1.Studies on the Resistance of seed of Ural Licorice(Glycyrrhiza uralensis)

Several resistant characteristics of Glycyrrhiza uralensis seeds were studied. The results showed that hard seed can resist attack from fungi or bacteria. It can endure high temperature for 4h at 80℃ and 10 min at 100℃, but when at 100℃ for 1 h it loses its germinating power. The seeds stored for 4 years do not reduce viability. After long period storage for 13 years,about 60% of the seed still retains its germinating power.
2.Effect of PRL on the expression of CD154 in Jurkat cells

Expression of PRL receptor mRNA in Jurkat D1.1 cell line was confirmed by RT PCR. CD154 expression of Jurkat cells was significantly increased when these cells were incubated with PHA and recombinant human PRL, suggesting that binding of PRL to its receptor promotes CD154 expression which plays a role in immune regulation.
3.Expression of PRLr mRNA in human immune system

Objective:To investigate the expression of prolactin receptor(PRLr) in human immune system.Methods:Specimens of human immune system were obtained from both central and peripheral immune organs, including thymus(thymoma), bone marrow, lymph node and peripheral blood mononuclear cells. Specific fragments of PRLr mRNA were obtained through RNA isolation and RT-PCR amplification and confirmed by DNA sequencing.Results:PRLr mRNA was detected from the thymoma, bone marrow, lymph node and peripheral blood mononuclear cells. All the PRLr cDNA fragments generated by PCR were as long as the expected length, which was 276 bp. The sequencing result of the cDNA was identical to the sequence of PRLr cDNA in GeneBank.Conclusion:The study confirmed that prolactin receptor mRNA was expressed in human central immune organs such as thymus and bone marrow and peripheral immune organs such as lymph node and peripheral blood mononuclear cells. This phenomenon provided a biologic structural evidence that neuro-endocrine hormone PRL might play a modulating role on immune cells from receptor's point of view.
4.A Model Establishment of CombinedPancreas-Kidney Transplantation in Dog

Objective To explore a best operative method in order to improve operative manipulated skill of simultaneous pancreas-kidney transplantation(SPKT),a model of SPKT was established in dogs.Methods 12 dogs of SPKT were performed in animal model.A renoportal end-to-end anastomoses between the renal and the spleen vein.Only two vascular end-to-side anastomoses between the donor portal vein and recipient ilio-vein,and between the donor celiac arteries and recipient abdominal aorta were constructed.Pancrease was placed in the right iliac fossa and kidney in the left.Pancreatic exocretion goes through bladder drainage.Results Combined resection was successfully performed in 6 dogs and the other underwent simultaneous pancreas-kidney transplantation.Satisfactory result was obtained in 5 survival dog for a mean of (1 5?0 8)days,although 1 dog died from hypopiesis.Conclusions The model is practically feasible and might be used in studying the problems involved in SPKT,which is effective for the treatment of end-stage diabetic nephropathy.
5.Nitric oxide synthases expression in human bladder cancer and their relationship with angiogenesis

Objective Evaluated NOS expression in bladder tissue from the patients with transitional cell carcinoma (TCC) of the bladder and studied its relationship with angiogenesis. Methods Bladder carcinoma tissue specimens were procured from 58 patients with TCC and 14 cases of benign bladder tissue as contrast group. NOS immunohistochemistry was performed on all tissue specimens and microvessal density (MVD) was counted by endothelial cells immunostained. Results Inducible NOS specific proteins were found in 47 of 58 bladder cancer specimens but not in control bladder tissue. The malignant cells and inflammatory cells within the carcinomas were highly iNOS positive whereas specimens of bladder mucosa outside the malignant regions showed only a weak positive iNOS immunostaining. The endothelial cells in both normal urothlium and tumor tissue showed a highly positive eNOS immunostaining but its immunoreaction was not detected in either malignant or benign epithelium. A chemiluminescence enzyme immunoassay based on magnetic microparicles (MmPsCLEIA) was developed to evaluate serum α-fetoprotein (AFP) in parallel with tramional colorimetric enzyme-linked immunsorbrnt assay (ELISA).A sestematic comparison between the MmPs-CLEIA and colorimetric ELISA concluded that the MPs-CLEIA exhibited fewer of immunoreagents,less total assay time,and better linearity,recovery,precision,senitivity and validity.AFP was detected in forty human serum samples by the proposed MPs-CLEIA and ELISA,and the results werecompared with commercial electrochemilunminescence immunoassay (ECLIA) kit.The correlation coefficient between MPs-CLEIA and ELISA was obtained with R2=0.6703; however,the correlation between MPs-CLEIA and ECLIA (R2=0.9582) was obviously better than that between colorimetric ELISA and ECLIA (R2=0.6866).

Glypican-3 (GPC3) is reported as a great promising tumor marker for hepatocellular carcinoma (HCC) diagnosis.Highly sensitive and accurate analysis of serum GPC3 (sGPC3),in combination with or instead of traditional HCC marker alpha-fetoprotein (AFP),is essential for early diagnosis of HCC.Biomaterial-functionalized magnetic particles have been utilized as solid supports with good biological compatibility for sensitive immunoassay.Here,the magnetic nanoparticles (MnPs) and magnetic microparticles (MmPs) with carboxyl groups were further modified with streptavidin,and applied for the development of chemiluminescence enzyme immunoassay (CLEIA).After comparing between MnPs- and MmPs-based CLEIA,MnPs-based CLEIA was proved to be a better method with less assay time,greater sensitivity,better linearity and longer chemiluminescence platform.MnPs-based CLEIA was applied for detection of sGPC3 in normal liver,hepatoeirrhosis,secondary liver cancer and HCC serum samples.The results indicated that sGPC3 was effective in diagnosis of HCC with high performance.

9.Evaluation of soft tissue profile in 20 adults with unoperated isolated cleft palate

Objective To evaluate the soft tissue profile in adults with unoperated isolated cleft palate. Methods Twenty patients older than 16 years(median age,18.4) with unoperated isolated cleft palate were enrolled.Soft tissue lateral cephalograms were traced and compared with those of cleft operated patients and class-Ⅰocclusion individuals.The results of various measurements were analyzed with ANOVA by SPSS11.5. Results In adults with unoperated isolated cleft palate,there was restriction at the top of the upper lip,and the antelabrum of upper lip and the lower lip were found normal. Conclusion The soft tissue profile in adults with unoperated isolated cleft palate exhibits almost the same characteristics as the corresponding hard tissue,and there is covering over effect of the soft tissue to some extent.
10.Study on correlation between homocysteine and diabetes mellitus

Objective To explore the correlation between homocysteine(Hcy),lipids amd type 2 diabetes mellitus (T2DM). Methods The laboratory test results of 533 cases of patients with T2DM(T2DM group)and 362 cases of healthy individuals (healthy control group)were retrospectively analyzed.Results The serum levels of Hcy and rates of abnormal serum cholesterol (TC),triacylglycerol(TG),low density lipoprotein cholesterol(LDL-C),high density lipoprotein cholesterol(HDL-C)levels in the T2DM group were higher than those in the healthy control group,had statistically significant differences(P <0.05).Conclusion The high serum level of Hcy and abnormal lipid metabolism are correlated with T2DM,which might be risk factors of diabetes mel-litus and cardiovascular disease.
