1.Effects of exogenous fragile histidine triad gene on apoptosis of MGC-803 gastric cancer cells induced by octreotide

Objective:To investigate the effect of exogenous extopic fragile hisdidine triad (FHIT) gene on the apoptosis of gastric cancer cells induced by octreotide and elucidate the underlying mechanism. Methods: Human gastric cancer cell line MGC-803, which was loss of FHIT gene, was transfected with plasmid pRcCMV-FHIT and pRcCMV via lipofectamine mediation. After transfection, Western blotting was used to analyse the expression of FHIT protein in positive cells screened out by G418. The cells were treated with octreotide 10~(-10), 10~(-9), 10~(-8), 10-7, and 10~(-6) mol/L for 24, 48 and 72 h. Cell proliferation was evaluated by MTT assay and apoptosis by flow cytometry. The expression of bcl-2 and bax protein was detected with Western blotting.Results:FHIT protein expression was detected in MGC-803 gastric cancer cells after FHIT gene transfection. After treatment with octreotide 10~(-8) mol/L for 48 h, the apoptotic rate of MGC-803 cells transfected with FHIT gene was (26.777±1.702)%, significantly higher than that in empty vector group and untransfected group [(13.800±0.511)% vs (12.634±0.479)%, F=245.789, P<0.05]. The expression of bcl-2 protein was decreased while the expression of bax protein was increased in FHIT gene transfection group after octreotide treatment. Conclusion:The exogenous FHIT gene and octreotide had strong synergistic effects on apoptosis of MGC-803 cell lines, and their action mechanism may be related to the alteration of the expression of apoptosis-related proteins of bcl-2 family.
2.Effects of total flavonoids of litchi and rosiglitazone on expression of PPAR-γ and CTGF in HSC-T6 rat hepatic stellate cells

Objective To investigate effects of total flavonoids of litchi (TFL) on the proliferation of rat hepatic stellate cells (HSC-T6) in comparison with western medicine rosiglitazone, and to explore the mechanism of anti hepatic fibrosis of TFL. Methods Effect of TFL on proliferation of HSC-T6 was examined by MTT. The expression of peroxisome proliferator-activated receptor-γ (PPAR-γ) mRNA, connective tissue growth factor (CTGF) mRNA in HSC-T6 cells exposured were examined by real-time quantitative PCR. Effects on HSC-T6 CTGF protein from TFL and rosiglitazone were detected by Western bloting. Results The expression of PPAR-γ mRNA was upregulated and the expression of CTGF mRNA and protein was downregulated after exposure to TFL and rosiglitazone for 72 hours. And the effect of TFL increased with the increase of concentration. Conclusion TFL can inhibit the proliferation of HSC-T6 and antagonizing liver fibrosis. This mechanism may be associated with the upregulation of PPAR-γ expression and the downregulation of CTGF expression.
3.Effects of RNA interference targeting CDC25a gene on proliferation of human liver cancer HepG2 cells

[ ABSTRACT] AIM:To investigate the effect of silencing cell division cycle 25a ( CDC25a) gene on the prolifera-tion of human hepatoma HepG2 cells.METHODS:CDC25a gene in human hepatoma HepG2 cells was silenced by RNA interference.Real-time PCR was applied to detect the expression of CDC25a, cyclin E and CDK2 at mRNA levels in the HepG2 cells.Western blotting was applied to detect the expression of CDC25a at protein level.In addition, MTT assay, Giemsa staining and flow cytometry were used to measure the proliferation of human hepatoma HepG2 cells.RESULTS:The expression of CDC25a at mRNA and protein levels in RNA silence group was lower than those in negative control group and normal control group (P<0.05).The mRNA expression of cyclin E and CDK2 in silence group was lower than that in negative control group and normal control group (P<0.05).The cell proliferation in silence group was lower than that in negative control group and normal control group ( P<0.05) .The results of flow cytometry revealed that the cells in silence group were blocked in G1 phase.CONCLUSION:Infection of LV-CDC25a-RNAi recombinant to the HepG2 cells effec-tively inhibits the CDC25a gene expression and the proliferation of human hepatoma cells, and arrests the cells in G1 phase, suggesting that CDC25a gene may be a key target for the treatment of liver cancer.
4.High-energy shockwave for the chronic pain of muscle and bone joint

BACKGROUND: At present, there are some reports regarding the treatment of high-energy shockwave to chronic pain of muscle and born joint, however,the therapeutic effects and mechanism are still uncertain and need further discussion.OBJECTIVE: To explore the curative effects, mechanism and clinical application of high energy shockwave to bone joint myofascitis.DESIGN: Non-randomized case controlled study based on diagnosis SETTING: Rehabilitation Medicine Department of First Affiliated Hospital of Sun Yat-sen University.PARTICIPANTS: Totally 90 patients with chronic pain of muscle and bone joint treated in Rehabilitation Medicine Department of First Affiliated Hospital of Sun Yat-sen University from July 2001 to April 2002 were divided into treatment group and control group each with 45 cases by the order of visiting time. There were 17 males and 28 females in the treatment group with mean age of 54 years and 15 males and 30 females in the control group with mean age of 63 years.METHODS: Shockwave therapy was applied to the treatment group while routine physical treatment was conducted to the control group. Simple McGill pain questionnaire(MPQ) was used to evaluate the general reactions of patients to pain and assess the shoulder joint territory and clinical effects.therapeutic effects.RESULTS: There was significant difference on sensory, affective and pain score, visual analogous scale(VAS) and pain presentation inventory(PPI) before and after treatment in the treatment group( t =5.69, 5.67, 7.06, 8.37,6. 21, P < 0.01 ). When compared with control group, there was significant difference on sensory, pain score, VAS between the treatment group and control group(t =4. 66, P < 0.01; t =2.52,3.40, P <0.05).CONCLUSION: Extracorporeal high-energy shockwave has reliable effects on treating chronic pain caused by bone joint myofascitis, which is characterized by its effective, quick and safe features.
5.Investigate the Prevalent Feature of Children with Mycoplasma Pneumoniae Infection in Xi'an Region from Jan.2015 to Dec.2016

Objective To investigate the prevalent feature of children with Mycoplasma pneumoniae (MP) infection in Xi'an region from Jan.2015 to Dec.2016.Methods From Jan.2015 to Dec.2016,9 648 cases with acute lower respiratory tract infection in xijing hospital were given serum test of the antibody of Mycoplasma pneumoniae by ELISA.Results A total of 9 648 cases with lower respiratory tract infection in children in xijing hospital received MP antibody detection,the positive cases were 2 971,the total positive rate was 30.79 %.MP-IgM antibody positive rates between different age groups had significant differences,in which infants' group has the lowest(7.00 %),and with the age growing,the positive rate increased.The positive rates in different age groups of toddler,preschool age,and school age were 27.29 %,36.10 % and 37.76 % respec tively.The positive rates of MP-IgM antibody in different quarter were different.The highest antibody positive rate was found in the first quarter (January-March),followed by the fourth quarter,the third quarter and the second quarter.The difference was statistically significant (P<0.01).Conclusion There was MP infection prevalence in Xi'an from 2015 to 2016.The positive rate in children increased with the age,the peak of infection was in the first quarter,so the detection and prevention of MP should be strengthened at that time of the year.
6.Comparison of two common insemination methods on in-vitro fertilization and embryo transfer

The in-vitro fertilization and embryo transfer technique has been widely applied in human insemination. The rate of successful insemination is gradual y rising, and the in-vitro fertilization directly determine the insemination outcome.
OBJECTIVE:To evaluate the difference between the two common using insemination methods, microdrop and open, in in-vitro fertilization and embryo development.
METHODS:A randomized study was conducted to compare microdrop and open insemination methods among non-male factor patients undergoing in-vitro fertilization and embryo transfer. A total of 1 175 cases were enrol ed in the research. There were 573 cases in the microdrop group, and 602 cases in open insemination group. The fertilization rate and embryo development in the two groups were compared.
RESULTS AND CONCLUSION:The fertilization failure rate [total fertilization failure rate+low fertilization rate (<25%oocytes fertilized)] in the microdrop insemination group was higher than in the open insemination group (11.9%, 3.3%, P<0.001), while the good quality embryo rate and pregnancy rate did not differ significantly between the two groups (al P>0.05). The open insemination method is a simple insemination method with a lower fertilization failure rate. As the fertilization is a highly complicated process involving many extrinsic and intrinsic factors, further study is needed to confirm the effects of the two insemination methods on in-vitro fertilization outcome.
8.The association between metalloestrogens in seminal plasma and sperm quality in infertile patients

Objective:
To analyze the association between the concentration of metalloestrogens ( MEs ) in seminal plasma and sperm quality in infertile patients, so as to provide the basis for the study of MEs on human sperm quality.
Methods:
The spermatozoa concentration, progressive rate ( PR ), normal morphology rate ( NMR ) and DNA fragmentation index ( DFI ) were determined in the infertile male patients from Zhejiang Provincial People's Hospital from March to April, 2020. The contents of As, Cd, Co, Cr, Cu, Hg and Ni in seminal plasma were determined by inductively coupled plasma mass spectrometry ( ICP-MS ). The distribution of MEs in seminal plasma and the sperm quality in different concentration of MEs ( grouped by quartiles ) were analyzed.
Results:
Among 105 cases recruited, 28 cases were normal in sperm parameters and 77 cases were abnormal, including 22 cases of oligospermia, 47 cases of asthenospermia, 54 cases of dysspermia and 30 cases of abnormal DFI. As, Cr, Cu, Hg and Ni were detected in all samples, the detection rates of Cd and Co were 78.57%-90.91%. Compared with the normal group, the concentration of As in oligospermia group was higher [( 18.96±12.56 ) µg/L vs. (13.67±4.19) µg/L, P<0.05],the concentration of Cr was higher in asthenospermia, dysspermia and abnormal DFI in Q3 and Q4 groups groups [( 386.62±81.92 ), ( 378.02±81.46 ), ( 393.88±77.03 ) µg/L vs. ( 343.12±55.08 ) µg/L,P<0.05]. The PRs in Q3 and Q4 groups of Cr were lower than that in Q2 group [( 32.95±18.22 )%, ( 27.74±22.77 )% vs. ( 54.18±24.64 )%, P<0.05], the DFI in Q3 and Q4 groups was higher than that in Q2 group [( 26.91±14.77 )%, ( 29.91±16.93 )% vs. ( 9.87±10.93 )%, P<0.05], and the NMR in Q4 group was lower than that in Q2 group [( 1.62±1.72 )% vs. ( 3.36±1.97 )%, P<0.05]. The concentration of sperm in Q3 group of Cu was higher than that in Q2 group [( 115.87±88.22 )×106/mL vs. (61.91±66.16)×106/mL, P<0.05].
Conclusion
As and Cu in seminal plasma are associated with abnormal sperm concentration, and Cr is associated with NMR, PR and DFI.
9.Analgestic Effect of Scalp Nerve Block with Ropivacaine Hydrochloride at Different Time Points After Craniotomy

Objective To observe the effect of scalp nerve block ( SNB ) with ropivacaine hydrochloride at different time points on pain management after craniotomy. Methods Ninety patients undergoing craniotomy were randomly divided into 3 groups:group A, SNB conducted before surgery;group B, SNB conducted after surgery;group C, SNB conducted both before and after surgery, with 0. 5% of ropivacaine hydrochloride in each group. All patients received the same general anesthesia and diclofenac sodium were administered rectally as rescue analgesics. Sites and duration of surgeries, end-tidal sevoflurane concentration during incision, HR and SBP levels during the course of surgery and postoperative period, the VAS scores, GCS and Ramsay scores at 0. 5, 2, 4, 6, 12, 24, 48 h postoperatively, time of the first rescue appication analgesics and total consumption of rescue analgesics, the adverse effects, awareness under anesthesia were analyzed respectively, as well as local anesthesia relevant adverse events and time of wound healing. Results The end-tidal sevoflurane concentration was significantly decreased in group B (3. 19±0. 36)% as compared with group A (1. 81±0. 24)% and C (1. 77±0. 33)% (P<0. 05);The VAS scores of group A (3. 77±2. 27, 4. 20±2. 09) at 2 and 4 h were higher than those in group B (2. 77±1. 98, 3. 20±2. 20) and C (2. 97±1. 77,2. 27±1. 93) (P<0. 05), while at other time points the differences were not significant (P>0. 05);Compared with group A (600 mg), the consumption of rescue analgesics of group B (300 mg) and C (250 mg) were statistically lower (P<0. 05);Vital signs, GCS, Ramsay scores, time of the first rescue analgesics postoperatively used, and time of wound healing among the three groups were not various significantly (P>0. 05);The relevant side effects were not different statistically, and there were no patients suffering from obvious awareness under anesthesia, pruritus, respiratory depression or local anesthesia relevant adverse effects. Conclusion SNB conducted before surgery can decrease the consumption of sevoflurane during incision, but has limited analgesic effects postoperatively. SNB conducted after surgery may provide transitional analgesia for neurosurgical patients undergoing craniotomy, while SNB conducted both before and after surgery does not show significantly longer analgesic time in postoperative pain management.
10.Prevention of subcutaneous effusion after breast cancer surgery and curative effect analysis on two different drainage methods

Objective To investigate the cause,prevention methods and curative effect on two different drainage methods of subcutaneous effusion after breast cancer surgery.Methods The clinical data of 68 cases subcutaneous effusion among 2 100 cases underwent breast cancer modified radical surgery in our hospitial were analyzed retrospectively from January 2010 to December 2016.The vacuum sealing drainage was performed in 1 032 cases and wound high negative pressure drainage system was performed in 1068 casess.Both the elastic bandage compresstion dressing didn't used.Results In 2 100 cases,68 cases of subcutaneous effusion occured (3.2%),including longitudinal incision 48 cases and transverse incision 20 cases.Among 1 068 cases of high negative pressure drainagesystem,subcutaneous effusion occured in 36 cases including 28 cases of incisional infection and 8 cases of skin flap necrosis.The extuhation time was 8-36 days,average 12 days.Among 1 032 cases of vacuum sealing drainage,subcutaneous effusion occured in 32 cases including 22 cases of incisional infection,10 cases of skin flap necrosis.The extubation time was 6-12 days,average 8 days.All cases were followed up for 3 months,no subcutaneous effusion and axillary effusion occurred.Conclusions Adequate drainage is the key to prevent subcutaneous effusion.Both vacuum sealing drainage and wound high negative pressure drainage can effectively reduce the occurrence of subcutaneous effusion,however,vacuum sealing drainage is more suitable for wounds with more exudation,larger lacuna and deeper incisions,especially the infected wound and abscess.
