1.Effect of high viral hepatitis B virus DNA loads on vertical transmission of hepatitis B virus in late-pregnant women

Objective To investigate the effect of high viral loads(HBV DNA concentration in blood >2.0 copy/ml)on the vertical transmission of hepatitis B virus in mothers with HBV DNA positivity.Method Forty pregnant women with HBV DNA positivity were divided randomly.double-blindly into 2 groups:at 28 weeks of pregnancy,one group received oral lamivudine(100 mg/d)and the other received oral placebo.The serum HBV DNA loads were tested at 28 and 40 weeks'gestation in mothers,and serum HBV DNA,HBsAg,HBeAg and anti-HBs were examined in infants at 12 month follow up.Result Thirtynine infants finished(one twins)the follow up,and 2 infants lost(5%).Among them 4 infants were confirmed to be HBV infection(10%,4/39),2 in the treatment group(10%,2/20)and 2 in the control group(11%,2/19)(P>0.05).The serum HBV DNA levels of 40 weeks'gestation in the treatment group,compared with the levels of 28 weeks'gestation in the treatment group and 40 weeks'gestation in the control group,showed a significant decline(P<0.01).The HBV DNA levels of the mothers whose infants were infected,were(3.1±3.4) copy/ml,(3.1±3.2) copy/ml during 28 and 40 weeks'gestation,and for mothers whose infants were non-infected,the levels were(3.4±2.2) copy/ml,(2.6±1.5) copy/ml respectively(P>0.05).The mean values of anti-HBs of 18 infants in the treatment group showed no significant difference as compared to 17 infants in the control group,(594±416)U/L vs (458±398) U/L (P>0.05).Conclusion The pregnant women's HBV DNA loads could be obviously decreased from high viral loads (HBV DNA concentrations in blood>2.0 copy/ml)after they take lamivudine from 36 weeks'gestation.But it might not reduce the maternal-fetal vertical transmission of HBV infection.
2.Inhibitory effect of arsenic trioxide on human glioma cell growth

AIM: To investigate the inhibitory effect of arsenic trioxide on human glioma cell line U251 growth, change of gene expression and intracellular calcium content. METHODS: MTT method was used to observe the growth inhibition effect. Cell cycle, positive rate of proliferation cell nuclear antigen (PCNA), apoptosis associated protein Fas and Bcl-2, and intracellular calcium ion (IECa~ 2+ ) levels were measured by flow cytometry in U251 cells treated with different doses of As_2O_3. Apoptosis was detected with annexin V-FITC+PI dual parameter. RESULTS: As_2O_3 inhibited the growth of U251 cells dramatically. There were obvious dosage-effect and time-effect correlations (P0.05). The cell cycle was arrested in G_2M phase. Apoptosis occurred in U251 cells treated with As_2O_3 by annexin V-FITC+PI dual parameter detection. CONCLUSION: As_2O_3 inhibits the growth of U251 cells in vitro dramatically and induces apoptosis. The mechanism is probably associated with the improvement of Fas expression and IECa~ 2+ levels, decrease in PCNA protein expression and cell cycle arrest.
3.Distinguishing Vogt-Koyanagi-Harada disease from multiple central serous chorioretinopathy with OCT and FFA

AIM:To observe the image features of Vogt-Koyanagi-Harada disease (VKH) and multiple central serous chorioretinopathy (CSC) by fundus fluorescein angiography (FFA) and optical coherence tomography (OCT).METHODS:Thirty-two eyes of 17 patients with VKH and thirty-five eyes of 30 patients with multiple CSC were collected from 2009 to 2016 in my hospital.RESULTS:All the eyes with VKH were found small and dense fluorescein leakage in the early stage.The 17 eyes (53%) with VKH were found fluorescein accumulation in the final stage;24 eyes (75%) with VKH were found high fluorescence of optic disc.All of eyes with multiple CSC were found multifocal leakage in the early stage.And 2 eyes (6%) with multiple CSC were found high fluorescence of optic disc.There were 28 eyes (14 patients) with VKH and 25 eyes (22 patients) with multiple CSC had been done OCT in my hospital.Retinal pigment epithelial fold was only found in VKH.Fluctuation of internal limiting membrane (ILM) and membrane structure had higher sensitivity to diagnostic VKH from to multiple CSC, with sensitivity of 54% and 68% respectively.CONCLUSION:There are some similarities as well as differences between FFA and OCT in diagnosis of VKH and multiple CSC.A combination usage of FFA and OCT can be more effective in distinguishing VKH from multiple CSC.
4.Factors of Depression in Korean-Chinese Elders in the Yanbian Korean Autonomous Prefecture in China: With Reference to Han-Chinese Living in the Yanbian Korean Autonomous Prefecture.

PURPOSE: This study was to investigate depression in Korean-Chinese elder living in the Yanbian Korean Autonomous Prefecture in China. METHODS: A cross-sectional community-based survey was conducted using face to face private interviews for elders aged over 59, who have been dwelling in the Yanbian Korean Autonomous Prefecture. The samples consisted of 183 Korean-Chinese and 182 Han-Chinese with the latter as a reference group. Data were collected from August 25 to September 20, 2011 and analyzed with the SPSS 18.0 program. The GDS (Geriatric Depression Scale) was used to measure elderly depression in the subjects. RESULTS: In Korean-Chinese, the rate of depression was higher in those who had lower educational levels, and were economically supported by the government. And those who had depression showed lower scores in Chinese language proficiency, health status, and social supports, and had more chronic diseases. Factors having effects on Korean-Chinese elderly depression included perceived health status and subjective support. CONCLUSION: According to the results, for preventing the depression of Korean-Chinese, it is necessary to develop health management programs and social support networks, which were easy to approach.

5.Intervention effects of tiaozhi jiangtang tablet on insulin resistance in rats with diabetes mellitus type 2.

OBJECTIVETo observe the effect of Tiaozhi Jiangtang Tablet (TJT) on insulin resistance (IR) in rats with diebetes mellitus type 2.
METHODSThe model rats of diebetes mellitus were induced by intraperitoneal injection of streptozotocin (30mg/kg) and feeding with high lipid forage. The rats in the TJT group were treated with TJT and those in the metformin group treated with metformin as positive controls. The glucose infusion rate (GIR) was detected by glucose clamp technique after treatment for 8 weeks. At the same time, fasting blood glucose ( FBG), fasting insulin ( FINS), free fatty acids ( FFA), total cholesterol ( TC), triglyceride (TG), and high-density lipoprotein cholesterol (HDL-C) were measured respectively, and insulin sensitivity index (ISI) and HDL-C/TC calculated. The changes of insulin sensitivity and lipid metabolism were evaluated.
RESULTSTC, TG, HDL-C/TC, FINS, and FFA significantly reduced in the TJT group as compared with those in the control group, while ISI and GIR significantly increased, the effects of TJT were similar to those of metformin.
CONCLUSIONTJT is effective in increasing insulin sensitivity and improving glucose and lipid metabolisms in rats with diebetes mellitus type 2.

Objective To evaluate the correlation between cerebral blood volume and permeability surface by using muhislice CT perfusion imaging with glioma grade.Methods Ninteen patients with gliomas underwent conventional MR and multislice CT perfusion imaging preoperatively.These patients were divided into low grade and high grade groups which were correspond to WHO Ⅱ grade gliomas and WHO Ⅲ or Ⅳ grade gliomas respectively.CT data were transferred to on-line working station and processed to obtain time-signal curves,color perfusion maps and calculated perfusion parameters,including cerebral blood volume(CBV),cerebral blood flow(CBF),mean transit time(MTT)and permeability surfaces (PS)in tumoral parenchyma.Kruskal-Wallis test and correlation of CBV and PS was assessed by using SPSS 11.0 software.Results The median of CBV and PS in low-grade and high-grade glioma were 2.7, 6.5 ml/100 g;0.389,12.810 ml?100 g~(-1)?min~(-1),respectively,corresponding t value were 12.907, 13.500 with P

Background Researches demonstrated that dendritic cells(DCs) are uniformly immature in the central cornea but mature in the peripheral region of cornea.So an important question is which factor impact the maturation of DCs,especially in terms of corneal transplant rejection and the known roles of DCs in the development and persistence of some corneal diseases.Objective This study aimed to examine whether corneal stroma cells (CSCs) inhibit DCs maturation through secreting transforming growth factor beta 2 (TGF-β2) and prostaglandin E2 (PGE2).Methods DCs,T cells and CSCs were isolated and cultured from clean BALB/c and C57BL/6 mice.The level of PGE2 and TGF-β2in CSCs culture supernatant and the fresh RPMI 1640 medium were then analyzed by enzyme linked immunosorbent assay (ELISA).During the DCs maturation stage,the neutralizing TGF-β2 antibody and the EP2 receptor antagonist AH6809 were added in the CSCs culture supernatant respectively.According to the different treatment,cultured cells were assigned to different groups as follows:control group,CSCs culture supernatant group,AH6809 group,TGF-β2 antibody group,AH6809 +TGF-β2 antibody group.Subsequently,the cellular surface markers for DCs,including CD11c,CD80,CD86,and MHC- Ⅱ,were analyzed by flow cytometry.The capability of stimulating the proliferation of T lymphocytes was evaluated by allogeneic mixed lymphocyte reactions,and the function of endocytosis was assessed by fluorescein isothiocyanate-dextran(FITC) uptake.Results The data of ELISA showed a higher concentration of TGF-β2 and PGE2 in murine CSCs culture supernatant than in the fresh RPMI 1640 medium.Compared with the CSCs culture supernatant group,the expression of CD80,CD86,and MHC- Ⅱ was up-regulated ( P < 0.05 ),the expression of dextran was down-regulated ( P < 0.05 ),and the stimulate index was increased( P< 0.05 ) in the TGF-β2 antibody group; the expression of CD86,and MHC-Ⅱ was up-regulated (P<0.05),the expression of dextran was down-regulated ( F =13.740,P =0.006 ),and the stimulate index was increased(P<0.05) in the AH6809 group;the expression of MHC-Ⅱ was up-regulated and the stimulate index was increased with statistical difference in interaction(P<0.05 ) in the AH6809+TGF-β2 antibody group.Compared with the control group,the expression of CD80 and CD86,and the stimulate index was still lower(P<0.05 ).Conclusions TGF-β2 and PGE2 contribute to the inhibitory effects on DCs maturation mediated by murine CSCs in vitro and further have additive effect on the immunosuppression of DCs.
9.Screening differentially expressed genes in denucleated K562 cells with restriction display technique.

OBJECTIVETo screen differentially expressed genes in cytochalasin B (CB)-induced denucleated K562 cells by restriction display (RD) technique.
METHODSThe total RNA was isolated and purified from K562 cells before and after CB (10 mug/ml) treatment. The mRNA from both treated and untreated K562 cells were reversely transcribed into cDNA, and the differentially expressed genes were separated using RD technique combined with polyacrylamide gel electrophoresis and sliver staining, followed by cloning, sequencing and homology analysis against GenBank database of these genes.
RESULTSSeven differentially expressed genes were identified in CB-treated cells including aquaporin 1 (AQP1) gene, which was verified to be up-regulated after CB treatment by RT-PCR.
CONCLUSIONAQP1 gene might be in close association with the regulation of denucleation processes and CB-induced proliferation inhibition of K562 cells.

10.Allotransplantation of spermatogonial stem cells in KM mice.

OBJECTIVETo study the feasibility of spermatogonial stem cell allotransplantation.
METHODSThe spermatogonial stem cell allotransplantation was performed, without the use of immune inhibitor, in KM mice of similar gene types, and the spermatogenesis in recipients' testes was evaluated. The right testes were pierced for transplantation while the left ones were taken as control.
RESULTSAllotransplant germ cells in KM mice can recover normal function of spermatogenesis in the transplanted testis without any immune suppression.
CONCLUSIONAllospermatogonial stem cells can be transplanted successfully among KM mice.
