7.Glucagon-Like PeptideⅠand Targeted Differentiation of Stem Cells
Lei YAN ; Xiao-Ling MU ;
China Biotechnology 2006;0(04):-
Diabetes mellitus has become one of the diseases which threaten the heath of human being in the 21st century.A goal of research in diabetes is to find a way to increase the number of functional insulin-producing cells. Islet transplantation has been considered to be the most effective approach to cure type Ⅰ and part of type Ⅱ diabetes mellitus.This approach, however, is severely limited by an inadequate supply of donor islets available for transplantation.Moreover, recent progress of stem cells research has shown that stem cells may act as a new source of islet transplantation in diabetes mellitus treatment. Recent evidence indicates that Glucagon-Like PeptideⅠ(GLP-1) plays a very important role in targeted differentiation of stem cells into Insulin-Producing Cells and pancreatic development. GLP-1 is an intestine-derived insulinotropic hormone that stimulates glucose dependent insulin production and secretion. GLP-1 can induce differentiation of stem cells into insulin-producing cells, which is achieved by up regulation of PDX-1 expression.PDX-1 is a transcription factor critical for pancreatic development and endocrine cell neogenesis and a marker for pancreatic stem cells. These new findings suggest an approach to create Insulin-Producing cells in vitro by expanding stem/progenitor cells and then to convert them into Insulin-Producing cells by treatment with GLP-1. Thus GLP-1 may be a means by which to create Insulin-Producing cells ex vivo for transplantation into patients with insulinopenic type Ⅰ diabetes and severe forms of type Ⅱ diabetes. This article reviews recent progress about GLP-1 and targeted differentiation of stem cells induced by GLP-1.

International Journal of Laboratory Medicine 2014;(16):2154-2155,2158
Objective To investigate the clinical significance of the combination detection of blood lactic acid,B-type brain natri-uretic peptide(BNP)and homocysteine(Hcy)in elderly acute cerebral infarction.Methods The arterial and venous blood during a-cute onset stage,at 24,48,72 h after thrombolysis and at 12 h after fasting in 85 cases of acute cerebral infarction was collected for detecting blood lactic acid,BNP and Hcy,performing the dynamic monitoring analysis and the comparative observation with the healthy control group.Results The blood lactic acid,BNP and Hcy levels in the acute onset stage group and the recovery group were significantly increased compared with the healthy control group(P <0.05).The blood lactic acid,BNP and Hcy levels during the onset stage in the acute cerebral infarction group were(6.47 ±3.92)mmol/L,(100.52 ±48.96 )pg/mL and(48.96 ±15.13 )μmol/L respectively,which were significantly higher than those at 48,72 h of postoperative recovery,the differences between them were statistically significant(P <0.05).The blood lactic acid,BNP and Hcy levels during postoperative convalescence in the cere-bral infarction group were gradually decreased and declined to the minimal level until postoperative 72 h,which were(2.16±1.83) mmol/L,(33.61±10.42)pg/mL and(18.87±8.27)μmol/L respectively.Conclusion Blood lactic acid,BNP and Hcy are closely related with the occurrence and development of cerebral infarction,the joint detection of these three indicators provide a reliable ba-sis for the early diagnosis of elderly acute cerebral infarction.The active and effective treatment on the patient should be timely a-dopted and the dynamic monitoring should be performed in order to correctly assess the prognosis.

Journal of Regional Anatomy and Operative Surgery 2014;(6):578-581,582
Objective To study the effect of hepatectomy operation on circulating tumor cells of liver cancer patients during surgery. Methods Circulating blood was draw from 18 liver cancer patients before and after hepatectomy and 20 healthy volunteers. Blood routine ex-amination for leukocyte enumeration and karyote counting by microscope were taken for every blood samples to compare with the number of leukocytes and karyotes between pre-hepatectomy and post-hepatectomy blood of liver cancer patients. Immunofluorescence and cell culture techniques were used to identify the tumor cells and compare with the numbers and characteristics of the circulating tumor cells in blood sam-ples among all groups. Fluorescent antibodies,AE1/AE3 and CD133 were used to mark tumor cells. Results The number of leukocytes and karyotes in post-hepatectomy blood samples of liver cancer patients increased compared with pre-hepatectomy blood samples. The AE1/AE3 positive cells-samples were more than CD133 positive cells-samples and double positive cells-samples(P<0. 001). AE1/AE3 positive cells of healthy volunteers’ blood were more than post-hepatectomy blood of liver cancer patients(P<0. 05). No significant differences in CD133 positive cells and double positive cells were found among all groups(P>0. 05). There were no significant differences in clone formation rate after cell culture among all groups(P>0. 05). For 3 clone formed samples of liver cancer patients,there were no differences in tumor source, pathologic type and differentiation level. Conclusion The circulating blood of liver cancer patients,non-cancer patients and healthy people all exist circulating tumor cells. Hepatoma resection does not affect the number and characteristics of circulating tumor cells in liver cancer pa-tients’ blood.
