1.Analysis of allergens in 167 pateints with allergic contact dermatitis

Objective To assess the environmental contact allergens in patients with allergic contact dermatitis (ACD).Methods Totally,167 patients with ACD were included in this study.All the patients underwent patch test.Results Of these patients,92 (55.1%) were diagnosed as facial ACD,and 148 showed positive patch test results (88.6%).The six most common allergens in a decreasing order were nickel sulfate,fragrance mix,paraphenylenediamine,thimerosal,octanoates and amerchol L 101.Conclusion Patch test may be an efficient way to confirm the cause of ACD.
2.Effect of panretinal photocoagulation on the expression of cyclooxygenase-2, vascular endothelial cell growth factor in epiretinal membrane of proliferative diabetic retinopathy

Objective To observe the effect of panretinal photocoagulation (PRP) on the expression of cyclooxygenase-2 (COX-2),vascular endothelial cell growth factor (VEGF) in epiretinal membrane of proliferative diabetic retinopathy (PDR).Methods A total of 35 patients (35 eyes) with PDR and underwent plana vitrectomy were enrolled in this study.The patients were divided into non-PRP group (19 patients,19 eyes) and PRP group (16 patients,16 eyes) depends on if they had received PRP before surgery.The epiretinal membranes stripped during operation were collected for pathological examination.The histopathological features was observed by haematoxylin and eosin stain.The expression of CD34,COX-2 and VEGF,and microvessel density (MVD) were measured by immunohistochemistry method.Results Many new dispersed capillary blood vessels were found in the thick epiretinal membranes of nonPRP group,while scattered small blood vessels were found in the relatively thin epiretinal membranes of PRP group.MVD value was (7.42± 1.39) in the non-PRP group and (4.56± 1.22) in the PRP group,which was lower than the non-PRP group (t=6.41,P<0.01).The expression of CD34,COX-2 and VEGF in the tissues of epiretinal membrane in PRP group were obviously lower than the non-PRP group (t=6.147,5.944,7.445;P<0.01).Conclusion PRP can effectively inhibit the expression of COX-2 and VEGF in epiretinal membrane of PRP patients.
3.Multiple-slice spiral CT evaluation of occipital condyle fractures

Objective To explore the MSCT findings of occipital condyle fracture (OCF) and improve its diagnostic accuracy. Methods Nineteen patients with OCF, selected from 110 patients suffering high energy injuries at the craniocervical junction, were enrolled into the study. The MSCT appearances of OCFs were retrospectively analyzed by two experienced radiologists. OCF had four types: type Ⅰ was a comminuted fracture, type Ⅱ was a extension of basilar skull fracture, type Ⅲ was an avulsion fracture at the attachment site of alar ligament on occipital condyle, type Ⅳ was a fracture of mixed pattern consisting of two or more above fracture types. Results In 19 patients, the left, right and bilateral OCFs were seen in 7,11 cases, and 1 case, respectively. Type Ⅰ was found in one case, which was a comminution of the left occipital condyle. Type Ⅱ was found in 5 cases, which involved the middle and posterior parts of occipital condyles with 2 on the left and 3 on the right, Type Ⅲ was found in 12 cases which showed various degree of fragment displacement with 3 occuring on the left, 8 on the right, 1 involving bilateral sides, 6 involving articular surfaces and 7 accompaning by enlargment of alar ligaments. Type Ⅳ was found in one case, with coexistence of Type Ⅱ and type Ⅲ. In addition, OCFs were accompanied by head and (or) cervical spine injuries in 14 cases, which included cranial fracture in 8 cases, epidural hematoma in 4 cases, subarachnoid hemorrhage in one case, cerebral contusion and laceration in one case, subfalcial hernia in one case, cervical spine fracture and dislocation in 9 cases, and so on. Conclusion OCFs can be accurately diagnosed by MSCT, which is important for selection of treatment protocols.
4.Direct generation of pluripotent stem cells from differentiated somatic cells.

Embryonic stem (ES) cells have the unique capacity to proliferate extensively and maintain the potential to differentiate into advanced derivatives of all three primary germ layers. ES cell lines can also be generated from human blastocyst embryos and are considered promising donor sources for cell transplantation therapies for diseases such as juvenile diabetes, Parkinson's disease, and heart failure. However, as for organ transplants, tissue rejection remains a significant concern for ES cell transplantation. Another concern is the use of human embryos. One possible means to avoid these issues is by reprogramming the nuclei of differentiated cells to ES cell-like, pluripotent cells. This review discusses the potential of these strategies to generate tailor-made pluripotent stem cells and the role of transcription factors in the reprogramming process.

5.The clinical significance and expression of serum IGF-I and IGFBP-3 in the colorectal cancer

Objective To study the expression of insulin-like growth factor I(IGF-I)and insulin like growth factor binding protein -3(IGFBP-3)in the colorectal cancer and the relations between them. Methods A case-control study was conducted.We selected 113 patients with a pathologically conformed diagnostic colorectal cancer as the colorectal cancer(CRC)group, 37 patients with colorectal benign diseases as benign control(N-CRC)group, and other 76 healthy subjects as normal control(NC)group. CRC was divided into lymphatic metastasis(M-CRC)subgroup and no lymphatic metastasis(NM-CRC) subgroup and was also categorized by TNM staging.Concentrations of serum IGF-I and IGFBP-3 were measured by chemiluminescence method.SNK test and one-way ANOVA were used for comparison between the two groups and among multi-groups, respectively.Results A slightly decreasing trend of the concentrations of serum IGF-I was observed following the sequences of the NC, N-CRC and CRC groups. Moreover,the concentrations of serum IGF-I in T4 were significantly lower than that in(T1+T2)and T3 (F=6.279,P=0.003).The concentration of serum IGFBP-3 in the CRC group(3.13 ±1.57)μg/ml was significantly lower than that both in the N-CRC(3.42 ±1.32)μg/ml(F=3.04,P=0.09)and the Normal group(4.62 ±1.10)μg/ml(F=10.88,P=0.001).The concentration of serum IGFBP-3 in the M-CRC was higher than that in NM-CRC(F=4.44,P<0.05).The ratio of IGF-I/IGFBP-3 in the CRC group (48.85 ±29.14)was remarkably higher than that both in the N-CRC group(42.38 ±12.58)(F=5.05, P=0.02)and the Normal group(42.46 ±16.24)(F=5.68,P=0.02).The value of diagnosis is general. The area under the ROC curve(AUC)of IGF-I was 0.63(95% CI 0.56 -0.70), the sensitivity and specificity of IGF-I were 85.8%and 40.7%respectively when the critical value was 102.5 μg/ml.The area under the ROC curve(AUC)of IGFBP-3 was 0.72(95%CI 0.64-0.78),the sensitivity and specificity of IGFBP-3 were 78.8%and 62.8%respectively when the critical value was 3.32 μg/ml.Conclusions The concentrations of IGF-I and IGFBP-3 were low expressed in serum of patients with colorectal cancer. Meanwhile,they are correlated with the lymphatic metastasis and TNM staging.
6.Quantitative analysis of biochemical components in the suprachoroidal space fluid of choriodal detachment associated with rhegmatogenous retinal detachment

Objective To investigate the nature of the suprachoroidal fluid by detecting the concentration of total protein (TP), lactate dehydrogenase (LDH), albumin (ALB), total cholesterol (CHOL), total bilirubin (TBIL) in suprachoroidal liquid of patients who have rhegmatogenous retinal detachment with choroid detachment (RRDCD). Methods Eighteen RRDCD patients (18 eyes) who underwent vitrectomy were enrolled in this study. There were 10 males (10 eyes) and 8 females (8 eyes), 8 right eyes and 10 left eyes. There were 8 patients with age of ≤55 years, 10 patients with age of >55 years. There were 7 patients with duration of≤30 days, 11 patients with duration of >30 days. There were 7 eyes with diopters of ≥-6.0 D, 11 eyes with diopters of <-6.0 D. There were 11 eyes with class C proliferative vitreoretinopathy (PVR), 7 eyes with class D PVR. Suprachoroidal fluid samples were collected from all the patients, and took preoperative serum samples as RRDCD group. Ten serum samples of normal people were set as control group. The concentration of TP, LDH, ALB, CHOL, TBIL in all the subjects were measured. The properties of the suprachoroidal fluid were identified by Light standard and concentration standard of ALB, CHOL, TBIL. Results There was no difference on the concentration of TP, LDH, ALB, CHOL, TBIL from suprachoroidal fluid samples in the patients with different age, sex, eyes, diopter, PVR grade (P>0.05). There was no difference on the concentration of TP, LDH, ALB, CHOL, TBIL from preoperative serum samples in the patients between RRDCD group and control group (P>0.05). There was no difference on the concentration of ALB and CHOL from suprachoroidal fluid samples and preoperative serum samples in the RRDCD patients (P>0.05), but there were significant differences on the concentration of TP, LDH, TBIL (P<0.05). According to the Light standard, there were 17 cases of exudates and 1 case of transudate. According to the concentration standard of ALB, CHOL and TBIL, there were 14, 18, and 16 cases of exudates, and 4, 0, and 2 cases of transudate, respectively. There was no difference on the identification result of Light standard and concentration standard of ALB, CHOL, TBIL (χ2=2.090, 1.029, 0.364;P>0.05). Conclusion The suprachoroidal fluid of RRDCD patients composed of TP, LDH, CHOL and TBIL. The suprachoroidal fluid is more likely to be exudate.
7.RGD-modified iron oxide nanoparticles for targeted molecular imaging of hepatocellular carcinoma

Objective To synthesize a molecular probe targeted to human hepatoma HepG2 cells with high expression of integrin αvβ3 (RGD-PEG-VSOP) and evaluate its MRI efficacy in vitro.Methods RGD-PEG-VSOP was characterized and analyzed by 1H NMR and TEM.MTT test was used to evaluate its biological safety.In vitro experiments at the cellular level,the targeting effect of RGD-PEG-VSOP to integrin was assessed,meanwhile the nontargeted nanoparticles were used as controls.Results TEM showed that the nanoparticles were spherical and uniform in size,with a relatively high r1 relaxivity of 1.37 mM-1S-1.MRI showed the signal intensity of the HepG2 cells treated with RGD-PEG--VSOP was significantly higher than that of the HepG2 cells treated with PEG-VSOP (P<0.05).Conclusion RGD-PEG-VSOP has positive T1 contrast effect.At the cellular level,the RGD-PEG-VSOP nanoparticles have the characteristics targeted to integrin αvβ3.
8.AIDS discrimination in junior college students and the effect of AIDS knowledge on discrimination

Objective:
To analyze the situation of AIDS knowledge and discrimination among freshmen in Chengdu city, and to explore possible effects of AIDS knowledge on discrimination.
Methods:
A cluster random sampling was employed to investigate 1 053 college students from 11 universities in Chengdu about their HIV/AIDS knowledge and discrimination. The scores of AIDS knowledge and discrimination of students with different characteristics were analyzed, and the influence path of AIDS knowledge on AIDS discrimination were further analyzed based on different peer relationships.
Results:
The total scores of AIDS knowledge was negatively correlated to AIDS discrimination( r s =-0.13, P <0.01). After adjusting for confounding factors, the total score of AIDS knowledge was associated with the total score of AIDS discrimination( β =-0.12, P <0.01). AIDS knowledge played a role in AIDS discrimination in intimate, general and unfamiliar peer relationships, with standardized path coefficients of -0.20, -0.24 and -0.18 respectively( P <0.01).
Conclusion
AIDS knowledge are correlated with discrimination among freshmen under different peer relationships. More anti-AIDS discrimination courses should be added to AIDS education to reduce the students’ fear and stigma of HIV/AIDS patients and related risk groups.
9.Gastrodin Inhibits Non-alcoholic Fatty Liver Disease via Mediating SREBP1c Signaling Pathway

ObjectiveTo observe the effect of gastrodin on the steroid regulatory element-binding protein 1c (SREBP1c) signaling pathway in high-fat high-cholesterol diet (HFHC)-induced mice and explore the mechanism of gastrodin in the treatment of non-alcoholic fatty liver disease (NAFLD). MethodEight-week-old male C57BL/6J mice were used in vivo and divided into the following four groups, with six mice in each group: normal group, gastrodin group (50 mg·kg-1), model group, and model + gastrodin group (50 mg·kg-1). NAFLD model was established by feeding mice with HFHC for four weeks, and the mice were euthanized and the liver tissues were collected after four weeks. In vitro experiments were performed using Huh7 cells which were divided into five groups, and induced with free fatty acids (FFA, 200 μmol·L-1, oleic acid-palmitic acid 2∶1) to establish an NAFLD cell model. After 24 h, different concentrations of gastrodin (0, 5, 10, 20, and 40 μmol·L-1) were added to each group and cultured for another 24 h. Oil red O staining was used to detect lipid accumulation in mouse liver and Huh7 cells. Hematoxylin-eosin (HE) staining was used to observe pathological changes in liver tissue. Levels of serum total cholesterol (TC), low-density lipoprotein cholesterol (LDL-C), and triglycerides (TG) were measured using an automatic biochemical analyzer. Relevant assay kits were used to detect liver TC, TG, and FFA levels. Real-time quantitative polymerase chain reaction (Real-time PCR) and Western blot were used to detect the expression of lipid synthesis-related proteins fatty acid synthase (FASN), acetyl-CoA carboxylase 1 (ACC1), and stearoyl-CoA desaturase 1 (SCD1). ResultCompared with the normal group, the model group showed significantly increased serum TC, LDL-C, and TG levels (P<0.01), liver TC, TG, and FFA levels (P<0.01), increased lipid accumulation in Huh7 cells (P<0.01), and significantly increased expression levels of lipid synthesis-related genes SREBP1c, FASN, ACC1, and SCD1 in mice and Huh7 cells (P<0.01). Compared with the model group, after gastrodin treatment, the serum TC, LDL-C, and TG levels in mice significantly decreased (P<0.05, P<0.01), the severity of fatty liver disease improved significantly, liver TC, TG, and FFA levels decreased significantly (P<0.05, P<0.01), lipid accumulation in Huh7 cells decreased significantly (P<0.05, P<0.01), the expression levels of lipid synthesis-related genes SREBP1c, FASN, ACC1, and SCD1 in mice and Huh7 cells decreased significantly (P<0.05, P<0.01). ConclusionGastrodin can reduce hepatic lipid accumulation and blood lipid levels, improve HFHC-induced NAFLD, and its mechanism of action may be related to the regulation of the SREBP1c lipid synthesis-related signaling pathway.
