1.Significance of CD117 and CD34 expression in gastrointestinal stromal tumors

AIM: In this article, gastrointestinal tumor-related historical archives of pathology in our hospital were reviewed in order to acknowledge gastrointestinal stromal tumors (GIST). METHODS: These cases were rediagnosed correctly , these tissues were labeled with some antibodies such as CD117, CD34, ?-SMA, S-100, and their clinical characteristics were explored. RESULTS: CD117, CD34 were expressed widely in GISTs ,the percentage of positive expression was CD117 (50/56, 89.3%), CD34 (37/56, 66.1%), respectively, ?-SMA, S-100 were (17/56, 30.4%), (4/56, 7.1%), respectively, desmin was negative in these cases. Among them, 29 cases were benign and borderline, 27 cases were malignant. The percentage of occurring in stomach and intestine was 91.1%. CONCLUSION: CD117, CD34 are expressed significantly in GIST, these might be assistant markers for GIST diagnosis. GISTs mostly occurred in stomach and intestine.
2.Study on patient safety status and influencing factors of knowledge, attitude, behavior and practice

Objective To investigate cognitive status of nursing students for patient safety and analyze its influencing factors,and to supply reference for the patient safety education of nursing students.Methods Using convenience sampling method,617 nursing students in Tianjin and Fuzhou were investigated by self-designed the nursing students for patient safety KABP questionnaire.The results underwent analysis.Results The patient safety KABP total score was 3.34±0.44,all dimensions from high to low were attitude 3.82±0.51,behaviour and practice 3.42±0.60,knowledge 2.92±0.67.Undergraduate nursing students overall cognition was lower than those of college nursing students,t=5.194,P<0.01,the overall cognitive situation of students who had entered the graduation practice was higher than nursing students who had not entered the graduation practice,t=3.264,P<0.01,however attitude was negative,t=-3.813,P<0.01.Regression analysis showed that factors influencing cognition of nursing students for patient safety were mainly education degree and whether entering graduation practice or not.Conclusions The cognition of patient safety in nursing students was in the medium level,mainly affected by the education degree and whether entering graduation practice or not.
3.Research of siRNA Inhibition Effect in HIV-1 Gene Expression

RNA interfering (RNAi) is a process of sequence-specific, posttranscriptional gene silencing in animals and plants. In mammalian cells, duplexes of 19~25 nts (nucleotides) RNAs efficiently inhibit gene expression. The pBS/H1SP vector expressing siRNA is used which inhibit specific HIV-1 gene expression. To assess the intracellular effect of this H1 promoter-driven siRNA, a reporter plasmid pEGFP-C1-HIV protein which expresses fusion protein of enhanced green fluorescent protein (EGFP) and HIV protein was used. The expression of the reporters can be easily visualized by fluorescence microscopy in living cells. siRNA-generating vectors targeted to several HIV-1 genomes were constructed and then co-transfected with respond reporter expression vectors into HEK293 cells. Cells transfected with pHIV-siRNA exhibited a significant inhibition of pEGFP-HIV expression compared with cells transfected with control vectors. By this way, it is successfully to select effective siRNA for silencing target HIV-1 genes. Then two or three siRNA transcripts targeted to different HIV genes were expressed by one plasmid, and a relative strong inhibition effect was observed.
4.Design of method comparison and bias estimation of multi tests on multi instruments

Objective To design an experiment for method-comparison and bias estimation of multi tests on multi instruments.Methods According to the procedure described by the NCCLS approved guideline EP9-A and improving the method of sample collection,we took 8 patient mixing samples each day to analyze all comparison tests on 11 auto-chemistry analyzer within following 5 days.The duplicates were assessed within the same run.The coefficient of correlation and average bias% were calculated,and the system errors at medical decided levels were assessed.Results Taking ALT as an example,the coefficients of correlation were between 0.994-1.000,and the average bias% were between-0.460%-4.927%,SE at 40U/L was-1.510-1.834 and SE at 300 U/L was-3.101-9.188.Conclusion In all tests that joined the comparison among the different instruments,28 tests were acceptable,2 tests were acceptable after modifying the coefficients,and AMY and LIP were not acceptable.
5.Effect of electroacupuncturing Shuigou and Baihui on activation of microglia in brain of ischemia-reperfusion rats

BACKGROUND: Microglia is the immune effect cell in central nervous system, if it is activated, it will release neuron poisoning factors and inflammatory factors, and bring fatal injury.OBJECTIVE: To investigated the effect of focal cerebral reperfusion on activation of microglia and the effect of the regulation of electroacupuncturing Shuigou and Baihui.DESIGN: A randomized and controlled study.SETTING: The department of Physioltherapy, the 458 Hospital of Chinese PLA; Dongfang Hospital Affiliated to Shanghai Tongji University; The department of Histology-embryology, Tongji Medical College of Huazhong University of Science and Technology.MATERIALS: This experiment was processed at Histology-embryology Center of Tongji Medical College of Huazhong University of Science and Technology in March 2002. Total 80 Wistar rats were randomly divided into 8 groups with 10 in each group.METHODS: ① Normal control group: The rats were not given any treatment and killed next day. ② Sham operation group: Only artery was separated, no suture inserted, killed next day. ③ 6, 12 and 24-hour reperfusion groups: The rats' right middle cerebral artery (MCA) ischemia-reperfusion model was made by embolus suture technique, then the rat was killed after 30 minutes' occlusion at MCA and reperfusing respectively 6, 12, 24hours. ④ 6-hour reperfusion + electroacupuncture group: Electroacupuncture was carried out immediately after the model was made, then the rat was killedafter 30 minutes' occlusion at MCA and reperfusion 6 hours (The frequency was 4 Hz-16 Hz. Stimulus intensity increased 1 V per 10minutes and the final intensity was 3V. The stimulus was lasted 30 minutes). ⑤ 12-hour reperfusion + electroacupuncture B group: Electroacupuncture was carried out immediately after the model was made and 8 hours later, then the rat was killed after 30 minutes' occlusion at MCA and reperfusion 12 hours. ⑥ 24-hour reperfusion + electroacupuncture group: Electroacupuncture was carried out immediately after the model was made and 8, 16 hours later, then the rat was killed after 30 minutes' occlusion at MCA and reperfusion 24 hours.MAIN OUTCOME MEASURES: To calculate numbers of microglia and to observe its morphological changes.RESULTS: Data of totally 67 rats was entered the results analysis. No microglia was seen in the normal and sham operation groups. Large quantity of microglia was activated at the border of ischemic area in 6, 12 and 24reperfusion group, their quantity was largely increased, reaching apex at reperfusion 12 hours [(35.38±1.77), (54.25±1.67), (49.29±2.21)/200sights]. The quantity in every 6, 12 and 24 reperfusion + electroacupuncture group was less than that in model group [(32.11±2.80), (50.88±2.64),(45.45±3.95)/200 sights, P < 0.05].CONCLUSION: The microglia in brain is activated after the cerebral ischemia-reperfusion and induces toxic effect on neurons. The electroacupuncture can decrease activation of microglia so as to protect the neurons.

Objective To investigate the protective effect of Gingko Biloba extract (GBE) on acute lung hemorrhage induced by Lipopolysaccharide(LPS) in newborn rats. Methods 1. Acute lung hemorrhage models were reproduced by intraperitoneal injection with LPS (5 mg/kg). 2. Thirty two rats were randomly divided into 4 groups,GBE groups (4 mg/kg,8 mg/kg, 16 mg/kg) and LPS group 5 mg/kg. Results In group LPS, extensive lung hemorrhage was observed after 4 hours of injection . TNF - ? iung content was obvious in LPS group. The expression of lung nuclear factor(NF-kB )immunohistochemistry wasobvious. While the parameters were obviously attenuated by GBE before LPS. Conclusion GBE may be useful in the treatment of acute pulmonary inflammatory disease.

It is confirmed that chronic atrophic gastritis (CAG) caused by Helicobacter pylori is the main cause of gastric precancerous lesions.CAG is also the key determinant in gastric cancer risk assessment,which affects pepsinogen and gastrin-17 secretion.Most of the gastric cancer patients have poor prognosis,and non-invasive tools for gastric cancer screening and diagnosis are lacking.Therefore,the early detection of gastric cancer in order to reduce the disease mortality is necessary.Pepsinogen and gastrin-17 are biomarkers of gastric mucosa and gastric antra.The serological testing for the stomach-specific biomarkers offers the possibility to know preneoplastic gastric mucosal conditions.
