1.Influence of Qiangxin Tongmai decotion on heart function and plasma brain natriuretic peptide level in patients with chronic heart failure

Objective To explore the effect of conventional western medicine combined with Qiangxin Tongmai decotion in the treatment of patients with chronic heart failure (CHF).Methods According to the digital table,60 patients with CHF were randomly divided into two groups,30 cases in each group.The control group was treated with conventional western medicine,while the treatment group received combination therapy of conventional western medicine and Qiangxin Tongmai decotion.The heart function and plasma brain natriuretic peptide (BNP) level were observed.Results The total effective rate of the treatment group was 93.3%,which was significantly higher than 70.0% of the control group (x2 =5.455,P < 0.05).After treatment,the level of BNP in the treatment group was (211.3 ± 87.7) ng/L,which was significantly lower than that of the control group [(410.8 ± 94.3) ng/L] (t =8.485,P < 0.01).Conclusion Qiangxin Tongmai decotion can obviously improve the heart function and reduce the level of BNP in patients with CHF.
2.Influence of the estradiol on the thyroid iodine uptake and the expression of NIS in the thyroid gland of rats

Objective To identify the impact of estradiol on the goiter and the function of the thyroid gland with different dosages of estradiol.Methods 48 Wistar rats(female,normal state) were divided into 4 groups: Ⅰ,ovx,Ⅱ,sham;Ⅲ,ovx+E_(2)-low and Ⅳ ovx +E_(2)-large.In each group,we applied nuclide scanning of()~(131)I and()~(99)mTcO4 to detect the thyroid iodine uptake and adopted the technique of RT-PCR to test the expression of NIS.Quantity of TSH、FT_(3)、FT_(4) and the estradiol were examined.Results The expression of NIS was inhibited in group Ⅰ and group Ⅳ.Conclutions Estradiol can enhance the capability of the thyroid glands uptaking iodine and inhibit the expression of NIS and so estradiol is a very important component in the pathogenesis of goiter.
3.Effects of mesenchymal stem cells transplantation on the brain-derived neurotrophic factor and growth associated protein-43 after the spinal cord injury of rats

[Objective]To observe the effects of mesenchymal stem cells(MSCs) transplantation on the brain-derived neurotrophic factor(BDNF) and growth associated protein-43(GAP-43) after the spinal cord injury(SCI) of rats, and to investigate the mechanism of repairing the SCI by MSCs transplantation. [Method]Seven days after the operation of SCI,the MSCs were transplanted into the injured site. Then GAP-43 and BDNF were tested by RT-PCR. By means of above,the mechanism of repairing the SCI after the cell transplantation was investigated.[Result]Compared with group B, transplantation of MSCs enhanced the expression of BDNF mRNA and GAP-43 mRNA in the spinal cord of group A, improved the neuron regeneration.[Conclusion] The transplantation of MSCs can repair the injuried site and promote the regeneration of axon by enhanced the expression of BDNF mRNA and GAP-43 mRNA. It is one of the mechanism of repairing the SCI by MSCs transplantation.
4.The Mentation of Patients with Stroke

Objective To analyze the mentation of patients with stroke and the risk factors.MethodsThe mentation of 309 patients(178 male cases and 131 female cases) with stroke was evaluated with special scales.ResultsAmong the male cases,the incidence of anxiety was 30%;the incidence of depression was 37%.Among the female cases,the incidence of anxiety was 43%;the incidence of depression was 51%.The incidences of anxiety and depression of female were higher than that of male(P<0.05).ConclusionThe incidences of anxiety and depression in stroke patients are higher than common people.The degree of stroke,family relations,gender and age of the patients are the risk factors of anxiety and depression after stroke.
5.Characteristics of Sleep Disorders in Patients with Stroke

Objective To study the characteristics of sleep disorders in patients with stroke.Methods 350 patients with stroke were evaluated with Pittsburgh Sleep Quality Index (PSQI) and Neurofunction Deficit Score (NDS).Results The familiar sleep disorders in stroke patients were difficulty to fall asleep, early to awake, hard to keep the sleep long, with inordinate sleep structure, excess sleep in daytime, paroxysmal nona. The incidence rate of sleep disorder in female cases was higher than that of the male cases ( P<0.05).Conclusion The incidence of dyssomnia in stroke patients is higher than that of common people. As a whole,the incidence of dyssomnia in female stroke patients is higher than male. The incidence of sleep disorders such as difficulty to fall asleep, early to awake, hard to keep the sleep long, with inordinate sleep structure in female stroke patients is higher than that of male patients. The incidence of sleep disorders such as excess sleep in daytime, paroxysmal nona in male stroke patients is higher than that of female patients.
6.Hyperbaric oxygenation on infantile cerebral palsy

ObjectiveTo evaluate effects of hyperbaric oxygenation on motor function in children with cerebral palsy. Methods49 infants with cerebral palsy were classified into two groups, hyperbaric oxygenation group(29cases) and control group (20 cases). Two groups were treated with cerebrolysin and received family rehabilitation drill. In additional, the hyperbaric oxygenation group was treated with hyperbaric oxygenation therapy. The children's motor function of two groups was evaluated before and after the treatment.ResultsThe effect in the hyperbaric oxygenation group was better than that in the control group(P<0.01). Conclusions Hyperbaric oxygenation is an effective therapy on infantile cerebral palsy.
7.New perspectives on medical treatment of gastric cancer.

Perioperative chemotherapy for gastric cancer has made steady progress with the application of chemotherapeutic agents and conduct of relevant clinical trials. However, palliative chemotherapy has only made marginal progress, and the median survival remains between 8-10 months. An emerging understanding of tumor biology, cellular and molecular mechanisms has revealed novel targets in gastric cancer therapy, such as trastuzumab, bevacizumab, cetuximab, etc. There are still some problems in clinical practice due to the high heterogeneity of gastric cancer including optimization of perioperative chemotherapy regimen, determination of right strategy to personalize chemotherapy or target therapy for advanced gastric cancer. In order to practically achieve individualized therapy, multicenter, prospective, randomized clinical trials according specific matters of gastric cancer should be performed in China. A deeper understanding of the biological characteristics of gastric cancer, and more translational medicine research and multi-disciplinary team collaboration should be carried out to achieve personalized medicine.

10.Efficient Secretion of Recombinant PEX in COS7 Cells Directed by MMP-9 Signal Peptide

With the rapidly development of the biotechnology industry,large quantities of recombinant proteins are needed for specific therapeutic and diagnostic applications.Bacterial cells are most often used for the production of recombinant proteins.However,recombinant proteins expressed in the cytoplasm of bacteria are often misfolded as insoluble inclusion bodies and therefore inactive.To circumvent this problem,several eukaryotic expression systems have also been developed over the years,ranging from yeast to mammalian cell-based technologies.For many mammalian proteins,especially those secreted and modified posttranslationally,a more compatible expression system is highly desirable because proper folding or modification can only be provided with closely related cells,i.e.,mammalian cells.Large scale transient transfection of mammalian cells is a recent and powerful technology for the fast production of milligram amounts of recombinant proteins.Transient expression by means of extrachromosomal replication in COS cells is frequently used to check the functional integrity of genes/plasmids and to produce small quantities of cell supernatants containing the protein of interest.As it is allowed for easy and efficient purification,many recombinant proteins used for therapeutic and structural studies are naturally secreted or engineered to be secreted.The use of a proper signal peptide is one of the major determinants for the efficient secretion of heterologous proteins from mammalian cells.The noncatalytic C-terminal hemopexin-like domain of MMP-2,PEX,can block angiogenesis and tumor growth in vivo.Large quantities of biochemically active recombinant PEX are required for the study of their functions and biochemical properties,as well as for their industrial applications.For this purpose,the rat growth hormone,mouse IgG? chain and MMP-9 signal peptides were used for expression of PEX in COS7 cells,and their secretion efficiencies were compared by Western blotting and ELISA.Western-blotting of PEX protein from culture media,resulted in detection of proteins with the predicted molecular mass,which indicate that all of the signal sequences could direct PEX secretion successfully.The MMP-9 signal peptide seems to be superior to the signal peptides from IgG and rGH both in terms of extracellular yield and in terms of secretion efficiency.Thus,expression of pM9PEX construct resulted in higher yields of extracellular PEX and the majority of the produced PEX was secreted and not trapped intracellularly.To examine whether the observed difference in secretion yields is promoted at the transcriptional level,a RT-PCR analysis was performed at 6 h after transfection.The presence of mRNA transcripts of PEX was observed in all the DNA constructs.Moreover,semiquantitative reverse transcription(RT-PCR)results show that there were no significant differences in the expression levels of PEX among the constructs at 6 h after transfection.Though there was no difference in the expression levels of PEX at an early time point after transfection,the presence of an ER-targeting signal peptide sequence in the expression vector affected the trafficking of expressed proteins in the cells.Hence,the described difference in exported yields is probably promoted at the secretion level,rather than at the transcriptional level.Chick chorioallantoic membrane(CAM)bioassay show that the PEX protein purified from cell culture had biological activity to inhibit the angiogenesis.The MMP-9's signal peptide is used for the first time as leader sequence for secretion of foreign proteins.The results revealed that higher amounts of secreted PEX were obtained when vectors containing MMP-9 signal peptide were used and it is also indicated that MMP-9 signal sequence could be effective on promoting the secretion of other heterologous proteins in eukaryotic cells.
