2.Influence of Asphyxia on Nitric Oxide and Nitric Oxide Synthase Levels in Renal of Neonatal Rats

Objective To study the effects of nitric oxide synthase(NOS) and nitric oxide(NO) in post-asphyxial renal injury in neonatal rats.Methods Forty-eight Wistar neonatal rats were randomly divided into 4 groups:controls,2 h,24 h and 48 h post-asphyxia groups (12 in each group).The rats were decapitated in different times(2 h,24 h and 48 h) after asphyxia for 30 minutes.The renals were dissected to determined the concentrations of NO and NOS.And the scores of renal tubules were measured under light microscope.Results Compared with control group,the levels of NO and NOS significantly increased at 2 h and 24 h after asphyxia.The scores of renal tubules were significant difference at 24 h and 48 h after asphyxia compared to controls.Conclusion These findings suggest NOS and NO may play an important role in the development of post-asphyxia renal injury.
3.Diffusion Tensor Imaging of White Matter in Early Stage of Parkinson''s Disease Based on Tract-based Spatial Statistics Analysis

Purpose To observe white matter structure features of patients with early stage (Hoehn-Yahr 1-2 phase) Parkinson''s disease (PD) by using diffusion tensor imaging (DTI) based on the fiber bundle analysis tract-based spatial statistics (TBSS); and to explore the brain regions of PD patients in which DTI parameters are significantly correlated with unified Parkinson''s disease rating scale (UPDRS) score elevation. Materials and Methods DTI sequence was performed on 27 cases of PD and 30 cases of healthy volunteers. DTI parameters including fractional anisotropy (FA), mean diffusivity (MD), axial diffusivity (AD), and radial diffusivity (RD) were analyzed in all participants by using TBSS technique, and the parameters of two groups were compared. The correlation of clinical UPDRS score with FA value, MD and RD value in the PD group were analyzed.Results Compared with that in the control group, FA values of several brain regions in PD group decreased, while MD and RD value increased (P<0.05). AD showed no obvious change (P>0.05). UPDRS score of PD group was correlated with FA, MD and RD value (P=0.001). The brain regions that showed FA value decreased, MD and RD value increased included corpus callosum, left anterior limb of internal capsule, bilateral anterior radial crown, bilateral superior radial crown and left posterior thalamic radiation (P=0.001).Conclusion There is some changes in white matter structure of the patients with early stage Parkinson''s disease, which may due to demyelination or fiber integrity damaged.

This article reviews various rehabilitation treatments for dysfunction after burn injury to promote rehabilitation of burn patients.

5.Excretion of ligustrazine hydrochloride in Salviae Miltiorrhiza and Ligustrazine Hydrochloride Injection in rats

Objective To study the excretion characteristics of ligus-trazine hydrochloride in Salviae Miltiorrhiza and Ligustrazine Hydrochlo-ride Injection in rats.Methods Rats were injected 20 , 40 and 80 mg· kg-1 Salvia Miltiorrhiza and Ligustrazine Hydrochloride Injection by caudal vein, respectively.The urine and bile were collected at 0 -4, 4-8, 8-12, 12-24 h and 0-2, 2-4, 4-6, 6-12, 12-24 h, respectively.The concentrations of ligustrazine hydrochloride in urine and bile were measured by HPLC.Results After intravenous injection different doses of Salviae Miltiorrhiza and Ligustrazine Hydrochloride Injection in rats, few ligustrazine hydrochloride was excreted in urine and bile as prototype.Conclusion The results indicate that prototype was not the main elimination form of ligustrazine hydrochloride in rats.
6.The effect of calorie restriction on the expression of liver's gluconeogenesis genes of rats fed a high fat diet.

OBJECTIVETo observe the effect of calorie restriction on the high fat diet rats mRNA expressions of liver forkhead box O1(FoxO1), phosphoenolpyruvate carboxykinase (PEPCK), glucose-6-phosphatase (G-6-P) and to explore the possible mechanisms.
METHODS24 normal 6-week-old male Wistar rats were randomly divided into three groups: normal chow group (NC, n = 7), high fat diet group (HF, n = 9) and calorie restriction group (CR, n = 8). They were fed for 12 weeks. At the end of the experiment, the rats were sacrificed and their fasting blood glucose (FBG), insulin (INS), triglycerides (TG), total cholesterol (TC) were measured. Their visceral fat (VF) and body weight (BW) were also measured and VF/BW was calculated. Gene expression was investigated by using semi-quantitative RT-PCR methods. Liver histology was studied with HE stained slides.
RESULTSCompared with the NC group, HF group rats developed visceral obesity which was accompanied by higher FBG, plasma INS, TG, and TC. The levels of FoxO1, PEPCK, and G-6-P increased by 18.9%, 33.8%, and 24.6%, respectively (P less than 0.01). Liver steatosis was observed with microscopy. The BW, VF FBG, INS, TG and TC of the CR group rats were lower in comparison to those of the HF group. The levels of FoxO1, PEPCK and G-6-P were lower by 26.6%, 35.0%, 34.3% (P less than 0.01). Meanwhile, liver steatosis was also milder.
CONCLUSIONCalorie restriction can inhibit the expressions of FoxO1, PEPCK and G-6-P, strengthen insulin signal conduction, suppress gluconeogenesis and thus regulate glycometabolism.

7.Soft and hard tissue changes in Class II division 1 patients treated with Tip-Edge plus appliance.

OBJECTIVETo investigate the soft and hard tissue changes in Class II division 1 patients treated with Tip-Edge plus technique.
METHODSSixteen Class II division 1 patients (7 boys and 9 girls) with mandibular retrusion in permanent dentition were selected and treated with Tip-Edge plus appliance. Lateral cephalometric films were analyzed before and after treatment. The effects were evaluated with Holdaway soft tissues analysis and routine cephalometric analysis methods. The arithmetic mean and standard deviation were calculated for each variable. Paired t-test was performed.
RESULTSThe average treatment time was 16 months. Normal overjet and overbite were established with retroclination of upper incisors and proclination of lower incisors. U1-NA(°) and U1-NA (mm) decreaed by (15.40 ± 5.31)° and (4.16 ± 1.82) mm (P < 0.01). NLA showed an average increase of (-16.60 ± 5.29)° (P < 0.01). Remarkable soft tissue change was noted after the treatment.
CONCLUSIONSThe profile in Class II division 1 patients could be quickly and efficiently improved after treatment with Tip-Edge plus technique.

8.Expression and Antigenic Analysis of the Recombinant Epitope of Herpes Simplex Virus Type 2 Glycoprotein G

A fragment containing amino acid residues 561~578 of HSV-2 glycoprotein G(gG2) was obtained by PCR assembling technique,and doubly cloned into vector pET-KDO.The recombinant plasmid was transformed to BL21(DE3)plysS.Fusion protein,of molecular weight about 39kDa was highly expressed by induction of IPTG.Western blot result showed the fusion protein had good antigenicity.After putification and digestion,the purity reached 95%.The digested purified protein was analysed by ELISA and showed good sensitivity and specificity.The recombinant protein should be useful for type-specific serodiagnosis of HSV-2.
9.Clinical trial of voriconazole tablets combined with caspofungin injection in the treatment of hematological disease with invasive pulmonary aspergillosis

Objective To observe the clinical efficacy and safety of voriconazole tablets combined with caspofungin injection in the treatment of hematological disease with invasive pulmonary aspergillosis.Methods A total of 100 patients of hematologic disease with invasive pulmonary aspergillosis were randomly divided into control group and treatment group with 50 cases per group.Control group was given amphotericin B,the first dose was 5 mg,the maximum dose of 25 mg at a speed of 5 mg · d-1,and then the dose of 25 mg · d-1 for 10 days,qd,intravenous drip,which was treated for 15 d.Treatment group was given caspofungin,the first dose of 70 mg,adjusted to 50 mg,qd,intravenous drip + voriconazole,the first day of 6 mg · kg-1,the second day adjusted to 4 mg · kg-1,bid,orally,which was treated for 15-28 d (treatment to full effect).The clinical efficacy and adverse drug reactions were compared between two groups.Results After treatment,the main indexes in treatment and control groups were compared:the total effective rates were 88.00% (44/50 cases) and 68.00% (34/50 cases),the total effective rates with empirical treatment were 92.31% (24/26 cases) and 81.48 % (22/27 cases),the total effective rates with proactive treatment were 83.33% (20/24 cases) and 52.17% (12/23 cases),the differences were statistically significant (all P < 0.05).The adverse drug reactions in treatment group were based on nervous system damage and liver and kidney dysfunction,which in control group were based on nervous system damage,peripheral edema,liver and kidney dysfunction and visual abnormalities.The incidences of adverse drug reactions in treatment and control groups were 4.00% and 14.00% with significant difference (P < 0.05).Conclusion Voriconazole tablet combined with caspofungin injection has definitive clinical efficacy and safety in the treatment of hematological disease with invasive pulmonary aspergillosiss.
10.Application of succinct teaching and more practicing mode in standardized nursing skill training

Objective To explore effect of succinct teaching and more practicing mode in standardized nursing skill training.Methods 120 nurses were randomly selected and divided into experimental group and the control group with 60 nurses in each group.Control group was trained with traditional hands-on training mode.Succinct teaching and more practicing mode was applied to the experimental group.The effects were evaluated with the training mode effect questionnaire.Results Theoretical examination results for the experimental group and control group were (84.52 ± 4.92) and ( 82.83 ± 3.76).There was significant difference between two groups (t =-2.103,P < 0.05 ).For the practical examination results,significant difference was also observed between experimental group and control group [ ( 83.33 ± 3.94) vs ( 81.30 ± 3.29),respectively; t =- 3.067,P <0.05].95% of the nurses in the experimental group were very satisfied with the succinct teaching and more practicing mode.90% nurses were very satisfied with the training administration,training effect and attitude.Conclusions Application of succinct teaching and more practicing mode in standardized nursing skill training was more targeted and personalized.It could feedback quickly and correct the shortcomings in the operation.Interests and motivation could be stimulated and the quality of nursing skill training,could be improved.
