2.Construction of prokaryotic expression vector of mouse CD25 extracellular domain

Objective To construct the prokaryotic expression vector of mouse CD25 extracellular domain and to express it in E coli.Methods Total RNA was isolated from splenocytes of Balb/c mice.The CD25 extracellular domain gene was amplified by RT-PCR and cloned into the PET-32a vector.A positive recombinant,PET-32a-CD25e,was identified by enzyme cleaving and sequencing before its expression in E.coli,and transferred into E.coli BL21(DE3) plysS for its expression.After purification with Ni+ resin and renaturation in vitro,a relative molecular mass(Mr) of the interesting protein was detected by SDS-PAGE and Wes-tern blotting.Effect of the purified interesting protein on the proliferation of splenocytes from T cell vaccine-immunized syngeneic mice was detected by MTT assay.Results The cloned CD25 extracellular domain gene was identified to be functional by sequencing and expression.The purified interesting protein could significantly induce the proliferation and IL-4 secretion of splenocytes from T cell vaccine-immunized mice in vitro.Conclusion Mouse CD25 extracellular domain gene can be successfully expressed in prokaryotic cells with biological activity,which lays a foundation for further relative studies.
3.CT Study of the Internal Occipital Protuberance

Objective To study the development and shape of the occipital protuberance and its clinical significance with normal cranial CT scans.Methods 3000 cases of normal cranial CT images were selected.There were 1500 males and 1500 females,which were divided into nine age groups.The internal occipital protuberances were observed and measured on bone-window.Results The internal occipital protuberance was seen earlist at four-month-old-infant,and growth with age.The shape of the internal occipital protuberance was classified into four types:single-peak-type,plane-protusion-type,tropezoid-type and irregular-type.The single-peak-type was seen most commonly.Big semilunar depression of the internal occipital protuberance could be seen in patient with magna citern arachnoid cyst.Conclusion The height and shape of normal internal occipital protuberance have many diversities.Big semilunar depression of the internal protuberance is a important sign in diagnosis of magna cistern arachnoid cyst.
4. Advance in components with antitumor effect of Panax ginseng and their mechanisms

Panax ginseng C. A. Mey is one of precious traditional Chinese herbal medicine for two thousand years history, due to its various pharmacological effects and wide utilization in the clinical treatment of tumors and other diseases. Presently tumor has become an important factor threatening human health that the antitumor effect of P. ginseng is attracted great attention. In this paper, the effective components of antitumor action by P. ginseng and its molecular mechanism and structure-activity relationship are summarized. Studies have shown that the main effective components of P. ginseng for antitumor effect are ginsenosides and its metabolic products of intestinal bacteria, ginseng polysaccharides, and ginseng polyacetylenes. The functional mechanism of which are clear relatively now and their main mechanisms including induction of cycle arrest, apoptosis and differentiation of tumor cells, enhancement of immunity to tumor cells, inhibition of tumor cell proliferation, invasion and metastasis, etc. And the molecular mechanism is involved in the regulation of many related genes, proteins, proteases, immune cells, cytokines, and signaling pathways, etc. In addition, the active ingredients of P. ginseng exert antitumor effect in a dose-dependent manner, and the different chemical structures of which lead to different antitumor activity. In conclusion, P. ginseng is abundant with antitumor active ingredients, which is expected to provide safe and effective natural medicine and its preparation for clinical treatment of various tumors in the future.
5.Analysis on the effects of health education and promotion on osteoporosis in middle-aged and elderly residents in Jiangsu province

Objective To study the effects of community comprehensive intervention model on osteoporosis, in order to provides a reference for osteoporosis health education work. Methods A multistage stratified random cluster sampling was applied to conduct a survey in Jiangsu province, in accordance with the economic level and geographic location. Nanjing, Nantong and Xuzhou cities were sampled, three urban streets were sampled in every district , respectively. A total of 1 500 40-69 years old community residents were screened, 619 high-risk participants for osteoporosis were arranged into the intervention group, and a total of 590 residents from the same communities were chosen as control group, who were gender- and age-matched with the intervention group. The intervention group residents received health management and comprehensive intervention, after half a year, a self-designed questionnaire was used to investigate the effects. Results Compared with control group, after six months, knowledge of intervention group related to osteoporosis awareness was improved significantly, such as the most serious harm to human body for osteoporosis (χ2=24.27), characteristic of osteoporosis (χ2=11.05), basic supplements to improve bone health (χ2=60.65) , the recommended daily calcium intake to postmenopausal women and the elderly (χ2=26.80), the difference was statistically significant (P<0.05); daily behavior related to diet and exercise of intervention group has improved significantly, such as drinking milk or itsproducts less than 3 days a week (χ2=25.83), eating beans and products for less than 3 days a week (χ2=19.20), drinking coffee for more than three days a week (χ2=7.77), not often taking outdoor activities (χ2=18.55) ,(P<0.05);Diseases and symptoms related to osteoporosis in intervention group was significantly reduced (P<0.001). Conclusions The osteoporosis is one of the most important public health problem for Jiangsu province, the work for osteoporosis prevention and control should be emphasized in community.
6.Develop of a New Wide-range X-ray Intensifying Screen

Objective To display space occupying lesions of spine,mediastina and lungs behind heart and small lesions in bilateral pulmonary fields in the same chest film without changing the exposure doses which were now used for routine chest images.Methods The distribution of current intensifying screens' intensifying materials were changed and reasonable technical processes were given.High-velocity and middle-velocity intensifying materials were placed in center and in two sides of the screen respectively.Results Tests in seven volunteers using the new kind intensifying screens demonstrated that diseases could be better shown than normal intensifying screens in chest images.Conclusion The renovation can provide more and clearer and richer stratifications of image information to help chest X-ray diagnoses.
7.The mechanism of rosiglitazone compound based on network pharmacology.

Applications of network pharmacology are increasingly widespread and methods abound in the field of drug development and pharmacological research. In this study, we choose rosiglitazone compound as the object to predict the targets and to discuss the mechanism based on three kinds of prediction methods of network pharmacology. Comparison of the prediction result has identified that the three kinds of prediction methods had their own characteristics: targets and pathways predicted were not in accordance with each other. However, the calcium signaling pathway could be predicted in the three kinds of methods, which associated with diabetes and cognitive impairment caused by diabetes by bioinformatics analysis. The above conclusion indicates that the calcium signaling pathway is important in signal pathway regulation of rosiglitazone compound, which provides a clue to further explain the mechanism of the compound and also provides a reference for the selection and application of methods of network pharmacology in the actual research.

8.Meta-analysis of the methylenetetrahydrofolate reductase C677T polymorphism and susceptibility to depression

Objective To evaluate the association between methylenetetrahydrofolate reductase (MTHFR) C677T gene polymorphism and depression risk.Methods Available studies containing genotype frequencies of MTHFR C677T were chosen,and pooled odds ratio (OR) with 95% confidence interval (CI) was used to assess the strength of the association.Results Twenty-five eligible studies including 4 048 cases and 12 827 controls were identified.Meta-analysis showed a significant effect in the co-dominant model (T vs.C:OR 0 =1.25; Z=3.82,P=0.000;TTvs.CC:OR1=1.55,Z=3.79,P=0.000 ; except CT vs.CC:OR2=1.13; Z=1.78,P=0.076).Stratified analysis denoted that there was significantly higher risk in depression onset before 60 years old than depression onset after 60 years old.In subgroup analysis stratified by regressive meta-analysis,MTHFR C677T variant was statistically significantly relevant to depression risk in Chinese populations (OR 0 =1.58,Z=3.95,P=0.000; OR 1 =2.55,Z=3.92,P=0.000; OR 2 =1.43,Z=2.72,P=0.007),but not in no-Chinese populations (OR 0 =1.12,Z=2.10,P=0.035 ; OR 1 =1.23,Z=2.23,P=0.025 ; OR 2 =1.06,Z=0.74,P=0.456).Evidentiary strength rated by GRADE showed that 1 study moderate,3 studies low and 7 stdudies very low.Conclusion Our study provides evidence for a hereditary association between the MTHFR C677T variant and depression susceptibility,whereas there is an obvious variety in different regions,ethnicities and onset age.
9.Study on inhibitory effects and mechanism of lipophilic components in Salvia miltiorrhiza on angiogenesis in vitro.

In this study, the human umbilical vein endothelial cell model was used to study the regulating effect of lipophilic components in Salvia miltiorrhiza on angiogenesis, and explore its possible mechanism. The cell model was established to determine the effect of lipophilic components in S. miltiorrhiza on the proliferative activity and migration capacity of endothelial cells. Then the realtime fluorescence quantification PCR technology was applied to detect the changes in the gene expressions of angiogenesis-related cytokines VEGF-A, VEGF-C and MMP-9. The results showed that 5 mg x L(-1) lipophilic components in S. miltiorrhiza could inhibit the proliferation and migration of endothelial cells, and reduce the expression of VEGF-A and MMP-9 genes. It indicated that lipophilic components in S. miltiorrhiza may inhibit the proliferation and migration of endothelial cells by inhibiting the expression of VEGF-A and MMP-9 genes, so as to show the inhibitory effect on angiogenesis.

10.Multiple-electrode radiofrequency ablation via switching system in treating early-stage hepatocellular carcinoma

Objective To retrospectively compare the safety and local efficacy of multiple-electrode switching based radiofrequency ablation (RFA) and the conventional RFA in treating single early-stage hepatocellular carcinoma (HCC).Methods A total of 82 patients with single early-stage HCC received either RFA with a multiple-electrode switching system (n =43) or conventional RFA (n =39) as the first-line treatment.The rate of initial local complete response,major complications and local tumor progression (LTP) were compared between two groups.Results The total ablation time was significantly shorter in the switching-RFA group [(16.7 ± 3.4) mins] than in the conventional RFA group [(29.8 ± 10.4) mins] (P < 0.05).The rate of initial local complete response was 100% (43/43) in the switching-RFA group and 94.9% (37/39) in the conventional RFA group (P >0.05).After a mean follow-up period of (26.4 ± 21.8)months (ranging 3.0-91.6 months),the rates of LTP in the switching-RFA group and conventional RFA group were 16.3% (7/43) and 17.9% (7/39),respectively.The LTP rates in two groups were 16.1% versus 11.2% atyear1,and20.5% versus 20.6% at year2 (P=0.666).Conclusions The multiple-electrode switching based RFA is safe and effective with shorter ablation time in treating single early-stage hepatocellular carcinoma.
