Objective To investigate the polymorphism of glutathione S-transferease P1 gene(GSTP1) and the association between the mutation and susceptibility in childhood asthma.Methods The distribute frequency of Ile105/Ile105,Ile105/Val105 and Val105/Val105 ge-notype in GSTP1 of 51 children with asthmatic and 40 normal children were studied with polymerase chain reaction-restriction tragment length polymorphism(PCR-RFLP).Results The frequencies of Ile/Ile Ile/Val,Val/Val genotype were 66.7%,27.4% and 5.9%,the frequencies of Ile,Val allele were 80.4% and 19.6% in the asthmatic group.But the frequencies of Ile/Ile,Ile/Val,Val/Val genotype were 90.0%,7.5% and 2.5%,the frequencies of Ile,Val allele were 93.8%,6.2% in control group.The frequencies Ile/Val,Val/Val genotype and Val allele in asthmatic group were more than that in control group.A significant difference was found in the frequency distribution of GSTP1 genotypes between two groups(?2=6.947 P
2.Establishment of pharmacodynamic model of sevoflurane in pediatric patients: bispectral index

Objective To establish the pharmacodynamic model of sevoflurane with bispectral index (BIS) as the effective index in pediatric patients.Methods Thirteen ASA physical status Ⅰ or Ⅱ pediatric patients,aged 4-9 yr,weighing 12-39 kg,undergoing non-cardiac surgery,were selected in the study.The pediatric patients sequentially inhaled 1%,5 % and 1% sevoflurane via a face mask and each concentration was inhaled for 15 min.BIS value,HR,BP and SpO2 were automatically recorded every 10 s.Based on nonlinear mixed effect modeling,the population pharmacodynamic model of sevoflurane was established using NONMEM software.The effect of age on the pharmacodynamic parameters was evaluated using a stepwise forward addition then backward elimination modeling approach.The standard for model improvement was defined as a decrease in the value of the objective function by more than 3.84.Results Twelve pediatric patients,aged 4.0-8.5 yr,weighing 12.8-38.0 kg,with body height of 92-135 cm,were enrolled in this study and the data which were enrolled comprised 2964 effective concentration-time-BIS points.The model was not improved significantly with any covariates (age,body height,and body weight) introduced (P > 0.05).The estimated parameters of the final pharmacodynamic model of sevoflurane were as follows:ke0 =O.516/min ; EC50 (BIS50) =2.11% ; γ =2.46 ; E0 =74.6 ; EMAx =11.2.Conclusion The pharmacodynamic model of sevoflurane is successfully established with BIS as the effective index in pediatric patients,and the analysis for each parameter of the model indicates that the sensitivity to sevoflurane is lower,but the blood-brain equilibration time of the drug is shorter and the onset and recovery are faster in children than in adults.
3.In vitro hematopoietic stem cells from embryonic stem cells reconstruct hematopoiesis in mice

AIM:To investigate the potential of hematopoietic stem cells(HSCs)derived from mouse embryonic stem cells(ESCs)to reconstruct hematopoiesis in vivo.METHODS:Using a three-step method,a mice embryonic stem cell line,E14.1 was induced into hematopoietic stem cells.The cell markers with CD34+/Sca-1+ were identified by flow cytometry analysis,then HSCs(1?109 cells/L)from third-step were injected into SCID mice for observing teratoma formation.To validate function of HSCs,colonogenic cell assay was conducted and the hematopoiesis in lethally irradiated mice was reconstituted.RESULTS:The method of three-step differentiation,combined to use more hematopoietic stimulating factor promoted the E14.1 cell differentiation into HSCs with highest percent of CD34+/Sca-1+ cells(as high as 58.64%?4.20%)with more CFU-E,CFU-GM and CFU-GEMM populations.The cells showed the character of hematopoietic progenitors by Wright-Giemsa staining.Positive selected CD34+/Sca-1+ cells by magnetic sorting from third-step differentiation were transplanted into 7 lethally irradiated female mice while predominant hematopoietic reconstitution were observed in 10 d after transplantation,with 71.4%(5/7)successful engraftment rate.Three recipients showed that the cell population of the peripheral blood leukocytes,red blood cells and hemoglobin approached to normal index at 40 d after transplantation,but followed relative slow renew in platelet count.Survival rate in transplant group was 43%,compared to 100% mortality in control mice.Karyotyping assays confirmed the female mice with XY.CONCLUSION:The three-step differentiation and the culture conditions described here support the differentiation of mouse ESCs into HSCs.HSCs derived from mouse ESCs can reconstruct hematopoiesis.

5.Study on quality assurance system of grade 3 laminar operation room

ObjectiveTo explore the features of the cleanliness change with time during the use of laminar operating rooms in order to establish reliable quality assurance system. MethodsTwo grade 3 lami-nar operation rooms were chosen to be tested after one and a half year and two and a half years, laser counter was used to detect the static particulates after cleaning for 30 minutes and 45 minutes. ResultsAfter the laminar operation room was used for one and a half years the amount of 0.5 μm particulates in indoor air was less than 350 per liter, and after two and a half years it's above 350 per liter after cleaning for 30 minutes, an obvious difference than the previous one. The anaount of 1 μm and 2 pan particulates also increase obviously, but ff cleaning for 45 minutes the amount of 0.5 μm particulates in indoor air will fulfill the qualification. Al-though the amount of 0.5 μm, 1 pan and 2 μm particulates after 2 years using was higher than the one and a half yesrs'using after cleaning for 45 minutes, but the difference had no statistic meaning. CondusionsThe elesnliness of laminar operation rooms will drop as time prolonged, Amount of particulates in indoor air will in-crease too but the cleanliness can be ensured by extending cleaning time. After the quality assurance sys-tem was established the amount of smile particulates is still within the standard level

Objective To investigated the clinical value at pathogenetie features of spontaneous hemopneu-mothorax in aged people and therapeutic investigation of VATS.Methods 32 aged patients with spontaneous hemo-pneumothorax who underwent VATS.all used small chest wound asssociated with video-assisted thoraeoscopy.Results All procedures were carried OUt salty,recruitments were satisfactary,there were no operative death,all patients cured and had no sepuetae.There were 2 cases had gas leak last for a long time after operation,there Were 7 eBses had reexpansion pulmonary edema,arrhythmias,pulmonary infection.Conclusion VATS is a safe and effeetlve technique with smaller wound and shorter hospital stay for the treatments of spontaneous hemopneumothorax in aged people.
7.Association between skin prick test of dust mites and serum specific IgE in diagnosis of mite allergy

Objective To investigate the association between the results of Dermatophagoides pteronyssinus ( Der p) and Dermatophagoides farinae ( Der f) allergen SPT and serum sIgE in diagnosis of IgE-mediated mite allergy.Methods A total of 166 allergic outpatients with positive expression of serum sIgE of Der p and Der f diagnosed in Beijing Tongren Hospital,the General Hospital of Shenyang Military Region and Guangzhou Institute of Respiratory Disease were enrolled in the study.The SPT was performed by injecting liquid containing the allergen of Der p and Der f.One hundred ninety healthy subjects with negative serum sIgE of Der p and Der f were assigned as controls according to paired design ratio,1 to 1.The sensitivity and specificity of SPT used Der p and Der f allergen were evaluated in the diagnosis of mite allergy.The association between the results of SPT and serum sIgE was analyzed by linear regression method.Results In the allergic group,the positive rates of SPT for Der p and Der f were 98.80% ( 164/166) and 95.18% (158/166) respectively.In the control group,the positive rate of STP for Der p and Der f were 2.11% (4/190) and 1.58% (3/190) respectively.The SPT results of Der p and Der f showed significant statistical difference between healthy subjects and allergic patients (x2 =285.5,386.3;P<0.01 ).The sensitivity and specificity of the SPT were 98.80% (164/166) and 97.89% (186/190) for Der p and 95.18% (158/166) and 98.42% (187/190) for Der f,respectively.There was a significant association between SPT and sIgE with linear correlation analysis( r =0.737,0.708;P < 0.O1 ).Conclusions Both SPT of Der p and Der f is highly sensitive and specific.At the same time,SPT has a significant positive correlation with sIgE.It can be used as an accurate,convenient and fast test in diagnosis of mite allergic diseases.
8.Long-term efficacy and safety of fentanyl transdermal system

BACKGROUND:Fentanyl transdermal system has analgesic effect similar to oral sustained-release morphine and has been widely used in advanced cancer pain management in several years. However, recent literatures about some serious adverse events associated with fentanyl transdermal system have been published, and the long-term safety of fentanyl transdermal system treatment is stil chalenged. OBJECTIVE:To observe the long-term clinical efficacy and safety of fentanyl transdermal system for pain management in patients with advanced cancer. METHODS:A total of 309 patients with advanced cancer pain were enroled, including 166 females and 143 males. The age ranged from 26-72 years old. Patients received oral sustained-release morphine for 2 weeks, and then were subject to fentanyl transdermal system for pain management at the 3rd week until the 12th week. A prospective study with self-contrast method was conducted to compare the analgesic effects of these two drugs, as wel as patient's acceptability, adverse events and toxicity in the administration course. RESULTS AND CONCLUSION: Stable pain relief was harvested throughout the oral administration of both sustained-release morphine and fentanyl transdermal system therapy. During the use of oral sustained-release morphine, adverse reactions appearing in sequence were constipation, nausea, fatigue and anorexia. After converting to fentanyl transdermal system, the symptoms of constipation (χ2=5.22,P=0.02) and nausea (χ2=4.38, P=0.04) significantly reduced, and vomiting was abated but showed no significant difference (χ2=2.7,P=0.10). 2.3% of patients had skin reactions to the patches, and regressed after replacing the patch area. Skin reactions were aleviated at 2-10 weeks after oral administration of fentanyl transdermal system. Some uncommon adverse events including headache, diarrhea, dyspnoea, excessive sweating or other symptoms often occurred at the time of the initial dosage increase. Preference or strong preference for fentanyl transdermal system in comparison to previous oral sustained-release morphine was reported by 91% of patients. These results demonstrate that fentanyl transdermal patches can provide stable pain relief for advanced cancer patients with good acceptability, and mitigate the incidence of adverse events due to oral drugs.
9.Treatment of tracheobronchial tuberculosis by bronchoscopic intervention

Objective To discuss the clinical value of electronic bronchoscopic intervention in treatment of tracheobronchial tuberculosis. Methods Clinical features of 45 patients with tracheal and bronchial tuberculosis which were confirmed by electronic bronchoscope and treated by bronchoscopic intervention were retrospectively analyzed from January 2007 to December 2013 in our hospital. Results The efficiency of bronchoscopic intervention is 88.9%, of which 28 cases achieved a significant effect (accounting for 62.2%). Conclusion Electronic bronchoscopic intervention is a preferred way in treatment of tracheobronchial tuberculosis.
10.Study on antitumor activity of allotri-tridecyl diethylamine in vitro and in vitro

AIM: To investigate antitumor effect of allotri-tridecyl diethylamine (D-108) in vivo and in vitro. METHODS: The cytotoxic effects of D-108 on various tumor cell lines, human gingival fibroblast and marrow stromal cell cultured in vitro were determined with trypan blue dye exclusion test and MTT method. The acute toxicity of mice by administration of D-108 was evaluated by Bliss method. At a tolerable dose level, D-108 was administrated to treat transplanted solid tumor U14, and tumor weight inhibition was observed. Apoptosis morphological transformation of HL 60 cell induced by D-108 was detected by the Giemsa staining. RESULTS: The cytotoxic effects in vitro of D-108 on various tumor cell lines (IC_ 50 : 0.22 to 2.19 mg?L~ -1 ) were more powerful than both human gingival fibroblast and marrow stromal cell (IC_ 50 : 5.55 and 3.57 mg?L~ -1 ). LD_ 50 of D-108 was 36.49 mg?kg~ -1 (mice, i.g.). D-108 inhibited in vivo growth of implanted solid tumor U14 of mice effectually. The inhibition rate of tumor weight of D-108 (100 mg?kg~ -1 ?d~ -1 i.g.) was 45.27 %. HL 60 cell appearanced typical apoptosis morphological transformation induced by D-108. CONCLUSION: D-108 had obvious antitumor activity in vivo and in vitro and little toxicity. D-108 could induce the apoptosis of HL 60 cell.
