1.Establishment of a guinea pig model of dermatophytosis caused by Trichophyton verrucosum

Objective To establish a guinea pig model of dermatophytosis caused by Trichophyton verrucosum.Methods Thirty-two guinea pigs were randomly and equally divided into four groups to remain untreated (group 1 ),receive depilation and skin abrasion in the back (group 2 and 4),or only depilation (group 3).The back was smeared with a suspension (100 μl) of Trichophyton verrucosum in group 1,2 and 3,and with that of Trichophyton mentagrophytes in group 4.Direct microscopy,fungal culture and histopathology were performed to validate the infection of guinea pigs by Trichophyton.Results Direct microscopy was persistently negative in group 1,turned to be positive from the 7th to 9th day in group 2,3,and 4.Colonies began to appear 7 days after the inoculation of scrapings onto Sabouraud's dextrose agar (SDA) in group 2 and 3,and were observed at all the inoculated sites 2 weeks after the inoculation,with a morphological similarity to Trichophyton verrucosum before inoculation into guinea pigs.Hematoxylin and eosin staining revealed strongly refractive hyphae and spores in stratum corneum epidermidis and infundibular regions of some hair follicles in group 2 and 3.Conclusion A guinea pig model of dermatophytosis caused by Trichophyton verrucosum is successfully constructed.

6.Preparation and performance of biologic antimicrobial materials for pelvic tissue repair

BACKGROUND:The biological extracellular matrix materials become the focus of pelvic floor repair materials research because of its excellent biocompatibility and mechanical compatibility. However, bacterial infection can damage the function of biological repair materials. <br> OBJECTIVE:To prepare the biologic antimicrobial materials for pelvic tissue function repair. <br> METHODS:Chitosan and tigecycline-loaded chitosan nanoparticles suspension was prepared by the electrostatic adsorption and self-aggregation of nanoparticles preparation techniques. Then the suspension was coated onto the surface of extracellular matrix materials. The morphological examination was performed by scanning electron microscope. The antibacterial property was detected by solid plate microbial culture method. <br> RESULTS AND CONCLUSION:Infrared spectra displayed that aromatic ring skeleton vibration peak of chitosan occurred on the biologic antimicrobial materials, and it was significantly widened at about 3 359 cm-1, indicating the composite coating was successful to modify the surface of extracellular matrix materials. As a very smal dose of tigecycline, there was no characteristic absorption peak on the infrared spectra. Scanning electron microscopy showed the porous structure of the material surface with some nanoparticles adhesion. The prepared materials had good antibacterial properties on Staphylococcus aureus and Escherichia coli, and the inhibition zone diameter was significantly increased with the increasing concentration of antibacterial agent. 7.Prevention and treatment of severe complications of thoracoscopy

Objective Study the prevention and early diagnosis of severe complications of thoracoscopy. Methods 400 patients,(18~81)years,ASA Ⅰ-Ⅲ,underwent thoracoscoy,including 320 pulmonary bubble resections and 80 thorax,mediastinum or lung tumor biopsy or resections.All patients were anesthetized with one-lung ventilation using a double-lumen tube and monitored continuously in circulatory and respiratory functions with a multi-runctional monitor. Results Severe intraoperative complications occurred in 8 patients,including 5 reexpansion pulmonary edema,1 complete occlusion of double-lumen tube due to massive pleural effusion,1 massive bleeding and 1 cardiac arrest due to bilateral pneumothorax.All complications were successfully treated and the patients were cured and discharged. Conclusions Thoracoscopy must be well prepared.Applying one lung anesthesia with double-lumen tube,careful observation during operation,early recognition and treatment of complications may contribute to assuring the safety of this procedure.
8.The clinical significance of dynamic changes of CD4~+ CD25~+ regulatory T cells in patients subject to allogeneic hematopoietic stem cell transplantation

Objective To investigate the clinical significance of dynamic changes of CD4~+ CD25~+ regulatory T cells(Treg)in patients subject to allogeneic hematopoietic stem cell transplanta- tion(alIo-HSCT).Methods Forty-five patients received allo-HSCT.The graft-vesus-host disease (GVHD)was prevented by cyclosporine A and short-term MTX regimen in 31 patients.Fourteen of all the patients received Zenapox(CD25MAb)at the day of transplantation and day 4 after transplan- tation.The levels of Treg in peripheral blood were detected by flow cytometry from 45 patients at 2nd,4th,8th and 12th week after allo-HSCT and the time of aGVHD development,respectively.Re- suits Anti-CD25 could suppress the peripheral blood levels of Treg significantly.The Treg levels were significantly higher in patients with grade 0-1 aGVHD than those with 2-4 aGVHD at 8th and 12th week after transplantation.Among patients with 2-4 aGVHD,Treg levels were significantly low- er after development of aGVHD than before.Conclusions Treg are important for the aGVHD preven- tion and can be a useful clinical surveillant index for the development of aGVHD.It can significantly decrease the levels of Treg in the peripheral blood with anti-CD25.
9.Comparison of laboratory detection between infectious mononucleosis and mucocutaneous lymphnode syndrome

Objective To analyze the laboratory detection findings in infectious mononucleosis(IM) and mucocutaneous lymphnode syndrome(MCLS)and find the main points of the differential diagnosis and direct the treatment. Methods Five hundreds and twenty-five children were in-patient department over these past 10 years .These patients were divided into 2 groups,IM group (225 patients) and MCLS group(280 patients). EB-virus antibody,Paul-Bunnell test,the circumference blood numeration ,classification of leucocytes,the numeration of atypical lymphocytes were detected and compared,as well as blood platelet of these patients.Results The positive rate of EB-virus antibody in IM and MCLS were high, and had no discrepancy. However, the positive rate of Paul-Bunnell test and numeration of atypical lymphocyte in IM group were significantly higher than those in MCLS group. The total circumference blood leucocyte numeration of the groups increased,but the numeration neutrophil increased significantly in MCLS group, the rising numeration of lymphocyte in IM; the blood platelet numeration was elevated significantly in MCLS group and continued when the total circumference blood leucocyte numeration tended normally.Conclusions The causes of IM and MCLS are related to the infection of EB-virus. The laboratory characteristics of IM and MCLS are different,understanding the main points of the differential diagnosis contributes to the clear diagnose and early treatment.
