Platelet-derived growth factor (PDGF), a potent chemotactic and mitogenic factor, is involved in the regulation of hematopoiesis and platelet production. Our studies demonstrate the presence of functional PDGF receptors (PDGFR) on human megakaryocytes/platelets and CD34(+) cells, and their ability to mediate a mitogenic response. PDGF promotes the ex vivo expansion of human hematopoietic stem (CD34(+)) and progenitor (CD41(+)) cells. More significantly, PDGF enhances the engraftment of human CD45(+) cells and their myeloid subsets (CD33(+), CD14(+) cells) in NOD/SCID mice. PDGF also stimulates in vitro megakaryocytopoiesis via PDGFR and/or the indirect effect on bone marrow microenvironment to produce TPO and other cytokines. It also shows a direct stimulatory effect of PDGF on c-Fos, GATA-1 and NF-E2 expressions in megakaryocytes. We speculate that these transcription factors may be involved in the signal transduction of PDGF on the regulation of megakaryocytopoiesis. PDGF also enhances platelet recovery in mouse model with radiation-induced thrombocytopenia. This radioprotective effect is likely to be mediated via PDGFR with subsequent activation of the PI3K/Akt pathway. It provides a possible explanation that blockage of PDGFR may reduce thrombopoiesis and play a role in imatinib mesylate-induced thrombocytopenia.

Objective:To provide scientific basis for the clinical diagnosis and treatment of allergic rhinitis by observation of curative effect of treatment to allergic rhinitis by azelastine hydrochloride nasal spray combined with desloratadine .Methods: One hundred and twenty-nine patients with allergic rhinitis were randomly divided into two groups: an experimental group and a control group.The control group was treated by azelastine hydrochloride nasal spray .The experimental group was treated by azelastine hydrochloride nasal spray and desloratadine 5 mg per night.One period of treatment for each group is 4 weeks.Results:the cure rate of the experimental group was 93.85%and 81.25%in control group, with a great difference between them (P<0.05);The incidence rate of adverse drug reactions (ADR)of the former and the latter were 10.77%and 7.81%(P>0.05).Conclusion:azelastine hydro-chloride nasal spray combined with desloratadine can treat patients with allergic rhinitis effectively and safely .It can increase the total effective rate and improve present ’s symptoms.It has great value in clinical application .

Objective To investigate the infection status of hepatitis virus among the perinatal women in Nanjing.Methods 523 samples of perinatal women's serum were selected.The hepatitis virus markers were detected by the ELISA kit,the quantitive detection of HBV surface-antibody was detected by electrochemiluminescence immunoassay.Results The positive rate of anti-HAV IgM was 0.19％,47 cases were HBV surface antigen-positive,accounted for 8.99％,183 cases were HBV surface antibody-positive,accounted for 35.00％,224 cases were all negative HBV markers,accounted for 42.83％.The quantitive detection of HBV surface-antibody in high concentration was significant higher than middle concentration and low concentration (all P ＜ 0.05).The positive rate of anti-HCV IgG was 1.34％,anti-HDV IgG was all negative.The positive rate of anti-HEV IgM was 0.57％.The positive rate of anti-HEV IgG was 24.28％.With the advance of age,the positive rate of anti-HEV IgG had an increasing trend,with significant difference among groups (P ＜ 0.05).Conclusion There are important significances in the detection of hepatitis virus markers in pregnancy monitoring,it can improve the quality of the population.

Objective To discuss the nursing care after the treatment of lower limb deep venous thrombosis by using thrombolytic drug from lateral dorsum of foot vein.Methods 32 patients with lower limb deep venous thrombosis were treated by using micropump pumping thrombolytic drug from lateral dorsum of foot vein,and administration of heparin sodium,706 artificial plasma and danshen root,in combination with nursing care,healthy guidance,observing the signs,symptoms and the effect of treatment.Results Of 32 cases,12 cases were complete recovery,19 cases were partial recovery and 1 case was noneffective.Conclusion Correct nursing care is important in the patients with lower limb deep venous thrombosis after thrombolytic treatment.

Objective To investigate the clinical efficacy and safety of radiofrequency ablation (RFA)combined with low frequency ultrasound radiation with microbubble for the treatment of malignant liver neoplasms.Methods The clinical data of 25 patients with malignant liver neoplasms who received RFA combined with low frequency ultrasound radiation with microbubble at the Kunming General Hospital from January 2010 to June 2011 were retrospectively analyzed.RFA was firstly performed with ultrasonic guidance,and then low frequency ultrasound radiation with microbubble was performed according to the results of ultrasound contrast examination 1 week after RFA.Ultrasound contrast examination was performed and the hepatic function and serum alpha fetoprotein (AFP) were assayed before operation,3 and 6 months after operation,respectively.All data were analyzed using the t test or chi-square test.Results The gross tumor volumes of 16 patients reduced by 30％ at 3 months after operation when compared with that before operation.The gross tumor volumes of 17 patients reduced by 50％ at 6 months after operation when compared with that before operation.The gross tumor volume of 7 patients reduced,but tissues surrounding tumors showed the signal of bloodstream.The gross tumor volume of 1 patient did not reduce.The levels of aspartate aminotransferase (AST),total bilirubin (TBil) and alpha fetoprotein (AFP) at 3 months after operation were significantly lower than those before operation (t =23.51,13.79,257.07,P ＜0.05).The levels of alanine aminotransferase (ALT),AST,TBil and AFP at 6 months after operation were significantly lower than those before operation (t =5.67,27.19,21.02,333.46,P ＜ 0.05).No local recurrence was detected during a follow-up of 6 months.The scores of quality of life (QOL) were very good in 21 (84.0％) patients,good in 2 (8.0％) patients,fair in 1 (4.0％) patient and poor in 1 (4.0％) patient.The scores of Karnofsky (KPS)were improved in 19 (76.0％) patients,stable in 5 (20.0％) patients and aggravated in 1 (4.0％) patient.There were significant differences in QOL and KPS between pre-and postoperation (x2=41.600,18.960,P ＜0.05).Conclusion RFA combined with low frequency ultrasound radiation with microbubble is effective in reducing the tumor size and protect the hepatic function for patients with malignant liver neoplasms.

ObjectiveTo investigate the role of overexpressed proinflammatory cytokines in the pathogenesis of cerebral palsy (CP).MethodsLevels of tumor necrotic factor-α (TNF-α) and interleukin-6 (IL-6) in serum of 31 CP children, 20 healthy children (as controls), 37 neonates with CP risk factors such as hypoxic-ischemic injury and/or perinatal infection, and 20 healthy neonates (as controls) were measured by enzyme-linked immunosorbent assays (ELISA) retrospectively.ResultsLevels of TNF-α and IL-6 of CP children and neonates with CP risk factors were significantly higher than that of healthy controls (P<0.05). TNF-α level of CP children was significantly higher than that of neonates with CP risk factors (P<0.05), but there was no significant difference in IL-6 level between two groups.ConclusionOverexpressed proinflammatory cytokines play an important role in the pathogenesis of CP and may be an independent risk factor of CP.
