Objective To determine the effectivity of 5-azacytidine(5-Aza) inducing the cardiomyogenic SCs were isolated and purified from Sprague-Dawley (SD) rats. Some BMSCs were induced with 5-Aza,group Ⅰ was not induced with 5-Aza and the third generation of cells were used for the next experiment; group Ⅱ were cultured and induced with 5-Aza( 10 μmol/L) at the third day of the culture for 24 h; group Ⅲ were induced with 5-Aza( 10 μmol/L) for 24 h when the third generation of cell were fused; group Ⅳ were induced with 5-Aza( 10 μmol/L) for 24h at the third day of primary cuhure, which were induced with 5-Aza( 10 μmol/L) for 24 h before the third gener-phosis was found in the BMSCs among groups. CD34 was negatively expressed and CD29 and CD44 were positively gle 5-Aza inducing group(group Ⅲ ) was much higher than that in no 5-Aza inducing group( group Ⅰ ) [ MHC : (6. 82±2.05 ) % vs (3.61±1.14) % ], cTnI: (5.63±1.86) % vs (2.76±0.82) %, P < 0.05 ], but was significantly lower than the percentage in the double 5-Aza inducing group ( group Ⅳ ) [ MHC : ( 12.18±3.16) % vs ( 6.82±2. 05)% ] ,cTni:(9.93±2.79)% vs(5.63±1.86)% ,P<0.05]. Conclusions 5-Aza could promote the cardio-myogenic differentiation of BMSCs in a "Cardiac-like" Milieu. 5-Aza double induing is better than the single indu-cing.

Objective To investigate the effects of dexamethasone(DEX)on the secretion of interleukin (IL)-17 and interferon(IFN)-γ and the proportion of Th17,Tc17,Th1 ,Tc1 cells in peripheral blood mononuclear cells(PBMCs)of patients with systemic lupus erythematosus(SLE). Methods Thirty hospitalized SLE patients were recruited and twenty-two healthy volunteers were recruited as healthy controls. PBMCs were separated from SLE patients and healthy controls and then was cultured in vitro by medium or PMA/Ionomycin or PMA/Ionomycin +dexamethasone for six hours. Four- color immunofluorescent staining and flow cytometric assay were used to analyze the percentage of Th17,Tc17,Th1,Tc1 cells in PBMCs. Concentrations of IL-17 and IFN-γ in plasma and the supernatants of PBMCs which were cultured for 24 hours were measured by enzyme linked immunosorbent assay (ELISA). Results The plasma concentrations of IL-17 and IFN-γwere elevated in SLE patients as compared to the controls(P < 0.05). No significant differences were observed between patients and controls for the spontaneous production of IL-17 and IFN-γ or percentage of T subsets expressed by PBMCs. After the stimulation of PMA,compared with the controls, the level of IL-17 was significantly elevated in the supematants of PBMCs and the percentages of Th17 and Tc1 in SLE patients increased significantly(P < 0. 05). However, there showed no significant differences between SLE patients and the controls for the percentages of Th1 and Tc17 cells. DEX could significantly decrease the production of IL-17(P < 0. 01)and the percentages of Th17,Tc1 cells by the active PBMCs(P < 0. 05). Conclusions There is abnormal expression of T subset cells and their cytokines in vivo of SLE patients. DEX can interfere with immunological pathological process in the cytokine network imbalance of SLE patients and shows powerful inhibition of IL - 17. Our results may provide some laboratory evidence for the clinical application of corticosteroids.

Journal of Experimental Hematology 2010;18(6):1548-1551
The study was aimed to explore the effects of recombination human granulocyte colony-stimulating factor (rhG-CSF) on Th17 cells in donors' peripheral blood (GPB) and bone marrow grafts (GBM). 25 healthy donors were injected subcutaneously with rhG-CSF 5 µg/(kg·d) for 5 consecutive days. GBM and GPB were harvested after injection on day 4 and 5 respectively. Some of these donors' steady-state bone marrow (SSBM) and steady-state peripheral blood (SSPB) were harvested before rhG-CSF injection. The changes of IL-17 secreted by T cells in donor BM and PB before and after mobilization were detected by flow cytometry. The results showed that the ability to secrete IL-17 from CD4(+) T cells and CD8(+) T cells in GBM was significantly lower than those in SSBM (GBM vs SSBM Th17/CD4(+) T, 0.74% ± 0.27% vs 1.78% ± 1.19%, p < 0.05; Tc17/CD8(+)T, 0.19% ± 0.16% vs 0.36% ± 0.37%, p < 0.05), changes in peripheral blood and bone marrow were same (GPB vs SSPB Th17/CD4(+) T, 1.82% ± 0.91% vs 3.26% ± 1.89%, p < 0.01; Tc17/CD8(+) T, 0.21% ± 0.17% vs 0.44% ± 0.28%, p < 0.01). The ratios of Th17/CD4(+) T and Tc17/CD8(+) T in GPB were higher than in GBM (p < 0.05, p < 0.01). It is concluded that rhG-CSF in vivo can inhibit the generation of Th17 cells both in bone marrow and peripheral blood grafts, and it may be partial reason for GPB/GBM mixed transplantation without increasing the GVHD incidence.
