1.The application of temporal temperature gradient gel electrophoresis in the detection of mitochondrial gene mutations

Objective To evaluate the potential utility of temporal temperature gradient gel electrophoresis (TTGE) in detecting homoplasmic and heteroplasmic mtDNA mutations in patients with cancers. Methods The entire mtDNA of tumor and corresponding normal tissues from total 117 patients were amplified by PCR, and the somatic mutations were screened by TTGE. DNA fragments showing different band patterns in normal and tumor tissues in TTGE were sequened to identify the exact mutations. Results 160 fragments harbouring mtDNA mutations were found by TTGE in total 3 654 fragments. Compared with the direct squencing results, the sensitivity and specificity of TTGE in mutation detection were 96.27% and 94.05% respectively. Conclusion TTGE is an effective method for screening acquired homoplasmic mitochondrial gene mutation and its different percentage.
2.Imaging findings of posterior reversible encephalopathy syndrome caused by pregnancy-induced hypertension

Objective To investigate the computer tomography (CT) and magnetic res-onance imaging (MRI) features of posterior reversible encephalopathy syndrome (PRES) caused by pregnancy-indueed hypertension (PIH). Methods CT scan and MRI scan + diffusion-weighted imaging (DWI) were used in 31 patients with PIH. Results CT scan found low density lesions in parietal-occipital lobe of watershed regions in 31 patients with PIH. CT value was 19 to 23Hu. Among them, MRI showed slight hypointensity lesions on T1-weighted images and slight hyperintensity lesions on T2-weight images in 11 patients, fluid-attenuated inversion recovery (FLAIR) sequence showed hyperintensity lesions, DWI revealed isointense, and apparent diffusion coefficient- did not decrease. The recovery time of clinical symptoms was earlier than that of MRI. Conclusions The lesion sites on cr and MRI had certain characteristics in PIH occurred PRES at late pregnancy, combining clinical history a clear diagnosis could be made.
3.Birth Weight Prediction for Ultrasonic Measurement of Fetal Thigh Soft Tissue Thickness

Objective To evaluate ultrasonic measurement of fetal thigh soft tissue thickness in the estimation of fetal weight . Methods Fetal biparietal (BPD), head cirumference (HC), abdominal cirumference (AC) , femur length (FL) and fetal thigh soft tissue thickness (TSTT) were measured by ultrasonography in 196 pregnant women during five days before dilivery and the relation with neonatal birth weight were analyzed.Results There was significantly correlation between TSTT and neonatal weight (r=0.865) , and it was better to estimate fetal weight with TSTT than with the other parameters. The prospective study showed that the predictive value of fetal TSTT was 80 1%.Conclusions The ultrasonic measurement of fetal TSTT is a simple and valuable index in estimation of fetal weight. [
4.Effect of Creatine Phosphate Sodium on Postoperative Fatigue in Gastric Carcinoma Patients

Objective To investigate the effect of creatine phosphate sodium on postoperative fatigue syndrome (POFS) in gastric carcinoma patients. Methods Nighty cases of radical resection in gastric cancer were randomly divided into control group (n=45) and treatment group (n=45).Normal saline (100 mL) and phosphocreatine sodium (2 g) was admin-istrated intravenously after anaesthesia induction in treatment group while only normal saline was given in control group . After operation, the two groups of patients were all treated with enteral and parenteral nutrition. General performance was ob-served in both groups. Fatigue score (VAS score) was measured on the day before the operation,the first,third and fifth day after operation. Body weight, triceps skinfold thickness (TSF), mid upper arm circumference (AMC), plasma total protein (TP), plasma albumin (ALB) and prealbumin (PA) were examined at the same time. Results The total effective rate of the control group and the study group were 73.3%and 95.6%respectively and the difference was of statistically significant. Fa-tigue scores (VAS score) was significantly difference in both groups pre- and post-operation time points, also between 2 groups. Body mass, TSF and AMC in time points postoperation were all lower than those in the time point of preoperation. TP, ALB and PA of two groups in postoperative time points were significantly different compared with those in preoperative time point. The above indexes were all decreased with postoperation time, and these indexes in treatment group were signifi-cantly better than those in control group. Conclusion Creatine phosphate sodium can alleviate postoperative fatigue, im-prove nutritious status and promote early rehabilitation of patients.
5.Effect ofIllicium verum Water Extract on Excess-cold Syndrome in Rats

This study was aimed to observe the effect ofIllicium verum water extract on excess-cold syndrome in rats. The intragastric administration of the decoction of gypsum, gentian, phellodendron andrhizoma anemarrhenae (2:1.2:1:1.5) was used to establish the excess-cold rat model. Effects ofI. verum water extract on general body signs of excess-cold syndrome rats were observed. The detection was made on the contents of LD, PA, TG, TSH, T4 in the serum and LD, PA, glycogen in liver tissues. The vitality of the LDH, SDH and ATPas was also detected. The results showed thatI. verum water extract can improve the general body signs of excess-cold syndrome rats. It reduced the content of TG; increased the content of PA, T4, TSH; reduced the content of LD, glycogen, and the vitality of LDH;increased the vitality of LDH, Ca2+-Mg2+-ATPase in liver tissues. It was concluded thatI. verum water extract can improve some general body signs of excess-cold syndrome rats, improve the substance and energy metabolism, and regulate the thyroid axis function, in order to achieve the warmingyang for dispelling cold effect.
6.Influence of sleep deprivation on serum interleukin-1β level in rats with coronary atherosclerosis

Objective: To explore influence of sleep deprivation (SDP) on serum interleukin-1β (IL-1β) level in rats with coronary atherosclerosis (CAS). Methods: A total of 96 healthy SD rats were randomly and equally divided into normal control group, CAS group, SDP group and SDP + CAS group according to number table; rats in CAS group and SDP + CAS group were fed with high cholesterol diet to establish CAS animal model; SDP group and SDP + CAS group received SDP through small platform with water environment. Radioimmunoassay was used to measure serum concentration of IL-1β; and serum concentrations of total cholesterol (TC), triglyceride (TG), high density lipoprotein-cholesterol (HDL-C) and low density lipoprotein-cholesterol (LDL-C) were measured respectively; above indexes were compared among four groups. Results: (1) Compared with serum IL-1β level of normal control group, there were significant increase in SDP group, CAS group and SDP + CAS group[(13.33±3.20) ng/L vs. (14.40±4.41) ng/L vs. (22.21±5.94) ng/L vs. (30.17±7.45) ng/L] (P<0.05~<0.01); (2) Levels of TC, TG and LDL-C in CAS group and SDP + CAS group were significantly higher, level of HDL were significantly less than those of normal control group and SDP group (P<0.05~<0.01), levels of above indexes in SDP + CAS group were significantly higher or less than those of CAS group (P<0.05 all). Conclusion: Sleep deprivation can significantly heighten serum interleukin-1β level and total cholesterol, triglyceride, low density lipoprotein-cholesterol levels, decrease HDL level in rats with coronary atherosclerosis or sleep deprivation.
7.Simultaneous Determination of Six Quaternary Ammonium Alkaloids in Coptidis Rhizoma by UPLC

ObjectiveTo establish a new,rapid,and reliable reversed-phase ultra performance liquid chromatography (RP-UPLC) method for the simultaneous determination of six quaternary ammonium alkaloids (QAAs) in Coptidis Rhizoma.MethodsThe effect of different experimental parameters on the analysis of QAAs by RP-UPLC was evaluatcd.ResultsOptimal resolution was achieved with an Acquity UPLC BEH C18 column using a gradient elution profile and a mobile phase consisting of water spiked with 10 mmol/L ammonium bicarbonate (A,pH adjusted to 10.0 by ammonia water) and acetonitrile (B),at a flow rate of 0.30 rnL/min and wavelength of 345 nm.The column temperature was set at 30 ℃.The proposed method was found to be reproducible,precise,and rapid according to the method validation.Conclusion The proposed method,which is compatible with MS analysis and the preparation of QAA,provides some helpful insights into the quality control of Coptidis Rhizoma.
8.Comparison of Bayesian interim analysis and classical interim analysis in group sequential design.

OBJECTIVETo explore the differences between the Bayesian interim analysis and the classical interim analysis.
METHODSTo compare the means of two independent samples between control and treatment, superior hypothesis test was established. In line with the data requirements for group sequential design, Type Iota error of Bayesian interim analysis based on various prior distributions, Power, Average Sample Size and Average Stage were estimated in the interim analysis.
RESULTSIn the Pocock and O' Brien & Fleming designs, the Type Iota errors in the Bayesian interim analysis based on the skeptical prior distribution and the handicap prior distribution were controlled at around 0.05. When the powers of these two classical designs were both 80%, Bayesian powers of the skeptical prior distribution and the handicap prior distribution were markedly lower. The powers of the non-informative prior distribution and the enthusiastic prior distribution were distinctly higher than 80%.
CONCLUSIONIn the Bayesian interim analysis based on the skeptical prior distribution and the handicap Prior distribution, the Type Iota errors can be well controlled. Bayesian interim analyses using these two prior distributions, compared with the analysis adopting the O' Brien & Fleming method, can markedly increase the possibility of ending the clinical trials ahead of time. The Bayesian interim analyses based on these two distributions do not have practical value for group sequential design of the Pocock method.

9.Somatic mutation detection in complete mitochondrial DNA of lung cancer patients.

BACKGROUNDTo detect the somatic mutations in complete mitochondrial genome and to investigate the role of mtDNA mutation in the tumorigenesis of lung cancer.
METHODSDNA were extracted from sixteen lung cancer and corresponding normal tissues. The entire mitochondrial genome was amplified with 32 pairs of overlapping primers. mtDNA mutations were screened by temporal temperature gradient gel electrophoresis. mtDNA fragments showing different banding patterns between tumor and paracancerous tissues were sequenced to identify the exact mutations. The common 4 977 bp deletion was also analyzed in all sixteen tumor tissues as well as the matched paracancerous samples by PCR.
RESULTSTen out of sixteen (62.5%) tumor tissues showed a total of 29 mutations. Half (5/10) of tumors with somatic mutation presented one mutation and the rests had multiple ones. Out of 29 mutations, 17 (58.62%) were in D-loop region, 2 (6.90%) in rRNA and 10 (34.48%) in mRNA. Among 10 mRNA mutations, 7 were silence and 3 were missense mutations. Five out of twenty-nine alterations were heteroplasmic to heteroplasmic change, one was homoplasmic to heteroplasmic and the remains (23/29, 79.3%) were homoplasmic to homoplasmic change. There were five common deletions found, two in tumor tissues and three in paracancerous tissues. There was no mitochondrial microsatellite instability, except for the short deletion or insertion in np303-309.
CONCLUSIONSThe high incidence of mtDNA mutations found in patients with lung cancer suggests that mtDNA alterations might play an important role in tumorigenesis of lung cancer. Further studies should be needed to determine the pathological effects of somatic mtDNA mutations in lung cancer.
10.Study on the motivation and path of the formation of breastfeeding education behavior of nurses in NICU based on grounded theory

Objective:To explore the motivation of the formation of the breastfeeding mission and education behavior of nurses in NICU, analyze the deep reasons, and construct the motivation and path theoretical model of the formation of the breastfeeding mission and education behavior of nurses in NICU.Methods:Through semi-structured interviews, the qualitative research method of grounded theory was used to study 20 nurses in the Second Affiliated Hospital, University of South China from July to September 2020, to construct the motivation and path theoretical model of the formation of the breastfeeding mission and education behavior of nurses in NICU.Results:The research identified five main categories: attitude factor, ability factor, behavior factor, internalization and externalization. Under the influence of three dimensions of attitude level, ability level and behavior level and their cross-layer interaction effects, the NICU nurses were driven to form stable breastfeeding mission and education behaviors through internalization and externalization.Conclusions:Based on the evolutionary context of "multi-dimensional factors and their interaction drivers-internalization-externalization", the relationship between attitude factors, ability factors, behavioral factors, internalization, and externalization are clarified. It provides a theoretical reference basis for managers in managing the breastfeeding propaganda and education of nurses in the NICU, and enriches the theoretical framework of breastfeeding behavior in the NICU.
