1.The effect of anti-CD4-epirubicin to T lymphoma in vitro and in vivo

Objective:To detect the specificity killing effect of anti-CD4-epirubicin immunoconjugate on T origin lymphoma cells.Methods:The immunoconjugate was made by linking anti-CD4 monoclonal antibody with epirubicin using Dextran T10 as linkage. The affinity and the killing effect of the immunoconjugate in vitro and in vivo were detected by complement independent cytotoxicity, immuno-histochemistry, CFU-GM and ~ 125 I-anti-CD4 radio-immuno-imaging in nude mice.Results:Anti-CD4 immunoconjugate showed a strong affinity and specific killing effect to T origin lymphoma CEM cells.Conclusion:Anti-CD4-epirubicin and ~ 125 I-anti-CD4 could be used for diagnosis and treatment of T origin lymphoma.
2.Progress in study of spectrum-effect relationship of traditional Chinese medicine and discussions.

Spectrum-effect relationship of traditional Chinese medicine is a scientific method based on fingerprint of traditional Chinese medicine, which studied the correlations between fingerprint and activity. The method revealed the activity related peaks and clarified the active components. It provided directions and thoughts for the clarification of pharmacodynamic material basis and establishment of evaluation method to reflect the inherent quality of traditional Chinese medicine. In this text we would make a systematic review about the progress in the study of spectrum-effect relationship of traditional Chinese medicine after summarized the latest years of investigations from researchers at home and abroad, including the establishment of fingerprint, efficacy evaluation, and data processing. The key problems in each part were clarified and corresponding discussions were made, providing thoughts and advices for the following study of spectrum-effect relationship of traditional Chinese medicine. At last we made a expecting on the development trend of spectrum-effect relationship of traditional Chinese medicine.

3.Correlation between CTRP-3 level and hypertension

Objective:To study expression of complement C1q tumor necrosis factor-related protein 3 (CTRP-3)in patients with hypertension and its significance.Methods:A total of 562 patients with hypertension were enrolled as hypertension group,and 570 normal subjects undergoing physical examination were regarded as normal control group. General data,plasma levels of adiponectin (APN),leptin and CTRP-3 etc. were measured and compared be- tween two groups. Correlation among CTRP-3 level and related factors of hypertension were analyzed. Results:Compared with normal control group,there were significant reductions in plasma levels of APN [(12.1±0.4)μg/ml vs. (7.3±0.5)μg/ml],leptin [(10.1±0.4)ng/ml vs. (6.2±0.8)ng/ml]and CTRP-3 [(429±15)ng/ml vs. (314±13)ng/ml]in hypertension group,P<0.05 all. Spearman correlation analysis indicated that after correcting age and gender,plasma CTRP-3 level was significant inversely correlated with body mass index,systolic blood pres- sure,diastolic blood pressure,waist circumference,waist hip ratio,levels of total cholesterol,low density lipopro- tein cholesterol (LDL-C),triglyceride,fasting blood glucose,insulin,homeostasis model-insulin resistance index (HOMA-IR),glycosylated hemoglobin,high sensitivity C reactive protein (hsCRP)(r=-0.852~-0.011,P<0.05 or <0.01),and significant positively correlated with levels of high density lipoprotein cholesterol (HDL-C), APN and leptin (r=0.654~0.962,P<0.05 all). Multi-factor regression analysis indicated that compared with high tertile CTRP-3 group,the OR=14.17 (95%CI:3.62~28.34),P=0.001 in low tertile CTRP-3 group,sug- gesting that low plasma CTRP-3 level was independent risk factor for hypertension.Conclusion:Complement C1q tumor necrosis factor-related protein 3 level significantly correlated with hypertension,and it's an independent risk factor for hypertension.
4.Correlation between CTRP-3 level and obesity

Objective:To study expression of complement C1q tumor necrosis factor related protein 3 (CTRP‐3 ) in obese patients and its significance .Methods :A total of 402 obese patients were enrolled as obesity group and 405 normal people undergoing physical examination were regarded as normal control group . The correlation among CTRP‐3 level and related indexes were analyzed ,and multi-factor regression analysis was used to study independent risk factors for obesity .Results:Compared with normal control group ,there were significant rise in body mass index (BMI) ,homeostasis model -insulin resistance index (HOMA-IR) ,levels of systolic blood pressure (SBP) ,diastolic blood pressure (DBP) ,total cholesterol (TC) ,low density lipoprotein cholesterol (LDL‐C) ,triglyceride (TG) ,fasting blood glucose (FBG) ,insulin ,glycosylated hemoglobin (HbA1c) and high sensitive C reactive protein (hsCRP) ,and significant reductions in levels of high density lipoprotein cholesterol (HDL‐C) ,adiponectin (APN) ,leptin and CTRP‐3 in obesity group , P<0.05 or <0.01 . Spearman correlation analysis indicated that after age and gender correction ,CTRP‐3 level was significant inversely correlated with BMI (r= -0.221) ,SBP (r= -0.031) ,DBP (r= -0.043) ,TC (r= -0.147) , LDL‐C (r= -0.051) ,TG (r= -0.743) ,FBG (r= -0.238) ,insulin (r= -0.053) ,HOMA -IR (r= -0.281) , HbA1c (r= -0.741) and hsCRP levels (r= -0.216) ,P<0.05 or <0.01 ,and significant positively correlated with lev‐els of HDL‐C (r=0.351) ,APN (r= 0.852) and leptin (r=0.641) ,P<0.05 all .Multi-factor regression analysis indi‐cated that after correcting other influencing factors ,compared with high tertile CTRP‐3 group ,there were significant rise in OR value in middle tertile CTRP‐3 group (OR=6.47 ,95% CI 3.58 -12.18) and low tertile CTRP‐3 group (OR=12.39 ,95% CI 3.58-29.15) , P<0.01 all .Conclusion:CTRP‐3 level is significantly correlated with obesity -related fac‐tors ,and it′s an independent risk factor for obesity .
5.Dynamic study of ocular hemodynamic changes on DR before and after panretinal photocoagulation

AIM: To explored the treatment effects of retinal laser photocoagulation and the applications of color Doppler flow imaging(CDFI) in the diabetic retinopathy.METHODS: We collected 60 patients(120 eyes) with diabetic retinopathy(Ⅲ~Ⅳstage) from February 2013 to February 2014 in Anyang Eye Hospital admitted in fundus disease department.The health control (HC) group of 55 normal people was established simultaneously.Ocular blood flow velocity of the 55 normal people (110 eyes) and the 60 patients (120 eyes) was examined by CDFI of central retinal artery (CRA) and posterior ciliary artery (PCAs) using the PHILIPS HD6.Peak systolic velocity(PSV) was recorded.The examinations of CDFI, electroretinogram(ERG) and the vision were performed in pre-operation, 1,7d, 1, 3 and 6mo postoperatively.RESULTS: Compared the PSV of CRA of the DR group before and after photocoagulation to the data of the HC group, there was a significant difference(P<0.05);compared the PSV of CRA of the DR group after photocoagulation with the data before, there was a significant difference(P<0.05).Compared the PSV of PCAs of the DR group before and after photocoagulation to the data of the HC group, there was a significant difference(P<0.05);compared the PSV of PCAs of the DR group after photocoagulation with the data before, there was a significant difference at 1d postoperatively(P<0.05), there was no significant difference at 7d, 1, 3 and 6mo(P>0.05).Compared the aA and bA of ERG of the DR group before and after photocoagulation to the data of the HC group, there was a significant difference(P<0.05).Compared the aA and bA of ERG of the DR group after photocoagulation with the data before, there was a significant difference(P<0.05).Compared the aT and bT of ERG of the DR group before and after photocoagulation to the data of the HC group, there was a significant difference(P<0.05);compared the aT and bT of ERG of the group DR after photocoagulation with the data before, there was no significant difference(P>0.05).The vision of 49 cases(98 eyes) was improved 1 to 3 rows, the effective rate was 82%.The vision of 11 cases(22 eyes) remained constant.CONCLUSION: Retinal laser photocoagulation is an effective treatment to diabetic retinopathy patients, which can significantly reduce the peak systolic velocity of the central retinal artery, improve and stabilize the condition, protect visual function in long-term clinical effect.Color doppler flow imaging can observe the changes of ocular vessel flow velocity in diabetic` eyes noninvasively, repeatedly and in real time, providing a basis for clinical treatments.
6.The effects of laminin and fibronectin on human lens epithelial cells growth characters and vimentin expression

AIM: To observe the morphologic characters and the expression of vimentin on human lens epithelial cells (hLECs)cultured on laminin and fibronectin.METHODS: The primarily and secondarily hLECs were cultured on plates treated with laminin or fibronectin. The feature of hLECS was observed and recorded using an inverse microscope with digital camera and statistical analysis was applied. MTT assay was used to show the growth curve of subcultured cells (the 3rd passage). Immunofluorescent staining was used to demonstrate the morphologic change in vimentin expression in the 3rd passaged hLECs.RESULTS: The hLECs in laminin or fibronectin group grew out significant earlier than the untreated group. The cells could be subcultured fluendly in those two treated groups,while only a few survival cells were found in the untreated group. But the hLECs could undergo apoptosis in laminin group at the terminal phase of their life cycle, while lentoid bodies emerged in fibronectin treated group. There were more viable cells in the fibronectin group from the 4th to the 7th day by MTT reaction. Immunofluorescent staining showed a clear morphological difference between the two groups.CONCLUSION: Laminin provide a suitable growth microenvironment for the hLECs while fibronectin promotes the cells proliferation and differentiation.
7.TURP plus endocrine therapy (ET) versus α1A-blockers plus ET for bladder outlet obstruction in advanced prostate cancer.

OBJECTIVETo compare the effect of transurethral resection of the prostate combined with endocrine therapy (TURP + ET) with that of αlA-blockers combined with ET ((αlA-b + ET) in the treatment of bladder outlet obstruction (BOO) in patients with advanced prostate cancer (PCa), and to investigate the safety of the TURP + ET for the treatment of PCa with BOO.
METHODSWe retrospectively analyzed 63 cases of PCa with BOO, 28 treated by αlA-b + ET and the other 35 by TURP + ET. We obtained the residual urine volume (RV), maximum urinary flow rate (Qmax), International Prostate Symptom Score (IPSS), and quality of life score (QoL) before and after treatment along with the overall survival rate of the patients, followed by comparison of the parameters between the two methods.
RESULTSAt 3 months after treatment, RV, IPSS, and QoL in the TURP + ET group were significantly decreased from (137.8 ± 27.6) ml, (22.3 ± 3.6), and (4.2 ± 0.8) to (29 ± 13.6) ml, (7.8 ± 2.1), and (1.6 ± 0.5) respectively (P < 0.05), while Qmax increased from (5.6 ± 2.1) ml/s to (17.6 ± 2.7) ml/s (P < 0.05); the former three parameters in the αlA-b + ET group decreased from (133.6 ± 24.9) ml, (21.5 ± 3.2), and (4.7 ± 1.1) to (42 ± 18.3) ml, (12.8 ± 2.6), and (2.5 ± 0.7) respectively (P < 0.05), while the latter one increased from (6.3 ± 2.4) ml/s to (11.7 ± 2.3) ml/s (P < 0.05), all with statistically significant differences between the two groups (P < 0.05). The overall survival rate of the TURP + ET group was not significantly different from that of the αlA-b + ET group (51.4% vs 46.4% , P > 0.05).
CONCLUSIONTURP + ET is preferable to αlA-b + ET for its advantage of relieving BOO symptoms in advanced PCa without affecting the overall survival rate of the patients.

9.Clinical study of 23G vs 20G vitreous surgery combined phacoemulsification and IOL implantation for macular epiretinal membrane with cataract with

AIM: To evaluate the effects of 23G vs 20G pars plana vitrectomy ( PPV ) combined with internal limiting membrane peeling, phacoemulsification and intraocular lens implantation for macular epiretinal membrane with cataract. ·METHODS: Totally 45 eyes of 45 patients with macular epiretinal membrane and cataract were enrolled in this retrospective non-randomized controlled clinical study. All eyes were treated with PPV combined with internal limiting membrane peeling, phacoemulsification and intraocular lens implantation. There were 20 eyes in 23G PPV group, and 25 eyes in 20G PPV group. The best corrected visual acuity ( BCVA ) , intraocular pressure (IOP), counting of corneal endothelial cells ( CEC) and central retinal thickness ( CRT ) were examined before surgery. BCVA results were converted to the logarithm of the minimum angle of resolution ( LogMAR ) visual acuity. All operations were performed by the same doctor. Operation time for vitrectomy and membrane peeling, average ultrasound energy ( AVE) and effective phacoemulsification time ( EPT ) were recorded. BCVA and CRT were observed postoperatively at 30d and 90d, counting of CEC was observed postoperatively at 90d. IOP was observed postoperatively at 1d and 7d. ·RESULTS:The mean operation time for vitrectomy were 12. 57± 1. 35min in 23G group and 17. 30 ± 1. 19min in 20G group. The difference was statistically significant ( t =-12. 488, P<0. 01). There were no statistical significances in operation time for membrane peeling, AVE and EPT between 23G and 20G groups ( t=-0. 68,-1. 186,-0. 737, P=0. 500, 0. 242,0. 465). On 1d after surgery, IOP in 23G group was lower than that in 20G group, the difference was statistically significant (t= -2. 345, P=0. 024). The BCVA and CRT of the two groups both improved after operations. There were no statistically significant differences between two groups in terms of IOP, BCVA, and CRT ( F = 0. 465, 1. 895, 0. 689; P = 0. 499, 0. 176, 0. 411). IOP, BCVA and CRT were significant statistical different in different time-point within each group ( F=291. 245, 103. 06, 665. 402, P<0. 01 ). Different surgical methods of 23G and 20G had interactive effects on IOP with different time points ( F = 13. 245, P<0. 01 ), but different surgeries had no interactive effects on BCVA and CRT with different time points (F=1. 212, 2. 293;P=0. 283, 0. 129). The counting CEC in 23G group was more than that in 20G group postoperatively at 90d, the difference was statistically significant (t=2. 049, P=0. 048). ·CONCLUSION: The 23G PPV combined with internal limiting membrane peeling, phacoemulsification, intraocular lens implantation for macular epiretinal membrane with cataract is effective. Compared with 20G PPV, 23G PPV has advantages in operation time for vitrectomy and counting CEC. But lower IOP is likely in 23G PPV on 1d after surgery
10.Inhibition effect of c-Met inhibitor on proliferation of lens epithelial cells

AIM: To observe the inhibition of c-Met inhibitor on proliferation of lens epithelial cells (LECs).METHODS: Human's LECs were cultured and hepatocyte growth factor (HGF) and K252a were added to second passage of cells supplied with Dulbecco's modified eagle's medium (DMEM). MTT assay was used to examine the proliferation of LECs, and Western-blot was used to detect the expression change of Bcl-2 and Caspase-3.RESULTS: The photodensity (A) of HGF (50nmol/L) + K252a (30nmol/L) was not significantly different from that of DMEM control (P>0.05). The expression of Bcl-2 and Caspase-3 were not significantly different from that in the control group.CONCLUSION: K252a, the inhibitor of c-Met, can effectively inhibit the proliferation of LECs.
