1.Protective effect of EPO on early diabetic rat's kidney
Journal of Xi'an Jiaotong University(Medical Sciences) 2003;0(06):-
Objective To study the effect of sub-clinical dosage of EPO on early renal lesions of diabetic rats. Methods The rats were randomly divided into normal control group (NC), diabetic group (DM), diabetic rats treated with A-dosage EPO (DMTA), and diabetic rats treated with B-dosage EPO (DMTB). Diabetic rats induced by STZ were given EPO intraperitoneally for 10 weeks; then blood glucose, blood pressure, renal function indexes, renal morphological parameter, the expression of cell apoptosis associated protein Bcl-2/Bax and VEGF were measured. Results After treatment of EPO, renal functional and morphological indexes were improved significantly and the ratio of Bcl-2/Bax and VEGF increased, both in a dose-dependent manner. Blood sugar, blood pressure and hemoglobin concentration had no significant differences. Conclusion Sub-clinical dosage of EPO could play an advantageous role in diabetic nephropathy. Inhibition of cell apoptosis by upregulation of Bcl-2/Bax expression rate may be responsible for renal protective effects despite the upregulation of VEGF.

Objective To study the effect of hepatectomy operation on circulating tumor cells of liver cancer patients during surgery. Methods Circulating blood was draw from 18 liver cancer patients before and after hepatectomy and 20 healthy volunteers. Blood routine ex-amination for leukocyte enumeration and karyote counting by microscope were taken for every blood samples to compare with the number of leukocytes and karyotes between pre-hepatectomy and post-hepatectomy blood of liver cancer patients. Immunofluorescence and cell culture techniques were used to identify the tumor cells and compare with the numbers and characteristics of the circulating tumor cells in blood sam-ples among all groups. Fluorescent antibodies,AE1/AE3 and CD133 were used to mark tumor cells. Results The number of leukocytes and karyotes in post-hepatectomy blood samples of liver cancer patients increased compared with pre-hepatectomy blood samples. The AE1/AE3 positive cells-samples were more than CD133 positive cells-samples and double positive cells-samples(P<0. 001). AE1/AE3 positive cells of healthy volunteers’ blood were more than post-hepatectomy blood of liver cancer patients(P<0. 05). No significant differences in CD133 positive cells and double positive cells were found among all groups(P>0. 05). There were no significant differences in clone formation rate after cell culture among all groups(P>0. 05). For 3 clone formed samples of liver cancer patients, there were no differences in tumor source, pathologic type and differentiation level. Conclusion The circulating blood of liver cancer patients,non-cancer patients and healthy people all exist circulating tumor cells. Hepatoma resection does not affect the number and characteristics of circulating tumor cells in liver cancer pa-tients’ blood.

Journal of Geriatric Cardiology 2009;6(2):99-101
Background and Objective Hyperlipidemia is one of the most potent and best substantiated risk factors for coronary heart disease (CHD). Purified Salvia miltiorrhiza extract (PSME) had been shown to have hypolipidemic effect in animal experiments. This study aimed to evaluate its iipids modulating effects in hyperlipidemic patients. Methods In this single-blind, placebo controlled study, lipid profiles of 80 hyperlipidemic patients were checked at same conditions. They were divided into two equal groups randomly (each composing of 40 patients). They were given PSME tablet (800 mg) three times per day, or placebo tablet. All patients were put on NCEP type Ⅱ diet and six weeks later, lipid profiles were checked. Results In PSME group, total cholesterol decreased by 27.32 mg/dl (12.3% reduction), LDL-cholesterol decreased by 23.13 mg/dl (16.8% reduction) and HDL-cholesterol increased by 9.06 mg/dl (11.1%), all were statistically significant. Although triglyceride dropped by 12.12 mg/dl (5.1%) but this was not significant statistically (P=-0.34). There were no significant changes oflipids levels in the placebo group. Conclusions PSME has significant favorable effect on total cholesterol, LDL-cholesterol, and HDL-cholesterol and may be a potential agent for the treatment ofatherogenic dyslipidemia.

Chinese Journal of Internal Medicine 2013;(7):574-577
Objective To investigate whether unfolded protein response (UPR) plays a role in the pathogenesis of spondyloarthritis (SpA),and to assess UPR-related gene expression in SpA and other arthritis patients. Methods Eighteen patients with SpA,12 with rheumatoid arthritis (RA) and 6 with osteoarthritis (OA) were recruited.Macrophages were isolated from synovial fluid samples by immunomagnetic separation. The expression of UPR-regulated genes, including binding immunoglobulin protein (BiP),glucose-regulated protein 94 (GRP94),C/EBP homologous protein (CHOP),growth arrested and DNA damage-inducible 34 (GADD34),X-box binding protein 1 (XBP-1) and endoplasmic reticulum DnaJ homolog 4 (ERdj4),was tested by real time polymerase chain reaction (PCR).Results Compared with macrophages in OA patients, the expression of BiP and GRP94 mRNA [(6.06 ± 2.08) × 10-2 vs (1.11 ±0.72) × 10-2 for BiP mRNA,11.80(7.30-38.40) × 10-3 vs 1.27(1.02-4.18) × 10-3 for GRP94 mRNA,both P values <0.01] was significantly increased in macrophages in SpA patients.XBP1 mRNA was up-regulated [(12.70 ± 5.20) × 10-3 vs (4.14 ± 2.56) × 10-3,p < 0.01] in SpA group as well.UPR-regulated gene expression in SpA patients with HLA-B27 positive or HLA-B27 negative was similar. However, none of UPR-regulated genes showed different expression between the SpA group and RA group except for GADD34 mRNA [7.30 (5.56-15.40) × 10-3 vs 21.30 (12.20-27.60) × 10-3,P =0.009].Conclusions Our data suggest that UPR possibly participates in the pathogenesis of SpA,although the relationship between HLA-B27 and UPR still needs further investigation.
