1.Effects of Akt signal pathway on the expression of NF-κB in renal tubular epithelial cells

Objective To investigate the role of Akt signal pathway on the expression of monocyte chemoattractant protein-1 ( MCP-1 ) and nuclear transcription factor-κB (NF-κB) in renal tubular epithelial cells (HK-2) stimulated by albumin and to explore the mechanisms of action. Method The HK-2 cells were incubated in the presence of albumin (5,15,30 mg/mL) with or without Ly294002 (an inhibitor of Akt). Expression of mRNA was analyzed by reverse transcription-polymerase chain reaction (RT-PCR).Expression of Akt and protein MCP-1 were assessed by Western blot. Electrophoretic mobility shift assay (EMSA) was used to detect the activation of NF-κB. q-test was used to evaluate the differences in means between groups. Results Compared with control group, the expression of MCP-1 mRNA remarkly increased. [Control group: 0.233 ±0.01; BSA(5 mg/mL) group: 0.285 ±0.04; BSA( 15 mg/mL) group:0.387 ± 0.02; BSA ( 30 mg/mL) group: 0.473 ± 0.05; BSA ( 30 mg/mL) + Ly294002 group: 0. 325 ±0.05, P < 0.05 ]. The expression of MCP-1 protein in renal interstitum of operation group were remarkly increased too. [ Control group: 100 ± 15.1; BSA ( 5 mg/mL) group: 148 ± 19.3; BSA ( 15 mg/mL) group: 176±20.7; BSA(30 mg/mL) group: 263 ± 18.1; BSA(30 mg/mL) + Ly294002 group: 175 ± 18.0, P <0.05 ]. Albumin stimulated the expression of MCP-1mRNA and protein in a dose-dependent manner. Albumin remarkably increased the activity of NF-κB. Albumin enhanced the expression of Akt. Ly294002 inhibited albumin-induced the expression of NF-κB and partially decreased the level of MCP-1. Apositive correlation was noted between NF-κB activation and MCP-1 expression( r = 0.68 ,P < 0.01 ). Conclusions Albumin-induces MCP-1 and NF-κB production via Akt signal pathway in renal tubular epithelial cells.
2.Analysis of phakic before intraocular lens implantation for fundus examination

AIM:To investigate the findings of the eyes which were examined preoperatively by three mirror contact lens before the implantation of implantable collamer lens ( ICL) . To analysis the retinal pathological changes and to explore the clinical analysis of early diagnosis and treatment in retinopathy on fundus examination before operation.
METHODS:The retrospective case series study included 127 eyes of 64 patients who underwent phakic intraocular lens implantation were received the fundus examination by three mirror from April 2011 to April 2012 in our hospital. The age, refractive diopter, the findings of Goldmann three mirror examination and the condition of retinal photocoagulation were analysed and concluded.RESULTS:A total of 34 eyes (26. 8%) out of all 127 eyes ( 64 cases ) were found to have peripheral retinal pathological changes. Eight eyes ( 6. 3%) with retinal holes, 15 eyes(11. 8%) with retinal lattice degeneration, 5 eyes ( 3.9%) with retina cream degeneration, 3 eyes (2.4%)with retinal paving stone degeneration,2 eyes with vitreoretinal adhesion and traction, 1 eye ( 0. 8%) with retinal hemorrhage. Twenty-five cases were given retinal photocoagulation and then received the ICL implantation after 3mo. The follow - up time was 1a. No retinal detachment happened.
CONCLUSION: Phakic before intraocular lens implantation for fundus examination by three mirror is contributed to find the peripheral retinal pathological changes and abnormity. And make the appropriate treatment before operation for improving the security of operation, it can also give help to the postoperative follow-up of the fundus of these patients.
3.The Effect of New Photosensitizer Combined with Adriamycin on Cell Proliferation and Cell Cycle

Objective: To study the effect of new photosensitizer Chlorophyl-derivative (CPD4) combined with Adriamycin on cell cycle and cell proliferation of MCF-7 breast cancer cell line and to investigate the mechanism of combination therapy. Methods: A new type of photosensitizer and traditional chemotherapy drug Adriamycin (ADM) were used to treat breast cancer cell line MCF-7. Flow cytometry was employed to detect apoptosis rate and cell cycle distribution in ADM group, group with photodynamic effect and the combined group. The influence of ADM on the mean fluorescence intensity and the changes in the mean fluorescence intensity after CPO4 (1.5μg/mL) treatment were analyzed. Results: The apoptosis rate of the combination group was higher than that in the other two groups, with statistical significance (P<0.01). Photodynamic effect caused G_0/G_1 phase arrest in MCF-7 cells. Low concentration of ADM increased the number of G_2/M phase cells. The percentage of G_2/M phase cells was increased in the combination group. No significant difference was found in the mean fluorescence intensity between ADM pretreated MCF-7 cells for 24 hours and 48 hours and the control group (P>0.05). Pretreatment of MCF-7 cells with ADM increased the volume of photosensitizer CPD4 into the cells. The mean fluorescence intensity at 2 hours after CPD4 incubation was the highest. Conclusion: ADM can increase the amount of CPD4 into the MCF-7 cells. Photodynamic therapy com-bined with Adriamycin has a synergistic effect on MCF-7 cells.
4.Research in analgesic effect of two intervention methods on the newborns' pain

Objective To compare the effect of music and music plus non- nutritive sucking interventions on the newboms' pain caused by heel lance. Methods 75 neonates from the neonatal intensive care unit (NICU) whose gestational age=28 weeks were randomly separated into the music group,the music plus non-nutritive sucking group and the control group. The newboms' crying and grimacing,heart rate (HR), oxygen 9at-uration(SpO2) changes and times of crying 90 seconds after heel lance were observed. Results The cry-ing and grimacing time,the convalescence time of HR and SpO2 in the two intervention groups were significantly shorter than the control group.And the amplitude of HR, SpO2 changes was also significantly smaller than the control group.The crying and grimacing time and the convalescence time of HR and SpO2 in the music plus non-nutritive sucking group were significantly shorter than the music group. Conclusions Music or music plus non-nutritive sucking can ease newboms' pain caused by heellance.Music plus non-nutritive sucking can strengthen the music ther-apy's analgesic effect.
5.Quantitative Analysis of Feature Parameter in Brain Tumor CT Perfusion Image

Objective:The abstraction of characteristics in CT perfusion image is crucial in the clinical diagnosis of brain infarction and cerebral tumor.Methods:After describing the relevant parameters of CT cerebral perfusion imaging,such as cerebral blood flow (CBF),cerebral blood volume (CBV),the mean transition time (MTT) and the permeability surface (PS).With using Visual C ++ programming and CT scan images sequence obtained,the time-density curves of the brain tumor region were visualized.The more accurate hemodynamic parameters were calculated through an improved algorithm basing on the deconvolution and measuring method of the length,angle,area and other parameters.Conclusion:The quantitative analysis of feature parameter is of great benefit to clinical doctor in making timely diagnosis and treatment of brain tumor.
8.Allogeneic adipose-derived stem cells combined with nano collagen-based bone for repair of ulna bone defects

BACKGROUND:With the continuous development of medical model, high-level treatment and rehabilitation for bone defects are required. Therefore, bone tissue engineering technique based on stem cel culture has become a more important component of bone defect repair.
OBJECTIVE:To explore and analyze the feasibility of alogeneic adipose-derived stem cels combined with nano colagen-based bone for repair of ulna bone defects.
METHODS:Seventy adult female New Zealand rabbits were selected, 10 of which were used to isolate and culture adipose-derived stem cels, another 60 were randomized into scaffold group, scaffold+bone autograft group, scaffold+alogeneic adipose-derived stem cels group, with 20 rabbits in each group. The middle part of the ulna was resected to construct 12-mm bone defect models in the three groups, and then, corresponding materials were implanted. At 6, 9 and 12 weeks after implantation, bone mineral density was detected; at 14 weeks after implantation, the rates of solid fusion rate, non-solid fusion, and false joint were observed.
RESULTS AND CONCLUSION:Compared with the other two groups, the rate of solid fusion was significantly higher and the rates of non-solid fusion and false joint were lower in the scaffold+alogeneic adipose-derived stem cels group (P < 0.05). Compared with the scaffold group, the rate of solid fusion was higher and the rates of non-solid fusion and false joint were significantly lower in the scaffold+bone autograft group (P< 0.05). The bone mineral density increased dramaticaly at 6, 9, 12 weeks after implantation of scaffold+alogeneic adipose-derived stem cels, which was significantly higher than that in the scaffold group and scaffold+bone autograft group (P < 0.05). Compared with the scaffold group, the bone mineral density in the scaffold+bone autograft group was significantly higher (P < 0.05). These findings indicate that alogeneic adipose-derived stem cels combined with nano colagen-based bone has exact fusion effects with a higher bone mineral density.
9.Evaluation of the application of regional citrate anticoagulation in sustained low efficiency hemodialysis

Objective To compare the anticoagulation effect of regional citrate and heparin in patients with sustained low-efficiency hemodialysis (SLED).Method This study was conducted in the teaching hospital of Sichuan University between November 2011 and January 2013.Sixty-three patients suffering from acute kidney injury or end-stage renal diseases (ESRD) were enrolled and further randomized to 2 groups:citrate and heparin anticoagulation treatment groups in SLED.SLED was conducted by Fresenius 4008sARrTplus dialyzer for 8 hours each session,and blood flow was set at 150 ml/min.Prothrombin time (PT),activated partial thromboplastin time (APTT) and platelet (PLT) count were analyzed.Result Sixty-three patients underwent 118 sessions of SLED.Among them,59 patients (93.7%) was discharged after treatment or converted to outpatient intermittent hemodialysis,and 4 patients died of multiple organ failure during hospitalization.Compared with that in the citrate group,both PT and APTT in heparin group was significantly higher [PT:(15.5 ± 2.0) s vs (12.3 ± 2.7) s,P < 0.001 ; APTF:(56.0 ± 10.9) s vs (32.8 ± 6.1) s,P < 0.001 ; respectively] at 2 h during SLED.However,the PT and APTT levels in heparin group decreased afterwards and were similar with those in the citrate groups at 5 h during treatment.There is no difference on PLT counts between these two groups after treatment.Conclusion The anticoagulation effect of regional citrate and heparin was similar in patients when receiving SLED.Regional citrate may be an alternative anticoagulant approach for the patients at high risk of bleeding who require the treatment of SLED.
10.Clinical characteristics and homology of Acinetobacter baumannii infection in patients in a respiratory intensive care unit

Objective To study transmission characteristics of Acinetobacter baumannii (A.baumannii )clone and clinical features of infected patients in a respiratory intensive care unit (RICU),so as to guide the effective pre-vention and control of A.baumannii transmission.Methods 40 A.baumannii strains isolated from RICU patients were performed homology analysis by pulsed-field gel electrophoresis (PFGE)and cluster analysis,relationship of strains was determined,antimicrobial resistance and clinical data were analyzed.Results 40 A.baumannii strains were divided into 11 genotypes(A-K),the main epidemic strains were type A,B and E.The resistant rate to imipen-em was the lowest(70.00%),the next was levofloxacin (77.50%).The average age of 40 infected patients was 67 years old,the average length of hospital stay was 41 days,12 patients died because of invalid treatment.There was overlapping hospitalization among patients infected with type A and E strains;among patients infected with type B strain,there was no overlapping hospitalization among the last 6 and first 6 infected patients.Conclusion The main epidemic strains of A.baumannii were type A,B and E,antimicrobial resistant rate is high;the infected patients are with high average age and long length of hospital stay.It is important to reduce the transmission of A.baumannii through rational use of antimicrobial agents,strict aseptic operation,and intensified disinfection and sterilization of hospital environment and medical devices.
