Objective:To investigate the impact of polysomy 17 of breast cancer on testing results of human epidermal growth factor receptor 2 (HER2) and its clinicopathologic significance. Methods:Seventy-one patients with primary invasive breast carcinoma were studied. The HER2 gene and chromosome 17 copy numbers were determined by dual-color fluorescence in situ hybridization (FISH). The testing results were expressed by absolute HER2 gene copy number or the ratio of HER2 to chromosome 17. Based on the FISH testing results and HER2 protein expression determined by immunohistochemistry the results were compared between different groups divided by related clinicopathologic parameters. Results:All patients who had doubtable FISH results, either by absolute HER2 copy number (14 of 71 patients; 19.7%) or by the ratio HER2/chromosome 17 (2 of 71 patients, 2.8%), displayed polysomy 17. Polysomy 17-positive patients had no significant difference with HER2-negative patients in tumor grade, lymph node metastasis, and estrogen receptor (ER) expression (all P>0.05); but compared with HER2-positive patients, they showed lower tumor grade (50.0% vs 81.5%, P=0.025), higher rate of negative lymph node (55.6% vs 25.9%, P=0.045), and higher rate of ER positive expression (83.3% vs 41.7%, P=0.005) and progesterone receptor(PR)positive expression (87.5% vs 44.4%, P=0.003).Conclusion:Compared with HER2 gene amplification group, polysomy 17-positive group tends to have negative HER2 gene expression. Polysomy 17 influences the testing results of HER2 and may be the main factor that caused doubtable results in FISH examination.

In this study we implanted magnetically labeled neural stem cells (NSCs) in PD rats and then monitored their survival and migration in the host brain by magnetic resonance imaging (MRI). The mesencephalic NSCs were obtained from the brain of SD rats. Superparamagnetic iron oxide (SPIO) was transferred to NSCs by Lipofectamine transfection. Eighteen PD lesioned rats were selected for transplantation by evaluation of their rotational behavior in response to amphetamine and randomly assigned to 3 groups, i.e., sham group, PBS group and NSCs transplanted group, with 6 rats in each group. MR scanning was performed at 1, 2, 4, 6, 8 and 10 week(s) following transplantation. At the meantime, rotational behavior was assessed in each group. Our results showed that SPIO particles were clearly visible with Prissian blue staining in neurospheres and cells derived from NSCs. The rotational behavior of the NSCs transplanted group was remarkably improved compared with that of sham group and PBS group (P < 0.05). In vivo MR tracking of NSCs showed that SPIO labeling led to a strong susceptibility change of signal 1 week after transplantation on T2 weighted images. And a large circular hypointense signal appeared in the transplanted area on T2* gradient echo images. Ten weeks following transplantation, the hypointense signal on T2 weighted and T2* gradient echo images was still displayed. It is concluded that SPIO particles could label NSCs effectively, and MRI detection of SPIO labeled cells is a promising method and novel approach to analyzing the NSCs following transplantation in the treatment of PD.

To investigate the advantage of using the medical sodium hyaluronate gel undergo goniosynechialysis, and to compare the difference of the viscoelastic agent and perfusion fluid by clinical control study.
●METHODS:Totally 103 cases (103 eyes) patients who had angle- closure glaucoma (ACG) with cataract all in Beijing Tongren Hospital affiliated to Capital Medical University, from September 2012 to February 2013. All the patients had primary glaucoma or recurrence ACG with cataract. Their intraocular pressure (lOP) could be controlled in normal value using one or two anti -glaucoma medications. We divided all the patients into two groups by chance. Experimental group had 53 cases (50 eyes), and control group had 50 cases (53 eyes). Two groups all had been performed phacoemulsification combined with goniosynechialysis by the same experiential operator. But experimental group did goniosynechialysis using the medical sodium hyaluronate gel, control group did goniosynechialysis using perfusion fluid. We observed the change of lOP, central anterior chamber depth and anterior chamber structure of two groups in postoperative 1mo.
●RESULTS: SPSS19. 0 statistics software was utilized to analyze the data, This study selected one- side test α =0. 05, adopted paired t - test or two- sample t - test. We obtained that experimental group lOP decreased 12. 42 ± 0. 04mmHg, anterior chamber depth increased 1. 276 ± 0. 201mm; that control group lOP decreased 6. 56 ± 0. 08mmHg, anterior chamber depth increased 0. 852 ± 0. 132mm. And experimental group anterior chamber angle opened more widely than the other group, the comparison had statistical significance (P<0. 05).
●CONCLUSlON: ln phacoemulsification combine with goniosynechialysis, use medical sodium hyaluronate gel do goniosynechialysis can more effectively decrease lOP. This because the medical sodium hyaluronate gel has high glutinousness, it can separate angle of anterior chamber and controlling lOP effectively.

Chinese Journal of Applied Physiology 2002;18(2):149-151
AIMTo explore the effect and mechanism of interleukin-2 (IL-2) on the rhythm of isolated rat heart and neonatal myocytes.
METHODSCultured neonatal rat cardiomyocytes and isolated perfused rat heart were used.
RESULTS(1) IL-2 (2.5-200 u/ml) reduced the spontaneous beating rate of cultured neonatal rat cardiomyocytes in a dose-dependent manner. (2) IL-2 at 50 u/ml increased both the heart rate and the number of premature ventricular excitation in the isolated heart. (3) Pretreatment with propranolol abolished the effect of 50 u/ml IL-2 on the isolated heart. (4) Heat inactivated IL-2 had no significant effect on the cultured cardiomyocytes and isolated hearts.
CONCLUSIONIL-2 inhibited the auto rhythmic of cultured cardiomyocyte directly while the positive chronotropic and arrhythmogenic effects of IL-2 in the isolated rat heart may be mediated by endogenous catecholamine.

Journal of Zhejiang University. Medical sciences 2002;31(1):19-22
OBJECTIVE: To explore the effects and mechanism of interleukin-2 on the isolated rat ventricular papillary muscle and cultured cardiomyocyte. METHODS: The isolated rat papillary muscle and cultured neonatal rat cardiomyocyte were used to determine the effects on contractile properties and spontaneous beating. RESULTS: IL-2 inhibited the contractile force of isolated rat papillary muscle and reduced the spontaneous beating rate of cultured neonatal rat cardiomyocytes in a dose-dependent manner. IL-2 at high concentration reduced the systolic duration and prolonged the diastolic duration. Pretreatment with L-NAME, an inhibitor of nitric oxide synthase, abolished the effects of IL-2. Heat inactivated IL-2 had no effect on papillary muscles and cardiomyocytes.CONCLUSION: Inhibitory effects of IL-2 on the contraction of isolated papillary muscle and spontaneous beating rate of cultured neonatal cardiomyocyte are mediated by nitric oxide.

In the process of vinification, the metabolic products of various yeasts are key to the sensory characteristics. Different species produce different volatile composition, which results in direct influences on wine's bouquet mostly. Metabolic characteristics of Saccharomyces and non-Saccharomyces and the different influences on odour by them as well as interactions among various species were described in this paper. Research progress on non-Saccharomyces was summarized.Studies confirmed that some characteristics of non-Saccharomyces play a positive role in the structure of bouquet of wine. Combined use of Saccharomyces and some of non-Saccharomyces will be beneficial to sensory characteristics of wine.
