1.Comparison of relative blood volume changes in dialysis-symptomatic hypotension and dialysis-refractory hypertension during hemodialysis

Objective To investigate and compare relative blood volume (RBV) changes in dialysis-symptomatic hypotension and dialysis-refractory hypertension during hemodialysis. Methods Fifteen patients with dialysis-symptomatic hypotension (SH group) and thirteen patients with dialysis refractory hypertension (RH group) on chronic haemodialysis were enrolled in this study. RBV, blood pressure, heart rate and ultrafiltration volume (UV) were measured before hemodialysis and at 1 hour intervals during hemodialysis. The culture conditions of Saccharomyces cerevisiae sp.strain by 1.1 b were optimized for the production of D-(-)-mandelate dehydrogenase which is useful for the asymmetric bioreduction of benzoylformate to form D-(-)-mandelate.The optimum medium(per liter)consistes of 60 g peptone,30 g maltose, 0.5 g MgSO_4,0.01 g ZnSO_4,1.0 g KCl.After optimization of the culture medium,the enzyme production in shake flasks is enhanced from 2.56 to 20.21 U/L.The optimum fermentation conditions were determined as follows:medium volume 100 mL(i.e.,40%for a 250-mL shake flask),pH 6.5,inoculum size 10%,temperature 30℃,and cultivation time 25 h.
3.Progress on the measurement of corneal biomechanical properties

Cornea is the major refractive components of the eye. As a viscoelastic tissue, cornea exhibits complicated biomechanical properties: non - linear elasticity, anisotropy and viscoelasticity. The biomechanical properties play an important role in keeping the normal structureand function. Changes in biomechanical properties are always earlier than the clinical symptoms. So quantitative measurement of the biomechanical properties benefits the early diagnosis and treatment of diseases. Different methods to measure the biomechanical properties of cornea were reviewed in detail, including classic ex vivo destructive tests, commercially available in vivo measuring methods and other emerging methods with the potential for clinical application but not validated for in vivo measurement. The operating principles, advantages as well as limitations of these methods were also described.
6.Regulation of rhIFN-? and rhIL-2 on vascular endothelia l cell in vitro

AIM: To study the effects of rhIFN-? and r hI L-2 on human umbilical vein endothelial cell (HUVEC) proliferation, migration, cell cycle and vascular endothelial growth factor (VEGF). METHODS: Cultured HUVECs were used as model to determine the cel l proliferation with MTT method. Cell cycle was analyzed by cytometry. The cell migration was investigated by agarose scraping method and vascular endothelial g rowth factor (VEGF) content in supernatant of cultured HUVEC was determined by e nzyme-linked immunosorbent assay (ELISA). RESULTS: The growth and migrating number of endothelial cells in rhIFN-? group were 0.199?0.009 and 75.750?23.330, in rhIL-2 group was 0 .217?0.005 and 49.250?8.140, and in combined group was 0.183?0.080 and 40.500?17.230, respectively. In comparison with control group (0.248?0.005 and 160.500?13.220), the effects showed more significant (all P0.05). CONCLUSIONS: rhIFN-? and rhIL-2 show inhibitory effect on vasc ular endothelial cell proliferation, migration and DNA synthesis. When used in c ombination, synergistic effect of rhIFN-? and rhIL-2 is observed, suggesting th at these cytokines play an important role in angiogenesis diseases.
7.Biological status and L1 protein expression of human papillomavirus 16 in patients with cervical lesions

Objective To evaluate the relationship of biological status of HPV 16 and expression of L1 protein with the degree of cervical tumorigenesis.Methods Sixty-one patients with cervical lessions were enrolled and divided into five groups according to pathology of cervical lesions,including chronic cervicitis(n = 27),cervical intraepithelial neoplasia(CIN) Ⅰ-Ⅱ(n = 10),carcinoma in situ(n = 8),early-stage(n =7)and mid/late-stage(n = 9)of cervical carcinoma.HPV type and its biological status were detected by PCR amplification,and L1 protein in HPV 16 positive tissues was detected by Western blot.Kruskal-Wallis was used to compare between the groups,and Pearson correlation analysis was used to evaluate the relationship of HPV biological status and L1 protain expression with cervical lesions.ResultsFifty-three out of 61 patients with cervical lesions were detected with HPV positive(86.9%).All 25 patients with chronic cervicitis were in HPV free mode; in CIN Ⅰ-Ⅱ group,6 patients were in free and 2 in integral mode; in carcinoma in situ group,1 patient in free,3 in mixed and 3 in integral mode; in the early-stage cervical cancer group,2 patients in mixed and 4 in integral mode; in mid/late-stage cancer group,1 patient in mixed and 6 in integral mode.A strong positive correlation was found between the HPV in integral mode and the severity of cervical lesions(r = 0.705,P <0.01).The expression of L1 protein was negatively correlated with the aggravation of cervical lesions in HPV 16-positive patients(r = -0.755,P <0.01).Conclusion The integral mode of HPV16 and low expression of L1 protein may have predictive value for the severity of cervical lesions.
8.Correlation study between lupus nephritis patients of rheumatism syndrome and SLEDAI--a clinical study.

OBJECTIVETo explore lupus nephritis (LN) patients' monocyte chemotactic protein 1 (MCP-1) and urinary IP-10 (ulP-10) levels, the correlation between each clinical activity index and rheumatism syndrome, thereby proving objective evidence for microscopic typing of rheumatism syndrome.
METHODSTotally 60 LN patients were assigned to the rheumatism group (31 cases) and the non-rheumatism group (29 cases). Besides, 20 healthy volunteers were recruited as the normal control group. Clinical data and renal pathology were collected, and urinary levels of MCP-1 and IP-10 detected by ELISA. The correlation between rheumatism syndrome and each activity index as well as manifestations of clinical activities was comprehensively analyzed. Results (1) Patients in the rheumatism group were more liable to occur fever, serositis, edema, and hypertension (P<0.05). (2) Compared with the non-rheumatism group, patients in the rheumatism group exhibited much higher levels of 24 h protein quantification and blood urea nitrogen, higher levels of uMCP-1 and ulP-10. Microscopic hematuria, anti-ds-DNA, anti-Sm, the positive rate of AnuA, scores of SLEDAI and BILAG were higher in the rheumatism group than in the non-rheumatism group (P<0.05). Levels of plasma albumin and complement C3 were lower in the rheumatism group than in the non-rheumatism group (P<0.05). (3) The average activity index (AI) of the renal pathology was higher in the rheumatism group than in the non-rheumatism group. The most frequent pathological type of rheumatism group was type IV of LN.
CONCLUSIONSMore severe renal damage and immune abnormality occurred in LN patients of rheumatism syndrome. Rheumatism syndrome is closely correlated to clinical activity indices.
