1.Investigation on residue of triadimefon and its metablites in ginseng

Objective To set up a clean-up method using gel permeation chromatography(GPC)and ENVI-Carb-SPE.The residues of triadimefon and its metablites,triadimenol A and triadimenol B in ginseng were detected by GC-MS with negative chemical ionization(NCI).Methods The sample was extracted with acetone and the extract was cleaned using GPC and ENVI-Carb-SPE.Based on GC-MS(NCI)the pesticides were separated on a DB-5MS column using a temperature program and were detected with a mass selective detector in selective ion monitoring(SIM)mode.The reference solution was prepared by the blank sample extract to overcome the matrix effect,the external reference method was used to detect.Results Three pesticides were separated within 10 min.The average spiked recoveries in three levels were 90%—105% with relative standard deviations(RSD)below 6%(n=6)in roots and stems.The limits of detection(LOD)of triadimefon and triadimenols were 0.1 and 10 ?g/L.The precision was below 2%(n=6).Conclusion The method is sensitive for the residue analysis of three pesticides and could be used to the triadimefon and triadimenols detection and security control in ginseng.

3.Analysis of Vancomycin-induced Neutropenia Adverse Reactions/Adverse Events in Children

OBJECTIVE:To provide reference for vancomycin-induced neutropenia adverse reactions/adverse events in clinical diagnosis. METHODS:With a case of children with neutropenia treated by long-course and large-dose vancomycin,PubMed and CNKI were retrieved to collect related literature and the literature was analyzed. RESULTS:Neutropenia may be associated with vancomycin,based on causality criterion of adverse reaction in China. CONCLUSIONS:Vancomycin-induced neutropenia in chil-dren is most likely associated with prolonged exposure induced by infusion,vancomycin dosage should be reduced or stopped,and routine blood and plasma concentration should be closely monitored.
4.A novel method for culture and identification of primary human retinal microvascular endothelial cells

Background To optimize the culture method of human retinal microvascular endothelial cells is very important for the study of retinal angiogenesis disease.Human retinal microvascular endothelial cells have been successfully cultured in previous studies,but further improvement of the culture method to harvest higher yields and purity cells is still needed.Objective This study was to design a modified method to isolate and purify human retinal microvascular endothelial cells much easily and quickly,and to compare the expression of specific markers of vascular endothelial cells,factor Ⅷ and CD31/CD34 in the cells.Methods The use of human donor eyeballs was approved by the Ethic Commission of Zhongshan Ophthalmic Center of Sun Yat-sen University.The retina tissue from healthy donor was isolated and digested by the two-step digestion method with 2% trypsin and 0.133% collagenase Ⅳ.Human retinal microvascular endothelial cells were collected and plated in 60 mm dishes coated by 0.1% fibronectin and cultured in endothelial cell-specialized medium supplemented with 10% fetal bovine serum,0.3 mg/L β-endothelial cell growth factor (ECGF) and 100 ng/L sodium heparin.During the culturing,the growth situation of the cells was monitored by morphological observation,and immunohistochemical staining was performed to probe vascular endothelial cell-specific membrane protein CD31,CD34 and factor Ⅷ for identification of the cell purity.Results Human retinal microvascular endothelial cells were isolated successfully from the retina by the twostep digestion method.The primary cultured cells adhered to well 72 hours later and achieved confluence with the typical cobblestone appearance 9 to 10 days after cultured.The cells exhibited the blue nuclei and reddish cytoplasm by regular haematoxylin and eosin stain and showed a strong positive response for CD31,CD34 and factor Ⅷ by immunohistochemistry.The positive dye of CD31 and CD34 was lower than Ⅷ factor in both endothelial cells.Conclusions Modified culture method of human retinal microvascular endothelial cells can improve cell culture result and purify target cells.
5.Selection of operation for old thoracolumbar fracture:anterior fixation versus posterior vertebral osteotomy

Objective To compare the surgical results between anterior fixation and posterior vertebral osteotomy in patients with old thoracolumbar fracture. Methods 39 cases of old thoracolumbar fracture with posttraumatic kyphosis and neurologic compromise underwent either anterior fixation (n=19) or posterior vertebral osteotomy(n=20). In the anterior group, the average patient age was 38.3 years (range:21 to 64), the mean time from injury to operation was 5.3 months (range:2 to 16), and the mean preoperative kyphotic angle was 25.2?(range:10?to 43?). In the posterior group, the average patient age was 39.9 years (range:18 to 68), the mean time from injury to operation was 5.6 years (range: 2months to 16 years), and the mean preoperative kyphotic angle was 27.6?(range:5?to 60?). Results In the anterior group, the mean operation time was 274 minutes (range:140 to 395) with a mean blood loss of 994 mL (range: 300 to 2000), the mean postoperative kyphotic angle was 14.7?(range: 0?to 35?), and the mean angle of correction was 10.5 ?(range: 5?to 16?). In the posterior group, the mean operation time was 283 minutes (range:190 to 390) with a mean blood loss of 1654 mL (range: 800 to 3800), the mean postoperative kyphotic angle was 4.4?(range:-10?to 35?), and the mean angle of correction was 23.2?(range: 7?to 40?). All the patients with incomplete neural injury had improvement of neurologic function. Conclusion The posterior vertebral osteotomy can produce better results in kyphotic correction, while it does not increase the trauma of operation.
6.Single-level surgery through anterior-posterior approach to treat dislocation of inferior cervical vertebra

Objective To introduce a single level fixation and reduction for cervical dislocation. Methods 15 patients who had old cervical dislocation were treated from May 2003 to April 2004. All the patients had facet locking that could not be reduced though traction. We used posterior approach to relax facet locking and thick Ethicon wire to fix spinal process. We used anterior poach to plug coralline hydroxyapatite (CHA) as the graft. AO plates and locking screws were used to fix the vertebral body. Results All the patients got satisfactory reduction and good alignment through lysis of facet locking and CHA plug. Conclusion The posterior decompression combined with anterior stabilization can achieve satisfactory reduction and alignment for dislocation of inferior cervical vertebra.
7.Effect of PNS on post hyperpolarization potential in rat stellate ganglion neurons

Aim To study the relationship between theeffects of saponins of panax notoginseng(PNS)on fir-ing frequency and hyperpolarization potential in neu-rons.Methods Applieation of a depolarizing currentpulse to stellate ganglion(SG) neurons of rat evokedaction potentials(AP).These neurons were classifiedas phasic or tonic neurons on the basis of their firingpatterns.Then we investigated the effects of PNS on fir-ing pattern and frequency,after hyperpolarization po-tential(AHP) and fastexcitatory postsynaptic potential(f-EPSP) in high Ca2 +Krebs’solution of SG neuronsin rat.Results The firing frequency of tonic neuronswas reduced by PNS.At the concentration of0.12 ~0.16 g.L-1,PNS reversibly depressed AHP in a dosedependentmanner.And the aggrandizing action of highCa2 +on f-EPSP was antagonized by PNS.Conclusion PNS can reduce the firing frequency of rat SG neu-rons,but this action was not caused by reinforcement ofAHP potential.The restraining regulation of excitabili-ty of neurons by PNS may underlie its inhibitory actionon calcium influx.

9.Effects of Bushen Wenyang Huayu Recipe on Expressions of HIF-1α, PHD2, and VHL in Endometriosis Rats with Shen Yang Deficiency Blood Stasis Syndrome.

OBJECTIVETo observe the effect of Bushen Wenyang Huayu Recipe (BWHR) on hypoxia inducible factor-1α (HIF-1α), proline hydroxylase2 (PHD2), von Hippel Lindau disease (VHL) suppressor gene expressions in endometriosis (EM) rats with Shen yang deficiency blood stasis syndrome (SYDBSS), and to explore the pathogenesis of EM and the mechanism of BWHR for treating EM.
METHODSTotally 50 SD rats were randomly divided into five groups, i.e., the blank control group, the sham-operation group, the model group, the Chinese medicine (CM) group, and the Western medicine (WM) group, 10 in each group. Rats in the blank control group and the sham-operation group were fed routinely. Rats in the rest 3 groups received 30-day "extended refrigerator freezing and ice water immersion" and combined with " autotransplantation" to establish EM rat model with SYDBSS. One Milliliter BWHR at 3.33 g/mL was administered to rats in the CM group by gastrogavage. Gestrinone at the daily dose of 0. 5 mg/kg was administered to rats in the WM group by gastrogavage. Equal volume of normal saline was administered to rats in the model group, the blank control group, and the sham-operation group. The size and morphology of ectopic foci in rats were observed after 4 weeks of medication. Expressions of serum CA125, plasma cyclic adenosine monophosphate (cAMP), and plasma cyclic guanosine monophosphate (cGMP) were detected by radioimmunoassay. Morphological changes of eutopic endometrium and ectopic tissue were observed under the optical microscope by HE staining. Protein expressions and contents of HIF-lα, PHD2, and VHL were detected by immunohistochemical SABC method and Western blot. mRNA expressions of HIF-1α, PHD2, and VHL were detected by RT-PCR.
RESULTSThe ectopic foci grew significantly in the model group. Their volumes were obviously contracted after treated by CM and WM. Compared with the blank control group and the sham-operation group, serum CA125 and plasma cGMP obviously increased, cAMP obviously decreased (P < 0.05); expressions and contents of HIF-1α mRNA and protein all decreased (P < 0.05); mRNA and protein expressions and contents of PHD2 and VHL all decreased in the model group (P < 0.05). Compared with model group, levels of CA125 and cGMP obviously decreased; cAMP levels obviously increased, expressions and contents of HIF-1α mRNA and protein all increased, mRNA and protein expressions and contents of PHD2 and VHL all increased in the WM group and the CM group (P < 0.05). Compared with the CM group, PHD2 protein contents were higher in the WM group (P < 0.05). HIF-1α was negatively correlated with PHD2 (r = -0.799, P = 0.00). HIF-1α was negatively correlated with VHL (r = -0. 625, P = 0.003).
CONCLUSIONSBWHR could effectively treat EM. Its mechanism might be associated with reducing contents of HIF-1α, serum CA125, and plasma cGMP, and up-regulating expressions of PHD2, VHL, and cAMP.

10.Characteristics of perioperative lumbar operation for the elderly.

OBJECTIVESTo study the characteristics of senile patients during perioperative lumbar operation and discuss matched surgical method.
METHODSFrom 1998 to 2000, lumbar operations were performed in 41 elderly patients (average age 68). 14 patients had lumbar disc herniation, 11 lumbar disc herniation with lumbar spinal stenosis, 12 lumbar instability and spondylolisthesis, 2 reoperation, and 2 combined compression of cervicle and thoracic spinal cord.
RESULTSMany patients were complicated by the following diseases: hypertension (14), coronary heart disease (9), diabetes mellitus (4), cerebral infarction (3), rheumatic heart disease (1), pulmonary heart disease (1), and complicated with 2 or 3 diseases (9). Discectomy was performed in 14 patients, decompression + discectomy in 11, posterior lateral fusion with pedicle instrument in 9, posterior lateral intravertebrae fusion with instrument in 3. Fixation of 2 levels was made in 5 patients, 3 levels in 3, and multi-levels in 2. Postoperation JOA amelioration rate was 81.6%.
CONCLUSIONThe condition of adjacent segment should be considered in the operative treatment of elderly patients. Caution should also be taken to coexisting diseases.
