1.Screening,identification,enzymatic characteristics and optimization of fermentation condition of a high alkaline xylanase-producing strain

ObjectiveTo screen a high alkaline xylanase-producing strain,subject to molecular identification and characterization of enzymatic property,and optimize its fermentation condition.MethodsThe farmland soil samples from Shanxi,Henan and Zhejiang Provinces were collected aseptically,from which the high xylanase-producing strains were screened by enrichment culture,isolation and identification of 16S r DNA,and determined for enzymatic properties.The carbon source(xylan,lactose,soluble starch,sucrose,bran and glucose),nitrogen source[ammonium oxalate,peptone,yeast powder,(NH_4)_2SO_4,NH_4Cl,Na NO_3,urea,beef extract,bean powder,KNO_3,(NH_4)_2HPO_4or random mixture of two of peptone,yeast powder,beef extract,(NH_4)_2HPO_4and bean powder],metal ion[CuCl_2,MgCl_2,ZnCl_2,Al_2(SO_4)_3,CoCl_2,MnCl_2,AgNO_3,NaCl,CaCl_2,FeCl_2,BaCl_2 and FeCl_3],pH value(3.0~10.0)in medium and the fermentation temperature(25,28,30,33,35,37 and 40℃)were optimized by single factor test,while the contents of components[xylan,(NH_4)_2HPO_4and bean powder]by response surface method.The high alkaline xylanase-producing strain was fermented by using the optimized medium under the optimized condition,and determined for xylanase activity,and the result was compared with that predicated in model.ResultsA high alkaline xylanase-producing strain named as SX6-18 was screened and identified as Paemibacillus based on 16s rDNA sequence analysis.The xylanase produced by the strain maintained more than 70%of relative activity at temperatures of 25~55℃and pH 6.0~10.0.Mg(2+)promoted while Fe(2+)promoted while Fe(3+),Mn(3+),Mn(2+)and Cu(2+)and Cu(2+)inhibited the activity of xylanase.The optimal carbon source,nitrogen source and metal ion of the medium were xylan,bean powder+(NH_4)_2HPO_4 and Fe(2+)inhibited the activity of xylanase.The optimal carbon source,nitrogen source and metal ion of the medium were xylan,bean powder+(NH_4)_2HPO_4 and Fe(3+),while the optimal p H value and temperature for fermentation were 8.0 and 30℃,respectively.However,the optimal component contents were 15.00 g/L xylan,3.03 g/L(NH_4)_2HPO_4and 3.28 g/L bean powder.The activity of xylanase cultured in optimized medium under opti-mized condition for fermentation reached 701.08 U/m L,which was closed to the expected value(693.96 U/m L).ConclusionA high alkaline xylanase-producing strain SX6-18 was successfully screened,which maintained relatively high enzyme activity in alkaline condition.This study laid a foundation of application of xylanase in papermaking and washing fields.
2.The study of tumor necrosis factor-alpla single nucleotide polymorphism in Chinese Han patients with polymyositis/dermatomyositis

Objective To investigate the tumor necrosis factor alpha gene single nucleotide polymorphism (SNP) in Chinese Han polymyositis/dennatomyositis (PM/DM) patients. Methods A casecontrol study of three TNF-α SNPs were undertaken and comparison between cases of PM/DM (n=69) and DM patients ( n=52 ) with normal subjects ( n=57 ) was performed. The genotype of every subject was determined by polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP). Results There was no significant difference in the genotypes and alleles frequencies of the three TNF-α SNPs(-238,-308, and -1031)between PM/DM or DM and normal subjects. The haplotype AGT (-238, -308, -1031 ) significantly increased in controls vs PM/DM and DM (PM/DM, P=0.01; DM, P=0.02) subjects. The haplotype AGC was a significant risk factor for DM (P=0.04, 0R=9.84, 95%CI 1.39~69.57 ). The haplotype AGC increased in PM/DM (4.3%)vs controls(0.6% ), but the difference was not significant (P=0.09, OR 7.22, 95%CI 1.02~50.90). The haplotype GGC was significantly decreased in the DM subgroup vs healthy control subjects (P=0.04, OR=0.47,95%CI 0.23~0.99). Conclusion The TNF gene (-238,-308,-1031) haplotype AGC is a risk factor for DM.
3.Research on effects of radiation on hippocampus neuron and the related molecular mechanism

Objective:To explore the effects of radiation on hippocampus neuron and the related damage mechanism in order to provide the theory base for treatment strategy of radiation brain injury. Methods:We looked up the scientific literature including following methods. Pregnant mice were irradiated with tritiated water, and then the contents of SOM in pars intermedia and AVP in hypothalamus of newborn mice were detected by radioimmunoassay. Pregnant rats were irradiated with tritiated water or X-ray, and the neuronal cells derived from the newborn rats were cultured. The morphology of cultured neuronal cells was compared with those of normal neuronal cells. The number of apoptotic cells in cortical fifth layer was measured by HE staining. Results:After radiation with tritiated water, the level of SOM in pars intermedia of newborn mouse and AVP content in hypothalamus content of male newborn mouse significantly increased and decreased, respectively. Radiation with tritiated water could result in the morphology change of neuronal cells and increase the percent of apoptosis cells in cortical fifth layer. Moreover, the percent of apoptosis cells in tritiated water group was higher than that in the X ray group. Conclusion: Radiation could impact on brain neuropeptide contents, and neurobehavioral, learning and memonic ability, result in the morphology change of hippocampal neurons, and induce the midbrain cells apoptosis.
4.Analysis of the health management results of 149 cases of chronic metabolic diseases

Objective To discuss an effective and practicable mode of health management service by preventing the dangerous factors from causing diseases for 149 sufferer of chronic metabolic diseases and analyzing the results of such prevention.Methods Set standard examining items for collecting health information,which will be input the health information managing system and then be quantitatively assessed.The number of those who have isolated systolic hypertension and chronic metabolic disturbance disease,including hyperlipemia,hyperglycemia,hyperuricemia and fatty liver is 149.These people will be divided into 5 groups and preventions will be conducted against those dangerous factors which could lead to diseases.The results of such prevention will be assessed later.Results Significant changes have been shown in the observation indexes including blood pressure,total cholesterol(CT),triglycerides(TG),low density lipoprotein cholesterol(HDL-ch),fasting blood glucose(FBG),uric acid(UA),alanine aminotransferase(ALT)and ultrasound scan after the abovementioned preventions.Conclusion The mode of health management service mentioned in this article iS effective and practicable.
5.Epidemiological characteristics of rotavirus infection in infants with diarrhea in Nanjing

Objective To investigate the epidemiologic features of infant rotavirus (RV) diarrhea in Nanjing area .Methods The detection of group A rotavirus were performed on 7 325 outpatients from intestinal clinic of Nanjing Children′s Hospital by using immune colloidal gold method ,and the epidemiological analysis were did according to the detection results .Results In the 7 325 pa-tients ,2 353 were found rotavirus positive ,the total positive rate was 32 .12% .The positive rate of 1 -2 years old group was the highest(43 .51% ) ,compared with other age groups (P< 0 .05) .In November ,December and January ,the positive rates were 52 .20% ,46 .98% ,46 .59% ,respectively .Compared with the positive rate in other months ,there was statistically significant differ-ence(P<0 .05) .The infection rate of RV in vaccine group and non-vaccine group were 9 .06% and 38 .74% ,respectively .There was significant difference between the two groups .Conclusion Infants of 6 months to less than 3 years old in Nanjing have higher de-tection rate of RV infection ,and the infection peak appears in autumn and winter .
6.Observations on the Efficacy of Acupuncture plus Tuina Therapy in Treating Supraspinatus Tendinitis

Objective:To investigate the clinical efficacy ofacupuncture plus Tuina therapy in treating supraspinatus tendinitis.Methods:One hundred patients with supraspinatus tendinitis were randomly allocated to two groups.The treatment group of 50 cases was treated with acupuncture plus Tuina therapy and the control group of 50 cases,with simple acupuncture.Treatment was given once daily,10 times as a course.The therapeutic effects were evaluated after two courses oftreatment.Results:The total efficacy rate was 96.0%in the treatment group and 74.0%in the control group.x2 test showed that there was a statistically significant difference in the total efficacy rate between the two groups(P<0.01).Conclusion:Acupuncture plus Tuina therapy is superior to simple acupuncture in treating supraspinatus tendinitis.
7.Correlation between colorectal cancer and Helicobacter pylori infection

Objective To explore the correlation between Helicobacter pylori (H.pylori) infection and colorectal cancer.Methods From January 2008 to January 2014,263 patients pathologically diagnosed as colorectal cancer were selected as colorectal cancer group,and at the same period 263 patients with normal colon mucosa under colonoscopy were set as control group.The rates of H.pylori infection,the rates of H.pylori infection in patients with different pathological types of colorectal cancer,the rates of H.pylori infection accompanied with atrophic gastritis and the rates of H.pylori infection accompanied with intestinal metaplasia of two groups were retrospectively analyzed.The difference of H.pylori infection between proximal colorectal cancer and distal colorectal cancer was compared.Chi-square test was performed for comparison and odds ratio (OR) value and 95% confidence interval (CI) were calculated.Results Among 263 patients with colorectal cancer,the H.pylori infection rate of colorectal cancer group was 63.50% (167/263),which was higher than that of control group (39.54%,104/263),and the difference was statistically significant (x2 =27.66,OR=2.66,95% CI 1.85 to 3.83,P<0.01).There was no statistical difference in rates of H.pylori infection among different pathological types of colorectal cancer (x2=0.15,P=0.93).The rate of H.pylori infection accompanied with atrophic gastritis in patients with colorectal cancer was 46.39% (122/263) which was higher than that of control group (23.57%,62/263),and the difference was statistically significant (x2=28.53,OR=2.94,95%CI 1.98 to 4.36,P<0.01).The rates of H.pylori infection accompanied with intestinal metaplasia in patients with colorectal cancer was 17.87% (47/263) which was higher than that of control group (4.18%,11/263),and the difference was statistically significant (x2=23.56,OR=5.50,95%CI 2.76 to 10.95,P<0.01).There was no significant difference in the rate of H.pylori infection between the proximal colorectal cancer and the distal colorectal cancer (x2 =1.48,P =0.22).Conclusion H.pylori infection,together with atrophic gastritis and intestinal metaplasia caused by it are correlated with colorectal cancer.
8.The status, problem and progress of diabetic retinopathy treatment

Laser photocoagulation,intravitreal injection of antibody against vascular endothelial growth factor (VEGF) or corticosteroids and pars plana vitrectomy are current popular therapeutic approaches for diabetic retinopathy (DR).However,some DR patients still progress to irreversible blindness even after the above treatments which do not aim at the pathological mechanisms and influence factors for DR.Thus,with the further elucidation on the molecular pathological mechanisms and overall understanding of the factors affecting DR development,more and more potential therapeutic interventions such as neuron protection,vascular reconstruction and protection,gene therapy,non-VEGF dependent antineovascularization agents have been explored.Individual precise therapy based on the potential therapeutic targets would provide the promising future for DR patients.
9.Technology and Application of Dual-source CT Urography

Objective To evaluate the technology and application of dual-source CT urography. Methods 56 cases with clinical symptoms and dual-source CT urography were analyzed retrospectively through conventional plain scan, arterial phase scan, venous phase scan, which gained data were reconstructed 3D images of MPR, MIP, CPR and VR, and the im- ages were analyzed. Results The 3D images of kidneys, ureters and bladder were reconstructed successfully, which showed clearly the detail of the entire urinary tract and the local fine structure, morphology, taking shape and the relationship with the surrounding tissue through rotating and zooming. Conclusion Through the dual-source CT to scan the entire urinary tract is suitable for a variety of data acquisition of three-dimensional reconstruction, which can greatly enhance the focal positive rate and diagnosis rate.
10.Effects of endodontic instrument taper on post retention of central or lateral maxillary incisors in vitro

Objective:To evaluate the effects of various tapers of canal preparations on the retention of cast posts in endodontically treated upper incisors in vitro.Methods: Forty human central or lateral maxillary incisors were prepared by removing the clinical crown at 1 mm above the approximal CEJ and treated the canals with instruments of varying tapers. The groups comprising 10 teeth of each group were divided as follows: Taper 0.02 /15~40#SS (stainless steel K-files) with step-back technique as control, PF (ProFile)OS 3#/0.06 with taper 0.06/PF 25#/ in apical third, PF OS 4#/0.07 with taper PF 0.06/30# in apical third, PF OS 5#/0.08 with PF taper 0.06/35# in apical third. Then, canals were filled with gutta percha and AH Plus sealer using vertical condensation technique with a level of 5 mm of gutta-percha left in the apical segment of the root. Metal cast posts were made by direct method and then the post was cemented with HY-BOND GLASIONMER CX (Shofu,Inc)cement. The roots were fixed in copper rings with mounting plaster and mounted on a WD-5A machine.The posts were subjected to gradually increasing vertical tensile force until dislodgment of the post occurred. Force (N) required to dislodge the post was obtained for each tooth, and the average force of the groups was compared using a one-way ANOVA and SNK multiple comparisons with SAS V8.1. Results:The highest resistance to dislodgment was obtained with PF OS 4#/0.07, whereas the lowest was obtained by the PF OS 3#/0.06. Nonstatistical difference was found between Taper 0.02 /15~40# and PF OS 3#/0.06. PF OS 4#/0.07 was statistically superior to Taper 0.02 /15~40# and PF OS 3#/0.06 but not PF OS 5#/0.08. Conclusion: The retention of posts is affected strongly with the increase in the diameter than with the increase in the taper angle. PF OS 4#/0.07 with taper PF 0.06/30# in apical third seems to be the best taper for maximal resistance to post dislodgment.
