1.Effect of humidified heating airway on the off-line of tracheotomy postoperative patients

Objective To investigate the nursing effect of humidified heating airway on the off-line of tracheostomy postoperative patients.Methods Fifty six cases of off-line of tracheostomy post operative patients were randomly divided into the experiment group and control group.The experiment group was given continuous warming humidification to airway,and control group was cared by the method of micro pump continuous drip.Comfort,sputum viscosity and phlegm scab formation incidence were compared between the two groups.Results The difference of between the two groups was statistically significant in the comfort and sputum viscosity(all P < 0?05)?Treatment effect in the experiment group was obviously better than that in the control group.There was no significant difference in phlegm scab formation between the experiment group and the control group(P > 0?05).Conclusion The application of humidified heating airway to the off-line of tracheostomy postoperative patients can increase comfort,reduce sputum viscosity,and the method ean be used in clinical nursing?
2.Target Antigen of Hyperacute Rejection in Pig-to-Human Xenotransplantation

Objective To investigate the aim antigen coursing the hyperacute rejection of xenotransplantation. Methods Documents about hyperacute rejection in xenotransplantation were reviewed and summarized in detail. Results Pig is thought to be one of the ideal donors of xenotransplantation, but the major obstacle is hyperacute rejection mediated by complement that is activated though human serum. ?-Gal is recognized as the major antigen and its expression is controlled by ?-1,3 galactosyltransferase. Immunoabsorption of pre-exsisted antibody, enzymatic digestion of ?-Gal, knockout ?-GT gene and transgenic technology have been used to solve this problem. Even so, there remain other antigens which can combine with natural antibodies in human serum, such as, 40?10 3 molecule in erythrocyte, 210?10 3, 10 5?10 3 and 50?10 3 antigen in pig embryo brain cell, etc. Conclusion ?-Gal is the major antigen which course the hyperacute rejection. Besides ?-Gal, many nonalpha-gal need further investigation.

4.A new indole derivative from endophyte Myrothecium roridum IFB-E091 in Artemisia annua.

Three compounds were isolated from solid culture of endophyte Myrothecium roridum IFB-E091 in Artemisia annua. Their structures were determined as (S)-(-)-N-[2-(3-hydroxy-2-oxo-2,3-dihydro-1H-indol-3-yl)-ethyl]-acetamide (1), N-(4-hydroxyphenethyl)acetamide (2) and asperfumoid (3), in which compound 1 was a new indole derivative. In cytotoxicity assay, the compound 1 had no obvious inhibition activity in human hepatoma cell line SMMC-7721 and human cervical carcinoma cell line HeLa.
5.Distribution and antimicrobial resistance of pathogens causing healthcare-associated infection in a hospital in 2011-2015

Objective To explore the distribution and antimicrobial resistance of pathogens causing healthcare-associated infection(HAI) in a hospital,and provide theoretical basis for rational use of antimicrobial agents and control of HAL Methods HAI occurred in patients in this hospital between 2011 and 2015 were chosen,distribution of major pathogens causing HAI and antimicrobial resistance were analyzed.Results A total of 852 pathogenic strains were isolated,including 458 (53.75 %) strains of gram-negative bacteria,259 (30.40 %) fungi,and 135 (15.85 %) gram-positive bacteria,the top 4 pathogens were Klebsiella pneumoniae (n =159,18.66 %),Candida albicans (n =119,13.97%),Acinetobacter baumannii (n =81,9.51%),and Escherichia coli (n =74,8.69 %).Isolation rate of gram-negative bacteria increased from 47.41 % in 2012 to 61.57% in 2015,fungi increased from 24.78% in 2011 to 34.45% in 2014 (2015 was 27.95%),gram-positive bacteria decreased from 26.55% in 2011 to 10.48% in 2015.Resistance rates of Escherichia coli and Klebsiella pneumoniae to imipenem and meropenem were 8.11%-12.58%;Acinetobacter baumannii had the lowest resistance rate to cefoperazone/sulbactam(23.46%),resistance rate of Candida albicans to fluconazole was up to 59.66%,vancomycin-resistant gram-positive bacteria was not found.Conclusion The major pathogens causing HAI are opportunistic pathogens,antimicrobial resistance is serious,the species and distribution varies with different years,it is necessary to enhance specimen detection rate,strengthen the monitoring on pathogen resistance,and use antimicrobial agents rationally.
6.Three-dimensional reconstruction technique in breast volume measurement and its artificial influencing factors

Objective To set up a method of three-dimensional breast volume measurement, to observe the effect of the artificial influencing factors on the final data, which are used for 3-D reconstruction of breast volume. Methods The CT data were used to reconstruct a digital prototype of the breast implant and the chest. The 3-D models were established firstly, and then the model was reasonablly cut to get single breast volume, breast implant volume, differences of the both sides of chest and breast volume. These methods were put up by the repeated test when the reference frame and the ranges were fixed. And the analysis of the artificial influencing factors was performed on 10 women whose breasts were unsym-metrical. The effects of the artificial influencing factors were analyzed by moving the coordinate axis, revolving the chest around the coordinate axis and changing the selected area. Results The repeatable test showed that the error was less than 4. 08 ml. The average volume differences between both sides of the breast before and after the given influencing factors were set, were 10. 339 ml and 7. 73 ml, as moved ± 4mm along the axis X; as revolving ±2° along the axis Z, that were 8. 654 ml and 7. 971 ml; as increasing or decreasing of the selected diameter of ± 4 mm, that were 6. 764 ml and 4. 267 ml, respectively. Conclusion Using the 3-D reconstruction technique for breast volume measurement is more simple, accurate and repeatable.
7.Prevention and treatment of acute graft-versus-host disease in mice by bortezomib

Objective To observe the effect of bortezomib on acute graft-versus-host disease (aGVHD) in an aGVHD model of mice and investigate the related mechanism. Methods Male C57BL/6( H-2Kb)mice were used as donors and female Balb/c (H-2Kd) mice used as recipients. Balb/c mice received total body irradiation (TBI) by 7.0 Gy X-radiation, and randomly divided into five groups. normal (group A), TBI (group B), TBI + bortezomib (group C), TBI + bone marrow cells (BMC) + spleen cells (SC) (group D) and TBI + bortezomib + BMC + SC (group E). The physical signs and the pathological damage of aGVHD, mean survival time, and chimerism were observed in recipients. The NF-κB p65 levels in nuclei of the liver and small intestine tissues of groups A,B and C were analyzed by Western blot. Results ( 1 ) The clinical aGVHD score in group D was (7.37±0. 32), significantly higher than in group E (5.85 ± 0.40) (P<0. 05). Histopathology of the gut, liver and skin illuminated that the Ⅲ-Ⅳ degree GVHD occurred in group D. The occurrence of aGVHD in group E was later than in group D. The symptoms and the pathological damage of aGVHD in group E were milder than in group D. The average survival time in group E was significantly longer than that in group D (P<0.05). The percentage of donor-derived cells in recipient mice was above 90% at day 12 after transplantation; (2) NF-κB p65 levels in nuclei of the liver and small intestine tissues in group B was significantly higher than in group C on the day 1,3 and 5 (P<0. 05). Conclusion Bortezomib can inhibit the activation and expression of NF-κB,which may be the underlying mechanism for it to relieve aGVHD.
8.Asymmetric Morita-Baylis-Hillman Reaction of Conjugated Nitroalkene with Activated Aldehyde Catalyzed by Cinchona Alkaloids

Objective:To develop a catalytic system for the asymmetric Morita-Baylis-Hillman ( MBH) reaction of conjugated ni-troalkene with activated aldehyde, and screen out the chiral catalysts with high activity and enantioselectivity. Methods: Totally 21 chiral organocatalysts were applied in the asymmetric MBH reaction ofβ-nitrostyrene with ethyl glyoxylate, and the ee value was deter-mined by chiral HPLC. The effects of temperature, solvent and substrate ratio on the catalytic reaction were investigated. Results: In the presence of cinchona alkaloid catalyst (DHQ)2AQN, β-nitrostyrene reacted with ethyl glyoxylate in toluene at 0℃ affording the MBH adduct in 60% yield with good enantioselectivity (up to 56.9% ee). Conclusion: The bis-cinchona alkaloids with aromatic bridging group are the efficient catalysts for the asymmetric MBH reaction ofβ-nitrostyrene with ethyl glyoxylate, and moderate isolated yield and enantioselectivity are obtained.
9.Clinical efficacy of fine fat grafting of the body on facial rejuvenation

Objective To investigate how to improve the survival rate,and to avoid the serious complications in fat grafting.Methods Under comprehensive design of facial filling area,the fat was harvested by concentration in mesh;under the micro autologous fat transplantation gun with blunt needle injection technique,the fat was filled into the face to achieve the effect of improving the facial contour and the rejuvenation.From June 2014 to July 2015,78 patients with facial contour and facial soft tissue relaxation depression were treatment by the technique.Results The follow-up of 2 months to 1 year showed that 64 patients had the soft,young,significantly enhanced facial contour;7 patients had second times grafts and the remaining were satisfied.7 patients gave up the second times grafts because the other reasons.The surgical complications such as facial nerve injury and infection and embolism were not seen.Conclusions This technique by comprehensive design and purification of autogenous fat granules achieves a better effect on facial rejuvenation with fewer complications.
10.Influence of the lentiviral vectors mediated mouse genetic engineering Tr after allogeneic bone marrow transplantation in mice

Objective To explore the influence of the lentiviral vectors mediated mouse genetic engineering regulatory T cells(Tr) infused after allogeneic bone marrow transplantation(allo-BMT) on graft-versushost disease(GVHD) in mice. Methods Lentivirus-mediated expression of forkhead box P3 (Foxp3) converted CD4 + CD25 - T cells from BALB/c mice into engineered Tr in vitro. An allo-BMT model of BALB/c→C57BL/6 mice was established. After irradiation, the recipients were injected with donor cells along with genetic engineering Tr. Survival time, histopathological analysis, serum levels of inflammatory cytokines were observed after allo-BMT. Results The mean survival times in radiation group, transplantation control group, engineering Tr group and empty vector control group were ( 8.8 ± 0.6 ) d, ( 36.7 ± 2.5 ) d, ( 51.6 ± 4.0 ) d and ( 34.1 ± 2. 3 ) d. The survival time in engineering Tr group was significantly increased as compared to other groups as judged by the log-rank test ( P <0.05 ). Histopathological analysis in several target organs( skin, liver and small intestine) confirmed the presence of severe GVHD in transplantation control group and empty vector control group. No histological signs of GVHD were observed in recipients in engineering Tr group. The serum levels of IFN-γ, IL-2 and TNF-α were all increased after transplantation in above groups. The peaks of concentrations of IFN-γ, IL-2 and TNF-α in engineering Tr group were significantly decreased compared to transplantation control group and empty vector control group at day 21 ( P < 0. 05 ). Conclusion Co-injection of genetic engineering Tr can efficiently prevent recipients from lethal GVHD during allo-BMT in mice by reducing the serum levels of inflammatory cytokines.
