1.Expression of hepatocyte growth factor and vascular endothelial growth factor in acute myeloid leukemia and their relationship with angiogenesis

Objective To investigate the expression of hepatocyte growth factor (HGF)and vascular endothelial growth factor (VEGF)in acute myeloid leukemia(AML) and its significance in the progress of AML.Methods Serum levels of HGF and VEGF in AML patients,including 25 de novo cases,17 cases with complete remission( CR),16 cases without remission and 12 cases with relapse,were measured by enzyme-linked immunosorbent assay( ELISA ).The data were compared with those of normal controls.Results The serum levels of HGF were( 1357.29 ± 358.64)ng/L in de novo patients,(1175.93 ± 306.71 )ng/L in patients without remission and( 1261.21 ± 340.83 )ng/L in patients with relapse,which were significantly higher than those in normal control(232.62 ± 99.13 ) ng/L and CR patients ( 256.65 ± 94.32 ) ng/L( F =78.35,P < 0.01 ).The serum levels of VEGF were ( 253.84 ± 49.14 ) ng/L in de novo patients,( 245.87 ± 54.68 ) ng/L in patients without remission and(264.75 ± 62.52) ng/L in patients with relapse,which were significantly higher than those in normal control ( 97.61 ± 16.19 ) ng/L and CR patients ( 99.76 ± 15.93 ) ng/L ( F =68.65,P < 0.01 ).Compared with de novo group,the serum levels of HGF and VEGF in CR patients decreased significantly but showed no significant difference from those in normal controls(P >0.05).The expression of HGF and VEGF in acute myeloid leukemia was positively correlated with each other ( r =0.49,P < 0.05 ).Conclusion The expression of HGF and VEGF was closely related with the incidence and development of acute myeloid leukemia.Preventing HGF expression may be a possible strategy for leukemia therapy in the future.
2.Clinical study of plasma exchange in the treatment of refractory idiopathic thrombocytopenic purpur with 2∶2∶1 exchange liquid.

Objective To study the effect of the plasma exchange in the treatment of RITP with 2∶2∶1 exchange liquid.Methods 32 cases of RITP were treated with plasma exchange using CS 3000 blood cell separator.Results Plasma exchange could reduce the levels of harmful substances like PA Ig,PA C 3,CIC and raise the levels of platelet by 3 times.Conclusion Plasma exchange is effective in the treatment of RITP with 2∶2∶1 exchange liquid,with reliable outcome.
3.Study on immune function and mechanism of T lymphocyte proliferation in patients with acute leukemia MSC

Objective:To study the inhibitory effect of bone marrow mesenchymal stem cells (MSC) on the proliferation of allogeneic T lymphocytes in patients with acute leukemia and its mechanism. Methods: 30 patients with acute myeloid leukemia ( AML) ,30 patients with acute lymphoblastic leukemia ( ALL) ,and 30 healthy subjects ( healthy group) were selected. MSC cells were isolated and cultured in three groups. The expression level of MSC cells in three groups were determined by MSC assay. The inhibitory effect of MSC on the proliferation of T cells was detected by Transwell assay. Results: AML group supernatant TGF-β1,HGF levels were significantly lower than the ALL group and the healthy group (P<0. 05),significantly higher than the levels of IL-11 ALL group of AML group,healthy group ( P<0. 05 ); three groups MSC supernatant IL-6 levels difference was not statistically significant ( P>0. 05). AML,ALL,MSC cells secrete cytokines healthy group of T lymphocyte proliferation still had a significant inhibitory effect, inhibition of differences in the case of contact and not in direct contact co-culture of T lymphocyte proliferation was not statistically sig-nificant (P> 0. 05); after joining the MSC,T cell proliferation in the three groups was significantly inhibited upon addition of anti-TGF-β1 antibody,anti-HGF antibody after T cell proliferation effective than a simple reversal join MSC (P<0. 05). Conclusion:MSC for acute leukemia patients inhibit allogeneic T lymphocyte proliferation,functioning principle may be related to the secretion of cyto-kines.
4.The quantification and significance of muscle segment homeobox gene Msx2, human topoisomerase II-α, HPV16 and VEGF in sinonasal inverted papilloma.

OBJECTIVE:
To investigate the quantification and significance of Msx2, topoII-α; HPV16 and VEGF in sinonasal inverted papilloma(SNIP), to study the correlation among the four factors,and to discover the relationship between Msx2 and topoII-α in the process of SNIP malignant transfomation.
METHOD:
Real-time quantitative Polymerase Chain Reaction (RT-qPCR) was used to detect the expression of Msx2, topoII-α, HPV16 and VEGF in 13 cases of sinonasal inverted papilloma (SNIP), 10 cases of sinonasal squamous cell carcinoma(NSCC) and 10 cases of inflammatory nasal polyp paraffin (INP)tissues. According to the pathology results SNIP were divided into mild dysplasia, moderate dysplasia and severe dysplasia. All the data were analysised by SPSS17. 0, P<0. 05 was refered to statistically significant difference.
RESULT:
The mRNA level of Msx2, topoII-α, VEGF and HPV16 in SNIP, NSCC tissues were significantly higher than in the INP tissues (P<0. 05). The expression differences of Msx2, topoII-α, HPV16 and VEGF mRNA level in SNIP tissues which were divided into three groups according to their pathological results,were all statistically significantly different between any two of the three groups (P< 0. 05). Using Pearson correlation coefficient analysis,we found positive correlation between any two of the mRNA level of Msx2, topoII-α, VEGF and HPV16 (P<0. 05).
CONCLUSION
Msx2 and topoII-α may play an important role in the process of SNIP Malignant transformation,which may be new targets for gene therapy of SNIP and NSCC.

5.Expression and significance of Msx2 and topo II-alpha in sinonasal inverted papilloma.

OBJECTIVE:
To investigate the expression and significance of muscle segment homeobox2 (Msx2) and topo II-alpha in sinonasal inverted papilloma (SNIP), and the relationship in the process of malignant transformation of SNIP.
METHOD:
Immunohistochemical method was used to detect the expression of Msx2 and topo II-alpha in 32 cases of SNIP, 30 cases of inflammatory nasal polyp (INP) and 30 cases of SNIP with carcinoma. According to the pathology results, SNIP were divided into mild atypical hyperplasia, moderate atypical hyperplasia and severe atypical hyperplasia.
RESULT:
The mean optical density of Msx2 in SNIP and SNIP with carcinoma tissues were 0.2183 +/- 0.0598 and 0.2521 +/- 0.0761,which were significantly higher than 0.1878 +/- 0. 0372 in the INP tissue (P<0.05 or 0.01). The mean optical density of topo II-alpha in SNIP and SNIP with carcinoma tissues were 0.2303 +/- 0.0397 and 0.2666 +/- 0.0483, which were significantly higher than 0.1978 +/- 0.0388 in the NIP tissue (P<0.01). There were significant difference of Msx2 and topo II-alpha in SNIP between any two of the three groups divided according to pathological morphology (P<0.01 or 0.05). The expression of Msx2 and topo II-alpha in SNIP were positively correlated (P<0.05).
CONCLUSION
Msx2 and topo II-alpha may play an important role in the occurrence and development of SNIP. So it can be used as new therapeutic targets.

6.Clinical evaluation of wedge-shaped defect restored by gingival retraction technique.

OBJECTIVETo evaluate the periodontal conditions after the wedge-shaped defect was restored by gingival retraction technique.
METHODSA total of 138 mandibular premolars with wedge-shaped defect were selected and divided into A, B groups. Group A was restored with Dyract after using retraction cord. Group B was directly restored with Dyract. Clinical parameters including plaque index (PLI), gingival index (GI), sulcus bleeding index (SBI), probing depth (PD), volumes of gingival crevicular fluid (GCF) and levels of aspartate aminotransferases (AST) of gingival crevicular fluid were measured at baseline, 1 week, 1 month, 3 months and 6 months after operation.
RESULTSThere was no difference in PLI, GI, SBI, PD between group A and B during 6 months after operation, while the difference of GCF and AST was significant between group A and B at 3 months and 6 months after operation (P < 0.05, P < 0.01).
CONCLUSIONSGingival retraction technique applied in wedge-shaped defect restoration can reduce the damage to the periodontal tissue.

7.Prevalence and epidemic characteristic of overweight, obesity, and central obesity in Shanghai Pudong New Area

Objective To explore the prevalence and epidemic characteristics of overweight, obesity, and central obesity in Shanghai Pudong New Area. Methods Multi-stage stratified random sampling method was conducted in Pudong New Area in 2013. Residents over 15 years old were randomly selected from 13 communities and participated in the cross-sectional study. A structured questionnaire was used to collect the information and physical examination, biochemical assays were also conducted. SPSS 20. 0 was used to analyze the results. Results The averagebodymassindexofparticipantswas(25.04±3.79)kg/m2andtheaveragewaistcircumferencewas(82.44± 9. 51) cm. The prevalences of overweight, obesity, and central obesity were 41. 67%, 17. 70%, and 31. 32%respectively. Standardized prevalences of overweight, obesity, and central obesity were 34. 39%, 16. 96%, and 25. 66% respectively, according to 2010 national age composition of population. Statistically significant sex differences were found among the prevalence of overweight, obesity, and central obesity groups(all P<0. 05). The prevalence of central obesity in female was raised by increasing age. Residents with hypertension, hyperglycemia, dyslipidemia, and metabolic disorders had higher prevalences in overweight, obesity, and central obesity ( all P<0. 05). Conclusion The prevalences of overweight, obesity, and central obesity among the residents in Shanghai Pudong New Area were relatively high. Relevant risk factors should be explored to promote health education that may enhance people′s awareness of weight management.
