2.Reliability and validity of the Chinese version of the Venous Leg Ulcer Quality of Life Questionnaire

Objective To translate the Venous Leg Ulcer Quality of life Questionnaire (VLU-QoL) into Chinese and evaluate the reliability and validity of Chinese version.Methods VLU-QoL was translated into Chinese according to the standards setted by ISPOR.Totally 232 patients with venous leg ulcer were recruited and investigated by the Chinese version of VLU-QoL.Results The Chinese version of VLU-QoL comprised 34 items,Cronbach α was 0.940,inter-rater reliability was 0.841;test-retest reliability was 0.825.I-CVI was 0.83-1.00,S-CVI/Ave was 0.955;Criterion-related validity were 0.714 and 0.541;factor analysis got 3 factors,which explained 54.367% of the total variance.Conclusions The Chinese version of VLU-QoL has been proved to be reliable and valid.It can be used as a valid tool for the measurement of quality of life for patients with venous leg ulcer.

Objective:To study the effect of melatonin(Mel),a potent antioxidant,on the oxidative stress and the activity of hypothalamo-pituitary-adrenal axis in diabetic rats.Methods:Male SD rats were assigned to the following 4 groups:normal control(treated with 2% ethyl alcohol lavage\[NC,n=6\]),diabetes mellitus control(treated with 2% ethyl alcohol lavage \[DM,n=7\]),diabetes mellitus model group 1(treated with Mel lavage \[10 mg?kg-1?d-1,DM+Mel1,n=6\]),diabetes mellitus model group 2(treated with Mel lavage \[0.2 mg?kg-1?d-1,DM+Mel2,n=6\]).Diabetes in DM group,DM+Mel1 group and DM+Mel2 group was induced by intraperitoneal injection of streptozotacin(65 mg/kg).Blood glucose,body weight and adrenal gland weight/body weight(AW/BW)were determined in the 4 groups 12 weeks later;malondialdehyde(MDA)was examined with visible spectrophotometry;blood total cholesterol(TC)and triglyceride(TG)were examined with enzymology method;blood corticotrophin-releasing hormone(CRH),adreno-corticotrophic hormone(ACTH)and corticosterone(CORT)were examined with radioimmune method.Results:Blood glucose,MDA,TC,TG,CRH,ACTH,CORT and AW/BW in DM group were significantly higher than those in NC group(P

Objective To explore the signal mechanism of proliferation stimulating effect of connec-tive tissue growth factor (CTGF) on hypertrophic scar (HS) derived fibroblasts. Methods <'3>H-TdR in-corporation technique was used to determine the proliferative effect of CTGF at different concentration. Western blot was applied to semi-quantitively analyze the expression of phosphorylated and total ERK1/2 protein after 0, 5, 10, 15, 30, and 60 min of CTGF stimulation, and the relative value of which was de-fined as AI to measure the activation of ERK1/2 signal pathway. PD98059 was admitted to specifically block the ERK1/2 pathway, and subsequently cell proliferation stimulated by CTGF was studied by MTT. Results CTGF could stimulate fibroblasts proliferation with a dose-dependant manner, and activa-ted the ERK1/2 signal pathway, and AI built up to 0.209±0.0201, reaching the apex at 15 min after stimulation performed. Inhibition of ERK1/2 activation by PD98059 suppressed CTGF-mediated HS fi-broblasts proliferation significantly, while OD significantly dropped. Conclusion CTGF induces a prolif-erative response in HS fibroblasts, and this action is mainly dependent on the activation of ERK1/2 signal pathway.
5.Prognosis of reducing pulmonary artery pressure in patients with congenital heart disease complicated with pulmonary hypertension

Objective To investigate the safety and efficacy of reducing pulmonary arterial pressure in patients with congeni‐tal heart disease(CHD) complicated with pulmonary hypertension(PAH) .Methods From September 2014 to January 2015 in our hospital ,34 patients with CHD complicated with PAH were selected ,all of them were treated with endothelin receptor antagonist , bosentan ,on the basis of conventional therapy .The hemodynamics observation ,cardiac function detection and six‐minute walking distance test(6 MWD)were performed after pulmonary artery pressure reducing therapy .Results After treatment ,the pulmonary vascular resistance (PVR) ,arterial oxygen saturation(SaO2 ) ,Qp/Qs and right cardiac output(CO) in these patients was (218 .30 ± 91 .25)dyn · s-1 · cm -5 ,(95 .16 ± 7 .84)% ,(1 .10 ± 0 .27) and (5 .20 ± 1 .36) L/min ,respectively ,which were better than those be‐fore treatment(P<0 .05) .There was no statistically significant difference in the pulmonary artery systolic pressure(sPAP) ,pulmo‐nary artery diastolic pressure pulse AI(dPAP) ,mean pulmonary arterial pressure(MPAP) ,heart rate(HR) and central venous pres‐sure(CVP) before and after the treatment(P>0 .05) .There was no statistically significant difference in the right ventricular diame‐ter(RVD) ,left ventricular ejection fraction(LVEF) ,three tricuspid regurgitation velocity(VP) ,three tricuspid valve transvalvular pressure difference(PGTV) ,left ventricular diastolic volume (LVEDV) and left ventricular diastolic diameter(LVEDD) before and after the treatment(P>0 .05) .After 3 months and 6 months of treatment ,6 MWD was (378 .09 ± 40 .12) and (423 .07 ± 35 .32) m respectively ,both of which were higher than those before treatment ,the differences were statistically significant (P<0 .05) .Conclu‐sion The application of bosentan therapy is safe and effective for patients with CHD complicated with PAH ,which could improve pulmonary hemodynamics and 6 MWD .
6.Relationship between Th17/Treg cells and monocyte chemoattractant protein-1 in KD children

Objective:To investigate the relationship between CD 4+CD25+regulatory T cells ( Treg) and monocyte chemoat-tractant protein-1 and its mechanism in KD pathogenesis .Methods:48 patients of KD (28 cases of recovery ,20 cases of acute phase ) were admitted from June 2014 to June 2015 and the other 40 cases of healthy children was admitted to control group .The levels of serum interleukin-15,17,23 (IL-15,17,23) and MCP-1 of two groups were measured with ELISA.The levels of Treg/Th17 cell ratio of two groups were measured with flow cytometry .Results:The levels of IL-15,IL-17,IL-23 and MCP-1 of KD group were higher than control groups(P<0.05) and the levels of IL-15,IL-17,IL-23 and MCP-1 of acute phase groups were higher than recovery groups .The levels of Th17,Treg cells,Th17/Treg of KD group were lower than the control group (P<0.05),while the levels of Th17,Treg cells,Th17/Treg cells in the acute phase of KD patients were higher than stable phase .The levels of IL-15,IL-17,IL-23 and MCP-1 and Treg/Th17 were negative correlation with univariate analysis by Pearson (P<0.05).Conclusion:The imbalance between Treg and Th17 cells and MCP-1 levels continue to rise may be associated with KD onset and progress of the disease is closely related to children in KD vivo by meas -uring children with KD Treg/Th17 cells and MCP-1 ratio of assessment Children with the condition and prognosis has some signifi-cance.
7.The Dose-Effect Relationship Between Water Fluoride Levels and Hepatic Damage in Children

Objective To explore the dose-effect relationship between water fluoride levels and hepatic damage in children and observe the difference in hepatic function between high-loaded fluoride people and dental fluorosis people in the same water fluoride level region. Methods 210 children were selected and divided into seven groups according to drinking water fluoride concentrations and whether they suffered from dental fluorosis. Urine and serum fluoride content total protein TP and albumin ALB content and activities of ALT AST and LDH in serum were determined. Results Both of urine and serum fluoride of high fluoride people and dental fluorosis people were higher than those of the control moreover fluoride contents in urine and serum increased gradually with the increase of fluoride level in drinking water. No significant differences were seen in serum TP ALB ALT and AST levels among groups. Serum LDH activities significantly increased in dental fluorosis people from area of 2.58 mg/L fluoride in drinking water and in two groups from area of 4.51 mg/L fluoride. Moreover there was an obvious dose-effect relationship between the drinking water fluoride concentration and LDH activity. Conclusion If the concentration of fluoride in drinking water exceed 2.0 mg/L it will cause hepatic damage in children with an remarkable dose-effect relationship. The degree of hepatic damage is related to not only water fluoride level but also the condition with or without dental fluorosis.
8.Determination of ephedrine hydrochloride and pseudoephedrine hydrochloride in Suxing Capsules by HPLC

AIM: To establish a determination for ephedrine hydrochloride and pseudoephedrine hydrochloride in Suxing Capsules(Herba Ephedrae, Semen Armeniacae Amarum, Fructus Perillal, etc.). METHODS:HPLC with Agilent ZORBAX Extend C_(18) (250 mm?4.6 mm, 5 ?m) column was used. Acetonitrile-0.2% H_3PO_4(5∶95) was used as a mobile phase. The flow rate was 1.0 mL?min~(-1). The detection wawelength was at 210 nm. RESULTS:The methodological evaluation showed that this method had a good repeatability and reproducibility; The average recovery of ephedrine hydrochloride was 99.65% and RSD was 1.6%. The average recovery of (pseudoephedr)ine hydrochloride was 98.90% and RSD was 2.3%. CONCLUSION: This method is simple, quick, accurate and with good reproducibility and can be used for the quality control of Suxing Capsules.
9.The correlation between red cell distribution width and chronic heart failure in children with congenital heart disease

Objective To observe the changes in red cell distribution width (RDW)and plasma N -terminal pro -brain natriuretic peptide(NT -proBNP)in children with chronic heart failure due to congenital heart disease (CHD),and to investigate their correlations with cardiac function.Methods Eighty -four children with chronic heart failure and pneumonia secondary to CHD who were hospitalized in Wuhan Health Care Center for Women and Children from January to December 201 4 were enrolled.According to the modified Ross scoring system,they were divided into 3 subgroups:mild,moderate and severe heart failure subgroups.Forty -five children with pneumonia who were hospitali-zed during the same period were randomly selected as the control group.Then the levels of RDW and plasma NT -proBNP were compared in each group,and the correlation between RDW and NT -proBNP was analyzed.Results The levels of RDW and plasma NT -proBNP in children with heart failure caused by CHD were significantly higher than those of the control group[(1 5.6 ±2.2)% vs (1 3.7 ±1 .5)%,(3 252.9 ±1 061 .5)ng/L vs (67.4 ±1 7.9)ng/L, t =5.51 8,7.838,all P <0.01 ].NT -proBNP levels were gradually increased along with increasing heart failure grade, and there were statistically significant differences[(1 71 8.3 ±456.2)ng/L vs (3 371 .4 ±834.2)ng/L vs (4 681 .2 ± 1 21 4.8)ng/L,F =1 5.961 ,P <0.01 ].RDW levels also increased gradually,while there were no statistically signifi-cant differences[(1 5.2 ±1 .5)% vs (1 5.7 ±2.0)% vs (1 6.0 ±2.8)%,F =2.246,P >0.05].There was no signifi-cant correlation between RDMand NT -proBNP (r =0.1 34,P >0.05).Conclusions For children with CHD,higher RDW and NT -proBNP levels can aid in the diagnosis of chronic heart failure,and NT -proBNP level is associated with the severity of heart failure,yet it is still unclear whether RDW level can reflect the severity of heart failure.
10.Protective Effect of Gingko Biloba Extract on Acute Lung Hemorrhage Induced by Lipopolysaccharide in Newborn Rats

Objective To investigate the protective effect of Gingko Biloba extract (GBE) on acute lung hemorrhage induced by Lipopolysaccharide(LPS) in newborn rats. Methods 1. Acute lung hemorrhage models were reproduced by intraperitoneal injection with LPS (5 mg/kg). 2. Thirty two rats were randomly divided into 4 groups,GBE groups (4 mg/kg,8 mg/kg, 16 mg/kg) and LPS group 5 mg/kg. Results In group LPS, extensive lung hemorrhage was observed after 4 hours of injection . TNF - ? iung content was obvious in LPS group. The expression of lung nuclear factor(NF-kB )immunohistochemistry wasobvious. While the parameters were obviously attenuated by GBE before LPS. Conclusion GBE may be useful in the treatment of acute pulmonary inflammatory disease.
