1.Effects of 2-methoxyl estradiol on the expression of L-selectin protein and its mRNA in K562 cells

Objective To investigate the effects of 2-methoxyl estradiol(2-ME)on the expression of L-selectin and its mRNA and the relationship between L-selectin and bcr/abl fusion gene expression in chronic myelocytic leukemia(CML)K562 cells.Methods The cultured K562 cells were divided into three groups.The control group,K562 cells were cultured without 2-ME treatment.The experimental group,K562 cells were cultured in the medium containing different concentrations of 2-ME(1,2,4,8 and 16?mol/L)for 36h.The negative control group,K562 cells were replaced by water without RNase in the medium containing different concentrations of 2-ME for 36h.The expression of L-selectin protein in K562 cells was determined by flow cytometry(FCM)method.The expression of L-selectin mRNA in K562 cells was detected by half-quantitative reverse transcription-polymerase chain reaction(RT-PCR).The bcr/abl fusion gene expression in K562 cells was detected by quantitative SYBR Green Ⅰ real-time RT-PCR.Results After being treated with 2-ME at different concentrations for 36 hours,the expression of L-selectin protein and its mRNA increased in K562 cells in a concentration-dependent manner,and the expression level in the experimental group was higher than that in the control group(P

Recent studies have shown that platelets have a variety of important functions, and play an important role in the pathophysiology of liver and renal diseases. The quantitative and functional changes of platelets, could affect the bleeding complications in patients with liver and renal diseases. This article reviews the progress of the effects of thrombocytopenia and platelet dysfunction on patients with liver and renal diseases, and the role of platelets in the pathophysiology of liver and renal diseases in the 58th American Society of Hematology Annual Meeting.
3.External effect of honokiol on U937 cell

AIM: To investigate the anti-proliferative and apoptosis-inducing effect of honokiol on U937 cell line in vitro. METHODS: Proliferation of U937 cells and PBMCs were analyzed by MTT assay. Flow Cytometry and cell morphological observation were performed to find out whether honokiol could affect cell cycle and induce apoptosis of U937 as well as PBMCs in vitro. RT-PCR and Western blotting techniques were used to detect the changes in mRNA expression and protein production of bcl-2 and bax in U937 cells after treated with honokiol. RESULTS: Honokiol could significantly inhibit the proliferation of U937 cells at IC_ 50 concentration of 11.8 ?g/mL, but slightly inhibit the proliferation of PBMCs, at IC_ 50 concentration of 40.3 ?g/mL, respectively. Most honokiol-treated cells were arrested at G_0/G_1 phase. CONCLUSION: Honokiol could inhibit the proliferation and induce apoptosis of U937 cells, while has little effect on the proliferation and survival of PBMCs. Bax might be involved in the gene regulation related to honokiol-induced apoptosis.
4.Development of Multiple ELISAs for the Detection of Antibodies against Classical Swine Fever Virus in Pig Sera

The major immunogenic proteins (Ems,E2 and NS3) of classical swine fever virus (CSFV) (Shimen strain) were expressed in E.coli and purified by affinity chromatography.The recombinant antigens were applied to develop multiple enzyme-linked immunosorbent assays (ELISAs) for the detection of specific antibodies in pig sera.Optimum cut-off values were determined by receiver operating characteristic (ROC) analysis after testing 201 sera of vaccinated pigs and 64 negative sera of unvaccinated piglets.The multiple ELISAs were validated with 265 pig sera yielding high sensitivity and specificity in comparison with the virus neutralization results.The results demonstrated that multiple ELISAs can be a valuable tool for the detection of CSFV infection and serological surveys in CSFV-free countries or for the evaluation of the antibody responses in pigs induced by a live attenuated C-strain vaccination.
5.Diagnostic and therapeutic advances in relapsed and refractory acute myeloid leukemia

Although globally recommended standardized stratification treatment of acute myeloid leukemia (AML) effectively improves its therapeutic effects and prognosis,due to unknown personalized differences of tumorous biology,most of the patients eventually develop to therapy failure,relapse,and even death.Additionally,a number of patients are naturally resistant to current used combined chemotherapy and with extremely poor prognosis,This review presents definitions of refractory and relapsed AML,then describes their pathogenetic mechanism and lastly summarizes three aspects of therapeutic principle,clinical drugs and hematopoietic stem cell transplantation (HSCT).To review advances in relapsed and refractory AML diagnosis and treatment will be helpful to master disease characteristics and to guide clinical treatment.

Acute myeloid leukemia (AML) is a group of heterogeneous diseases with diverse genetic abnormalities,variable responsiveness to therapy and prognosis.In recent years,a lot of information has become available regarding chromosome and gene mutations that occur in AML and their influence on prognosis.Improvements in the understanding of molecular biology of AML are critical for accurate diagnosis.risk stratification,monitoring of minimal residual disease and provides opportunities to develop targeted therapies and improve the clinical outcome.This article reviewed chromosome abnormalities and characteristic gene mutations,and discussed their clinical signiflcances and presented new drugs in clinical trials presented in the 55th ASH annual meeting.
7.Influence of visfatin in expressions of insulin receptor substrate and PI3K in fat cells in vitro

Objective:To investigate the relationship between visfatin and insulin resistance (IR)of type 2 diabetes mellitus(T2DM) through the classic insulin signaling pathway phosphatidy inositol-3 kinase (PI3K).Methods:The human T2DM preadipocyte cells were recoveried, extended and differentiated.The visfatin overexpression vectors were built, transformated, cultivated and extracted.The fat cells were transfected with different overexpression levels (0.0, 1.0, 2.5 and 5.0 μg);0.0 μg group was used as control group,and the remaining three groups as observation groups.The mRNA expression levels of visfatin, insulin receptor substrate-1(IRS-1), insulin receptor substrate-2(IRS-2) and PI3K (P85α) were detected by Q-PCR.The protein expression levels of visfatin, IRS-1, IRS-2, PI3K (P85α), IRS-1 and IRS-2 phosphorylation levels were measured by Western blotting method.The glucose uptake rates of fat cells were determined by [3H]-2-deoxidation-D-glucose uptake assay.Results:The expression levels of visfatin mRNA and protein in various groups were increased with the increase of transfection concentration gradient (P<0.01);and the constructed overexpression vector of visfatin was effective.With the increase of visfatin expression, the mRNA and protein expression levels of IRS-1 and PI3K (P85α) and IRS-1 phosphorylation degrees in various groups were increased (P<0.01), but the IRS-2 mRNA and protein expression levels had no obvious changes(P>0.05).The glucose uptake rates of fat cells were elevated with the increasing of visfatin expression (P<0.05).Conclusion:The visfatin overexpression of fat cells in vitro can increase the expression levels of IRS-1 and PI3K.
8.Lung cancer Ⅳ recipe combined with chemotherapy in treating non-small cell lung cancer in the elderly: a case-control study

Objective To explore the therapeutic effect of lung cancer Ⅳ recipe combined with chemotherapy on treating middle and advanced stages of non-small cell lung cancer (NSCLC) in the elderly.Methods A total of 77 NSCLC patients in middle and advanced stages aged 70 years and over who had heat-toxin syndrome in traditional Chinese medicine (TCM) were selected from Apr.2013 to Oct.2014 in our hospital.Patients were randomly divided into chemotherapy group (n=39) and chemotherapy + herbs group (n=38).Karnofsky assessment scale (KPS),effective rate,symptom and sign in traditional Chinese medicine,changes in the levels of high-sensitivity c-reactive protein (hs-CRP),carcino embryonic antigen (CEA),neuron specific enolase (NSE),tumor special growth factor (TSGF) were observed before and 1 and 56 days after treatment.Results There was no significant difference in effective rate between the two groups (P>0.05).The KPS and symptom and sign in TCM were improved significantly in chemotherapy + herbs group as compared with chemotherapy group (36.7 % vs.26.5 %,80.0 % vs.58.8 %,respectively (x2 =2.721,6.731,both P<0.05).There were no significant differences in hs-CRP,NSE,CEA,TSGF levels between two groups before and after treatment (P>0.05),but hs-CRP level in both groups was decreased after treatment as compared to pre-treatment and the decvease was more obvious in chemotherapy + herbs group (51.47% vs.23.77%,x2 =19.713,P<0.05).Conclusions Lung cancer Ⅳ recipe can improve the main clinical symptoms,quality of life and tolerance to chemotherapy in NSCLC patients with heat-toxin manifestation in TCM.There is a correlation between heat-toxin syndrome and hsCRP level in NSCLC patients,which shows that hs-CRP probably is the material basis of the heattoxin syndrome in TCM.
9.Expression of urotensinⅡreceptor GPR14 in cardiovasculature and brain of rats

Objective: To observe the expression of the G-protein-coupled-receptor 14 (GPR14), urotensinⅡreceptor, in the cardiovascular system and brain of SD rats. Methods: Semi-quantitative reverse transcription-polymerase chain reaction (RT-PCR) was used to detect the GPR14 mRNA. Results: In cardiovascular system, GPR14 mRNA was detected in the left ventricle, left atrium, thoratic aorta and carotid aorta. The highest level of expression was found in the left ventricle. In the brain, GPR14 mRNA was detected in cortex, hippocampus, hypothalamus and cerebellum, and higher level of expression was found in the cerebellum. Conclusion: GPR14 mRNA expression is found in the cardiovascular and neural tissues of tested rat, suggesting that urotensinⅡ may play an important role in cardiovasculature and central nervous activity.
10.Relation of body mass index to vitamin D, PTH, and bone turnover markers levels among women in Shanghai area

Objective To examine the relationship of body mass index (BMI) with levels of vitamin D,parathyroid hormone (PTH),and bone turnover markers among women in Shanghai area.Methods Altogether 810 Chinese women aged 45 year and older were enrolled to study the associations by multiple linear regression analyses.Bone mineral density was measured by a dual energy X-ray absorptiometry (Lunar,Prodigy,GE),Serum 25-hydroxyvitamin D [25 (OH) D],PTH,C-telopeptide of type Ⅰ collagen (CTX),osteocalcin (BGP),and bonespecific alkaline phosphatase (BSALP) were measured.Results (1) 25 (OH) D,BGP,BSALP,and CTX levels were significantly lower in obesity group than in control group ; PTH level was significantly higher in obesity group than in control group.(2) In multiple linear regression analyses,BMI was significantly associated with lower 25 (OH) D (β =-0.017,P =0.006),BGP (β =-0.077,P =0.019),and higher PTH (β =0.025,P =0.000).Conclusions (1) There were siginificant associations between higher BMI and lower 25 (OH) D among women in Shanghai.(2) Serum osteocalcin was associated not only with bone metabolism but also with energy metabolism.
