1.Study on the expression of a substrate-based polypeptide and its inhibition against HCV protease in vitro

Objective To synthesize an inhibitory peptide against serine protease of hepatitis C virus(HCV)and explore its inhibition ability in vitro. Methods Based on the sequence characteristics of four natural substrates of protease in HCV,the gene sequence of an inhibitory peptide against HCV serine protease was designed and directly synthesized by PCR. The segment was subcloned into prokaryotic expression vector pBVIL1,resulting in the construction of the recombinant plasmid pBVIL1/IP,which was then transformed into E. coli HB101 strain. Purified by ion exchange chromatography,the expressed protein was added into an in vitro system,which was comprised of the inhibitory peptide(expressed protein),protease and the substrate,i.e. a NS5A-B fragment,in phosphate buffer. Then SDS-PAGE was performed to test the inhibition effect of the polypeptide. Results The recombinant expression vector pBVIL1/IP containing target gene was successfully constructed. The peptide expressed as inclusion boay was identified by SDS-PAGE. The degradation of protease substrate NS5A-B fragment is inhibited proportionally with the increasing concentrations of the peptide. Conclusion The recombinant peptide shows inhibitory effect on HCV protease in vitro.
2.Experimental Study on the in Vivo Antibacterial Effects of Norfloxacin Injection in Animals

OBJECTIVE:To study the in vivo antibacterial effects of Norfloxacin injection in animals.METHODS:After infecting the animals by injection of Escherichia coli and Staphylococcus aureus suspension into the mice's abdominal cavity and rabbit's body,the animals were divided into therapy,prophylaxis and control groups to observe the effect of Norfloxacin.RESU-LTS:All mice of control group died within 24 hours after the injection of bacteria while those of the therapy and prophylaxis groups survived.The results of experiment on rabbits were similar to those on mice.CONCLUSION:The results show that Norfloxacin injection has obvious therapeutic and prophylactic effects on infections induced by E.coli and Staphylococcus aureus.
3.Computer-aided Diagnosis of Gastric Carcinoma B ased on Feature Selection and Probability Neural Network

Based on signal to noise ratio and probabilistic neural network method associated with experimental data,all analysis model in gastric carcinoma is presented.According to the available information,the samples of gastric carcinoma can be tested and ana.Lyzed.The signal to noise ratio is first calculated.Secondly,records in the database are chosen as a training set to build a probabilistie neural network model and the feature subset is selected according to accuracy.Finally,test set is to test accuracy of model.The model is implemented using MATLAB,and it can be generalized and applied to similar disease auxiliary diagnosis region.
4.Establishment of animal model with gastric cold syndrome.

This study is to establish the gastric cold model of rats. After gastric feeding with cold water for 5 weeks and extra iced water bath in the last 2 weeks, model group show distinct physical sign of gastric cold syndrome. The pathology of gastrics reveals gastricism of model group, while treatment group(treated with Fanzuojin Wan) show mild lesion. Elisa detection of model group show that the solution of interleukin-2 (IL-2) is higher than blank group. The difference with significance among model group, treatment group and blank group reveals the success of the establishment of gastric cold syndrome.

The pathology of gastrics reveals gastricism of model group, while treatment group (treat with Zuojin Wan) shows mild lesion. Elisa detection of model group show that the solution of interleukin-2 (IL-2) is higher than the blank group. The obvious difference among model group, treatment group and blank group reveals the success of the establishment of gastric hot model.

7.Diagnosis of primary neurogenic tumor on chest wall using CT imaging

Objective To discuss manifestations of neurogenic tumors on chest wall on CT imaging and pathology.Methods 7 patients with neurogenic tumors on chest wall confirmed surgically and pathologically were reviewed.To facilitate differential diagnosis,3 cases of malignant sarcoma were reviewed for their features on CT.Results 4 out of 5 cases of neurinoma were benign,with one case showing even density on plain CT scan,3 were poorly even,one case showed obvious evenness after enhancement,and 1 case was poorly evenly enhanced,2 benign cases had compression absorption of surrounding bone of scapula or ribs.One case of moderate malignancy was uneven in plain scan and slightly enhanced after enhancement procedure,and neighboring bone of scapula and rib were compressed and destroyed and absorbed,and neighboring muscular interspace and upper mediastinum were involved.One case showed single lesion of neurofibroma,displaying relatively even density on plain scan and moderate unevenness after enhancement procedure,as well as compression and absorption of neighboring ribs.One case of neurofibroma had multiple lesions,showing uneven density on plain scan.This case did not undergo enhanced scan.The remaining 3 cases were all sarcoma including 2 cases of fibrosarcoma and one case of synovial sarcoma.Conclusion Mass of soft tissues that are situated on chest wall or paravertebral area and with smooth edge are suggestive for benign neurogenic tumor.On the contrary,rough edge and coarse shape and infiltration into neighboring tissues are characteristic for malignant ones.

9.Efficacy observation of acupuncture for dry eye syndrome of lung-yin deficiency pattern

Objective: To observe the clinical efficacy of combining two needling manipulations, Er Long Xi Zhu (two dragons playing with a pearl) and Guo Yan Re (heat produced to reach the eyes), in treating dry eye syndrome (DES) of lung-yin deficiency pattern. Methods: Fifty-six eligible DES patients of lung-yin deficiency were randomized into an observation group and a control group, with 28 cases in each group. Same acupoints were selected in the two groups: Cuanzhu (BL 2), Sizhukong (TE 23), Taiyang (EX-HN 5) and Fengchi (GB 20) were chosen as the major points and Feishu (BL 13) and Chize (LU 5) as the adjuvant. Er Long Xi Zhu and Guo Yan Re needling manipulations were applied in the observation group while twirling reinforcing manipulation was used in the control group. Treatment was conducted once per day in both groups, for two sets of 15 consecutive days at a 2-day interval. Changes in the symptom score, tear break-up time (BUT) and tear production were observed afterwards, and the clinical efficacy was also compared between the two groups. Results: The total effective rate was 92.8% in the observation group, higher than 71.4% in the control group, and the between-group difference was statistically significant (P<0.05). After treatment, the symptom score, tear BUT and tear production showed significant improvements in both groups (all P<0.05); the symptom score, BUT and tear production in the observation group were significantly different from those in the control group (all P<0.05). Conclusion: Given the same acupoint selection, combining Er Long Xi Zhu and Guo Yan Re needling manipulations can produce more significant clinical efficacy than twirling reinforcing manipulation in treating DES of lung-yin deficiency pattern.
10.Effect of Nitroglycerin on Pulse Wave Velocity in Patients with Hypertension

Objective To observe the effect of nitroglycerin(NTG) on pulse wave velocity(PWV) in patients with hypertension(HTN).Methods36 volunteers,mean age 48.1±10.2 years,were divided into HTN group and non-hypertension(NHTN) group according to whether he or she had hypertension or not.The baseline PWV and PWV at 5th minute and 10th minute after sublingual NTG were detected.PWVs of the two groups were compared.ResultsThe basal PWV in HTN group was higher than that in NHTN group.PWV was reduced significantly after NTG were given sublingually 5 minutes or 10 minutes compared with baseline condition(both P<0.01).PWV 10 minutes after sublingual NTG raised a little compared with that after 5 minutes(P<0.01).PWVs 5 minutes or 10 minutes after sublingual NTG in HTN group were higher than those in NHTN group(both P<0.05).ConclusionNTG can reduce PWV in patients with HTN.
