Objective: To investigate the influence of 981208 agent on cell proliferaton of hepatocarcinoma induced by diethyl nitrosamine(DEN). Methods: Rat hepatocarcinogenesis model induced by DEN was established and 981208 agent was used during the stage. Influence of 981208 agent on hepatocytes positive expression of proliferating cell nuclear antigen (PCNA), H ras and bromodeoxyuridine labeling index (Brd U LI) during DEN induced rat hepatocarcinogenesis was examined by immunohistochemical method and analyzed by computerized image technique. Results: Positive expression of H ras, PCNA increased gradually and worked synergically. Overexpression of 2 proteins was inhibited significantly during DEN induced hepatcacinogenesis and Brd U LI was reduced strikingly in later stage of model by 981208 agent. Conclusion: 981208 agent can inhibit proliferating character of hepatocytes and slow down the process of hepatocarcinogenesis induced by DEN in rats.

Objective To investigate the relationship between nonalcoholic fatty liver disease (NAFLD) and benign prostatic hyperplasia (BPH) in elderly patients.Methods 50 elderly BPH men with NAFLD were enrolled in this study,while 30 age-matched BPH men without NAFLD were selected as the control group.Prostate volume (PV),body mass index (BMI),triglycerides (TG),fasting blood glucose (FBG),fasting plasma insulin (FINS) and homeostasis model of assessment for insulin resistanceindex (HOMA-IR) were compared between the two groups,and correlations of PV with FINS and HOMA-IR were analyzed in the NAFLD patients.Results The PV level was significantly higher in NAFLD patients than in control group [(48.2±16.3) ml vs.(33.2± 11.5) ml,P＜0.01].The incidence of severe BPH was higher in NAFLD group than in control group (42.2％ vs.13.3％,P＜0.05).BMI,TG,FBG,FINS and HOMA IR were higher in NAFLD group than in the control group (all P＜0.01).The PV was positively correlated with FINS and HOMA-IR levels in elderly NAFLD patients (r=0.814,0.601,respectively,both P＜ 0.01).Conclusions NAFLD is closely associated with BPH in elderly males,and NAFLD may be one of the risk factors for BPH.

Objective: To investigate the application of the calculated electrophysiological parameters in early diagnosis of carpal tunnel syndrome (CTS). Methods: 44 patients (60 hands) with a diagnosis of CTS and 31 healthy volunteers (44 hands) were enrolled. Acquired indicators include median nerve distal motor latency (DML), complex muscle action potential (CMAP) amplitude, conduction velocity (MCV), median nerve sensory nerve action potential (SNAP) amplitude, and conduction velocity (SCV). Then the terminal latency index (TLI), the residual latency (RL), and the difference in peak sensory latencies between the median and ulnar nerves (ΔPSL) were calculated. Results: The two groups were matched in age and gender distribution. The CTS group showed significant difference in SCV, DML, SNAP, and CMAP compared with the control group. The sensitivity, specificity, cutoff value, Youden index, and area under the curve of each indicator are respectively as follows: TLI (0.733, 0.932, 0.622, 0.629, and 0.877), RL (0.750, 0.977, 1.334, 0.727, and 0.907), ΔPSL (0.950, 0.841, 0.150, 0.791, and 0.942), SCV (0.950, 0.796, 56.5, 0.746, and 0.946), DML (0.867, 0.932, 3.55, 0.799, and 0.930), SNAP (0.683, 0.932, 21.68, 0.615, and 0.844), and CMAP (0.683, 0.773, 8.76, 0.456, and 0.758). Conclusion: The calculated electrophysiological parameters have higher sensitivities and specificities relative to a single electrophysiological parameter, which could greatly improve the accuracy of early diagnosis of CTS.

Objective : To investigate the status of occupational stress, burnout, and coping styles, and to explore the moderating effects of coping styles on the relationship between occupational stress and burnout among medical staff from one tertiary first-class hospital in Hubei Province, in order to provide the basis for mental health promotion of medical staff. Methods: From June to October in 2020, using stratified cluster sampling, doctors, nurses, and medical or pharmaceutical technicians on the ratio of 2∶2∶1 were selected from the tertiary first-class hospital. Chinese version of Burnout Questionnaire, Core Occupational Stress Scale (COSS) and Trait Coping Style Questionnaire were used to investigate the levels of burnout, occupational stress and coping styles. A linear stratified regression model was used to analyze the moderating effect of coping styles on the relationship between occupational stress and burnout. Results: Among 735 questionnaires issued, 679 valid ones were collected, with the response rate of 92.38%. The age of the respondents was ( 34.29±8.20 ) years old. There were 165 ( 24.30% ) males and 514 ( 75.70% ) females, including 241 ( 35.49% ) doctors, 358 ( 52.72% ) nurses and 80 (11.78%) medical or pharmaceutical technicians. The burnout score was 2.29±1.11 , and the positive rate of burnout was 44.33%; the COSS score was 47.32±8.31; the positive and negative coping style scores were 30.64±6.06 and 26.81±6.26, respectively. The linear stratified regression analysis suggested that the interactions between social support ( β=0.590, P<0.05 ), organization and reward ( β=0.523, P<0.05 ), and positive coping style were positively associated with burnout, and the interaction between organization and reward (β=-0.666, P<0.05) and negative coping style was negatively associated with burnout. Conclusions About 44.33% of medical staff investigated have burnout. Coping styles have a moderating effect on the relationship between occupational stress and burnout. Positive coping style helps to perceive social support and organizational support, to alleviate the adverse effects of occupational stress, and to reduce the risk of burnout.

OBJECTIVEThe study aimed to test if Paridis Rhizoma total saponins (PRTS) could induce apoptosis of human gastric cancer cell MKN-45.

METHODBased on the previous researches, PRTS was set by different concentrations to treat human gastric cancer cell for 12 h (5, 10, 20 mg x L(-1)). Fluorescent staining methods were adopted to observe apoptotic morphological changes of MKN-45. The apoptosis rates were analyzed by flow cytometry with Annexin V-FITC/PI staining. The enzymatic activities of caspase-3 and caspase-8 were measured by ELISA. The protein levels of Fas and FasL were detected by Western blotting.

RESULTUnder a fluorescence microscope, MKN-45 treated by PRTS was seen typical apoptotic morphological features. PRTS significantly increased the rate of apoptosis. Compared with the control group, there exsited significant differences in apoptosis rate of PRTS concentration of 20 mg x L(-1) (P < 0.01); besides, the enzymatic activities of caspase-3 and caspase-8 were promoted obviously after the effect of PRTS on MKN-45 cells for 12 h (P < 0.01). The protein levels of Fas and FasL in the MKN-45 were upgraded significantly.

CONCLUSIONPRTS can induce apoptosis of human gastric cancer cell MKN-45 , which is concerned with caspase-3 and caspase-8 and upgraded Fas and FasL.

Objective To establish embryonic stem cells (ESC) that can express green fluorescent protein (CFP) and differentiate them into neurons. It would provide tagging neurons for clinical transplantation to cure neural system diseases. Methods ESC (R1) was transfeeted with a plasmid containing the GFP by electroporation. A transgeuic cell line was obtained after selection with G418. The ESCs were characterized by AKP staining. Monolayer differentiation method was used to induce neural differentiation derived from GFP-ESC and immunofluorescence method was used to identify Tuj1 positive cells. Results There was no significant difference(X2=3.14,P0.05) in transfect rates between liposome and electroporation (65% vs 79%). The AKP staining of GFP-ESC was positive. GFP-ESC could be differentiated into neural cells. Conclusions These results show that ESC expressing GFP has been estabhshed, which can be differemiated into neurons.
