Aim The kinetics of nicotinic acetylcholine receptor (N-AChR) was stadied. MethodsThe saturation experiments of Torpedo electric organ N-AChR were taken with125I-?-BuTX. The results were calculated by Scatfit program. Experiments were thenperformed to study the kinetics of dissociation of ?-BuTX and nicotine from N-AChR,respectively. When their binding reached saturation, an excess of 1000 times ?-BuTX ornicotine was added. The saturation binding properties of the N-AChR extracted fromleg muscles of the 13th day chick embroys and optic lobes of the 20th day chick em-broys were studied. The results were calculated by Scatfit program. The othe experi-ments were taken to observe the competition between mcotine and 125I-?-BuTX forbinding with Torpedo, optic lobe and skeletal muscle of the chick N-AChRs. ResultsThe saturation experiments of Torpedo electric organ N-AChR resulted in a Scatchardplot of hyperbola which responded to the model of two kinds of receptor with on eli-gand.The difference of and B max between high affinity binding site and low affnitybineing site was significant. The dissaciation experiment showed that the fast dissocia-tion rate of tow ligands was 500 times more than that of the slow dissociation rate. Thisresult suggested that there may be two subtype N-AChRs.The saturation bindingproperties of N-AChR of leg muscles and optic lobe of the chick embroys revealed aScatchard plot of two kinds of N-AChR indicating a single type of site. The bindingaffinity of receptor of optic lobe was 100 times more than that of muscles. In competi-tion for Torpedo receptor by nicotine and 125I-?-BuTX, the values of IC50 were different:which suggested that two kinds of receptor sites were existent. In competition ofNicotine and 125I-?-BuTX for optic lobe and skeletal muscles of the chick N-AChR, thevalue IC50 of skeletal muscles N-AChR was 7. 7 times higher than that of optic lobe. Itindicated that two kinds of N-AChR subtype existed in the optic lobe and in theskeletal muscles of chick respectively. Conclusion N-AChR of Torpedo electric organcontains two kinds of subtype receptors. N-AChR of optic lobe of chick embroys is onesubtype receptor, and N-AChR of skeletal muscles of chick embroy is another subtypereceptor.

Availability of appropriate drugs for children has been a bottleneck both at home and abroad for years. As a result, drugs for adults that are market available emerge a choice of convenience for children's treatment instead. For unique physiological and psychological characteristics of children, these drugs must be dispensed on individual basis before usage to children in question.Such drugs play a key role in children's treatment, and incur many problems on the other hand in their use and practice, such as drug stability, dosage accuracy, potential risks, as well as legitimacy in drug dispensing and use.A safe, reasonable and normalized use of such drugs calls for an orchestrated effort among pharmacists, trade associations and drug regulators as well.

Objective To establish a gene diagnosis assay for spinal muscular atrophy(SMA) in children. Method Analysis of the survival motor neuron (SMN) gene in 19 SMA patients and in 21 normal controls were performed by using polymerase chain reaction - fragment length polymorphism (PCR - RFLP) method. Result Deletion of exon 7and 8 in SMNt gene were found in all 19 SMA patients, while no such changes were found in normal controls. Conclusion The SMNt gene exon 7 and 8 examine can be applied to SMA gene diagnosis, and the PCR- RFLP method have higher sensitivity and particularity to the SMA diagnosis.

Objective To evaluate the role of silent information regulator fac tor 2-related enzyme 1 (SIRT1)/Forkhead Box O3 (FoxO3a) signaling pathway in berberine pretreatment-induced reduction of hypoxia/reoxygenation (H/R)-caused injury to hepatic parenchymnal cells. Methods Hepatic parenchymal cells obtained from AML12 mice were cultured and seeded in 6-well plates (2 ml/well) and in 96-well plates (200 μl/well) at the density of l×l06 cells/ml.The cells were divided into 4 groups (n=36 each)using a randomn number table:control group (group C),group H/R,berberine pretreatment group (group BP) and SIRT1-siRNA group (group SS).The cells were cultured in normal culture atmosphere (5% CO2-21% O2-74% N2) in group C.In H/R,BP and SS groups, the cells were exposed to hypoxic air (5% CO2-1% O2-94% N2) for 12 h,followed by 6 h reoxygenation in normal culture atmosphere (5% CO2-21% O2-74% N2).In group SS,small interference RNA targeting SIRT1 (SIRT1-siRNA) was added to the culture medium at 24 h prior to hypoxia.Berberine (final concentration 5 μmol/L) was added at 2 h prior to hypoxia in BP and SS groups. At the end of reoxygenation, the cell viability was measured by methyl thiazolyl tetrazolium assay, the malondialdehyde (MDA) content and superoxide dismutase (SOD) activity were determined using enzyme-linked immunosorbent assay, cell apoptosis was detected by flow cytometry, the expression of SIRT1 and FoxO3α was detected by Western blot, and the acetylation of FoxO3α was measnred by using immunoprecipitation.Apoptotic rate was calculated. Results Compared with group C,the cell viability was significantly decreased, the MDA content was increased, the SOD activity was decreased, apoptotic rate was increased, the expression of SIRT1 and ratio of FoxO3α expression in nucleus/in cytoplasma were increased, and the acetylation of FoxO3α in the nucleus was increased in H/ R,BP and SS groups (P< 0.05).Compared with group H/R,the cell viability was significantly increased, the MDA content was decreased, the SOD activity was increased, apoptotic rate was decreased, the expression of SIRT1 and ratio of FoxO3α expression in nucleus/in cytoplasma were increased, and the acetylation of FoxO3α in the nucleus was increased in group BP (P<0.05).Compared with group BP,the cell viability was significantly decreased, the MDA content was increased, the SOD activity was decreased, apoptotic rate was increased, the expression of SIRT1 and ratio of FoxO3α expression in nucleus/in cytoplasma were decreased, and the acetylation of FoxO3α in the nucleus was decreased in group SS (P<O.05).Conclusion The mechanism by which berberine pretreatment attenuates H/R-caused injury to hepatic parenchymal cells is related to promotion of SIRT1 expression in cells and inhibition of FoxO3α acetylation in the nucleus.

It designs a way that can easily screen high-pyruvate-producing strain. It is a intelligently selected method which can highly improve the efficiency of strain screening. The principle can be described as the following:On the CaCO-3 medium,a transparent ring can be exhibited based on the reaction of PYR produced by the strain and CaCO-3 in the medium for pyruvate-calcium is a kind of soluble substance,it is obviously that the high-pyruvate-producing strain has a bigger dimension of the transparent ring.On the other hand, color reaction between TTC and ADH indicate the enzyme activities which have a proportional relation with color, our object strain is a weak-ADH-enzyme-activities type with a weak metabolic flux from PYR to alcohol. So the white color strain may be the right choice.
