Journal of Zhejiang Chinese Medical University 2013;(8):960-961,966
Today's society, many people are plagued by tinnitus and deafness. Chinese medicine on tinnitus and deafness disease and its therapies and for-mulas have discussed quite wel . It is believed that according to the features of the organs, treating tinnitus and deafness from the five internal organs is much better .The virtual is mostly related to the heart, spleen and kidney, but the real mostly related to the heart, liver and lungs. In the treatment of tin-nitus and deafness from the perspective of the kidney, attention should be paid to nourishing the kidneys and strengthening the essence. Treating from the spleen, attaches great importance to the spleen and replenishing qi, clearing material. Treatment from the aspects of liver, the first is to make qi run unob-structed, to prevent generating phlegm and fire. From pulmonary vessels, great importance should be attached to the recovery of dispersing the descending lung. Treated from heart, benefiting qi and nourishing blood should be given much attention not only to make up for its deficiency, but also attach great importance to the qi, send a fire on its reduction. Sick of longer time, compatibility on the basis of syndrome differentiation and treatment to invigorate the flux pulse, aromatic resuscitation of Chinese herbal medicine shal be used.

BACKGROUND: Nowadays, the scaffold for tissue engineering have low properties of physical mechanics, poor biocompatibility and has not free regulating degradation as matrix materials.OBJECTIVE: To develop a composite of scaffold for cartilage tissue engineering with higher porosity and good mechanical characteristic, biocompatibility and free regulating degradation as matrix materials.DESIGN, TIME AND SETTING: This experiment was conducted at the Research Institute of Engineering Materials, Lanzhou Jiaotong University from June 2006 to June 2008.MATERIALS: Gelatin, calcium polyphosphate fiber (diameter 10-20 μm) and rosin (size: 355-450μm) were used in the present study.METHODS: Calcium polyphosphate fiber was selected as reinforced materials and gelatin as matrix materials. The calcium polyphosphate/gelatin composite of the scaffold for cartilage tissue engineering were fabricated with a solvent-casting,particulate-leaching method.MAIN OUTCOME MEASURES: The microstructures, properties of physical mechanics and degradation were tested.RESULTS: ①Microcosmic observation: The composites had the 3-D, connectivity network microstructures. ②Physical and mechanical properties of scaffold composite: The experimental value of calcium polyphosphate/gelatin composite of the scaffold was consistent with calculated value. Experimental value of porosity was 60%-80%, meeting the requirement of porosity of tissue engineering scaffold composite. The compress modulus of scaffold would be increased with the crossllnkage increasing; ③The degradable rate of the scaffolds composites deteriorated quickly during 0-2 weeks, and slowly after 2 weeks and reduced with increased crosslinkage; the degradable liquid pH value was maintained between 5 and 7.CONCLUSION: The mechanical property and biodegradable property of calcium polyphosphate/gelatin composite can meet the demand of cartilage tissue engineering. This composite might be one of cartilage scaffold materials for cartilage tissue engineering with potentially broad applicability.

Objective To explore the role of intracellular reactive oxygen species (ROS) in the senescence of bone marrow mesenchymal stem cells(BMSCs)in patients with systemic lupus erythematosus(SLE) and the underlying mechanisms that controls the intracellular ROS levels in vitro. Methods Human bone marrow aspirates were collected from iliac of eight donors and eight SLE patients and cultured in vitro.Morphological appearance of BMSCs at different passages was examined by inverted microscope. Nuclear size was measured by fluorescence microscope. BMSCs were monitored using the senescence associated β-galacto-sidase (SAβ-gal) assay to characterize senescence in vitro. The quantification of intracellular ROS production was detected by flow cytometry. Real-time PCR technique was used to determine the gene expressions of PI3K, KRas, NRas and FoxO3 at transcription level. The expression of FoxO3, phospho-FoxO3 (p-FoxO3),AKT and phospho-AKT (p-AKT) protein were determined by Western blot analysis. Statistical analysis was conducted with t-test and Mann-Whitney rank test. Results There were no differences in morphology and nuclear size[(31.4±4.5) vs (28.2±4.8) μm, P=0.628] of BMSCs between SLE patients and normal controls. The percentage of SA β-gal positive BMSCs from SLE patients was higher than that from healthy controls [(31.8±9.0)% vs (12.4±0.7)%, P<0.05]. Intracellular ROS levels of BMSCs from SLE patients increased more significantly than healthy donors in vitro (34600±9600 vs 17 958±5400, P<0.05). No significant differences in the expression of PI3K, NRas, KRas and FoxO3 from SLE subjects were observed at mRNA levels compared with normal controls, though all showed a similar upward trend. The expression of p-FoxO3 and p-AKT of BMSCs from SLE patients increased significantly compared with healthy controls at protein levels. Conclusion These data suggest that BMSCs from SLE patients aged more quickly, with high SA β-gal activity and up-regulation of intracellular ROS, which is associated with up-regulation of p-FoxO3 and pAKT at protein levels. These results indicate that bone marrow mesenchymal cell senescence may be associated with the pathogcnesis of SLE by maintaining the lifespan of BMSCs.
