1.Glucagon-Like PeptideⅠand Targeted Differentiation of Stem Cells

Diabetes mellitus has become one of the diseases which threaten the heath of human being in the 21st century.A goal of research in diabetes is to find a way to increase the number of functional insulin-producing cells. Islet transplantation has been considered to be the most effective approach to cure type Ⅰ and part of type Ⅱ diabetes mellitus.This approach, however, is severely limited by an inadequate supply of donor islets available for transplantation.Moreover, recent progress of stem cells research has shown that stem cells may act as a new source of islet transplantation in diabetes mellitus treatment. Recent evidence indicates that Glucagon-Like PeptideⅠ(GLP-1) plays a very important role in targeted differentiation of stem cells into Insulin-Producing Cells and pancreatic development. GLP-1 is an intestine-derived insulinotropic hormone that stimulates glucose dependent insulin production and secretion. GLP-1 can induce differentiation of stem cells into insulin-producing cells, which is achieved by up regulation of PDX-1 expression.PDX-1 is a transcription factor critical for pancreatic development and endocrine cell neogenesis and a marker for pancreatic stem cells. These new findings suggest an approach to create Insulin-Producing cells in vitro by expanding stem/progenitor cells and then to convert them into Insulin-Producing cells by treatment with GLP-1. Thus GLP-1 may be a means by which to create Insulin-Producing cells ex vivo for transplantation into patients with insulinopenic type Ⅰ diabetes and severe forms of type Ⅱ diabetes. This article reviews recent progress about GLP-1 and targeted differentiation of stem cells induced by GLP-1.

3. Protective effect of methylamine irisolidone on cardiac function in rats with acute myocardial infarction

OBJECTIVE: To observe the effect of a new compound methylamine irisolidone on cardiac function in rats with experiment myocardial infarction. METHODS: The male Wistar rats were randomly divided into 6 groups: control group, ischemia model group, methylamine irisolidone preconditioning group, pretreated with different dosages of methylamine irisolidone (200,100,50 mg · kg-1, respectively) and 200 mg · kg-1 puerain(positive control). Coronary artery ligation was used to induce the acute myocardial ischemia model. After 30 d administration of methylamine irisolidone, the heart function parameters,including heart rate (HR), systolic blood pressure (SBP), diastolic blood pressure (DBP), left ventricular systolic pressure (LVSP), maximal velocity of increase and decrease of left ventriclar pressure (dp/dtmax), were analyzed and myocardial histology was analyzed by hematoxylin-eosin staining. RESULTS: Compared with model group, methylamine irisolidone could improve the heart function of rats in AMI injury, increased the systolic pressure and diastolic pressure, decreased the heart rate(P < 0.01). The myocardial ultrastructure injury was alleviate. CONCLUSION: Methylamine irisolidone had cardioprotective effects on acute myocardial ischemia. The mechanism may be related to slow down heart rate, prolong diastolic, so that an increase in the endocardial blood supply.

5.The effect of behavioral training on neural stem cell differentiation in the dentate gyrus of rats with hippocampal infarction

Objective To explore the effect of behavioral training on the differentiation of neural stem cells in the dental gyrus (DG) in rats with hippocampus infarction. Methods Seventy-eight Sprague-Dawley rats were randomized into infarction plus behavior training group, infarction group and control group. Photochemistry method was used to induce hippocampal infarction in rats of the infarction plus behavioral training group and infarc-tion group. At 1 day after surgery, Morris water maze training was used for infarction plus behavioral training group, free-movement without training was performed for infarction group. Double staining immunofluorescence was used to detect the co-expression of bromodeoxyuridine (BrdU) with neuronal nuclei ( NeuN ) or glia fibrillary acidic protein (GFAP) in the DG at different time points. Results Few BrdU/NeuN and BrdU/GFAP double staining cells were observed in the DG of control rats. In the infarction group and infarction plus behavioral training group the number of BrdU/NeuN and BrdU/GFAP double-stained cells increased in the DG on the opposite side compared with the control group on 14th, 21st, 28th and 35th days after surgery (P < 0.05 ). There observed significantly more BrdU/NeuN and BrdU/GFAP double-stained cells in the infarction plus behavioral training group than that in the infarction group on the 14th, 21st, 28th and 35th days after surgery ( P < 0.05 ). Conclusion Behavioral training can accelerate the differentiation of neural stem cells to neuron and astrocyte, by which to promote the re-covery of neural functions.
6.Expression and significance of CD24 in colorectal cancer

Objective To determine the expression of CD24 in colorectal carcinoma, and to explore the relationship between CD24 and the clinicopathological features of colorectal carcinoma.Methods The expression of CD24 in 62 cases of colorectal carcinoma, 47 cases of adenomas, 15 cases of colorectal polyps and 30 cases of the adjacent non-cancerous tissues were observed by immunohistochemical assay.The relationship between CD24 and the clinicopathological features was analyzed.Results The positive rates of CD24 in colorectal carcinoma 72.6% and adenomas 63.8% were significantly higher than those in colorectal hyperplastic polyps 13.3% (x2 =17.83, P =0.00;x2 =11.61, P =0.00) and adjacent non-cancerous tissues 6.7% (x2 =35.15, P =0.00;x2 =24.64, P =0.00).The expression of CD24 in colorectal carcinoma had a significant correlation with the tumor diameter (x2 =5.48, P =0.02), tumor differentiation (x2 =8.86, P =0.00), Duke staging (x2 =11.47, P =0.00) and lymph node metastasis (x2 =8.92, P =0.00).Conclusion The expression of CD24 is high in colorectal carcinoma, having a significant correlation with the size of tumor, degree of differentiation, Duke stage and lymph node metastasis.
7.Influence of Weight - Lost on Glucose Intolerance Test and Insulin Secretion in Obese Children

Objective To observe the influence of weight - lost therapy on obese children with impaired glucose tolerance( IGT), insulin resistance. Methods Fasting,post- prandail 2 hours blood glucose and insulin were measured in 14 obese children (age 8 - 15 years) with IGT before and after two - month weight - lost iherapy. Glucose were measured with enzymeoxidize assay, and insulin were measured with radio-immunity assay(RIA). Results Among 14 obese children with IGT, after two-month weight - lost the-rapy, there were 9 children becoming normal OGTT. There were significantly lower in the concentration of fasting insulin and post - prandail 2 hours blood insulin and glucose[(14.23?2.35) mIU/L,(47.20?10.26) mIU/L,(5.36?0.91) mmol/L] than before weight -lost therapy[ (32.54?7. 13) mIU/L,( 164.53?33.60) mIU/L, (8.75?1.09) mmol/L](P
8.Effects of low molecular heparin on cytokines in invasive pulmonary aspergillosis mice

Objective To investigate the effects of low molecular heparin(LWMH)on cytokines(TNF-?,IL-1? and IL-10)in blood plasma and bronchoalveolar lavage fluid of invasive pulmonary aspergillosis(IPA)mice.Methods The neutropenic IPA mouse models were established by administration of cyclophosphamide for immunologic function inhibition and intranasally challenge with Aspergillus fumigatus conidia(1?106 conidia/mouse).One hundred and twenty mice were randomly divided into 4 groups:normal control,IPA model,normal saline+LWMH and IPA+LWMH group.Normal saline+LWMH group and IPA+LWMH group received LWMH(subcutaneous injection,1 000 IU/kg,qd?2 d).Normal control and IPA model group received normal saline instedad of LWMH.At 4,8,12,24 and 48 h after inoculation,six mice were randomly taken from each group to be sacrificed.ELISA method was used to determine the concentrations of TNF-?,IL-1? and IL-10 in blood plasma and BALF.Results TNF-?,IL-1? and IL-10 in blood plasma and BALF increased significantly several hours after inoculation of conidia in IPA model and IPA+LWMH group.There were significant higher concentrations of TNF-? and IL-1? in blood plasma and BALF in IPA+LWMH group than in IPA model group(P
9.Investigation and Analysis of Absent Items in Package Inserts of Common Drugs for Elderly Patients in Our Hospital

OBJECTIVE:To provide reference for the standardization of package insert of common drugs for elderly patients. METHODS:Outpatient prescriptions in our hospital from 2013 to 2014 were collected,and common drugs of elderly patients(≥60 years old)were ordered by consumption sum,and package inserts of top 100 drugs were statistically analyzed for absent items. RESULTS:The 100 common drugs for elderly patients included 28 Chinese traditional drugs and 72 chemical drugs. The Chinese traditional drug had more absent items,mostly were pharmacokinetics and drug overdose(92.9%),medication of elderly patients (85.7%),drug interactions(78.6%)and pharmacological actions(50.0%);the proportion of contraindications(16/28),adverse re-actions(15/28) and precaution(9/28) with uncertainty was relatively high. Items of chemicals package insert were better than Chi-nese traditional drug,absent items were pharmacokinetics and drug overdose(5.6%),medication of elderly patients and pharmaco-logical actions (2.8%) and drug interactions (1.4%);the proportion of medication of elderly patients (16/72),drug interaction and drug overdose(12/72)with uncertainty was relatively high. CONCLUSIONS:The common problem existing in the drug manu-al is characterized by absence of usage for elderly,drug descriptions is not clear,etc. Package inserts should be further improved to strengthen its standardization.
10.Two cases of Avian Influenza A(HSNI) in Anhui province of China

Objective To analyze the clinical features,epidemiologic information and outcome in confirmed cases of human infection with a highly pathogenic influenza A(H5N1).Methods The clinical features and epidemiologic findings in 2 confirmed cases of avian influenza A(H5N1)in Anhui province,in November 2005.Clinical data on vital signs,physical findings,laboratory tests and roentgenology were obtained by means of retrospective review of the hospital records.Epidemiologic data were collected through interviews of the patients and their relatives.Results In both cases,the diagnosis of influenza A(H5N1)was confirmed by means of viral culture and real-time polymerase chain reaction with specific primers for H5 and N1 in samples obtained from tracheal aspiration.All patients were previously healthy young women and resided in village.They had a clear history of di- rect contact with sick/dead poultry and prepared dead chickens at home for eating(removed feathers, washed,cut)hut no report of confirmed HSN1 animals in the village.The time between exposure and onset of illness were 5 days,no one else in family sick.The time between the onset of illness and hos- pitalization were 5 days and 6 days,respectively.Two patients have initial symptoms of high fever (typically a temperature more than 38℃).The prominent clinical features were those of influenza syndrome,including fever,cough,and shortness of breath.Upper respiratory tract symptoms were absent.The platelet counts were decreased.In both patients,there were marked abnormalities on chest radiography,radiographic changes include bilateral and unilateral lobular consolidations with air bronchograms,and had dramatic worsening of findings.Two patients developed acute respiratory dis- tress syndrome(ARDS)complicated multiple organ dysfunction syndrome(MODS)and died of pro- gressive respiratory failure.Conclusions Exposure to dead poultry within a week before the onset of illness was associated with Influenza A(H5N1)infection in humans,but no definitive evidence of hu man-to-human transmission has been found yet.Influenza A(HSNI)infection,characterized by fe- ver,an influenza-like illness with lower respiratory tract symptoms,carries a high risk of death.
