1.EGFR RNAi inhibits bleomycin induced pulmonary fibrosis in mice

ObjectiveTo investigate the effect of down-regulating epidermal growth factor receptor (EGFR) expression on Bleomycin induced pulmonary fibrosis in mice.MethodsForty 4 ~6 week aged C57BL/c male mice were randomly divided into control,bleomycin,bleomycin plus EGFR RNAi groups and RNAi negative control group.Bleomycin group were treated with bleomycin (3 mg/kg) by endotracheal injection on day 0,control group were treated with PBS.And bleomycin plus EGFR RNAi group were received EGRF siRNA plus bleomycin intratracheal administration.RNAi negative control group received negative EGRF siRNA plus bleomycin intratracheal administration.Mice were sacrificed 10 days after the treatments.Hydroxyproline (HYP) assay was performed in the lung tissue.The lung tissue slides were examined pathologically with H.E.staining,and EGFR mRNA expression was detected by reverse transcriptasepolymerase chain reaction (RT-PCR) technique.Western blot were performed to identify the protein level.of total EGFR and phosphorylated EGFR.ResultsHistological examination of lung specimens demonstrated that EGFR siRNA administration lessened lung fibrosis induced by bleomycin and significantly reduced HYP content (543.00±25.89 vs 900.73±31.77,P<0.01).EGFR mRNA (0.31±0.05 vs 0.75±0.08,P <0.01) and protein expression(1.53±0.47 vs 2.56±0.37,P <0.01) in EGFR siRNA-treated mice was significantly decreased.The expression of phosphorylation of EGFR protein (1.78±0.35 vs 2.84±0.51,P <0.01) and EGFR protein in RNAi group was less than in bleomycin group.Conclusions EGFR RNAi reduced the BLM-induced lung fibrosis by inhabiting EGFR activation.

3.Clinical significance of thrombospondin-2 expression in gastric car-cinoma

Objective:To investigate the clinical significance of thrombospondin-2 (THBS2) mRNA expression in gastric carcino-ma and its relationships with clinicopathologic features, microvessel density (MVD), and matrix metalloproteinase-2 (MMP-2). Meth-ods:THBS2 mRNA expression was detected in 82 cases of gastric carcinomas and adjacent tissues using real-time quantitative fluores-cence polymerase chain reaction. The correlation of this expression with clinicopathologic features was also analyzed. Cluster of differ-entiation 34 (CD34) and MMP-2 protein expression was examined using an immunohistochemical Elivision method. MVD was deter-mined based on CD34-positive tubular structures. Results:The THBS2 mRNA expression level was significantly higher in the gastric carcinomas than in paraneoplastic tissues (P=0.002). The expression was associated with the depth of tumor invasion, MVD, and MMP-2 (P=0.02, r=0.35, P<0.01, and P=0.004, respectively) but not with patient gender, patient age, tumor size, histological type, and lymph node metastasis (P=0.53, P=0.53, P=0.21, P=0.84, and P=0.96, respectively). Conclusions: THBS2 may be significantly in-volved in the occurrence and progression of gastric carcinoma. The effects of THBS2 on gastric tumor growth and metastasis can be monitored by controlling the MMP-2 expression in the carcinoma. However, the specific functions and underlying mechanisms of TH-BS2 require further investigation.
4.Application of navigation-assited microsurgery resection of posterior fossa solid hemangioblastoma

objective To explore the application of navigation-assited microsurgery resection of posterior fossa solid hemangioblastoma.Methods The data of 16 patients with posterior fossa solid hemangioblastoma treated by navigation-assited microsurgery were analyzed retrospectively,compared with 19 patiems treated by microsurgery who suffered the same disease in the similar location(control group).Results The tumors were totally resected successfully acompanied by minimal blood loss and shortening of the operation time,compared with the control group.Manipulation and removal of these tumors were actually easier,in addition,adjacent structures can avoid damage.Conclusions Navigation-assited microsurgery is useful technique to surgical resection of fossa solid hemangioblastoma by significantly reducing blood loss at the time of surgery and operation procedure time,avoiding damage to the adjacent structures,and reducing the surgery complications and mortality.
5.Effects of prevention and treatment on atrophic gastritis in rats with Zengshengxiao Capsules

Objective:To study the protective and therapeutic effects of Zengshengxiao Capsules (ZSX) on atrophic gastritis in rats. Methods: The model was established by methods of active immunity and gastrogavage with bile and hot water. The method of acetic acid writhing was used. Results: (ZSX) had actions of promoting the blood flow of gastric mucosa, increasing in pH, inhibiting inflammatory cell infiltration, glandular atrophia and atypical hyperplasia. It also had action of alleviating pain. There were no toxic and side effects. Conclusion: (ZSX) had remarkably preventive and therapeutic effects on atrophic gastritis of rats.
6.Effects of Acorustatarinowii Schott on chronic restraint stress mice and its mechanisms

Objective To evaluate the effects of Acorustatarinowii Schott(ATS)on chronic restraint stress mice and its possible mechanisms. Methods Chronic stressed mice underwent a repeated 8h/d constraint in a specific behavior blocker ,however ,only CSA mice received once gavages of 4.5 g/(kg · d)ATS decoction for con-secutive 28 days. Morris water maze(MWM)task was conducted for evaluating learning and memory of the mice. ELISA was used to examine the levels of plasma corticosteroid. Results ATS dramatically ameliorated cognitive impairments and decreased serum corticosteroid level in the stressed mice(P<0.01). Conclusion ATS improves cognitive deficits provoked by chronic stress in mice ,which may attribute to decreasing plasma corticosteroid levels.
7.Nursing care in ultra-high pressure balloons angioplasty treatment of arteriovenous fistula stenosis in hemodialysis patients

Objective To investigate the nursing care in ultra-high pressure balloon dilatation for treating in patients with hemodialysis patients.Methods The patients with arteriovenous fistula were treated with ultra-high pressure balloon dilatation.Under fluoroscopy guidance the ultra high pressure balloon was used to dilate the stenosis.Preoperative preparation included psychological and routine care.Results Blood flow in all the 14 patients with hemodialysis was ≥ 250 ml/min,mean (270.00±27.33) ml / min.All patients were successfully completed the treatment,there were no angiorrhexis,bleeding,infection and embolism.Conclusion Careful nursing can promote the rehabilitation of patients,improve the effect of postoperative treatment.

Objective To investigate the suitable light intensity of plant growth of Dendrobium Sw.through study on photosynthetic characteristics.Methods The photosynthesis curves of light intensity, chlorophyll fluorescence parameter of different light intensity and inductivity of non-photochemical quenching of chlorophyll fluorescence in 3.0?104 lx on three species of Dendrobium Sw.in Huoshan (D.huo- shanens, D.candium, and D.moniliforme) were determined.Results Three species of Dendrobium Sw.were shadow plants for their response on light intensity, light saturation point, light compensate point, net photosynthesis rate, and apparent quantum efficiency are lower during photosynthesis.The photoinhibitory effect was formed on Dendrobium Sw.leaf when they grew in more than 4.0?104 lx.Conclusion Although three species of Dendrobium Sw.in Huoshan grow in (0.2-2.0)?104 lx normally, they are suitable to grow in (2.5-3.0)?104 lx instead of growing more than 4.0?104 lx.

Objective To explore the differences in clinical features between the latent autoimmune diabetes in adults (LADA) and type 2 diabetes mellitus (T2DM) in the elderly.Methods Clinical features of 41 cases with LADA and 49 cases with T2DM were retrospectively analyzed and compared.Results (1) Body mass index (BMI),fasting and postprandial levels of cpeptide,triglyceride (TG) and serum albumin were lower in LADA patients than in T2DM patients.(2) The levels of fasting and postprandial blood glucose were higher in LADA patients than in T2DM patients.(3) There were no statistically significant differences in the disease course,blood pressure (BP),and the functions of liver and kidney between LADA and T2DM patients.Conclusions As compared with T2DM patients,LADA patients have higher blood glucose level,lower BMI and TG level,and poorer function of islet B cell.But there is no significant difference in the chronic complications between two groups.
10.Improvement of the the thermostability of Penicillium expansum lipase by mutagenesis the random mutant ep8 at K55R.

In order to improve the thermostability of the Penicillium expansum Lipase (PEL), the lipase encoding genes was mutated by site-directed mutagenesis. A recombinant vector pAO815-ep8-K55R which contain double mutant genes was constructed by overlap extension PCR using the cDNA of a random-mutant lipase ep8 (a single site mutant) as the template and two special primers were used to generate another mutation site K55R. The recombinant vector was transformed into Pichia pastoris GS115 by electroporation and the recombinant mutant GS-pAO815-ep8- K55R can secret double-mutant lipase PEL-ep8-K55R-GS into the medium when it was induced by Methanol. The yield of the double-mutant lipase is 508 u/mL, which is 81% that of the wild type lipase PEL-GS (627 u/mL) and 55% that of random-mutant PEL-ep8-GS (924 u/mL). The specific activity of double-mutant lipase is 2309.1 u/mg, which is similar to random-mutant lipase PEL-ep8-GS and the wild type lipase PEL-GS. The optimum temperature of the double-mutant lipase is same with the wild type lipase PEL-GS and random-mutant lipase PEL-ep8-GS. While the Tm of the double-mutant lipase is 41.0 degrees C, 2.3 degrees C higher than the wild type lipase PEL-GS and 0.8% higher than the random-mutant lipase PEL-ep8-GS, indicating that the double-mutant lipase PEL-ep8-K55R-GS has higher thermostability.
