Objective:To study the inhibitory effect of bone marrow mesenchymal stem cells (MSC) on the proliferation of allogeneic T lymphocytes in patients with acute leukemia and its mechanism. Methods: 30 patients with acute myeloid leukemia ( AML) ,30 patients with acute lymphoblastic leukemia ( ALL) ,and 30 healthy subjects ( healthy group) were selected. MSC cells were isolated and cultured in three groups. The expression level of MSC cells in three groups were determined by MSC assay. The inhibitory effect of MSC on the proliferation of T cells was detected by Transwell assay. Results: AML group supernatant TGF-β1,HGF levels were significantly lower than the ALL group and the healthy group (P<0. 05),significantly higher than the levels of IL-11 ALL group of AML group, healthy group ( P<0. 05 ); three groups MSC supernatant IL-6 levels difference was not statistically significant ( P>0. 05). AML,ALL,MSC cells secrete cytokines healthy group of T lymphocyte proliferation still had a significant inhibitory effect, inhibition of differences in the case of contact and not in direct contact co-culture of T lymphocyte proliferation was not statistically sig-nificant (P> 0. 05); after joining the MSC,T cell proliferation in the three groups was significantly inhibited upon addition of anti-TGF-β1 antibody,anti-HGF antibody after T cell proliferation effective than a simple reversal join MSC (P<0. 05). Conclusion:MSC for acute leukemia patients inhibit allogeneic T lymphocyte proliferation, functioning principle may be related to the secretion of cyto-kines.

Paeonol has inhibitory effect on a variety of tumor cells, whereas cadherin is a kind of glycoprotein that is associated with the occurrence and development of different tumor. In this study, the interactions of paeonol and E-cadherin have been investigated by fluorescence spectrometry and atomic force microscopy (AFM).Fluorescence spectrometry results revealed that the addition of paeonol significantly quenched the fluorescence of E-cadherin.Based on the results of the quenching constant, it was inferred that the interaction of paeonol and E-cadherin was a static quenching process. The thermodynamic parameters ΔH and ΔS were calculated to be -4.3×10.5 J/mol and -1.3×10.3 J/(mol·K), respectively, which proved the involvement of weak interactive forces such as hydrogen bond van der Waals force.AFM results revealed that cadherin molecules were assembled into the long-chain structure. The addition of paeonol could significantly disrupt these assembling structures into short chains, which could be ascribed to the damage of the interdigitation model from the adjacent cadherin molecules. All these results reveal that cadherin is an important target of paeonol to modulate its activity.

Objective To evaluate the value of volumetric modulated arc therapy ( VMAT) in postopera-tive radiotherapy for cervical cancer .Methods Eighty four cases of cervical cancer patients were retrospectively analyzed after surgery .Thirty-nine patients received volumetric modulated arc therapy , while the other forty -five patients received three -dimensional conformal radiation therapy (3DCRT).The images were collected both before and after treatment. We recorded the setup errors including x (lift-right),y(ventral-dorsal),and z(an-terior-posterior)axes in thirty -nine patients with cone beam CT (CBCT).These images were compared with correponding planning images to calculate the position errors .We also analyzed the radiation dosimetry parameters and adverse reactions .Results The setup errors before and after radiation therapy in x ,y,and z axes were (1.07 ±0.91)cm,(0.76 ±1.07)cm,(0.74 ±0.81)cm and(0.11 ±0.04)cm,(0.11 ±0.09)cm,(0.08 ±0.20) cm,respectively. These data showed that the application of CBCT reduced the setup errors of treatment significant -ly.We analyzed the illuminated volume of bladder and rectum with dose volume histogram (DVH),and found there was no difference between VMAT and 3DCRT groups when 20 Gy irradiation dose was adopted .When the ir-radiation dose increased to 30 Gy and 40 Gy, the corresponding illuminated volume of bladder and rectum in VMAT group were 69.45 ±1.13,41.78 ±2.8 and 75.66 ±3.34,30.12 ±9.59,respctively.In the 3DCRT group, the corresponding illuminated volume of bladder and rectum were 93.37 ±6.59,88.32 ±12.13 and 95.26 ±8.95,90.32 ±6.98,respectively.VMAT can obviously decrease the illuminated volume of bladder and rectum when compared to 3DCRT.Conclusion VMAT can not only decrease the setup errors to improve the accuracy of radiotherapy ,but also reduce the reaction of normal tissues effectively in the cervical cancer patients who accepted postoperative radiotherapy .Therefore, the VMAT technology can improve both patients′treatment compliance and their quality of life .
