Journal of Chinese Physician 2010;12(7):910-914
Objective To study the expression regulation of GATA-3 on Th2 cytokines secretions in rats lung infected by respiratory syncytial virus (RSV).Methods 20 SD rates were random divided into two groups: Control group (10 rats) and RSV-infected group(10 rats, the model was established by RSV intravasation). The airway responses were observed after seven week.Lung tissue of SD rats were stained with HE, pathologic examinations of lung tissue- stained were made.GATA-3 expressions in lung tissue were detected by western-blots, respectively.Interleukin-4 (IL-4) and interleukin-5 (IL-5) in lung homogenate were detected by ELISA.Results Lung biopsy from RSV-infected group showed typical inflammation under light microscope.Airway resistance in RSV- infected group was higher than control group.Compared with control group( 1.231 ±0.31 ), the expression of GATA-3 significantly increased in RSV-infected group (1.743 ±0.21)( t =15.427, P <0.01).The levels of IL-4[(142.3 ± 12.3)ng/L] and IL-5[(167.5 ±14.2) ng/L] increased in RSV-infected group [(69.2±11.6) ng/L, ( 86.6 ± 14.3 ) ng/L] ( P<0.01 ).There was a positive correlation between protein levels of GATA-3 and IL-4 in lung tissues (r=0.864, P<0.01 ), and a positive correlation between GATA-3 and IL-5 in lung tissues(r=-0.756, P<0.01).Conclusion RSV-infection may up regulate the expression of GATA-3 in lung, which contributed to inflammation of Th2 type.
2.Microglial Toll-like receptor 2 and cerebral ischemia
International Journal of Cerebrovascular Diseases 2013;21(10):792-795
Microglia can be activated immediately after acute cerebral ischemia and play a role of double-edged sword of nerve repair and neurotoxicity.Studies in recent years have suggested that this dual role of microglia can be achieved by a Toll-like receptor 2 (TLR2) signal transduction pathway. Therefore, reasonable regulation of TLR2 expression of microglia may improve the therapeutic effect of cerebral ischemia to a certain extent. This article reviews the mechanisms of TLR2 in cerebral ischemia and the possible interventions.

BACKGROUND:Bone turnover markers are the products in the blood or urine in the process of bone reconstruction, which can be used to evaluate bone resorption and bone formation rate, thereby indicating potential bone diseases. OBJECTIVE:To analyze the effect of exercise on the levels of bone turnover markers in the blood and urine samples. METHODS:Databases of PubMed and Wangfang were retrieved with key words of“bone formation, bone resorption, alkaline phosphotase, osteocalcin PICP, PINP, hydroxyproline, pyridinoline, tartrate resistant acid phosphatase”by screening titles and abstracts to search papers related to exercise effects on the biological markers of bone formation. Final y, 48 papers were discussed. RESULTS AND CONCLUSION:The effect of exercise on bone turnover refers to the influence on bone formation and bone resorption. Exercise enhances or inhibits the activity of osteoblasts and osteoclasts, and accelerates or delays bone reconstruction by promoting or inhibiting the growth of osteoblasts and osteoclasts. In recent years, exercise exhibits more and more influence on the bone. Bone turnover markers compared with bone mineral density show changes earlier. By measuring the levels of bone turnover markers in blood and urine samples, it help us to understand the metabolism of bone tissue, to evaluate bone metabolic state, osteoporosis diagnosis classification, predicting of fracture risk, to observe the curative effect of drug treatment, and to diagnosis the metabolic bone differential disease.

Objective To study the type, characteristics, common complications of the cranio-cerebral injury and assessment for the body impairment in victims involved in road traffic accidents. Method Data from 862 cases with cranio-cerebral injury in road traffic accidents were collected and were statistically analyzed according to injured body part, complication, the type of complications and the severity of injury. Results Compound cranio-cerebral injuries were sustained in 276 cases, brain contusion in 192 cases, skull fracture in 79 cases, intracranial hemorrhage in 91 cases, and scalp injury in 224 cases. The chief complications were identified as hydrocephalus (19,2.20%), epilepsy (l6,1.86%), hemiplegia (14, 1.62%), decerebrate cortical syndrome (12,1.39%). Conclusion Among the cranio-cerebral injuries sustained in the road traffic accidents, compound injury was the commonest insult, in which serious cerebral contusion was the major type of injury. The complications following cranio-cerebral injury should be diagnosed strictly according to the diagnostic standard and identification time, which should be differentiated from the other related diseases.

To study the relationship between the position and the severity of injury and the rate of disbility in the road traffic accidents(RTA).3547 RTA cases occurred in the downtown area in Wuhan from 1997 to 1999 year were investigated and analyzed.1792(50 52%)cases with disability were assessed. The rates of disability caused by the peripheral nerve and ear injury were the highest,90 00% and 83 30%,respectively,while those caused by the soft tissue injury was the lowest. The rates of IX X and I V grade disablility were 78 74%,4 41%,respectively.Our study suggests that the rate of disability evaluation is not coincident with the position of injury. The degree of disability might be influenced directly by treatment.Degree of IX X grade of disability were the commonest, while the I V grade of disability was the least.Grade I disability was only seen in injury of brain and spinal cord.

Objective To establish a simple, efficient and low-cost method for the detection of Salmonella typhimurium carrying SSeC gene. Methods In this study, a nano-biosensor ( FAM-P/GO) was successfully established based on the noncovalent assembly of carboxy-fluorescein ( FAM)-labeled probe and graphene oxide ( GO) . The target gene at different concentrations and SSeC gene-harbored bacterium sam-ples were detected by the FAM-P/GO nano-biosensor to evaluate its sensitivity. The specificity of the estab-lished nano-biosensor was evaluated by using DNAs with mismatched base pairs and single-stranded DNAs ( ssDNAs) extracted from various species. Results The established strategy for SSeC gene detection showed a good linear range of 0. 05-1. 0 μmol/L (R2=0. 992 1) with a lower limit of 0. 05 μmol/L. Moreover, the lower detection limit for target bacterium samples was 103 CFU/ml and the fluorescence intensity increased linearly with the concentration from 103 CFU/ml to 108 CFU/ml. The signal-to-noise ( S/N) of the experi-mental group was much greater than that of the control group, which indicated that the establish method was highly specific. Conclusion The FAM-P/GO nano-biosensor was successfully established in this study, which provided a new and possible way for the rapid detection of Salmonella typhimurium harboring SSeC gene.
