/HP,t =13.350,P < 0.0001).The HIF-1 protein expression in PC group was (1.90 ± 0.25),remarkable higher than that in control group (0.54 ± 0.21,t =4.183,P =0.0058).Compared with control group,VEGF level in PC group increased((432.86 ± 13.00) μg/L vs.(326.68 ± 11.08) μg/L,t =6.216,P <0.0001).Conclusion Proanthcyaindin plays a positive role in angiogenesis after ischemia.PC may induce up-regulation of HIF-1 and VEGF,and then promote the formation of endovascular through multiple signaling pathways consequently.
2.The role of resting metabolic rate in evaluation of Kidney Yin or Kidney Yang deficiency animal model induced by glucocorticoids

Objective To observe the role of resting metabolic rate (RMR) in evaluation of animal model of glucocorticoid-induced Kidney Yin deficiency and Kidney Yang deficiency syndrome.Methods Male BALB/c mice were divided into control group,model group,Jinkuishenqi pill group,and Zhibaidihuang pill group.The later 3 groups were given drinking water containing corticosterone (first dissolved in 1% ethanol,with a final concentration of corticosterone 100 μg/mL).The control group was given drinking water containing 1% ethanol.RMR was measured by closed fluid pressure respirometer.At the end of the experiment,the mice were sacrificed to detect the weight index of perirenal fat,epididymal fat,quadriceps,and tibialis anterior muscle.ELISA assay was used to detect the level of serum hormones.Histological changes of the liver and kidney were examined by HE staining.Malondialdehyde (MDA) content,succinate dehydrogenase (SDH) activity,cytochrome c oxidase (COX) activity and ATP level were measured.Results Compared with the control group,the RMR of model group was significantly increased at the 2nd day of beginning of the experiment,reached the highest on the 4th day (P<0.01),then decreased gradually,and to the 66th day,the RMR was significantly reduced (P<0.05).Use of corticosterone resulted in decrease of the serum levels of thyroxine (T4),muscle mass index,SDH activity,COX activity and ATP level,while increase of fat mass index and MDA level.The two Kidney nourishing prescriptions reduced animal mortality,and reduced the content of MDA in liver tissue.But only Jinkuishenqi pill increased the RMR at the 4th and 66th days (P<0.05),and significantly improved the liver SDH activity,COX activity and ATP level (P<0.01).The Zhibaidihuang pill showed no such effects.Conclusions RMR can be used for evaluation of animal model of Kidney Yin or Kidney Yang deficiency induced by glucocorticoids.

Humans are exposed to the ubiquitous radiofrequency (RF, 100 kHz-300 GHz) electromagnetic fields because of the mushroom development of wireless communications,raising concerns over the possible hazards of RF radiations.Epidemiological investigation has showed that chronic use of cellphones increases the risk of brain tumors.Since genetic damage is closely related to tumors, researchers have been trying to find out whether cellphones and other RF devices are genotoxic.However, the investigations have yielded both negative and positive results.This review summarized the recent in vitro and in vivo researches about genotoxicity of RF radiations and proposed a possible mechanism by which of RF radiations cause genetic damage.
4.The effects of B. adolescentis and L. acidophilus on proportion of Thl/Th2 in acute and convalescent UC-mice

Objective To observe and analyze the effect of B. Adolescentis and L. Acidophilus on the proportion of Th cell Th1/Th2 in peripheral blood of UC-mice in acute stage and recovery stage. Method 40 BABL/c mice were induced by 3% DSS water for 7 clays free drinking and then with distilled water for 10 days. They were randomly allocated in 4 groups: NS group, SASP group, BF0624 group and LT0637 group, also the fifth group-10 normal animals. The blood of mice were collected by removing their eyes at day 8 and day 18, and then the mononuclear cells were iso]atecl. The proportion of Th1/Th2 was analyze through flow eytometry,hy labeling the specific antibody ot Th cellular membrane with the CD4 antibody, and the cytoplastie antigen of Thl or Th2 with IIA antibody or IF'N-γ antibody. Result The proportion of Th1/Th2 in normal mouse was 0. 84 -0. 94,and it raised up in DSS-mice at both acute stage and recovery stage. It decreased unequally after 7 or 17 days'B, adoleseentis, L. Acidophilus and SASP treatment, but that of all three groups were lower than NS group (2.21±0. 83). Even the proportion got close to the normal animals after 17 days'L. Acidophilus -treated. Conclusion The proportion of Thl/Th2 increased at the acute stage and recovery stage of DSS-mice. Both B. Adoleseentis and L. Acidophilus had more effective than SASP on decreasing the proportion of Th1/Th2 at two stages,ospecially L. Acidophilus.
5.The effects of rehabilitation training on the expression of Nogo-A in the area surrounding cerebral infarc-tion

Objective To study the effects of rehabilitation training on the expression of Nogo-A around the area of cerebral infraction using rats. Methods A total of 60 Wistar rats were randomly divided into a reha- bilitation group and a control group after an experimental cerebral infarction had been established in them. The ani- mals in the rehabilitation group were given exercise with a rotating bar, a balance beam and a rolling cage for one hour daily, while those in the control group were caged without any abnormal exercise. Nogo-A expression in the ar- ea surrounding the infarcts was detected by imunohistochemical techniques at the 3rd, 7th, 14th, 21st and 28th day after infarction. Meanwhile, neurobehavioral evaluations were also conducted. Results The animals in the rehabilitation group scored much lower than the controls in the behavioral evaluations at the 14th, 21st and 28th day. The expression of Nogo-A in tissues around the infracted area increased by the 7th day and peaked at the 21st day in both groups, but the expression of Nogo-A was significantly stronger in the rehabilitation group at the 14th, 21st and 28th days. Conclusion Rehabilitation training decreased the expression of Nogo-A in the brain of rats after infarction. This may have important implications for the functional recovery of the central nervous system.
6.Bioinformatics Analysis of Clustered Regularly Interspaced Short Palindromic Repeats in the Genomes of Shigella.

This study was aimed to explore the features of clustered regularly interspaced short palindromic repeats (CRISPR) structures in Shigella by using bioinformatics. We used bioinformatics methods, including BLAST, alignment and RNA structure prediction, to analyze the CRISPR structures of Shigella genomes. The results showed that the CRISPRs existed in the four groups of Shigella, and the flanking sequences of upstream CRISPRs could be classified into the same group with those of the downstream. We also found some relatively conserved palindromic motifs in the leader sequences. Repeat sequences had the same group with corresponding flanking sequences, and could be classified into two different types by their RNA secondary structures, which contain "stem" and "ring". Some spacers were found to homologize with part sequences of plasmids or phages. The study indicated that there were correlations between repeat sequences and flanking sequences, and the repeats might act as a kind of recognition mechanism to mediate the interaction between foreign genetic elements and Cas proteins.

7.Clinical observation after Roux-en-Y gastric bypass in type 2 diabetes mellitus

Objective To observe multiple metabolic changes within one year after Roux-en-Y gastric bypass (RYGB) surgery in obese patients with type 2 diabetes mellitus, and to evaluate the index of the gastric bypass surgery and the determination of applicable population. Methods The clinical and laboratory data before and after surgery in 112 patients diagnosed as type 2 diabetes mellitus by RYGB were included in this study. According to BMI, these patients were divided into two groups: normal group (≤ 25kg/m2), and the overweight group (> 25 kg/m2). The physiologic and biochemical indexes of the patients were followed up at 1, 3, 6, 12 months, respectively. The statistical analysis was performed by SPSS17.0 software. Results Compared with the preoperative period, the levels of HbA1c and HOMA-IR in the postoperative period were significantly increased in the two groups. Principal component analysis showed that the postoperatively contributions of BMI and HbA1c in 6 months was bigger than that of the other indexes, while HOMA-β, HOMA-IR had larger contributions after 6-month postoperative period. Conclusion Various metabolic indexes in patients with type 2 diabetes improve significantly within one year after surgery, and the complete remission rate is gradually increased. The indexes including weight, blood glucose, serum lipids, HOMA-βand HOMA-IR in overweight and obese group have more significant improvements than those in normal group. Obese patients are more appropriate for the surgical treatment. The indexes, such as BMI, HbA1c,HOMA-βand HOMA-IR, should help to evaluate the operating effectiveness and preoperative indications.
8.Effect of dasatinib on biological properties of human bone marrow mesenchymal stem cells

AIM:To explore the effect of dasatinib on the viability, migration, cell cycle and apoptosis of human bone marrow mesenchymal stem cells (hBMSCs), as well as the underlying signal pathway to evaluate the influence of dasatinib on hematopoietic microenvironment clinically.METHODS:The cell viability was measured by CCK-8 assay.The migration ability was detected by wound healing assay.The cell cycle and apoptosis were analyzed by flow cytometry.Acridine orange/ethidium bromide staining was also used to detected apoptosis.The secretion of transforming growth factor-β1 (TGF-β1) and tumor necrosis factor-α (TNF-α) were measured by ELISA.The protein levels of cleaved caspase-3, protein kinase B (Akt) and phosphorylated Akt were determined by Western blot.RESULTS:Compared with control group, dasatinib at 1~10 nmol/L suppressed the viability and migration ability of hBMSCs, and dasatinib at concentration of 7 nmol/L was adopted in the following assays.Dasatinib promoted apoptosis, and blocked the cell cycle in G1 phase.In addition, the secretion of TGF-β1 and TNF-α was increased markedly.The protein levels of cleaved caspase-3 was increased, but the protein levels of Akt and phosphorylated Akt were decreased.CONCLUSION:Dasatinib inhibits the viability and migration ability of hBMSCs in a dose-dependent manner, promotes the secretion TGF-β1 and TNF-α, and induces cell cycle arrest and apoptosis.Dasatinib might regulate the biological behaviors of hBMSCs observed above by modulating the expression and phosphorylation of Akt.
9.Association of blood pressure variability and cerebral infarction in elderly men with atherosclerosis

Objective , To investigate the influence of blood pressure variability on cerebral infarction in older men. Methods Ambulatory blood pressure was measured in 1527 elderly men ( older than 65 yrs) with atherosclerosis. All cases were divided into 2 groups: Six hundred and seven patients with cerebral infarction ( group A)and 920 patients without cerebral infarction ( group B). Smooth curve method was used to analyze each patient's ambulatory blood pressure data and the trend of each patient's blood pressure curve was portrayed. The differences between the actual blood pressure and the blood pressure on the curve was defined as blood pressure variability,and the blood pressure variability between the 2 groups was compared. Results The systolic blood pressure variability in 24 hours in group A was significantly higher than that in group B( [8.4'±2. 2]mm Hg vs [ 8.0 ± 2. 0 ] mm Hg, P < 0. 01 ), especially for the systolic blood pressure variability in daytime( [ 8. 2 ± 2. 2 ] mm Hg vs [ 7. 8 ± 2. 1 ] mm Hg, P < 0. 01 ). However, the systolic blood pressure variability at night was not significantly different between the 2 groups( [ 8.9 ± 3. 9 ] mm Hg vs [ 8. 7 ± 3.7 ] mm Hg,P > 0. 05 ). There were no significant difference between the diastolic blood pressure of 24 hours( [5. 5 ± 3.8 ] mm Hg vs [5.5 ± 1.5 ]mm Hg,P >0. 05),during daytime([5.4 ± 1.5]mm Hg vs [5.3 ± 1.4] mm Hg,P >0.05)and nighttime ( [ 6. 1 ± 2.7 ] mm Hg vs [ 6. 1 ± 2. 6 ] mm Hg, P > 0. 05 ). Conclusion In elderly men with atherosclerosis,cerebral infarction was closely related to systolic blood pressure variability,but independent of nighttime systolic blood pressure and diastolic blood pressure variability.
10.Exercise can promote cognition and memory after molar loss

Objective To observe the effect of wheel running and treadmill running on the cognitive functioning of rats after the extraction of a molar.Methods Thirty-two Sprague-Dawley rats were randomly divided into a control group (group C,n =8),a molar extraction group (ML group,n =8),a wheel running pretreatment group (W-ML group,n=8) and a treadmill running pretreatment group (T-ML group,n =8).The rats in the T-ML and W-ML groups practiced wheel running and treadmill running for 1 week,then they and the rats in the ML group received molar extraction surgery.One week later,a passive avoidance test was used to evaluate the cognitive functioning of all of the rats.Results No significant differences were found in the groups' average latency I times,but the average latency II time of the ML group was significantly shorter.There was no significant difference in the average latency II times of the T-ML and W-ML groups.Conclusion Either wheel running or treadmill running can promote learning and memory after molar loss by oral surgery,at least in rats.
