Objective To investigate the gene-expression profile in kidney of rats during late sepsis (24hours) by using microarray technology in order to offer some clue to revealing the pathogenetic mechanism of sepsis at gene level. Method A total of 30 Wistar rats were selected and divided into model group and control group randomly(random number). The rats of control group were sham operated and the rats of model group received cecal ligature and puncture (CLP) operation. The biomarkers of renal function were assayed and the histopathological changes of kidney in rats were observed under transmission electron microscope 24 hours after operation. Gene chips containing 22 107 rat-genes cDNA were used to exmine gene-expression in kidney of septic rats to sieve the genes with different expressions with software. Data were analyzed by using SPSS version 11.0 software package.Statistical analyses of two independent samples carried out by using t -test. Results Compared with the control group, the levels of blood urea nitrogen (BUN) and creatinine (Cr) of sepsis group were higher (P ＜ 0.01 ). The histopathological changes in kidney of rats demonstrated the establishment of sepsis model successful 24 hours later.Compared with the control group, there were 325 genes with differential expression in model group. Among the known-functional genes, there were 100 up-regulated and 64 down-regulated. Sorted by biological function, the genes were mainly related to metabolism, immunoresponse, cellular signal transduction, apoptosis, ion channel,growth factor and so on. Conclusions A sequence of genes expressed differentially in kidney of rats with late sepsis. Microarray technology played an important role in the research into sepsis mechanisms.

Objective This study was carried out in order to analyze genetic diversity in plants of Dendrobium Sw. and compare the marker index (MI) between SRAP and RAPD markers. Methods Using 40 SRAP primer combinations and 36 RAPD primers,the genetic diversity of the materials in Dendrobium Sw. was amplified. Results A total of 1 977 loci were generated by 40 SRAP primer combinations,of which 90.2% was polymorphic and the genetic similarity (GS) ranged from 0.330 2 to 0.789 2. As for RAPD,281 bands were obtained,in which 87.1% was polymorphic,and the GS was from 0.494 2 to 0.773 1. There were some differences existing in the two cluster results revealed by SRAP and RAPD markers,they were as follows:compared to that of RAPD,the result of SRAP could more comfortably reflect the kinship and geographical origin,simultaneously conform to the traditional classification. For the experiment materials,SRAP (16.46) got an MI value 4.5 folds higher than that of RAPD (3.67),which meant SRAP markers were more effective in detecting genomic polymorphisms in the plants of Dendrobium Sw. Conclusion These results show that SRAP and RAPD could be applied to detecting genetic diversity analysis of plants in Dendrobium Sw. SRAP Also shows great superiority and advantage in this study.

Objective To investigate the suitable light intensity of plant growth of Dendrobium Sw.through study on photosynthetic characteristics.Methods The photosynthesis curves of light intensity, chlorophyll fluorescence parameter of different light intensity and inductivity of non-photochemical quenching of chlorophyll fluorescence in 3.0?104 lx on three species of Dendrobium Sw.in Huoshan (D.huo- shanens, D.candium, and D.moniliforme) were determined.Results Three species of Dendrobium Sw.were shadow plants for their response on light intensity, light saturation point, light compensate point, net photosynthesis rate, and apparent quantum efficiency are lower during photosynthesis.The photoinhibitory effect was formed on Dendrobium Sw.leaf when they grew in more than 4.0?104 lx.Conclusion Although three species of Dendrobium Sw.in Huoshan grow in (0.2-2.0)?104 lx normally, they are suitable to grow in (2.5-3.0)?104 lx instead of growing more than 4.0?104 lx.

Objective To investigate the relationship of the expression characteristics of CD4 + CD25 + Foxp3 + Treg cells and CD4 + IL-17 + Th17 cells and the progress of the elderly patients with laryngeal cancer.Methods Sixty elderly patients diagnosed as laryngeal cancer in our hospital from January 2013 to June 2014 were chosen.Among them,28 cases had lymph node metastasis and 32 without.Thirty cases without laryngeal diseases were chosen as control.The peripheral blood of elderly patients with laryngeal cancer and normal controls was extracted.The expression levels of Treg and Th17 cells in all groups were detected with flow cytometry,and the expression changes of Foxp3,RORγt in peripheral blood mononuclear cell (PBMC) were detected with real time quantitative PCR.Results Compared with the control group,CD4 + CD25 + Foxp3 + Treg and CD4 + IL-17 + Th17 cell percentage,Treg and Th17 cells specific transcription factor Foxp3,RORγt expression levels in elderly laryngeal cancer patients without lymph node metastasis were significantly higher (t=9.817,P=0.018; t=12.120,P=0.009; t=6.090,P=0.023; t=7.130,P=0.028).Compared with elderly laryngeal cancer patients without lymph node metastasis,Treg and Th17 cell percentage and Foxp3,RORγt expression levels were significantly increased (t =9.070,P =0.014; t =12.140,P =0.009; t =10.130,P =0.009 ; t =13.070,P =0.008),and the ratios of Th17/Treg and Foxp3/RORγt were significantly lower (2.401 ± 0.614 vs 3.763 ± 0.959 ; 0.401 ± 0.075 vs 0.563 ± 0.091 ; t =9.070,P =0.014 ; t =11.140,P =0.007) in elderly laryngeal cancer patients with lymph node metastasis.Conclusion In elderly patients with laryngeal cancer,Treg cells and Th17 cells imbalance is positively correlated to disease progression.Therefore,monitoring and correcting the expression levels of Treg and Th17 cells may be important for the diagnosis,treatment and prognosis of the elderly patients with laryngeal cancer.

Purpose To observe the mutation of K-ras gene and expression of Fascin-1 protein in CRC tissues and their relationship with clinical pathological features, and then to analyze the correlation between mutation of K-ras and expression of Fascin-1. Methods In 86 cases of CRC tissues, K-ras mutation was detected by DNA sequencing analysis, and Fascin-1 expression was detected by im-munohistochemical method. Results In CRC tissues the mutation rate of K-ras was 34. 88%, the expression rate of Fascin-1 was 60. 47%. The mutation rate of K-ras in lymph node metastasis group was higher than that of without lymph node metastasis group, and that in distant metastasis group was higher than that of without distant metastasis group(P<0. 05). The expression rate of Fascin-1 in serosa invasion group was higher than that of without serosa invasion group, and that in lymph node metastasis group was higher than that of without lymph node metastasis group, and that in distant metastasis group was higher than that of without distant metastasis group (P<0. 01). There was a correlation between the mutation of K-ras gene and the expression of Fascin-1 in CRC tissues (rp =0. 236, P<0. 05). Conclusions The CRC tissues with mutation of K-ras are more likely to metastasize and the CRC tissues with expression of Fascin-1 are more likely to invade serosa and metastasize. The CRC tissues with mutation of K-ras are more likely to express Fascin-1.

Objective To discuss the effect of different methods of dropping the high temperature of the tubercular meningitis patients. Methods 35 cases with tubercular meningitis with high fever were divided into research group(11 cases) and control group(24 cases) ,24 patients in control group were treated by traditional cooling method, 11 patients in research group were rteated by mild hypothermia therapy, The self signs, the intracranial pressure,Glasgow scores, complications and the death rates of patients were assessed and compared. Results The effective rates of the research and control cooling method were 81.8% and 41.7% respectively. The difference between the two groups was significant;After the intervention,compared with the control group, the self scale is superior in research group,and the intracranial pressure and the death rates of patients were lower,and patients'Glasgow scores were higher,but the complications between the two groups had no significant differences. Conclusion Mild hypothermia can effectively control patients'temperature,at the same time, which can improve the patients conscious state and reduce the mortality.

Objective To optimize the best subculture cycle through studying the influence of subculture time on tube plantlet growth of Dendrobium huoshanense and the change of medium composition.Methods Height,tiller,fresh weight,and chlorophyll content of the tube plantlet and pH value,water content,and sugar content of the medium were measured after different cycles of the subculture,the cost of culture medium for subculture was calculated as well.Results The height of the tube plantlet increase 282.86%,the tiller increase by 3.5 times,fresh weight reaches its maximum,chlorophyll content of the tube plantlet almost reaches its maximum after 40 d subculture;while water content and sugar content of the medium are decreased to the lowest point,pH value of medium(

Objective To observe the surface electromyogram(sEMG)characteristics of the lower limbs muscles in stroke patients and healthy subjects during the passive and active-assisted anti-resistance movements.Methods Twenty four stroke patients and 17 healthy individuals(control group) participated in this study.sEMG of rectus femoris(RF),vastus lateralis(VL),ventralis medialis(VM)and biceps femoris(BF)was recorded during passive and active-assisted anti-resistance of knee joint in both groups.Average EMG(AEMG),mean power frequency(MPF)and median frequency(MF) were analyzed.Results Multiple factor ANOVA showed that very significant differences in the values of MF,MPF and AEMG between non-paretic and the paretic limbs appeared(P

AIM: To investigate the inhibitory effects of genistein(Gen) on mouse allergic contact dermatitis(ACD).METHODS: The animal model of ACD was induced by DNFB.The effects of different doses of Gen on mouse ear swelling,body weight,histopathological changes in mouse ear skin,thymus index and spleen index were observed.RESULTS: All groups of Gen inhibited mouse ear swelling induced by DNFB significantly.The infiltration of inflammatory cells and thymus index were also reduced.However,the increase in mouse body weight was not affected.Low dose of Gen increased spleen index,high dose of Gen decreased spleen index.CONCLUSION: Genistein has significant inhibitory effects on mouse ACD induced by DNFB.
