Objective:To explore the technique of two-flap in ear reconstruction.Method:Quantitative tissue expansions were used in the mastoid area in the first stage.After the final injection,there was 1 month of sustaining time.Expanded skin flap and unexpanded fascia flap were designed in the second stage,so thetwo-flaptechnique was used in the ear reconstruction.From January 2004 to December 2008,1 427 patients of microtia were treated using two-flap technque.Result:The expanded skin flap could show the fine structures of the reconstructed ears.The reconstructed ears had vivid cranioauricular angle after using the unexpanded fascia flap.Conclusion: Two-flap method was easily manipulated and the complications were rare.The reconstructed ears had lucid and three-dimensional contour.
2.Resource and pharmacognostical study on Aralia echinocaulis.

OBJECTIVETo provide fundamental information for its exploiting. Aralia echinocaulis by the resource and identification study on.
METHODResource survey and various identification were carried out.
RESULTThe county level distribution and ecological environment of A. echinocaulis were initially observed. It mainly distributed in the mountainous areas of the Yangtze River basin and the south, and was usually used as folk drug. This study also displayed its morphological, microscopic and chemophysical identification features.
CONCLUSIONThe morphological features of original plant and crude drug, and the anatomical and chemophysical characteristics of A. echinocaulis are of identification value, and the species are also of greater development and utilization potentiality, but the resource does not support the sustainable utilization. Therefore, artificial propagation is apparently crucial to its exploitation.

3.Isolation, culture and identification of guinea pig nasal mucosa fibroblasts

BACKGROUND: Nasal mucosa fibroblasts are reported to involve in inflammation and wound repair of various nasal diseases by secreting a variety of cytokines and chemokines. Guinea pig is a most suitable experimental animal for the study of allergic diseases. OBJECTIVE: To investigate the effective methods of isolation, culture, purification and identification of guinea pig nasal mucosa fibroblasts. METHODS: Guinea pig nasal mucosa fibroblasts were isolated by collagenase digestion, and purified by differential velocity adherence. The morphology of fibroblasts was observed by inverted phase contrast microscope, fibroblasts were identified by vimentin immunocytochemical staining, and the cell growth was detected by MTT assay to draw the growth curve. RESULTS AND CONCLUSION: (1) Under inverted phase contrast microscope, there were cells with different shapes in the primary nasal mucosa fibroblasts, most of which showed spindle-like and applanate shape, and cells were scattered distribution in cluster. The purified fibroblasts were homogeneous, mainly were long spindle-shaped, and distributed in fish shoal-like and radial-like. (2) Immunocytochemical staining indicated that fibroblasts were positive for vimentin. (3) The cell growth curve appeared to be typical S-shaped. (4) To conclude, the isolated and cultured cells exhibit typical biological characteristics of fibroblasts.
4.Effects of Intravenous Immunoglobulin on Apoptosis and Necrosis of Myocytes in Viral Myocarditis

Objective To investigate the effects of intravenous immunoglobulin(IVIG) on apoptosis and necrosis of myocytes in mice with viral myocarditis.Methods Three hundreds and twenty Balb/c mice were randomly divided into 8 groups.Different courses of IVIG were given in varying time after virus inoculation,Chinese medicine Huangqi given in control group.The virus titer in myocardium、percentage of apoptosis and necrosis of myocytes were detected, myocardial histopathologic scores were counted.Results In every IVIG treatment group,the above 3 items were all significantly lower than that in virus control group and Huangqi group,as IVIG early long course group had the best effect.Conclusion IVIG may reduce the percentage of apoptosis and necrosis of myocytes and virus titer in myocardium in mice with viral myocarditis,the effects are better than that of Huangqi.
5.The miss rate analysis of anti-HLA antibodies detection for kidney transplantation preoperative recipients with sensitization history based on luminex xMAP

Objective To interrogate the detection of anti-HLA antibodies using two methods of Luminex xMAP,and to compare their detection capacity and to analyze their misdetection rate for initial screening,providing more accurate results in clinical practice.Methods 214 serum samples from recipients with a history of sensitization before renal transplantation were collected and detected by LM (LABScreen Mixed) and LSA (LABScreen Single Antigen) respectively on the Luminex xMAP platform.Results For the LM detection,the positive rates of anti-HLA class Ⅰ and Ⅱ were 50.9% and 23.4% respectively,which were lower than those used by the LSA detection (58.9% and 46.7% respectively).The difference had statistical significance (P < 0.05).The sensitivity,specificity,miss rate and mistake rate of anti-HLA class Ⅰ and Ⅱ[detection were 80.2%,90.9%,19.8%,9.1% and 49.0%,99.1%,51.0%,0.9% respectively.The missed detection gene with the highest rate was Cw * 17:01,B * 15:12,B * 45:01 for anti-HLA class Ⅰ and DPA1 * 01:03,DPB1 * 06:01,DPA1 * 01:03,DPB1 * 01:01 for anti-HLA class Ⅱ.The highest MFI value was 10603 and 3659.For the recipients with only blood transfusion history or pregnancy history,LM and LSA detection showed no statistically significant difference when detecting anti-HLA class Ⅰ antibodies,but had statistically significant difference when testing anti-HLA class Ⅱ antibodies.For the patients with history of both blood transfusion and pregnancy history,LM and LSA showed no significant difference in the detection of anti-HLA class Ⅰ antibodies and anti-HLA class Ⅱ antibodies.The miss rate of anti-HLA class Ⅰ antibody detection was lower than that of anti-HLA class Ⅱ antibody detection.Conclusion LSA detection has the higher sensivity and lower miss rate than LM detection.In the light of the disadvantage of LM detection as diagnostic preliminary screening,it is suggested that LSA detection should be used directly for the recipients with a history of sensitization.By this process optimization,it is more likely to cause missent inspection and the occurrence of rejection,as well as a poor long-term survival rate.

OBJECTIVE:
To explore the role of vrk1 gene in two Chinese pedigrees of the first and second branchial arch syndrome.
METHOD:
Sixty members in 2 Chinese pedigrees were recruited. The exon 2 -13 were analyzed by polymerase chain reaction and direct sequencing.
RESULT:
We found a new SNP in proband of Shandong pedigree.
CONCLUSION
vrk1 gene mutation can be excluded in 2 Chinese pedigrees of the first and second branchial arch syndrome.

7.Surgical treatment and etiological analysis of polyotia.

OBJECTIVETo explore the surgical treatment and pathologic mechanism of polyotia.
METHODSThe operative methods for polyotia were flexible. The main procedures were aimed at the remodeling of tragus with the redundant tissue. Tissue transplantation was used to correct the depression deformity. Compound auricular tissue transplantation and Z-plasty were used for correction of the malformation at the posterior part of polyotia.
RESULTSFrom 2003 to 2008, 9 patients with polyotia were treated. The tragus was reconstructed after resection of redundant tissue. The depression deformity was corrected with tissue infilling in 6 cases. The compound auricular tissue transplantation was performed in 4 cases. Z-plasty at the lower ear lobe was performed in 2 cases. The appearance and the location of the reconstructed ears looked symmetric to the healthy ear.
CONCLUSIONSNatural and symmetric appearance of reconstructed ear can be achieved in polyotia. The abnormal migration of neural crest cell might be the pathologic mechanism of polyotia.

8.Determination of borneol in Fufang Danshen intestinal adhesion pellets and study its in vitro dissolution in different dosage form.

The borneol was included with β-CD and prepared Fufang Danshen intestinal adhesion pellets. GC method for determination of borneol in Fufang Danshen intestinal adhesion pellets was established to study its in vitro dissolution and make a comparison with the Fufang Danshen tablet, in this way, the rationality of dosage form was evaluated. The first method of dissolution determination was used for determining the in vitro dissolution of borneol in Fufang Danshen intestinal adhesion pellets in artificial intestinal juice, and Fufang Danshen tablet in artificial gastric juice and intestinal juice, respectively. Result shows: the concentration of borneol in Fufang Danshen intestinal adhesion pellets and Fufang Danshen tablet was 0.79% and 0.80%, respectively. Its in vitro dissolution was nearly 70% within 12 h in Fufang Danshen intestinal adhesion pellets, and in Fufang Danshen tablet, the dissolution was about 60% within 20 min and more than 90% within 40 min, and in artificial gastric juice, was less than 20% within 40 min but more than 80% till 150 min. Research suggests that in comparison with Fufang Danshen tablet, in vitro dissolution of borneol in the Fufang Danshen intestinal adhesion pellets showed an obvious sustained release behavior. The borneol in Fufang Danshen intestinal adhesion pellets was included with β-CD and prepared enteric preparations. To some extent, the stimulation on stomach and intestinal mucosa can be reduced and safety can be improved.

Objective To observe the effectiveness of different administration time and positions of Kaiselu for patients with acute myocardial infarction On prevention of their constipation.Methods 68 acute myocardial infarction patients without three days puzzles stool were randomly divided into two groups f 34 case8 in each group).The patients in the observation group received Kaiselu 40~50 ml without the awareness of defecation,by the anal insertion 7~10 cm,while the control group received Kaiselu 40~50 ml with the awareness of defecation, by the anal insertion 3~4 cm. Results The defecation effect in the observation group Wag significantly higher than the control group(P<0.01).Conclusions It shows better defecation effect for patients with acute myocardial infarction received Kaiselu without the awareness of defecation by the anal insertion 7~10 cm.
10.Experimental research on DNA methylation profile in congenital microtia.

OBJECTIVETo screen for abnormal methylation in CpG islands and CpG sites through whole genome of congenital microtia to identify their associated genes. To discuss the relationship between abnormal methylation level of genes and the etiology of congenital microtia.
METHODSResidual ear cartilage of 50 patients with microtia was collected with ear cartilage of 34 patients without ear malformations as control. Nimblegen CpG promoter array was chosen to screen the 28,226 CpG islands in the whole genome of both experimental and control groups. The genes with differential methylated CpG islands were selected. SpectroCHIP array was chosen to detect the methylation level of each CpG site in abnormal methyletion CpG islands of both experimental and control groups. The CpG sites with differential methylation level were selected.
RESULTSThere were 36 CpG islands with differential methylated level in whole genome between experimental group and control group, among which 29 CpG islands were connected with 29 named genes. In the abnormal methylated CpG islands of COL18A1, MYH14, RBMY1A1 and ZIC3, 6 differentially methylated CpG sites were found with statistical significance. The methylation level of these 6 CpG sites in experimental group and control group were COL18A1_2_CpG_170.9783 +/- 0.0235 and 0.9526 +/- 0.0589; MYH14_CpG_170.9600 +/- 0.0414 and 0.9284 +/- 0.0655; RBMY1A1_1_CpG_3.40.9966 +/- 0.0055 and 0.9914 +/- 0.0069; RBMY1A1_1_CpG_130.9648 +/- 0.0118 and 0.9757 +/- 0.0127; ZIC3_3_CpG_150.0867 +/- 0.0212 and 0.0543 +/- 0.0399; ZIC3_2_CpG_270.3775 +/- 0.1816 and 0.472 3 +/- 0.0439.
CONCLUSIONSThe DNA methylation profile of the entire genome is initially established. The abnormal methylated CpG islands of COL18A1, MYH14, RBMY1A1 and ZIC3 might be related to the pathogenesis of microtia.
