OBJECTIVE:To study the effects of 5-hydroxymethylfurfural on learning-memory and cerebral free radical metabolism in cerebral ischemia and reperfusion model mice. METHODS: The mice were divided into sham operation group, model group, 5-HMF low dose group, 5-HMF high dose group, and dihydroergotamine mesilate positive control group. After administration of the corresponding drugs, all groups except the sham operation group were subjected to construction of cerebral ischemia and reperfusion model. The learning-memory of mice was examined by Morris water maze and step-down tests, and cerebral superoxide dismutase (SOD) activity and malondialdehyde (MDA) content were determined as well. To explore the effect of high glucose on the NF-κB/IκB signal pathway in cultured rat glomerular mesangial cells. The results showed that high glucose increased the degradation of IκB-α and the translocation to nucleus of NF-κB. These changes could be reverted mostly by MG132, a proteasome inhibitor. It suggests that the activation of the NF-κB signal pathway by high glucose concentration may probably be via the ubiquitin-proteasome pathway.

Objective To investigate the effect of pigment epithelium- derived factor (PEDF) on p38MAPK-CREB pathway and the expression of fibronectin (FN) in human mesangial cells (HMCs) cultured with high glucose. Methods HMCs were treated with different concentrations of glucose and the osmotic control respectively in the presence or absence of PEDF for 24 h:normal glucose (5.6 mmol/L),24.4 mmol/L mannitol,high glucose (30 mmol/L),high glucose+PEDF(30 mmol/L glucose with 10 nmol/L PEDF,40 nmol/L PEDF or 100 nmol/L PEDF).After samples were collected,the expression of phospho-p38MAPK (p-p38) and p-CREB was assessed by Western blotting,while FN mRNA and protein expression was assessed with RT-PCR and ELISA respectively. Results In contrast to normal glucose and mannitol treatments,the expression of p-p38MAPK,p-CREB and FN increased significantly in high glucose group (all P＜ 0.01).However,PEDF abolished the up-regulation of p-p38MAPK,p-CREB and FN induced by high glucose (all P＜0.05). Conclusion PEDF may inhibit fibrosis through P38MAPK-CREB pathway in diabetic nephropathy.

The present study was performed to explore the role of corticotropin releasing factor (CRF) in the nucleus basalis of Meynert (NBM) in spatial learning and memory of rats. The latency, distance and swimming path to find the platform were measured by Morris water maze after intra-NBM injections of 0, 0.01, 0.1 and 0.4 nmol of CRF. Intra-NBM injections of 0.1 or 0.4 nmol of CRF induced significant increase of the latency for spatial learning and memory, and there were no significant changes in the swimming speed in Morris water maze test. The results suggest that CRF plays an inhibitory role in spatial learning and memory consolidation in the NBM of rats.

The study is aimed to establish a method to determine the content of Vinorelbine in liposomes by HPLC.The experiment was carried out on Kromasil C18(4.6 mm×250 mm,5 μm) colum with the mobile phase consistedof acetonitrile-0.06 mol·L-1 KDP buffer(pH 3.0,35:65) at a flow rate of 1.0 mL·min-1.The detection wavelength was at 215 nm and the column temperature was at 30℃.The result showed that the linear range of Vinorelbine was from 2.08 μg·mL-1 to 20.8 μg·mL-1(r=0.9999).The average recovery rate was 98.64％(RSD =1.09％).It is con cluded that the method used in this analysis is simple,precise and replicable with high recovery and accuracy.It can be used to determine the content of total Vinorelbine in liposomes.

Objective To explore the in vitro isolation of ?2m-/Thy-1+ bone marrow-derived liver stem cells(BDLSCs) which bear double features of stem and liver cells from bone marrow stem cells(BMSCs)as so to provide suitable donor cells for the treatment of liver diseases by cellular transplant. Methods ?2m-/Thy-1+ BDLSCs were isolated by magnetic bead cell sorting(MACS) method from cholestatic rats in vitro,and cell purity was detected using flow cytometry.Liver associated phenotype markers were characterized by RT-PCR and immunofluorescence staining. Results BDLSCs isolated by MACS were purified and viable,and possessed hepatocyte-like features at gene and protein levels. Conclusion ?2m-/Thy-1+ BDLSCs are special subsets of BMSCs which may have promising potentials in the stem cell-based treatment of liver diseases.

Objective To identify the isolated rat bone marrow derived mesenchymal stem cells(MSCs),and evaluate the efficiency of adenoviral vector expressing human urokinase type plasminogen activator(uPA) in transfection of rat MSCs and its effect on proliferation of MSCs. Methods MSCs were isolated and purified by pasted wall purification,and were identified by immunicytochemistry.The transfection efficiency of uPA was detected by fluorescent microscopy,the expression of uPA in MSCs was detected by Western blotting,and the proliferation of MSCs was evaluated by MTT. Results The harvested MSCs exhibited the typical appearance of MSCs,and it was revealed by immunohistochemistry that the expression of MSCs markers CD29 and CD90 was positive,while that of CD34 and CD45 was negative.A tendency of increase in expression of green fluorescent protein(GFP) was observed with increase of multiplicity of infection(MOI).After transfection with AduPA for 72 h,the transfection efficiency reached(94.0?1.5)% at MOI of 80,and positive GFP cells could still be observed even after 7 d.The transfected uPA had no effect on the proliferation of MSCs. Conclusion MSCs are favourable genetic vectors to express uPA,and can be used for treatment of liver fibrosis.

ObjectiveTo investigate the sleep status and effect of night shift work on sleep in nurses.MethodsSleep characteristics and qualities of 348 nurses, 50 communication persons and 100 night-guards were analyzed with Pittsburgh Sleep Quality Index (PSQI), and insomnia was diagnosed based on DSM-IV criteria.Results71.86% of nurses had bad sleep quality (PSQI>7); the rate of insomnia was 38.22%. The same data were 74% and 48% respectively in communication persons, and 26.76% and 7% in night-guards. The sleep quality of nurses was related to age and years of night shift work. The sleep quality of nurses was similar to communication persons (P>0.05), but significantly different from that of night-guards (P<0.01).ConclusionShift work manner influences sleep status of nurses and makes them having disturbances on falling sleep time, the time of sleeping, sleep efficiency and daytime function. But it doesn't need medication.
