1.Effects of oxidized low density lipoprotein and lysophosphatidylcholine on cholesterol efflux from mouse peritoneal macrophage foam cells

AIM: To explore the effects of oxidized low density lipoprotein (OxLDL) and one of its component— lysophosphatidylcholine (LPC) on cholesterol efflux from mouse macrophage foam cells. METHODS: (1) Cholesterol efflux induced by apoAI from mouse peritoneal macrophage foam cells loaded with OxLDL or acylated LDL(AcLDL) was measured. (2) Cholesterol efflux induced by LPC and apoAI from macrophage foam cells separated from normal or apoE gene deficient (E 0) mouse loaded with AcLDL were measured. RESULTS: (1) When the macrophage foam cells were incubated with apoAI, cholesterol efflux from AcLDL-induced macrophage foam cells increased significantly compared to that of OxLDL-induced macrophage foam cells. (2) LPC promoted cholesterol efflux from macrophage foam cells in relation to both dosage and time. When LPC was incubated with E 0 mouse macrophage foam cells, the released cholesterol mass was significantly lower than that of normal mouse macrophage foam cells. It was also found that cholesterol efflux induced by apoAI normally occurred in E 0 mouse macrophage foam cells. CONCLUSIONS: (1) OxLDL accumulated cholesterol in macrophages and impair cholesterol efflux. (2) LPC induced cholesterol efflux from macrophage foam cells, which may occur via apoE pathway.
2.Analysis of intraperitoneal hyperthermic chemoperfusion combined with interleukin-2 in the treatment of malignant ascites

Objective To evaluate the efficacy of intraperitoneal hyperthcrmic chemoperfusion in combination with interleukin 2 in the treatment of malignant ascites.Methods A total of 47 patients aged 47-75 years with malignant ascites were randomly divided into two groups:the experimental group and the control group.All patients received a closed drainage and were treated with paclitaxel liposome,cisplatin (DDP),interleukin-2 and dexamethasone by intraperitonealinfusion.Patients were asked to change the body position.In addition,the patients in experimental group were received the high frequency hyperthermia immediately at a temperature of 42℃ to 43℃ for 90 minutes.Results The remission rate (RR) was higher in the experimental group than in the control group (75.0 % vs.43.5%,x2 =4.85,P<0.05).The median survival time (MST) was longer in the experimental group than in the control group (34 weeks vs.21 weeks,P<0.05).There were no significant differences in adverse reaction incidences between groups (P > 0.05).Conclusions Compared with intraperitoneal chemoperfusion alone,the intraperitoneal hyperthermic chemoperfusion has a higher RR and longer MST with no increased adverse reaction incidence,which can be in the clinical application.
3.The study of tumor necrosis factor-alpla single nucleotide polymorphism in Chinese Han patients with polymyositis/dermatomyositis

Objective To investigate the tumor necrosis factor alpha gene single nucleotide polymorphism (SNP) in Chinese Han polymyositis/dennatomyositis (PM/DM) patients. Methods A casecontrol study of three TNF-α SNPs were undertaken and comparison between cases of PM/DM (n=69) and DM patients ( n=52 ) with normal subjects ( n=57 ) was performed. The genotype of every subject was determined by polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP). Results There was no significant difference in the genotypes and alleles frequencies of the three TNF-α SNPs(-238,-308, and -1031)between PM/DM or DM and normal subjects. The haplotype AGT (-238, -308, -1031 ) significantly increased in controls vs PM/DM and DM (PM/DM, P=0.01; DM, P=0.02) subjects. The haplotype AGC was a significant risk factor for DM (P=0.04, 0R=9.84, 95%CI 1.39~69.57 ). The haplotype AGC increased in PM/DM (4.3%)vs controls(0.6% ), but the difference was not significant (P=0.09, OR 7.22, 95%CI 1.02~50.90). The haplotype GGC was significantly decreased in the DM subgroup vs healthy control subjects (P=0.04, OR=0.47,95%CI 0.23~0.99). Conclusion The TNF gene (-238,-308,-1031) haplotype AGC is a risk factor for DM.
4.Internal standard method to determine the recovery of nicotine in microdialysis

The paper reports the evaluation of the feasibility of using internal standard method for the determination of nicotine recovery in microdialysis in vitro. This in vitro experiment included two conditions. Nicotine and codeine phosphate were dissolved in Ringer's solution. Nicotine, codeine phosphate and the mixture of them were perfused through the CMA30 linear probe separately to calculate the proportion of the recovery (or delivery) of nicotine to that of codeine phosphate. And then codeine was perfused through the probe which was immersed in nicotine solution with different concentrations to calculate the proportion, too. In another condition nicotine was dissolved in rat plasma. The rat plasma protein binding rate was determined by using retrodialysis and internal standard method in vitro. The results are as follows: the proportion of the recovery (or delivery) of nicotine to that of codeine phosphate was fairly stable. The delivery of codeine was independent of nicotine concentration in the external medium. Protein binding rate determined by retrodialysis was almost the same as that determined by internal standard method. It suggests that the internal standard method is an effective way in the determination of nicotine recovery and codeine phosphate can be used as the internal standard.
5.Effects of IDH1 and IDH2 genes mutations on tumors

Isocitrate dehydrogenases(IDHs)are considered as key enzymes in the tricarboxylic acid cycle. Recurrent mutations in the IDH1 and IDH2 genes are recently found in several human cancers. Those point mutations specifically affect IDH1 and IDH2 active site arginine residues and confer a neomorphic enzyme function of directly catalyzing α-ketoglutarate(α-KG)to R-2-hydroxyglutarate(R-2-HG). R-2-HG can com-petitively inhibits α-KG-dependent enzymes and may therefore contribute to the occurrence and development of tumor. In addition,Mutation status of IDH1 and IDH2 are closely relative to the progress and prognosis of cer-tain tumor. Thus IDH1 and IDH2 are considered to be promising biomarkers for early diagnosis and prognosis and targeted therapy.
6.Analysis on prescription evaluation and drug use in a hospital

Objective To promote rational drug use by investigating quality of the outpatient and emergency prescriptions in the hospital .Methods We statistically summarized the evaluations of prescriptions and analyzed the drug use from January to Novem-ber ,2012 .Results The outpatient and emergency prescriptions were compliant to relevant writing standards ,without missing i-tems .The inaptitude of prescriptions were mainly in forms of inappropriate indication ,usage ,and dosage of drug and menstruum . The irrational prescriptions accounted for 5 .5% of those evaluated .Conclusion Rational drug use has been basically ensured through implementation of prescription evaluations and interventions .Irrational prescriptions exist in relatively small scale and are to be eliminated .

Objective To investigate the variation of corticotropin-releasing hormone(CRH)in plasma,the mRNA and protein expression of c-Fos and gap junction gene connexin-43(Cx43)in the myometrium of term pregnancy women,and to study the correlations among CRH,Cx43 and c-Fos at onset of labor.Methods 30 cases in labor(L group),30 cases not in labor(NL group)but in term pregnancy, and 30 cases of non-pregnant(NP group)women undergoing hysterectomy due to cervical intraepithelial neoplasia were recruited into this clinical study.Radioimmunoassay was employed to measure the concentration of CRH in plasma;in situ hybridization assay and streptravidin-biotin peroxidase(SP) immunohistoehemical techniques were respectively used to detect the expression levels of c-Fos mRNA,Cx43 mRNA and the corresponding proteins.Results(1)The concentration of CRH in L group(81.8?11.9) pmol/L was significantly higher than that in NL group(34.5?18.6)pmol/L(P

9.Study on the In-vitro Microdialysis Recovery of Nicotine and Its Influence Factors

Objective To detect the in-vitro microdialysis recovery of nicotine by decrement method(i.e.retrodialysis) and to investigate its influence factors,thus to supply experimental evidence for the in-vivo microdialysis of nicotine.Methods The in-vitro recovery of nicotine was detected by concentration difference method(increment method and decrement method).The stability of recovery was observed.The effects of the pretreatment with perfusate,and the flow rates and concentrations of the perfusate on nicotine recovery were studied.Results Filtering and stirring treatment increased the recovery,but the time of ultrasound degassing showed no effect on the recovery.The recovery detected by increment method was as the same as that detected by decrement method.Nicotine recovery was independent of nicotine concentration in the external medium,showing good stability.Conclusion It is necessary to filter and degas the perfusate before microdialysis.Retrodialysis can be used for the determination of in-vivo nicotine recovery.

Objective To establish the method of extraction and separation of formononetin in Mucuna sempervirens Hemsl..Methods The extraction and separation conditions of formononetin was studied by orthogonal test.Results Solid-liquid ratio was 1:40(g:mL),with methanol refluxing and extracting for 3 times at 85 ℃,1.5 h once,the content of formononetin was 0.038 12%.Conclusion The optimal parameters for extraction and separation condition of formononetin in Mucuna sempervirens Hemsl.was established.
