1.Perioperative management of abdoimal surgery in over aged patients

Objective To study the perioperative management of abdominal surgery in patients aged over 80 years. Methods The cliuical data of 25 patients aged over 80 years undergoing abdominal surgery were analysed retrospectively. Results Of the 25 patients, 8 subjected to biliary surgery, 13 gastiic cancer or cardiac cancer surgery, 1 esophageal cancer surgery, 3 intestinal surgery. 24 patients(96%) were cured; one died postoperalively.Conclusions In over aged patieats, they have more coexite diseases, and more postoperative complications, so extensive perioperative management, choice of selective operation, reducing of emergent operation, and giving albumin to help the recovery of tissues are essential.
2.Effect of Dexamethasone on tumor volume and apoptosis of transplanted HeLa cells in mice

Objective To investigate the effects of dexamethasone on the growth of cervical cancer induced by HeLa cells transplanted into BALB/c mice. Methods HeLa cells were inoculated subcutaneously into BALB/c mice. Dexamethasone (15 mg?kg-1?d-1) was injected intraperitoneally from the 6th or the 16th day after tumor inoculation for 10 days continuously. Diameter of tumors was measured every two or three days. Apoptotic bodies were observed by transmission electron microscopy and counted by TUNEL. Objective To investigate the effects of over physiological dose of dexamethasone on cervical carcinoma cell xenografted tumors in nude mouse.Methods HeLa cells were inoculated subcutaneously in BALB/c female mice aged 5-6 weeks.Over physiological dose of dexamethasone(62.5 mg?kg-1?d-1) was injected into the peritoneum from the 16th day after inoculation for 10 d.Diameters of tumor were measured every two or three days.The volumes and the inhibition rates of tumor were calculated.Apoptotic bodies were observed by transmission electron microscopy and counted by TUNEL.VEGF and TNF-? were detected by immunohistochemistry.Results In vivo,over physiological dose of dexamethasone can repress the growth of tumor and induce cell apoptosis.Compared with the control group,the tumor sizes were significantly smaller(P0.05).Conclusion Over physiological dose of dexamethasone can repress cervical tumor growth in vivo.This action may be related to apoptosis and VEGF,but have no relationship with TNF-?.
4.Functional MRI and cognition assessment in subcortical ischemic vascular disease

Objective To evaluate the imaging features of patients with vascular cognitive impairment (VCI) induced by Subcortical Ischemic Vascular Disease (SIVD), through magnetic resonance diffusion tensor imaging ( DTI ) and proton spectroscopy ( MRS ) technology. Methods A total of 52 patients with SIVD were enrolled. After analysis of scale score, 32 patients with cognitive impairment were assigned to VCI group and 20 patients with no cognitive impairment were assigned to control group. Both group received DTI and MRS examination. The mean values of apparent diffusion coefficient (ADC) and fractional anisotropy (FA) of the bilateral temporal, frontal, parietal and occipital white matter regions as well as in the bilateral centrum semiovale were calculated. The peak value of MRS of N-acetyl aspartate (NAA) , choline ( Cho), creatine (Cr) and phaseomannite ( mI ) were calculated. Results Compared with control group, FA decreased in the region of temporal, frontal, parietal as well as in the centrum semiovale , and ADC increased in VC[group ( P < 0.05 ) . In the frontal regions and centrum semiovale,the VCI patients had a significant FA decrease. The ADC value increased obviously in the temperal lobe.Spectrum analysis results showed, NAA/Cr was lower than control group in VCI group in the frontal lobe (1.43 ±0.08 vs 1.53 ± 0. 92), while mI/Cr was higher than control group in the temporal lobe(0. 51 ±0. 06 vs 0. 46 ± 0. 07 ) ( P < 0. 05 ). Conclusion FA in the temporal and centrum semiovale regions of VCI group and NAA in the temporal white matter regions decreased obviously. DTI and MRS could provide a reference value for early diagnosis and assessment of VCI.
5.Immunocytochemistry analysis of human neutrophil defensins

Our laboratory isolated three defensin molecules from the circulating neutrophils of healthydonors.The three molecule mixture was conjugated with BSA and immunized New Zealand rab-bit.ELISA showed that the rabbit antisera was of a very high titters,which were specific for hu-man defensins.Immunoperoxidase staining with this rabbit antisera indicated that normal humanneutrophils,acute myeloid leukemia cell lines KG1 and HL-60 were highly positive.In con-trast,normal mononuclear leukocytes,erythrocytes,erythroleukemia cell line K562 were all neg-ative,even rabbit and guinea pig neutrophils had no response to this rabbit anti-human de-fensins antisera.Our preliminary experiments suggested that the rabbit antisera was highly spe-cific for human defensins.Probably,defensin is a differentiation antigen of granunocyte lineage,and antibody against human defensins would be a candidate immunological reagent specific for a-cute myeloid leukemia.Further work for this should be done.
6.Genetic characterization of measles virus caused the measles outbreak in Xinjiang

Objective To analyze the genetic characterizations of wild type measles viruses caused the measles outbreak in Xinjiang. Methods Vero/Slam cell were used for measles viruses isolation from the specimens collected from measles outbreaks patients. Fragment of 676 nucleotide acids of the carboxylend of nucleoprotein gene were amplified using reverse transcription-polymerase chain reation (RT-PCR) method and then the PCR products were directly sequenced and analyzed. Phylogenetic tree was constructed based on 450 nucleotide acids of the N-terminus of nucleoprotein gene, and homological analysis was performed at nucleotide acid level. Results 11 measles viruses were sequenced and all belonged to H1a subgenotype.The nucleotide difference was 0-0. 2% between 11 Xinjiang isolates. And the nucleotide difference was 2.2%-2.4% between Xinjiang isolates and H1 genotype reference strain. Conclusion The Measles viruses causing the measles outbreak in Xinjiang were H1a subgenotype.

8.Repair of excessive width of fold line and conglutination of upper eyelid after double eyelid operation

Objective To investigate the methods of rectifying the excessive width of fold line and conglutination of the upper eyelid following blepharoplasty. Methods The width of double eyelid was redesigned, ranging from 6 mm to 8 mm. Then the scar of upper eyelid was excised about 1 mm to 2 mm, and the orbital septum fat flap, orbicularis oculi flap, infraorbicularis oculi fat flap and injection of lipochondria were utilized to fill the depression of upper eyelid. Results In our series, there were 20 eyelids of 16 cases receiving rectification. Orbital septum fat flap was used to correct 2 eye-lids, orbicularis oculi flap 4 eyelids, infraorbicularis oculi fat flap 10 eyelids and injection of lipochon-dria 4 cases. Among 12 eases of following-up, 10 of them were satisfied with the postoperative effects. The results were acceptable in other 2 cases. Conclusion It is a good approach to correct the excessive width of fold line and conglutination of the upper eyelid with local flaps, including the orbital septum fat flap, orbicularis oculi flap, infraorbicularis oculi fat flap and injection of lipochondria.
9.The association between C-reactive protein-757T/C gene polymorphism and carotid artery artherosclerosis in patients with ischemic cerebrovascular disease

Objective To explore the relationship of a polymorphism c.757T > C in C-reactive protein gene (CRP) and carotid artery atherosclerosis in patients with ischemic cerebrovascular disease (ICVD).Methods A case-control study investigated 276 ICVD patients and 125 controls matched for age and sex.Genotypes of-757T/C in CRP were analyzed in patients with ICVD using polymerase chain reaction-restriction fragment length polymorphism.The level of serum C-reactive protein was detected by using immune scatter turbidimetry and the intima-media thickness of common carotid artery and internal carotid artery were measured by color Doppler ultrasonography.The relationship between the polymorphism -757T/C in CRP and carotid atherosclerosis was assessed.Results The genotypes of the polymorphism -757T/Cin CRP in the ICVD group (207/276(75.0%),65/276(23.6%),4/276(1.4%)) was significantly different from the control group (75/125 (60.0%),46/125 (36.8%),4/125 (3.2%) ; x2 =9.531,P =0.008),the frequence of T allele in patients with ICVD (479/552(86.8%)) was increased significantly compared to the control group (196/250 (78.4%)),the frequence of C allele in the ICVD group (73/552(13.2%)) was decreased significantly in contrast to the control group (54/250 (21.6%) ;x2 =9.056,P =0.004).Among the patients with ICVD,the level of serum C-reactive protein in patients with-757TC/CC genotype was elevated significantly than that with-757TC/CC ((4.2± 1.6) mg/L vs (2.7 ± 1.9) mg/L; t =5.900,P < 0.01).Logistic regression analysis demonstrated that the-757C allele in CRP was related to the carotid artery atherosclerosis (OR =1.379,95% CI 0.852-0.864,P =0.023).Conclusions The polymorphism-757T/C in CRP maybe related to the risk of developing ICVD.The frequency of the-757C allele can increase the level of serum C-reactive protein.And this polymorphism may be associated with carotid artery atheroselerosis in patients with ICVD.
