1.Effects of Banxia Xiexin Decoction and Its Components on Protein Expression of Cx43 in Gap Junction in Gastric Tissue of Electrogastric Dysrhythmias Rats

Objective To discuss compatibility regularity of Banxia Xiexin Decoction adjusting gastric mobility. Methods Ninty healthy SD male rats were divided into normal group (10 rats) and model group (80 rats). Rat models of electrogastric dysrhythmias were established. After the evaluation of gastric electrophysiology, Banxia Xiexin Decoction were divided into Xinkai, Kujiang, Ganbu, Xinkaikujiang, Xinkaiganbu, and Kujiangganbu groups according to the compatibility regularity of Banxia Xiexin Decoction, 10 rats for each group. Rats in each medication administration group received gavage with same concentration but different volume for 4 weeks. Each group was analyzed for gastric electrical parameters after administration. The expressions of Cx43 and Cx43 mRNA in the gastric tissue of rats in each group were analyzed by immunohistochemistry and real-time fluorescent quantitative PCR. Results Compare with the normal group, the expressions of Cx43 and Cx43 mRNA in the gastric tissue of rats increased significantly in the model group. Compared with the model group, variable coefficient of electrogastric slow wave frequency in all medication administration groups decreased obviously. Banxia Xiexin Decoction and its components had decreased the expression of Cx43 and Cx43 mRNA to some extent, among which Xinkaiganbu group had best effects. Conclusion The mechanism of Banxia Xiexin Decoction adjusting gastric motility maybe related to its function of decreasing the expressions of Cx43 and Cx43 mRNA, as to adjust disordered electrogastric rhythm.

AIM: The experiment was designed to study the effect of compound Danshen dripping pills (DSDP) on myocardium with anoxin/reoxygenation. METHODS: The myocardial anoxin/reoxygenation model was made in perfused isolated rat heart. DSDP and isosorbide dinitrate (ID) were given at the time of pre-perfusion and reperfusion, then HPLC and H-600 electron microscope were used to detect the change of high energy phosphate and the ultrastructure of myocardial cell. 4.Value of Brain Natriuretic Peptide for the Diagnosis and Treatment of Chronic Left Heart Failure and Cor Pulmonale

Objective:To investigate the variation of plasma braln natriuretic peptide(BNP) and its value for diagnosis and treatment in patients with hypertension accompanied by chronic left heart fallure or chronic obstructive pulmonary disease (COPD) accompanied by cor pulmonale(CP) .Methods:A total of 36 patients with hypertension accompanied by chronic left heart fallure and 34 patients with COPD accompanied by CP ,who were admitted to Shanghal Second People’s Hospital from Nov 2010 to Nov 2013 ,were enrolled as study subjects .Meanwhile ,30 normal control were enrolled .Quantitative detection of plasma BNP level was done in each group before and after the treatment .Results:The levels of plasma BNP in the hypertension accompanied by chronic left heart fallure group and the COPD accompanied by CP group before the treatment were higher than that in the control group(P<0 .05) .The levels of plasma BNP in the hypertension accompanied by chronic left heart fallure group was higher than that in the COPD accompanied by CP group before the treatment(P< 0 .05) .After the treatment ,the levels of plasma BNP in both the hypertension accompanied by chronic left heart fallure group and the COPD accompanied by CP group decreased (P<0 .05) .The level of plasma BNP in the hypertension accompanied by chronic left heart fallure group was higher than that in the COPD accompanied by CP group after the treatment(P<0 .05) .Conclusions:BNP can be used as the index for differential diagnosis and therapeutic effect assessment of CP and chronic left heart fallure .
5.Application effect of close-loop path management of aspiration risk screening in patients in Department of Neurosurgery

Objective:To construct a close-loop path management model for aspiration risk screening of patients in Department of Neurosurgery, and verify its application effect in the nursing of inpatients in department of neurosurgery.Methods:Through literature retrieval and expert consultation, the close-loop path management model of aspiration risk screening was constructed. The convenient sampling method was adopted to select inpatients in the Neurosurgery Department of a ClassⅢ Grade A hospital in Jiangmen of Guangdong Province from January to December 2019. The patients admitted from January to June 2019 were set as the control group and received routine aspiration assessment and management while the patients admitted from July to December 2019 were set as the observation group and received close-loop management of aspiration risk screening. The incidences of aspiration and aspiration pneumonia were compared between the two groups.Results:In the control group, there were 506 patients, of whom 92 (18.18%) had aspiration. In the observation group, there were 543 patients, of whom 74 (13.63%) had aspiration. There was a statistically significant difference in the incidence of aspiration between the two groups (χ 2=4.078, P=0.043) . The incidence of aspiration pneumonia was 8.10% (41/506) in the control group and 5.52% (30/543) in the observation group. There was no statistically significant difference between the two groups (χ 2=2.758, P=0.097) . Conclusions:The close-loop management of aspiration risk screening standardizes the process of aspiration risk screening, guarantees the implementation of dynamic assessment and effectively reduces the incidence of aspiration, which can provide a reference for improving the management level of prevention and treatment of aspiration.
6.Construction of a mutant strain of Nocardia farcinica with mce4A gene deletion and analysis of the role of mce4A gene

Objective To construct a mutant strain of Nocardia farcinica ( N. farcinica ) IFM10152 with mammalian cell entry 4A gene (mce4A) deletion and to analyze the function of that gene dur-ing infection. -ethods The mutant strain of N. farcinica was constructed through in-frame deletion without antibiotic labeling and verified by PCR and sequencing analysis. To analyze the function of mce4A gene in the interaction between N. farcinica and host cells, in vitro growth experiment, macrophage killing experi-ment using THP-1 ( a human leukemia mononuclear cell line) as the model and adhesion and invasion exper-iments using HeLa cells ( cervical cancer epithelial cells) were carried out. Results The mutant strain with mce4A gene deletion was successfully constructed and named △mce4A. No significant difference in growth rate was observed between the mutant and the wild-type strains. After knocking out the mce4A gene, the ability of N. farcinica to resist macrophage killing was obviously weakened as well as its ability to adhere and invade. Conclusions The mutant strain of N. farcinica with mce4A gene deletion was successfully construc-ted. The mce4A gene might play an important role in the adhesion and invasion of N. farcinica to host cells and its survival in macrophages.
7.Effects of high glucose and lysophosphatidylcholine (LPC) on RAW264.7 macrophages during No-cardia farcinica infection

Objective To investigate the effects of high glucose and lysophosphatidylcholine (LPC) on the immune function of in vitro cultured macrophages during Nocardia farcinica infection. Meth-ods RAW264.7 macrophages were cultured in vitro under different conditions as follows: routine culture (control group),50 mmol/L glucose (high glucose group),10 mg/L LPC(LPC groupⅠ),25 mg/L LPC (LPC groupⅡ) and 50 mmol/L glucose+25 mg/L LPC(high glucose and LPC group). The activity of mac-rophages in each group was tested after 6,12,24 and 36 h of culture. After 24 h of culture, macrophages were collected from every group and co-cultured with Nocardia farcinica. Dynamic phagocytosis rates were detected at 1,2,3,4,5 and 6 h after co-culture. Toxic effects of Nocardia farcinica on macrophages and concentrations of IL-10 and TNF-α were measured at 1,3 and 6 h after co-culture. Results Macrophages in all four experimental groups showed decreased activity as compared with those in the control group (P<0.01). Phagocytosis of Nocardia farcinica by macrophages was also reduced by high glucose and LPC. Phagocytosis rates of high glucose group and LPC groupⅡ at 1 and 2 h,LPC groupⅠat 1,2 and 3 h,and high glucose and LPC group at 1,2,3 and 4 h after co-culture were significantly lower than that of the con-trol group (P<0.05 or P<0.01). Compared with the control group, significantly reduced toxic effects on macrophages caused by Nocardia farcinica was observed in the experimental groups (P<0.05 or P<0.01). Compared with the control group,LPC groupsⅠand Ⅱ and high glucose and LPC group had decreased se-cretion of IL-10 at 3 h,and high glucose group and LPC groupⅠhad decreased secretion of TNF-α at 1 h(P<0.05). Conclusion Culture macrophages under the conditions of high glucose and LPC would reduce their activity and impair their ability to phagocytose Nocardia farcinica. Moreover, high glucose and LPC might have impacts on the toxic effects of Nocardia farcinica on macrophages and the secretion of IL-10 and TNF-α.
8.A two-dimensional photographic and three-dimensional digital dental model comparative analysis in maxillary anterior teeth

Objective:To assess the accuracy of two-dimensional (2D) photographs in measuring esthetic parameters of the maxillary anterior teeth by comparing them with measurements obtained from three-dimensional (3D) dental models.Methods:A total of one hundred volunteers (49 males, 51 females, aged 18-23 years) were recruited from School and Hospital of Stomatology, Wuhan University from January to February 2024. 3D digital models of their dentitions were obtained using an intraoral scanner, and standardized frontal 2D intraoral photographs were captured with a digital camera. The lengths, widths and width/length ratio of the bilateral incisors, lateral incisors and canines were measured on both the 3D digital models and the 2D intraoral photographs. The width ratios of adjacent maxillary anterior were also calculated on the 2D intraoral photographs and the frontal view of 3D digital models.Results:The widths of lateral incisors [(5.85±0.60) mm] and canines [(4.73±0.71) mm] and the lengths of canines [(8.72±0.96) mm] in the 2D intraoral photographs were significantly lower than those in 3D digital models [(6.65±0.59), (7.76±0.60), (8.90±0.86) mm] ( t=-18.24, P<0.001; t=-54.43, P<0.001; t=-4.40, P<0.001), while there were no significant differences in the lengths and widths of the other teeth ( P>0.05). The width/length ratios measured from the 2D intraoral photographs for the lateral incisors and canines (0.74±0.08, 0.55±0.08) were significantly lower than those measured in the 3D digital models (0.84±0.09, 0.88±0.09) ( t=-19.68, P<0.001; t=-50.21, P<0.001), and the width/length ratio of the central incisors showed no significant difference between the two groups ( P>0.05). The width ratios of canines/lateral incisors and lateral incisors/central incisors measured on the 2D intraoral photographs (0.72±0.06, 0.85±0.11) were significantly smaller than those measured in the frontal view of 3D digital models (0.75±0.06, 0.89±0.11) ( t=-9.31, P<0.001; t=-6.58, P<0.001). Conclusions:There is a difference between 2D and 3D measurement results of teeth in the esthetic area and the magnitude of the difference varies with their position in the dental arch. When analyzing the measurement of the anterior teeth, it is necessary to choose the appropriate method according to the target tooth position.
9.Radiomics features of ascending and descending nasopharyngeal carcinoma.

OBJECTIVES:
To evaluate the application value of CT-based radiomics features for the ascending and descending types of nasopharyngeal carcinoma (NPC).
METHODS:
A total of 217 NPC patients (48 ascending type and 169 descending type), who obtained CT images before radiotherapy in Affiliated Cancer Hospital of Xiangya School of Medicine, Central South University from February 2015 to October 2017, were analyzed retrospectively. All patients were randomly divided into a training set (=153) and a test set (=64). Gross tumor volume in the nasopharynx (GTVnx) was selected as regions of interest (ROI) and was analyzed by radiomics. A total of 1 300 radiomics features were extracted via IBEX. The least absolute shrinkage and selection operator (LASSO) logistic regression was performed to choose the significant features. Support vector machine (SVM) and random forest (RF) classifiers were built and verified.
RESULTS:
Six features were selected by the LASSO from 1 300 radiomics features. Compared with SVM classifier, RF classifier showed better classification performance. The area under curve (AUC) of the receiver operating characteristic (ROC) curve, accuracy, sensitivity, and specificity were 0.989, 0.941, 1.000, and 0.924, respectively for the training set; 0.994, 0.937, 1.000, and 0.924, respectively for the validation set.
CONCLUSIONS
CT-based radiomics features possess great potential in differentiating ascending and descending NPC. It provides a certain basis for accurate medical treatment of NPC, and may affect the treatment strategy of NPC in the future.
