1.The interference of common methods in determination creatinine

Objective To investigate the interference of common methods in determination of creatinine. Methods Standardized with Fuller′s earth, Jaffe′s method, enzymatic method (ultraviolet), enzymatic method (colour), enzymatic method (electrode) were compared and evaluated. Results In eight interference materials, Jaffe′s method was positively affected by pyruvic acid (≥0.186 mmol/L), α-ketoglutaric acid(≥0.0615 g/L), ascorbic acid(≥0.82 mmol/L) and negatively affected by bilirubin(≥165.5 μmol/L). Enzymatic method (ultraviolet) was not interfered. Enzymatic method (color) was positively affected by creatine(≥0.21 mmol/L) and negatively interfered by dopamine(≥0.28 mmol/L). At last, enzymatic method (electrode) was positively interfered by creatine(≥0.62 mmol/L). Conclusion Enzymatic method(ultraviolet) is the best in the above methods.
2.Effect of gastric bypass surgery on fasting blood glucose and pancreatic islet cell in type 2 diabetic rats

Objective To study the effect and mechanism of gastric bypass surgery on type 2 diabetic rats.Methods The models of type 2 diabetic rats were induced by stretozotocin and 20 diabetic rats were randomly divided into 2 groups:diabetes-operation group (DO group,n =10)and diabetes-control group(DC group,n =10).20 normal rats were randomly divided into 2 groups:normal-operation(NO group,n =10) and normalcontrol group(NC group,n =10).Rats in DO and NO group underwent GBP and rats in DC group and NC group underwent sham operation.Fasting blood glucose(FBG) levels of rats in each group were detected before operation and on 72 h,1th week,4th week,8th week after operation.On the 8th week after operation,pancreas tissues were harvested for HE staining and immunofluorescence,histological changes observed.Results The FBG levels of rats were not statistically significant different before operation between DO group and DC group or between NO group and NC group (P > 0.05).After operation,the FBG levels of rats in DO group gradually declined (P < 0.05).FBG levels of rats in DO group were lower after operation than before operation(P <0.05) ; After operation FBG levels of rats were higher in DO group than in NO group and NC group at the same time point (P <0.05).In DC group,the difference of FBG levels of rats at different time point was not statistically significant(P > 0.05).The difference of FBG had no statistically significance between the different time points of the same group or between the same time point of different groups (P > 0.05).HE staining showed that,in DO group,newborn small islets appeared in pancreas which increased the number of islet.The new islets were smaller,mostly around the pancreatic duct and the structure was similar to that of the normal islets.Immunofluorescence staining also showed that the number of islets increased.Insulin immunofluorescence found more isolated small islets composed of two or three insulin positive cells.Insulin and glucagon double immunofluorescence found insulin and glucagon double positive(INS +/GLU +)cells in some islets.Conclusions GBP has obvious hypoglycemic effects on FBG levels of type 2 diabetic rats,in which the regeneration of pancreas islets may play an important role,while on normal rats GBP has no hypoglycemic effects.
3.Effects of Shengjiang Capsule on Gastric Evacuation in Rats

ive] To observe the effects of Shengjiang Capsule (SC) on gastric evacuation and to study its mechanism. [Methods] Eight-week-old SD rats were randomly allocated to five groups. Group A served as normal control, and Group B were treated with high dosage of SC, Group C with low dosage of SC, Group D with Zhike Baizhu Decotion and Group E with cisapride. After ten days of treatment, gas tric evacuation of rats was detected by isotope tracing method, erythrocyte-acetycholinesterase activity by micro-hydroxylamine method and plasma motilin (Mot) and somatostatin (SS) levels by radioimmunoas-say. [Results] Thirty-minute gastric evacuation rate was (51.44?.38)%, (40.82 + 7.24)% and (40.22?.16)% in Group B, Group C and Group D respectively, which was higher than that in Group A [ (33.18?.32)%]. Erythrocyte-acetycholinesterase activity, Mot and SS levels were (0.856?.128) umol/h, (124.26?5.94) ng/L and (39.42?.96) ng/L, and (0.726?.164 ) umol/h, (119.86 ?9.38) ng/L and (38.33 ?.64) ng/L in Group B and Group C respectively, which were higher than those in Group A [ (0.576 ?.150)/umol/h, (91.28 + 26.84) ng/L and (28.22 ?7.68)ng/L]. [Conclusion] SC can promote gastric evacuation, and its mechanism may be re lated to the increase of the cholinergic nerve function and plasma Mot and SS levels.
4.Design and application of cross contamination experiment for Beckman AU5800 automatic biochemical analyzer

Objective To design and verify an analytical method for the cross contamination of Beckman AU5800 automatic bio‐chemical analyzer′s reagent needle and stir bar .Methods The outer ring items (AST ,ALP ,TP ,LDH ,CK‐MB ,HBDH ,Urea ,UA , TG ,LDL‐C ,APOB ,Mg ,P ,Zn ,AMY ,CHE ,LPS ,ADA) were tested as the experiment objects .The cross contamination experiment was proceeded according to the test order .The impact rate range of 95% -105% was the criteria for cross contamination of items . Results Among all of the outer ring items ,TP ,HBDH ,APOB and TG were found to have cross contamination effect on CK‐MB , Mg ,Mg and LPS respectively .Conclusion The method established in this experiment can effectively analyze the cross contamina‐tion of all the items in Beckman AU5800 automatic biochemical analyzer .In addition ,it can help to adopt corresponding measures to reduce cross contamination and assure the accuracy of test results .

40 patients,20 males and 20 females,with 52 miniscrews were included.The concentration of IL-1β,IL-6 and IL-8 in the gingival crevicular fluid around the miniscrews were measured.The results showed that the concentrations of IL-1β,IL-6 and IL-8 increased during orthodintic tooth movement(P <0.05),indicating that orthodontic force had an effect on the cytokines around the miniscrews,which may be related to orthodontic bone resorption.

Objective To evaluate the efficacy and safety of continuous erythropoietin receptor activator (C.E.R.A.) once every 4 weeks by subcutaneous administration on hemoglobin (Hb)maintenance in dialytic patients with chronic renal anemia who had been treated with stable dose of erythropoietin (EPO).Methods This was an open,randomized,controlled,multi-center trial.All the hemodialysis or peritoneal dialytic patients in EPO maintenance treatment received subcutaneous EPO-β during the 6-week pre-treatment period to maintain Hb level between 100 g/L and 120 g/L.Eligible patients were randomized (2∶1 ) to accept either C.E.R.A.once every 4 weeks by subcutaneous administration ( C.E.R.A.group,n =187 ) or subcutaneous EPO-β 1-3 times weekly ( EPO group,n =94) for 28 weeks (including 20-week dose titration period and 8-week efficacy evaluation period ). The starting dose of C.E.R.A.was converted according to the dose of EPO-β administered in the week preceding the first study drug administration.The primary outcome was the change of Hb level between the baseline and that in the efficacy evaluation period.Results Totally 253 patients completed the whole 28-week treatment.The change of baseline-adjusted mean Hb was +2.57 g/L for C.E.R.A.group and + 1.23 g/L for EPO group,resulting in a treatment difference of 1.34 g/L (95% CI - 1.11-3.78 g/L).Since the lower limit of 95% CI was greater than the pre-defined non-inferiority margin -7.5 g/L( P < 0.0001 ),C.E.R.A.once every 4 weeks by subcutaneous administration was clinically non-inferior to EPO regarding the maintenance of stable Hb level.The proportion of patients maintaining Hb level within the range of 100-120 g/L through efficacy evaluation period was similar between the two groups ( 69.0% for C.E.R.A.group vs 68.9% for EPO group,P >0.05 ).The overall incidence of adverse events was similar between the C.E.R.A.(41.7%)and EPO (46.2% ) groups ( P > 0.05 ).The safety findings were in accordance with the patients' primary diseases rather than the administration.Conclusions Conversion from EPO to C.E.R.A.once every 4 weeks by subcutaneous injection could maintain the Hb in target level in dialytic patients with renal anemia,and it was non-inferior to EPO.In general,subcutaneous administration of C.E.R.A.is well tolerated in dialytic patients with chronic renal anemia.
