1.Study on a Grey Model for Evaluation of Anopheles minimus Density

The An. minimus density evaluation model was derived as follows: y=0^0578 e~0^0780(8X_10′+7X_12′+6X_11′+5X_15′+4X_9′+3X_4′+2X_8′+1X_7′) The correct rate of evaluation by the model was 92^0%. e_0^5=18%, with an average relative error of 21%. Conclusion A quasi-evaluation on the An. minimus density can be made by applying the grey model.
2.Advance in management of refractory/relapsing thrombotic thrombocytopenic purpura

Thrombotic thrombocytopenic purpura (TTP) is a kind of thrombotic microangiopathies which has low incidence but highly mortality.The outcome of TTP has been improved significantly since last decade due to the application of plasma exchange,however,early relapse occurs in some patients,and for relapsed/refractory patients the prognosis is still poor.This article reviews the recent progress in treatment and relevant influencing factors of TTP.

Breast cancer is one of the most common cancers.The clinical treatment of breast cancer has made great progress,but the inherent or acquired drug resistance,tumor migration and tumor infiltration,which lead to poor treatment efficiency and eventually death,are still the urgent problems to be solved.As with the occurrence and development of common tumors,the abnormal proliferation,migration and infiltration of breast tumor cells and muhidrug resistance (MDR) are closely related to the abnormal expression of specific genes in the cell.At present,many studies have found that the occurrence and development of tumor is closely related to the abnormal expression of microRNA.Therefore,in order to better understand the molecular mechanism of the occurrence and development of breast cancer,it is necessary to study the function of microRNA in breast tumor cells.In this paper,the application of microRNA in the treatment of breast cancer was reviewed,striving for providing effective ideas for the future selection of new strategies to control the development of tumors.
4.Determination of baicalin and naringin inQinbei mixture by HPLC

Objective To establish a high performance liquid chromatography (HPLC) method for the determination of baicalin and naringin inQinbei mixture.Methods The HPLC system consisted of the Fortis-C18(4.6 mm × 250 mm, 5μm) column, and the mobile phase consisted of MeOH:0.4% H3PO4 (42:58), and the flow rate was 1.0 ml/min, and the UV detector was set at 280 nm, and the column temperature was 30℃.Results The linear response range of baicalin was 0.062-0.930μg. The linear response range of naringin was 0.033-0.492μg. The average recovery of baicalin was 98.11% (RSD=1.62%). The average recovery of naringin was 96.78% (RSD=1.74%).Conclusions The method is simple, rapid, accurate and repeatable. It can be applied in determination of baicalin and naringin inQinbei mixture.
5.The significance of protein C in acute leukemia patients with disseminated intravascular coagulation

Objective To investigate diagnostic and prognostic significance of protein C in acute leukemia with disseminated intravaseular coagnlation(DIC). Methods APTT, P-T, D-Dimer, Fbg, antithrombin (AT) and protein C(PC) were determinated in plasma of 44 DIC patients with acute leukemia and 30 normal controls. Results PC was markedly lower in disseminated intravaseular coagulation with acute leukemia group(67.03±36.98) than that of control group (99.53±45.20), and significantly correlation DIC score( r = -0.57,P<0.01). PC of abnormality rate in DIC group was 86.36 %. APTT, PT, D-Dimer, Fbg were significantly increased and AT was decreased during the progress of DIC in relation to values observed in the control group. Conclusion DIC is related to the disorder of coagulation and fibfinolysis system. PC is a sensitive index to diagnose and prognose DIC in acute leukemia.
6.Observation on apoptosis of renal cell in the rat with obstructive jaundice and salvia miltiorrhiza's effect on apoptosis

Objective To investigate the function of apoptosis in the renal injury by observing change of renal pathology and ultrastructure in the rat with obstructive jaundice(OJ) and if salvia miltiorrhiza(SM) can lighten the renal dysfunction of obstructive jaundice.Methods A total of 100 adult Sprague-Dawley rats were studied.Among them,the 80 models of obstructive jaundice were established by ligating bile duct(BDL),then divided into two groups: the OJ rats administered daily abdomen injections of SM(1.7g per rat) after operation of bile duct ligation(the SM group,n= 40);the OJ rats receiving only the same normal saline(the OJ group,n=40).The other rats with sham operation receiving only normal saline(the control group,n=20).Three groups of rats were sacrificed in groups at postoperation 1,2,3 and 4 week,respectively.Then serum BUN & Cr were tested and renal change of histopathology and ultrastructure were observed;Apoptosis of renal tissue were assayed by TUNEL method.Results With the time of BDL extending,the value of serum BUN & Cr increased;Apoptotic cells increased in renal tissue.After treatment with SM,the injury degree of renal function and histopathologic changes decreased.Conclusion Obstructive jaundice can lead to renal injury.Apoptosis had an important effect on renal function injury in the rat with obstructive jaundice.Salvia miltiorrhiza can ease the degree of renal function injury.
7.Establishment of a modified model of adriamycin nephrosis in rats

Objective To study the adriamycin nephrosis model in different development pathological stages. Methods The rat adriamycin nephrosis model was established by injecting adriamycin into tail-vein twice every two weeks to detect blood and urina biochemical indicators and to observe the pathological changes of the renal tissues. Results The model showed serious edema,proteinuria,hypoproteinemia,and hyperlipidemia. Podocytes were swollen and mesangial cells developed mild hyperplasia at the end of the fourth week. The nephric tubule atrophied at the end of the eighth week accompanied with adhesion between glomeruli and Bowman's capsule. Glomeruli sclerosis of mild or medium degree was observed at the end of the twelfth week with obvious lymphocyte infiltration in the renal interstitium as well as the formation of collagen fibers. Conclusion The adriamycin nephrosis model was successfully developed by injecting adriamycin 4 mg/kg into tail-vein twice every two weeks. The acute model is similar to human minimal change disease,and the chronic model is similar to human focal segmental glomerulosclerosis.
8.Expression of nephrin, TGF-β1 and WT1 in adriamycin-induced-nephropathy rat model and its significance

Objective To investigate podocyte number, the expression of nephrin and transforming growth factor-β1(TGF-β1) in adriamycin-induced-nephropathy rat model and its significance. Methods The rat adriamycin nephrosis model was constructed to detect blood and urine biochemical indicators and observe the pathological changes of renal tissues by light microscope and electron microscope. The expression levels of nephrin and TGF-β1 as well as the podocyte number were examined at different time points by immunohistochemistry. Results The pathological changes of the renal tissues were obvious. Nephrin presented a weak signal at the end of the first week (P<0.05). TGF-β1 started to increase (P<0.05) while the podocyte number started to decrease at the end of the eighth week (P<0.05). Expression of nephrin was negatively correlated with the P<0.05) and serum creatinine (r=-0.71, P<0.05). Expression of TGF-β1 was blood urea nitrogen (r=0.62, P<0.05) and serum creatinine (r=0.59, urinary protein (r=-0.63, P<0.05), blood urea nitrogen (r=-0.72, P<0.05) and serum creatinine (r=-0.76, P<0.05); it was positively correlated with nephrin (r=0.78, P<0.01) but negatively correlated with TGF-β1 (r=-0.64, P<0.05). Conclusion The acute and chronic adriamycin nephrosis models were twice every two weeks. The genesis and development of proteinuria are closely related to the abnormal expression of nephrin. Focal segmental glomerulosclerosis occurs when the podocyte number decreases and TGF-β1 accelerates it.
9.Study on gene delivery system based on bisphosphonate-mediated gene-eluting stent

ObjectiveThe aim of the present study was to investigate the incorporation of plasmid DNA (pDNA) onto a coronary stent by chemo-immuno-conjugation for achieving site-specific gene delivery.MethodsA gene eluting stent was fabricated by reacting with polyallylamine bisphosphonate (PAA-BP) to introduce amine reactive groups on the surface.Then an anti-DNA antibody was chemically coupled and pDNA was immunologically tethered on the stent surface.Radioactive-labeled antibody was used to evaluate binding capacity and stability.ResultsThe presence of amine groups on the modified stent surface was confirmed by XPS and AFM analysis.The isotope label assay indicated that the amount of antibody chemically linked on the stents was 15-fold higher than that of the control stent and its retention time was also significantly longer.ConclusionThe results suggested that a large amount of reactive amine groups were introduced on the PAA-BP modified 316L coronary stent surface.This study provide a potential metal surface modification method that could facilitate coupling and tethering of biological molecules such as anti-DNA antibody and plasmid DNA (pDNA) to achieve sustained and highly localized gene delivery for substrate-mediated gene transfection.
