Mesenchymal stem cells (MSCs) have been the hot spot of rehabilitation and tissue engineered medicine.Due to their origination from the adult tissues,the multilineage differentiation potential and immune escape characteristics,MSCs become the ideal seed cells for cell transplant treatment.This article summarizes the biological characteristics of MSCs and its application in the treatment of liver disease.
2.Thioridazine-induced learning and memory decline is accompanied by elevation of ?-amyloid in rat brain

Aim To explore the molecular mechanism of interrelationship between thioridazine-induced learning and memory decline and the production of ?-amyloid (A?) in the rat brain. Methods Male Sprague-Dawley rats were injected intraperitoneally with thioridazine of 10 mg?kg-1?d-1 for 2 weeks in order to suppress cognition by inhibition of dopamine, acetylcholine and 5-hydroxytryptamine receptors. Morris water maze was used to measure spatial learning and memory performance. The A? content of brain was measured by radioimmunoassay. Immunohistochemistry was employed to determine ?-amyloid precursor protein (APP) level. The mRNA levels of APP,?-secretase and ?-secretase in brain were detected by RT-PCR. Results Thioridazine treatment to rats resulted in spatial learning and memory impairment shown by longer escape latency. Total A? was significantly increased by 1.3 times in thioridazine-treated rats. APP-immunoreactivities in cortex and hippocampus of thioridazine-treated rats were more pronounced than those of control rats. Levels of APP751 plus APP770 mRNA,?-secretase mRNA in brain increased nearly 2.5 and 2.6 folds respectively in thioridazine treated-rats, but no differences in mRNA levels of APP695 and ?-secretase were found between thioridazine-treated and control rats. Conclusion The thioridazine-induced cognitive decline is related to the increase of A? caused by elevation of APP751,APP770 and ?-secretase expression.
3. Influence of Intestinal Mucosal Barrier Dysfunction on Non-alcoholic Fatty Liver Disease

Nowadays, non-alcoholic fatty liver disease (NAFLD) is prevalent all over the world, and its incidence has been increasing year by year in China. The four aspects of intestinal mucosal barrier, including mechanical barrier, chemical barrier, biological barrier and immunological barrier are interrelated closely and related to NAFLD. This article reviewed the influence of intestinal mucosal barrier dysfunction on NAFLD.
4.Implantation of Homologous Cancellous Bone and Autologous Red Marrow under Endoscope for Bone Nonunion and Bone Defect

Objective To investigate the effectiveness of implantation of homologous cancellous and autologous red marrow under endoscope in the treatment of traumatic bone nonunion or bone defect.MethodsTotally 25 patients with traumatic bone nonunion or bone defect were enrolled in this study.The lesion involved the tibia bone in 9 cases,the femur in 13,and the humerus in 3.The grafts were sized 1 to 6 cm in length(mean,2.7 cm).Before the implantation of the cancellous bones,the scar fibrous tissues within the nonunion were removed completely by endoscopy,and then autologous red marrow obtained from the iliac crest was injected.ResultsAll the patients were followed up for 12 to 36 months(mean,25 months).The surgical wounds healed primarily in all of the cases without injuries to the blood vessels and nerves.Bony union was achieved within 4 to 9 months(mean,5.1 months) in 23 of the patients,the other 2 failed because of breaking and loosing of internal fixators.No patient developed re-nonunion and infection.ConclusionsImplantation of homologous cancellous and autologous red marrow under endoscope is effective for traumatic bone nonunion or bone defect with a low rate of complication and high rate of bony union.Since no need to harvest the ilium bone,the method is minimally invasive for the patients.
5.Chromones from Angelica morri Hayata

AIM:Bioactive constituents were expected to be obtained from the roots of Angelica morri Hayata. METHOD:They were extracted with 95% alcohol and isolated by using column chromatography and recrystallization methods. The structures were elucidated by means of physico-chemical data and UV,IR,1HNMR, 13CNMR,and EIMS. The inhibitory effect on the constriction of rat aortic rings was induced by K+ or Ca2+. RESULT:3′S-(-)-hamaudol,3′S-(-)-Ο-acetylhamaudol,3′R-(+)-hamaudol,and (±)-hamaudol were isolated from the pieces of Radix Angelica Morri. The inhibitory rate of 3′S-(-)-Ο-acetylhamaudol and (±)-hamaudol on the above pharmacologic model appears the relation of quantity response. CONCLUSION:All the above compounds were found in this species for the first time,and(±)-hamaudol is a new. One of effect mechanisms of 3′S-(-)-Ο-acetylhamaudol and (±)-hamaudol diluted aorta could contribute to be inhibiting Ca2+ influx of vascular smooth muscle.
6.Detection of clostridium perfringens with qPCR in mouse models and a clinical case

Objective To detect of clostridium perfringens by qPCR in mouse models and a clinical case in order to offer early diagnosis.Methods 40 Kunming mice were randomly grouped and intramuscular injected clostridium perfringens type A in leg 0.1 ml(3.5 × 109cfu/ml or 3.5 × 108cfu/ml or 3.5× 107cfu/ml,diluted with saline),while control group was injected with 9% sodium chloride 0.1ml.The mouse models and a clinical case were detected by qPCR.Results The death rate of 3.5 × 109,3.5 × 108,3.5 × 107cfu/ml and the blank group were 90%,70%,10% and 0% after intramuscular injection for 72 h spectively.The mean Ct values among these groups were 21.21 ±2.69,28.45 ±2.74,32.49 ±2.87 and 0.00 ± 0.00(P < 0.05).The Ct values of the patient were 30.67 and 30.44.Conclusions Cclostridium perfringens could be successful identified with qPCR in mouse models when the mice still did not show any symptoms.
7.Application of one-hand double-lumen simultaneous locking technique in the prevention of occlusion using high pressure resistant double-lumen PICCs

Objective To explore the effects of application of one-hand double-lumen simultaneous locking tech-nique on reduction of occlusion using high pressure resistant double-lumen PICCs. Methods Convenience sampling method was used to recruit 73 patients assigned into the experimental group,using one-hand double-lumen simulta-neous locking technique;66 patients were assigned into the control group,using routine locking method. The rate of PICC occlusion was recorded. Results The rate of incomplete occlusion was 8.22% in the experimental group,and 17.42% in the control group,the rate of complete occlusion in the experimental group was 0,and 0.76% in the control group,and the difference was statistically significant(P<0.05). Conclusion The one-hand double-lumen simul-taneous locking technique is easy and convenient to conduct. It can effectively reduce occlusion using high pres-sure resistant double-lumen PICCs.
8.Abnormal fat accumulation and changes in peroxisome fatty acids ?-oxidation in liver of obese rats with type 2 diabetes

Objective To investigate the metabolic changes in free fatty acids in the livers of obese rats with type 2 diabetes.Methods Thirty male SD rats were randomly divided into control group(Con group) and diabetes mellitus group(DM group),with 15 in each group.Rats in Con group and DM group were fed with normal diet and high-fat diet,respectively.Eight weeks later,OGTT and ISI test were performed to identify insulin resistance.Then the insulin-resistant rats received intraperitoneal injection of low-dose streptozocin(STZ) to induce type 2 diabetes.After giving high-fat diet further for six more weeks,8 rats of each group were sacrificed and artery blood and liver sample were obtained for further analysis.The mRNA levels of diacylglycerol O-acyltransferase 1(DGAT1),carnitine palmitoyltransferaseⅠ(CPTⅠ),palmitoyl-CoA oxidase(ACOX1),D-bifunctional protein(DBP) and L-bifunctional protein(LBP),which were involved in fatty acids metabolism,were evaluated by RT-PCR.The protein level of DBP was evaluated by Western blotting Peroxisome fatty acids ?-oxidation was measured by spectrophotometry.Free fatty acids in blood and liver were determined by gas chromatography.Oil red staining was used to determine the fat accumulation in liver.Results The mRNA expressions of DGAT1,CPTⅠ,ACOX1 and LBP increased(P

Objective To investigate the effect of pseudolaric acid B(PLAB) on growth of human gastric carcinoma cells in vitro.Methods The expression of PPAR? was detected by RT-PCR;the effect of PLAB on cell growth was tested by MTT;Hoechst33342/PI and DNA gel electrolysis were employed to examine apoptosis;cell cycle was checked by flow cytometry.Results When treated with 0.1~10 ?mmol/L PLAB for 72,the proliferation of MGC803 cells was significantly inhibited.The proportion of MGC803 cells at G2 phase was significantly increased when treated with 10 ?mmol/L PLAB after 48 h,and showed an apparent G2 phase arrest.After treatement with PLAB for 72,typical apoptotic changes were observed.The expression of PPAR? was at a low level in MGC803 cells and up-regulated when treated with 10 ?mmol/L PLAB for 48 h(P

Objective:To investigate the impact of Notch signaling pathway on the migration of human hepatic carcinoma cells and the expression of E-cadherin and COX-2 in these cells.Methods:Cultured hepatic carcinoma cell lines (SMMC-7721,MHCC97H),and normal non tumor liver cell line (HL-7702) in vitro.Transwell cell was used to measure the cell's capacity of invasion and migration.Western blot was used to measure the expression level ofNotch1,E-cadherin,COX-2 protein.DAPT was used to block the Notch signaling pathway,and compared the ability of invasion and migration between hepatic carcinoma cell lines and normal non tumor liver cell line,and the change of expression level of E-cadherin and COX-2 protein in hepatic carcinoma cells.Results:The migration ability of SMMC-7721 cells and MHCC97H cells were higher than HL-7702 cells,the difference was statistically significant (P<0.05);Compared to HL-7702 cells,the expression level of Notch1 and COX-2 in MHCC97H cells and SMMC-7721 cells significantly increased,the expression level of E-cadherin decreased significantly (P<0.05);After DAPT treatment,the migration ability of SMMC-7721 cells,MHCC97H cells were weaker than the control group,the difference was statistically significant (P<0.05);After DAPT treatment,the expression of COX-2 and Notch1 in SMMC-7721 and MHCC97H cells decreased significantly,while the expression of E-cadherin significantly increased (P<0.05).Conclusion:Notch signaling pathway plays an important role in the process of liver cancer cell migration and invasion,and its mechanism is related to the expression of E-cadherin and COX-2.
