Objective To screen serum proteomic marker of hepatic echinococcosis, establish a diagnotic model of serum protein fingerprint patterns, and evaluate its clinical application for hepatic echinococcosis. Methods Serum samples from 68 patients with hepatic echinococcosis matched with 73 controls composed of 33 patients with liver diseases other than hepatic echinococcosis and 40 healthy people were collected. All subjects were divided into training group (37) and testing group (67). Serum protein profiling of patients with hepatic echinococcosis and controls were detected using surface enhanced laser desorption/ionization time of flight mass spectrometry(SELDI-TOF-MS) and weak cation exchange protein chip(WCX2). Peak intensities were compared, in the training group, between 37 patients with hepatic echinococcosis and 37 controls, 5 patients with HCE and 5 patients with HAE, and 8 patients with hepatic echinococcosis before and after operation, respectively. ZJU-Protein Chip Data Analyze System(ZJU-PDAS) was used for data analysis and the model of serum protein fingerprint patterns was build by support vector machine (SVM). The sensitivity and specificity of the model for diagnosis of hepatic echinococcosis were verified by blind method on samples of testing group. Results There were nine different protein peak spectra between hepatic echinococcosis group and control group, of which eight protein peak spectra decreased in patient group, their relative molecular mass were 1044, 1047, 1073, 1075, 1338, 6453, 6649, 8714 m/z, respectively, while one protein peak spectrum(5651 m/z) increased(P < 0.05). The sensitivity, specificity, positive predictive and negative predictive value of the model validated by blind method were 77.4% (24/31), 66.7% (24/36), respectively. There were two different protein peak spectra between HCE group and HAE group, Their relative molecule mass were 8716 and 2751 m/z, respectively (P < 0.05). Six different proteins were detected from pre-operation group and post-operation group. Their relative molecular mass were 1297, 1505, 1525, 1534, 5921, 5941 m/z, respectively(P < 0.05). Conclusions It is a successful way to screen serum proteomic marker in patients with hepatic echinococcosis by SELDI-TOF-MS and Bio-informatics, and the marker has a potential clinical value in diagnosis and judging prognosis of hepatic echinococcosis.

Objective: To observe the effect of electroacupuncture (EA) pretreatment on the protein expression of c-fos in fastigial nucleus (FN) and lateral hypothalamus area (LHA) in rats with acute myocardial ischemia-reperfusion injury (MIRI), and to explore the role and mechanism of FN and LHA in EA at the Heart Meridian fighting against acute MIRI reaction. Methods: Seventy Sprague-Dawley rats were randomly divided into a sham operation group, a model group, an EA-Heart Meridian group and an EA-Lung Meridian group, with 14 rats in each group; an LHA lesion plus EA-Heart Meridian group (LHA+EA-Heart Meridian group) and a FN lesion plus EA-Heart Meridian group (FN+EA-Heart Meridian group), with 7 rats in each group. Except the sham operation group, the left anterior descending branch of coronary artery was ligated to establish acute MIRI rat models in the other 5 groups. In the three groups with EA-Heart Meridian treatment, Shenmen (HT 7) and Tongli (HT 5) were selected; Taiyuan (LU 9) and Lieque (LU 7) were selected in the EA-Lung Meridian group. All the EA groups received EA stimulation prior to modeling, with 1 mA in current intensity and 2 Hz in frequency, 20 min each time, once a day for a total of 7 d. The sham operation group and the model group did not receive EA stimulation. The electrocardiogram was observed in the rats to analyze the ST-segment deviation and cardiac arrhythmia score. The expression of c-fos protein in FN and LHA was detected by immunohistochemistry method. Results: Compared with the sham operation group, the ST-segment deviation, cardiac arrhythmia score and the expression of c-fos protein in the FN and LHA increased significantly in the model group (all P<0.05). Compared with the model group, the ST-segment deviation, cardiac arrhythmia score and the expression of c-fos protein in FN and LHA decreased significantly in the EA-Heart Meridian group (all P<0.05). Compared with the EA-Heart Meridian group, the ST-segment deviation and cardiac arrhythmia score increased significantly in the EA-Lung Meridian group, LHA+EA-Heart Meridian group and FN+EA-Heart Meridian group (all P<0.05); the expression of c-fos in FN increased significantly in the EA-Lung Meridian group and LHA+EA-Heart Meridian group (both P<0.05); the expression of c-fos in LHA increased significantly in the EA-Lung Meridian group and FN+EA-Heart Meridian group (both P<0.05). Conclusion: FN and LHA are involved in the mechanism of EA at Heart Meridian to improve the acute MIRI reactions, and the cerebellum may participate in the improvement of cardiac function by EA through the cerebellum-hypothalamus projection.

OBJECTIVETo observe the effects of Jingqianping Granule (JG) on mRNA and protein expressions of mu opioid receptor in the parietal cortex and the frontal cortex, the hypothalamus and hippocampus of premenstrual syndrome (PMS) Gan-qi invasion rats.
METHODSTwenty rats were selected to prepare the PMS Gan-qi invasion model. After modeling rats were divided into the model group and the Chinese herb treated group, ten in each group. Another 10 rats were selected as the normal control group. During the modeling, JG (1.6 g/kg) was given to rats in the Chinese herb treated group by gastrogavage, while equal volume of normal saline (1 mL/100 g) was given to rats in normal control group and the model group. All treatment was performed once daily for five successive days. The mRNA and protein expressions of mu opioid receptor in the parietal cortex and the frontal cortex, the hypothalamus and hippocampus were detected using RT-PCR and Western blot respectively.
RESULTSCompared with the normal control group, the bands of products of MOR mRNA and protein in the parietal cortex and the frontal cortex were relatively weaker in the model group, and the optical density value decreased. The MOR mRNA and protein expressions in the parietal cortex and the frontal cortex relatively decreased. But the bands of products of MOR mRNA and protein in the hypothalamus and hippocampus were relatively stronger and optic value increased. The MOR mRNA and protein expressions in the hypothalamus and hippocampus relatively increased with statistical difference (P<0.01, P<0.05). Compared with the model group, the bands of products of MOR mRNA and protein in the parietal cortex and the frontal cortex were relatively enhanced, the MOR mRNA expression in the parietal cortex increased, the MOR protein expression in the parietal cortex and the frontal cortex increased in the Chinese herb treated group. The bands of products of MOR mRNA and protein in the hypothalamus and hippocampus were relatively weaker. The MOR mRNA and protein expressions in the hypothalamus and hippocampus relatively decreased. The MOR protein expression in the hippocampus decreased relatively with statistical difference (P<0.01, P<0.05).
CONCLUSIONSExpression of mu opioid receptor in brains of PMS Gan-qi invasion rats was regionally specific. Administration of JG showed corresponding regulatory effects.
