1.Expression and significance of MMP-14 and EMMPRIN in human oral squamous cell carcinoma

Objective To investigate the expression and significance of MMP-14 and EMMPRIN and the correlation between them in oral squamous cell carcinoma ( OSCC) . Methods The expression of MMP-14 and EMMPRIN mR-NA in 43 cases of OSCC and 11 cases of normal oral tissue were detected by RT-PCR. Results The expression of MMP-14 and EMMPRIN mRNA in OSCC was significantly higher than those in normal oral tissue ( P <0.05 ) . Compared with those without lymph node metastasis in OSCC, the expression of MMP-14 and EMMPRIN mRNA with lymph node metastasis was significantly higher ( P <0.05 ) . Furthermore, the expression of MMP-14 and EMMPRIN was significantly associated with pathological stage and the lymph node metastasis,but neither of them was correlated with sex and age. Correlation analysis showed that MMP-14 expression level was positively related to EMMPRIN level(r=0.801,P<0.01). Conclusion The expression of MMP-14 and EMMPRIN in OSCC is signif-icantly correlated with occurrence, progress and metastasis of OSCC. Therefore, the combination of detecting the expression of MMP-14 and EMMPRIN mRNA may evaluate the degree of malignancy and predict the prognosis of patients with OSCC.
2.Differentiation of muscle-derived stem cells into neuron-like cells induced by ciliary neurotrophic factor and Compound Salvia Miltiorrhiza Injection in vitro

BACKGROUND: The low survival rate of neuron cells is one of the main mechanisms of stem cell allograft, which might lead to the failure of allograft. Nuclear factor-KB (NF-kB) is one of main transcription factors for cell signaling transduction and participates in call proliferation and differentiation.OBJECTIVE: To study the differentiation of muscle-derived stem cells (MDSCs) into neuron-like cells induced by ciliary neurotrophic factor (CNTF) and Compound Salvia Miltiorrhiza Injection in vitro, and the expression of NF- kB.DESIGN, TIME AND SETTING: The experiment at cell molecular level was performed at the Oral Science Laboratory of the Second Affiliated Hospital of Liaoning Medical College from March to May 2008.MATERIALS: A total of 10 Sprague Dawley neonatal rats aged 7 days were supplied by Experimental Animal Center, Liaoning Medical University. CNTF (Sigma, USA) and Compound Salvia Miltiorrhiza Injection (Chiatai Qingchunbao Pharmacantical Co.,Ltd., China) were used in this study.METHODS: Rat MDSCs were harvested in vitro, pudfied by differential adherence and enzyme digestion, and incubated in 6-well plate. Samples in the induction group were incubated in DMEM containing CNTF for 24 hours. The medium was changed.Subsequently, samples were dnsed three times, and then incubated in serum-free DMEM supplemented with Compound Salvia Miltiorrhiza Injection for 5 hours. Samples in the control group were treated with serum-free DMEM.MAIN OUTCOME MEASURES: Neurofilament protein and NF-KB inhibitor protein expression were detected using reverse transcription-polymerase chain reaction (RT-PCR) and Westem blotting.RESULTS: No neurofilament protein expression was found in MDSCs before induction, and neurofilament protein-positive MDSCs were detected following induction. 3.Risk of cervical cancer and precancerous diseases in the oral HPV carriers

Objective To evaluate the risk of the occurrence of cervical intraepithelial neoplasia (CIN) and invasive cervical cancer in the oral HPV carriers through a population-based investigation in Shanghai.Methods A total of 1200 cases of outpatients who attended the annual cervical examination and 50 preoperational cases of inpatients with CIN Ⅲ or invasive cervical cancer were enrolled from three clinical centers in Shanghai.The oral HPV infection was determined by real-time PCR.In 1200-case cross-sectional study,the incidence rate of CIN was compared between the oral HPV positive and negative cohort.In 1250-case case-control study,the positive rate of oral HPV DNA test was compared among normal control group,CIN Ⅰ-Ⅲ,and invasive cancer case groups,and all odds ratio (OR) values were calculated,respectively.The HPV transmission-related demographic and behavioral characters of the oral HPV carriers were also analyzed.Results The oral HPV carriers accounted for 5.9% (71/1200) of the investigated outpatients.The oral HPV DNA positive rates were gradually increased with the cervical disease grades,which were 5.8% (68/1182,normal),2/13 (CIN Ⅰ),1/5 (CIN Ⅱ),31.4% (11/35,CIN Ⅲ) and 5/15 (invasive cancer).In cross-sectional cohort studies,the relative risks (RR) of CIN Ⅰ,Ⅱ were 2.9 and 4.0 for oral HPV carriers,respectively.In case-control study,the OR values for CIN Ⅰ-Ⅲ and invasive cervical cancer were 3.1(95%CI:1.6-10.1),4.2(95%CI:1.7-28.4),7.1(95%CI:4.8-19.8) and 10.1 (95% CI:3.2-32.1),respectively.The oral sex and multi-sexual partner were two major risk factors for the oral and cervical HPV co-infection,HPV-related cervical cancer and precancerous diseases according to behavioral analysis.Conclusions There are complicated transmission pathways between oral and cervicalHPV.Oral HPV carriers should be intensively followed up and their oral HPV infection and HPV-related cervical diseases should be treated together.
4.The impact of changing the buccal position of the transported bone on early bone formation after alveolar vertical distraction.

OBJECTIVETo assess the effects of 1-step method of changing the buccal position of the transported bone after distraction on early bone formation.
METHODSThe custom-made bidirectional distraction devices were used. Total of 8 adult mongrel dogs were procured. The bilaterally mandible premolars were extracted. After 1 month, the complete osteotomy was performed and the devices were placed. The gradual distraction was started after 1 week latency at the rate of 1 mm per day, total 6 mm in height was achieved. The 1-step method of changing the buccal position of the transported bone was performed the day after the vertical distraction period in one side of the animals, and the other side as the controlled side. All animals were killed after a 7 days consolidation. The vascular system was stained post mortem carbon ink perfusion to assess possible damages. Quantitative evaluations of bone density were performed with dual-energy X-ray absorptiometry (DEXA), analysis of the ratio of vascular square was conducted in a computer.
RESULTSAll animals tolerated the procedures well. After vertical distraction, the tested side can be moved 3 mm in buccal direction. DEXA examination showed that the density of the distracted bone was no statistical difference in both the tested and the controlled side. Vascular damage was not observed, and there was no statistical difference in the ratio of vascular square by analyzing the histological section in computer.
CONCLUSIONIn some extent, 1-step method of changing the buccal position of the transported bone after distraction can mold the regenerate bone to correct the axial displacement, without endangering early callus formation and vascularization.
