1.A literature review on acupuncture and moxibustion for prevention and treatment of lung cancer in SCI iournals (2003-2013).

To explore the status of acupuncture and moxibustion for prevention and treatment of lung cancer in current years; literature regarding acupuncture and moxibustion for prevention and treatment of lung cancer between 2003 and 2013 from SCI journals was retrieved and analyzed. As a result, 20 papers were included, which were published in 17 journals including Journal of Clinical Oncology, Chest, Respirology and Lung Cancer, etc. Of them, 3 papers discussed the effects of acupuncture on progressing of lung cancer; 4 articles confirmed that acupuncture could reduce myelosuppression and digestive tract reactions induced by radiotherapy and chemotherapy; 6 papers showed that acupuncture could relieve pain or fatigue of lung cancer; 3 papers indicated that acupuncture could palliate dyspnea in lung cancer patients. It is concluded by domestic and overseas researches that acupuncture and moxibustion are effective and safe for symptoms of lung cancer, which is worthy of further study.

3.Rules of acupoints combination of ancient acupuncture for Xiaoke based on data mining technology.

The rules of acupoints combination of ancient acupuncture for Xiaoke are mainly explored. By retrieval on ancient literature, the database of acupuncture and moxibustion for Xiaoke is established; based on the database, association analysis between acupoints and symptoms is performed. According to the association analysis in 5 databases of Xiaoke database, Xiaoke database of kidney deficiency, Xiaoke-database of dry mnouth and thirst, Xiaoke database of difficult urination, Xiaoke database of drinking addiction, the results are mainly characterized with symptom differentiation combination, distal-local combination, local combination and front-back combination, which can nourish yin and clear heat. It is believed that establishment of TCM ancient literature database and exploration of data mining technology is a potential research orientation.

4.Clinical characteristics and prognosis of novel bunyavirus infection: 68-case report

Objective To retrospectively analyze the clinical characteristics,prognosis and risk factors of novel bunyavirus infection.Methods The clinical data of 68 patients with novel bunyavirus infection confirmed by laboratory diagnosis at Wendeng Central Hospital of Weihai were retrospectively collected.Epidemiological characteristics,clinical manifestations,physical signs and laboratory results were analyzed.Results Twenty two patients (32.4 %) had intimate contact with ermine (breeding ermine or ermine biting) ; 4 patients (5.9%) had been bitten by tick within 2 weeks,6 patients (7.4%) had intimate contact with patients with severe fever with thrombocytopenia syndrome (SFTS) ; and 25 patients (36.8 %) had a history of fieldwork before the onset of the disease.Thirty-four patients (50.0 %) were over 60 years old and 27 cases (39.7%) had underlying diseases.Initial symptoms in all patients were fever accompanied by loss of appetite,fatigue and other toxemic symptoms,followed by multi organ damage.Other clinical manifestations included nervous system damage (27 cases,39.7%),hemorrhage (4 cases,5.9%),rapid atrial fibrillation (10 cases,14.7%) and pneumonia (18 cases,26.5%).White blood cell count of 55 cases (80.9%) was less than or equal to 2.0 × 109/L,platelet count of 18 cases (26.5%) was less than or equal to 30 × 109/L.Abnormal hepatic function was found in 62 cases (91.2%); elevated myocardial enzymes was found in 68 cases (100.0%),prolonged activated partial thromboplastin time in 44 cases (64.7%),hyponatremia in 23 cases (33.8%),hypokalemia in 29 cases (42.6%),hypocalcemia in 36 cases (82.4%),hyperglycemia in 49 cases (72.1%).Serum nucleic acid quantitation of novel bunyavirus varied from 1.10 × 102 to 5.78 × 107 tissue culture infective dose (TCID)/ mL.Fifty five cases were cured,accounting for 80.9 %,while 13 (19.1%) died eventually.Conclusions High risk factors of novel bunyavirus infection included intimate contact with ermine and infected patients,tick biting and fieldwork.Patients with elder age,underlying diseases,nervous system symptoms,hemorrhage,pneumonia,low platelet,high viral load and elevated myocardial enzymes may have poor progonsis.
5.Effect of response to phenylephrine-mediatedsignal pathway in renal arteries of offspring rats inducedby maternal high-salt diet

Aim To study the effect of high salt diet during pregnancy on the development of renal vessels in offspring rats and its mechanism.Methods Natural pregnant Sprague-Dawley rats were randomly divided into high-salt group and control group.The pregnant rats in the high-salt group were given high-salt diet of 8% NaCl content , while the control group normal diet with 1% NaCl content.In both groups, pregnant rats were given normal drinking water.After delivery, all mothers returned to normal diet and all neonatal rats were breast-fed until one month old.The adult male off springs were used as experimental animals.The vessel tone of renal interlobar arteries and electrophysiological behavior of single vascular smooth muscle cells (VSMCs) were detected respectively.Results The contractile response of renal arteries to phenylephrine(Phe) in high-salt group was stronger than that in the control group(P<0.05).The effect of protein kinase C(PKC) non selective blocker GF109203X on Phe-induced contraction in two groups also showed significant difference(P<0.05).In electrophysiology experiments, Phe inhibited high-conductance Ca2+-activated K+ channel(BK channel) currents in renal arteriolar smooth muscle cells in two groups, and the inhibitory effect was more pronounced in high-salt offsprings (P<0.05).GF109203X could eliminate the response of Phe on BK currents in both groups(P>0.05).Conclusions High-salt diet during pregnancy could increase the sensitivity of renal interlobar arterial contractile response to Phe in adult male offsprings, which is associated with PKC-mediated BK channels pathway.Maternal high-salt diet during pregnancy may increase the risk of renal vascular diseases in adult offsprings.
6.Effect of epigallocatechin-3-gallate on radiosensitivity of CNE-1 cells

Objective To investigate the effect of EGCG on the radiosensitivity of human nasopharyngeal carcinoma CNE-1 cells.Methods CNE-1 cells were divided into four groups:control,EGCG treatment,UVC or X-ray exposure,and EGCG combined with UVC or X-rays.After treatment with different concentrations of EGCG for 24,48 and 72 h and UVC or X-rays,cell growth was determined with MTT assay,cell survival was measured with clonogenic assay,cell cycle was deteeted with flow cytometry,cell apoptosis was detected by the annexin V-FITC cell apoptosis kit,and protein expression was assayed by Westem blot.Results EGCG inhibited cell growth in a dose-and time-dependent manner(r =0.817and 0.364).Compared with UVC or X-ray irradiation alone,the radiosensitivity of CNE-1 cells was enhanced by 2 h pre-treatment of 50 μmol/L EGCG,which disrupted S phase arrest caused by UVC( t =18.68,P < 0.05 ) and increased the population of S and G2/M arrest caused by X-rays ( t =7.11 and 6.99,P <0.05 ).UVC could cause a significant increase of sub-G1 population( t =6.67,P < 0.05 ) and Annexin V-FITC assay indicated apoptosis was further elevated by EGCG ( t =10.28,P < 0.05 ).However,no significant induction of apoptosis was observed in the cells either irradiated with X-rays alone or combinationly treated with EGCG and X-rays.The combination treatment of EGCG and UVC significantly increased the expression of Bax and Caspase-3 proteins,but failed to affect Bcl-2 protein expression.Conclusions EGCG enhances the growth inhibition of CNE-1 cells caused by UVC or X-rays,which is relevant to apoptosis induction or cell cycle arrest.
7.Comparison of broth microdilution and agar dilution methods for antifungal susceptibility testing of Malassezia speciesin vitro

Objective To compare broth microdilution and agar dilution methods for in vitro testing of activities of fluconazole,ketoconazole and itraconazole against clinical Malassezia isolates.Methods Broth microdilution and agar dilution methods were used to determine the minimal inhibitory concentration(MIC)of fluconazole,ketoconazole and itraconazole for 27 clinical strains(5 species)of Malassezia.Results The minimal inhibitory concentration(MIC)ranges of fluconazole,ketoconazole and itraconazole were 0.25-≥64 mg/L,≤0.03-0.5 mg/L and ≤0.03-0.125 mg/L respectively as shown by broth microdilution method,2-≥64 mg/L,≤0.03-0.5 mg/L and ≤0.03-0.25 mg/L respectively as revealed by agar dilution method.Both methods demonstrated that itraconazole possessed the strongest activity against Malassezia species,followed by ketoconazole and fluconazole.The agreement rate in MICs between the two methods was 78.8%,85.2% and 88.9%,respectively for fluconazole,ketoconazole and itraconazole,with the intraclass correlation coefficients (ICCs)being 0.88,0.80 and 0.76 respectively.Conclusions Fluconazole,ketoconazole and itraconazole are highly active against Malassezia species in vitro,and itraconazole is the most active.Broth microdilution and agar dilution method coincide well in,and are applicable for,the antifungal susceptibility testing of Malassezia species in vitro.
8.Influence of desipramine on lipopolysaccharide-induced acute lung injury in mice

Aim To investigate the potential role of desipramine(DP) on lipoplysaccharide(LPS)-induced acute lung injury(ALI)and the mechanism of its action.Methods Kunming mice were divided into four groups randomly:NS group(NS),DP control group(DP),LPS group(LPS)and DP treatment group(DP+LPS).The model of ALI in mice was induced by lipoplysaccharidel(LPS,10 mg·kg~(-1),ip).Six hours after LPS challenged,the lung samples were taken for determination of lung wet-to-dry weight ratio(W/D),myeloperoxidase(MPO)activity,and malondialdehyde(MDA)content.The bronchoalveolar lavage fluid(BALF)samples were analyzed for total protein concentrateion and white blood cell(WBC)count.The levels of tumor necrosis factor-α(TNF-α)in lung were measured by ELISA.Results LPS could significantly increase the total protein concentration and WBC number in BALF.The lung W/D ration,MPO activity,MDA content and the levels of TNF-α in lungs all increased after ip injection of LPS.Pretreatment with DP decreased all the changes induced by the LPS.Conclusion Pretreatment with DP protects lung from LPS-induced lung injury in mice,which is,at least in part,through inhibiting the level of TNF-α and decreasing the sequestration of neutrophils and lipid peroxidation.

10.Impact of honokiol on growth and radiosensitivity in nasopharyngeal carcinoma cells

Objective To explore the influence of honokiol on growth,cell cycle and radiosensitivity in nasopharyngeal carcinoma cells,CNE-1 and CNE-2.Methods MTT assay and clonogenic assay were used to detect cell growth and survival respectively.Flow cytometry was employed to analyze cell cycle progression.Annexin V-FITC kit was used to detect cell apoptosis.Western blot assay was applied to examine protein expression.Results Honokiol signficantly inhibited proliferation of CNE-1 and CNE-2 cells in a dose and time dependent manner,the IC50 value was 2.84 and 2.68 μmol/L(24 h)and 2.50 and 2.20 μmol/L (48 h),respectively.After being treated with 2.5 μmol/L honokiol for 24 h,the ratios of early apoptosis,late apoptosis and necrosis were 24.53%,23.05% and 7.13% in CNE-1 cells compared with the control group(t =-41.17,-8.18,-6.08,P <0.05).The expression levels of pro-apoptotic proteins Caspase-3 and Bax were significantly increased to 2.31 and 1.89 times (t =-15.92,-17.15,P < 0.05),4.43 and 1.85 times (t =-29.39,-13.47,P < 0.05).Simultaneously the expression level of anti-apoptotic protein Bcl-2 was reduced by 2.22 and 2.74 times(t =26.94,66.14,P < 0.05) as compared with controls after being treated with 4 and 3 μmol/L honokiol.Additionally,honokiol at lower doses signicantly enhanced the senstivity of CNE-1 and CNE-2 cells to X-ray irradiation.The SER was 1.41 and 1.88 in CNE-1 and CNE-2 cells.3 Gy irradiation of X-rays increased the proportion at G2/M state in both cell lines (t =-14.96,-19.26,P < 0.05).Honokiol reduced the G2/M cell cycle arrest induced by irradiation significantly (t =7.65,4.98,P < 0.05).Simultaneously,cyclin B1 protein expression obviously elevated (t =-33.07,-73.49,P < 0.05).Conclusions Honokiol is a potent inhibitor of nasopharyngeal carcinoma cell growth by inducing cell apoptosis and necrosis and works as a radiosensitizer by disrupting G2/M cell cycle checkpoint.
