2.Setting Acute and Chronic Wards in Rehabilitation Medicine Department of General Hospital

Objective To investigate the significance to separate acute and chronic wards in a rehabilitation medicine department of generalhospital. Methods The patients discharged from acute wards from August 2012 to April 2013, when acute and chronic wards has beenseparated, were as group A (n=289), those from chronic wards were as group B (n=108). The patients discharged a year before the separation were as the control (group C, n=402). Their activities of daily living (ADL) when discharged, satisfaction and the time in the acute wards were observed. Results There was no significant difference in ADL between group A and group C (P>0.05), but it was between group B and group C (P<0.05). The patients more satisfied after separation (P<0.05), while the time in the acute wards decreased. Conclusion Separating the acute and chronic wards may improve the use of the medical resources and serve more patients. However, it improve the quality of care and satisfaction of the patients.
4.MicroRNA profiling on oxidative stress induced apoptosis of human lens epithelium

AIM: To identify the changes in microRNA ( miRNA ) profile of human lens epithelium ( HLE) induced by H2 O2 and the role of miRNA in oxidative stress induced apoptosis.
METHODS: HLE cell line HLE - B3 was treated by 100μmol/L H2 O2 for 24h and the total RNA were isolated by Trizol reagent. miRNA profile was generated by miRCURYTM LNA microRNA Array. The target genes of differentially expressed miRNAs were predicted by bioinformatics software.
RESULTS:Twenty-eight miRNAs showed significantly differential expression after H2 O2 treatment, 18 miRNAs upregulated and 10 miRNAs downregulated. The differentially expressed miRNAs may involve in apoptosis of lens epithetium and development of cataract through targeting BCL2L2 and MIP.
CONCLUSION: H2 O2 can induce dramatically changes in miRNA profile of HLE, which may play a pivotal role in the pathogenesis and development of cataract.
5.Research of the effective mechanism of rosiglitazone to biliary ischemia-reperfusion injury in autologous liver transplantation

Objective To explore the effective molecular mechanism of PPAR-γligands rosiglitazone to biliary ischemia-reperfusion injury in autologous liver transplantation. Method A total of 40 SD rats were randomly (random number) divided into sham operation group (SO), ischemia - reperfusion group (Ⅰ/R), rosiglitazone (ROS) and GW9662 group, with 10 ones in each. The models, rat biliary ischemiareperfusion injury of autologous liver transplantation, were made by modified two-cuff technique. Tissues of the liver and bile ducts and blood of those models were evaluated by pathological and biochemical methods to make sure the models were made successfully or not. SO group suffered autologous orthotopic liver transplantation, and L/R group suffered both that and ischemia-reperfusion. ROS group were injected rosiglitazone (0.3mg/kg) via portal vein after having been done all as I/R. GW9662 group suffered all as ROS, and 10min later ,they were injected GW9662(0.3mg/kg) via portal vein. 4h after the experiment, tissues of livers and bilary ducts were taken to be tested by immunohistochemistry method, and the blood punctured from the right ventricular were taken to be determined by ELISA. ANOVA was used for statistical analysis.Results IL-1β, TNF-α and IL-6 were mainly expressed in the cytoplasm of hepatocytes and bile duct cells,while NF-κB was expressed both in the cytoplasm and nuclei. Expression of those proteins in L/R and GW9662 group was increased, significantly higher when compared to the SO and ROS (P < 0.05). IL-1β,TNF-α and IL-6 in rat serum were simultaneously increased, and significantly higher than SO(P <0.05).Compared with the SO, expressions of the IL-1 β,TNF-α and IL-6 were not significantly changed in ROS (P> 0.05 )but significantly increased in GW9662. Conclusions PPAR-γ ligand rosiglitazone took protective role in biliary ischemia-reperfusion injury in autologous liver transplantation. The mechanism correlates with the release of the IL-lα, IL-1β and TNF-α and other inflammatory mediators, which decreased as the expression of NF-κB inhibited by its antagonist.
6.Analysis of 220 Cases of Drug Counseling in Inpatient Pharmacy of Our Hospital

OBJECTIVE:To provide reference for continuous improvement of drug counseling and pharmaceutical care. METH-ODS:220 cases of drug counseling answered by inpatient general pharmacist were collected from our hospital during 2012-2013, and the contents of drug counseling were classified and analyzed. RESULTS:Among consultants,73.18% of them were physi-cians;the type of drug involved mainly were antibiotics,accounting for 42.73%;main contents of drug counseling included drug selection,usage and dosage,accounting for 47.27%. CONCLUSIONS:Pharmacists provide drug counseling for physicians and nurses,answer various questions about usage,and guide rational drug use so as to enhance the communication between pharma-cists and physicians,nurses,and promote rational drug use in the clinic.

Objective To study the serum lipid(TC,TG,LDL-C) and the role of typeⅠcollagen,NF-?B,typeⅠcollagen mRNA and NF-?B mRNA levels and their mechanisms in the occurrence and progress of atherosclerosis in order to provide theoretical gist for its prevention.Methods Blood vessel wall pathological and the serum lipid changes,the expressions of typeⅠcollagen,NF-?B,typeⅠcollagen mRNA and NF-?B mRNA levels were observed in dietary induced atherosclerosis rabbit model by morphology study,immunohistochemistry,in situ hybridization technique and color image analyzer(CMAIS).Results The model of AS was established successfully after rabbits were fed with cholesterol,who developed the fatty streak lesion in the aortic intra-wall.In the rabbit model,TC and LDL were respectively increased 43 times and 37 times and there were no difference in TG and body weight.Type I collagen,NF-?B,type I collagen mRNA and NF-?B mRNA were 0.27?0.02,0.19?(0.01) and 0.30?0.03,0.35?0.03,respectively,which were higher than those of control group(0.08?0.01,0.09?0.01 and 0.11?0.01,0.10?0.09,respectively)(P
8.Preparation of paclitaxel-loaded nanoparticles targeting liver cancer stem cells and their effects on liver cancer Huh-7 and HepG2 cells

Objective:To investigate the preparation of paclitaxel-loaded nanoparticles targeting liver cancer stem cells and their effects on liver cancer HepG2 cells (CD133 positive subset accounting for 8%) and Huh-7 cells (CD133 positive subset accounting for 65%).Methods:Poly (lactic-co-glycolic acid)-loaded paclitaxel nanoparticles were prepared by using emulsification-solvent evaporation method. Paclitaxel-loaded nanoparticles decorated with anti-CD133 antibody, called targeted nanoparticles, were prepared by using 1- (3-dimethylaminopropyl)-3-ethylcarbodiimide/N-hydroxysuccinimide (EDC/NHS) cross-linking method. The manifestations and physicochemical characteristics of the nanoparticles including encapsulation efficiency, loading efficiency, particle size distribution, morphology and release in vitro were studied. Liver cancer Huh-7 and HepG2 cells accompanying with paclitaxel-loaded nanoparticles or targeted nanoparticles were cocultured. The uptake and accumulation of nanoparticles by liver cancer cells were analyzed by using flow cytometry, and positive cell proportion of CD133 was also detected. Cell survival was analyzed by using plate clonogenesis assay.Results:Scan electromicroscopy result showed particle size of targeted nanoparticles was (429.26±41.53) nm with zeta potential of -11.2 mV; targeted nanoparticles were possessed with spherical morphology and higher encapsulation efficiency [(87.53±5.90) %]. Flow cytometry showed that in Huh-7 cells at 37℃, the fluorescence intensity of targeted nanoparticles group (13 397±720) was higher than that of paclitaxel-loaded nanoparticles group (6 898±604), and the difference was statistically significant ( P < 0.05); there was no statistically difference in the fluorescence intensity of HepG2 cells in paclitaxel-loaded nanoparticles group and targeted nanoparticles group at 37 ℃ (7 899±343 vs. 8 432±516, P>0.05). CD133 positive cell proportion of Huh-7 cells in targeted nanoparticles group [(15.7±2.6)%] was lower than that in paclitaxel-loaded nanoparticles group [(54.9±7.4)%], and the difference was statistically significant ( t = 7.31, P = 0.008); there was no statistical difference of HepG2 cells between the two goups ( P > 0.05). Plate clonogenesis assay showed that the cell survival rate of Huh-7 cells in targeted nanoparticles group was lower than that in paclitaxel-loaded nanoparticles group at different time points, and the difference was statistically significant ( F = 5.56, P = 0.009); but there was no statistically significant difference in HepG2 cell survival rate between the two groups ( F = 1. 19, P = 0.142). Conclusion:Prepared nanoparticles targeting liver cancer stem cells have a good inhibitory effect on liver cancer Huh-7 cells.
9.Sonographic appearance of atypical breast cancer and needle biopsy

Objective To assess the value of color Doppler sonographic appearance and ultrasound-guided needle biopsy on confirmation of diagnosis of atypical breast cancer. Methods 12 patients suspected to be suffering from breast cancer were examined with color Doppler sonography and ultrasound-guided needle biopsy, the results were then analyzed and evaluated. Results Atypical breast cancer had small tumors (diameter less than 1.5cm), rounded or lobular in shape, and regular margin without obvious spicule. The tumors presented hypoecho and there was no acoustic attenuation behind the tumors. The intensity of vascular signals could not be taken as significant evidence for positive diagnosis. Results of ultrasound-guided needle biopsy was helpful for confirmation of the diagnosis. Conclusions Color Doppler sonography was important for the examination of tumor of atypical of breast cancer. The cancer could be confirmed in good time by the extended sonography and ultrasound-guided needle biopsy.
10.Hepatitis B virus antigen expression and Helicobacter pylori infection in gastric mucosa of patients with chronic liver disease
