China Pharmacy 2016;27(35):4904-4905,4906
OBJECTIVE:To compare economical efficiency of 4 therapeutic regimens for pediatric perianal abscess. METH-ODS:By retrospective study method,85 children were divided into group A(18 cases),B(10 cases),C(14 cases)and D(43 cases)according to different therapeutic regimens. They were given amoxicillin-flucloxacillin,flucloxacillin,cefathiamidine and ce-famandole 100 mg/(kg·d) intravenously for 5-7 d,respectively. Clinical efficacies and the occurrence of ADR were compared among 4 groups. The economical efficiency of 4 therapeutic regimens were analyzed by the method of cost-effectiveness analysis. RESULTS:For group A,B,C and D,the cure rates were 77.78%,80.00%,57.14% and 90.70%,that of group C was signifi-cantly lower than those of other 3 groups, with statistical significance(P<0.05). No significant ADR were found in 4 groups. The costs were 3 600.86,2 913.92,3 416.19 and 2 981.97 yuan, and the cost-effectiveness ratios were 46.30,36.42,59.79 and 32.88. The incremental cost-effectiveness ratios of group A,C and D to group B were -309.43,-21.97 and 6.36. The results of cost-ef-fectiveness analysis were supported by sensitivity analysis. CONCLUSIONS:For pediatric perianal abscess, cefamandole shows the best cost-effectiveness;but the therapeutic duration and total cost of flucloxacillin therapeutic regimen is shorter and lower. Thera-peutic regimen should be selected according to children’s condition.

Chinese Pharmacological Bulletin 2003;0(11):-
Aim To study effects of berberine on the proliferation a nd differentiation of HL-60 cells. Methods Cell inhibitory effec t was determined by growthcurve method and colony formation.Cell differentiation was analyzed by cytomorphological changes, NBT reduction and cell surface antig en.Cell cycle was analyzed by flow cytometry. Results The growth o f HL-60 cells was significantly inhibited by berberine in a time and dose-depe ndent manner.1,2,4,8 mg?L -1 berberine induced differentiation of HL-60 c ells: Morphological evaluation indicated that the berberine-treated cells exhibited granulation appearance; NBT reduction was significantly increase d; expression of CD11b increased obviously. Flow cytometry analysis showed that berberine arrested HL-60 cells in G 0/G 1 phase, the percent of the cells in S phase decreased. Conclusion Berberine can induce differentiatio n and growth inhibition of HL-60 cells
5.The synergistic antitumor effects of berberine alpha-hydroxy-beta-decanoylethyl sulfonate with hydroxycamptothecine and its effect on topoisomerase.
Acta Pharmaceutica Sinica 2011;46(4):390-4
Synergistic antitumor effects of HB (berberine alpha-hydroxy-beta-decanoylethyl sulfonate, houttuyn berberine) with HCPT (hydroxycamptothecine), and its correlative mechanism were studied in vitro. MTT assay was employed to determine the cytotoxicity of HB combined with HCPT in tumor cells culture in vitro, IC50 and combination index (CI value) were used to evaluate the synergistic effects. The supercoiled DNA relaxation mediated by topoisomerase I & II was measured by agarose gel electrophoresis assay, and influence of HB was detected. The results showed that HB could inhibit the proliferation of tumor cells (SGC-7901, SW1116 and SW480) in vitro, and the inhibition ratio was increased, IC50 was reduced when combining with HCPT. CI value of the two drugs was less than 1 in HepG2, SW480, SGC-7901 and SW1116 cells. The lowest value was 0.447, 0.626, 0.161 and 0.178 in these tumor cells, respectively, further indicating HB has synergistic action with HCPT on suppressing tumor proliferation. The agarose gel electrophoresis assay showed HB can inhibit topoisomerase I & II activity of SW480 cells at the concentration of 2.0-8.0 mg x L(-1). HCPT is a typical inhibitor of topoisomerase I , the synergistic action between HCPT and HB on suppressing tumor proliferation is perhaps related to the congenerous inhibition of topoisomerase.
6.CD4~+ CD25~+ regulatory T cells inhibit macrophage-derived foam cells formation by down-regulating scavenger receptor expression
Chinese Journal of Microbiology and Immunology 2010;30(2):99-103
Objective To investigate whether and how regulatory T cells(Tr) affect macrophages foam-cell formation, and thereby investigate the mechanism of Tr in the development of atherosclerosis. Methods Tr were isolated from lymphocyte suspensions by magnetic cell sorting-column and analyzed by flow cytometry. Macrophages were cultured alone, with CD4~+ CD25~+ T, or CD4~+ CD25~- T cells in the pres-ence of oxLDL for 48 h to transform macrophages into foam cells. Oil red O staining and cellular lipid meas-urement were used to identify macrophage foam cell formation. Semi-quantitative PCR, quantitative real-time PCR and Western blot analysis were carried out to explore the mechanism of Tr-mediated suppression on macrophages foam cell formation. Results Foam cell formation, as identified by oil red O staining, was readily apparent in cells treated with CD4~+ CD25~- T cells and without T cells. After treatment with Tr, a marked decrease(13.9% ± 5.6% ) in foam cell count was observed, compared with untreated cells(13.9% ±5.6% vs 52.9% ± 10.4%, P<0.001 ) or CD4~+ CD25~- T-treated cells(13.9% ± 5.6% vs 53. 1% ± 17.2%, P<0.001 ), 52.9% ± 10.4% and 53.1% ± 17.2%, respectively (P<0.001). The similar effect of Tr was obtained when extracted oil red O and measured by a spectrophotometer. Cholesteryl ester ac-cumulation also used to quantify foam cell formation. Compared with untreated (no T) and CD4~+ CD25~- Tr-treated macrophage cells (CD25~-), the lipids accumulation in CD4~+ CD25~+ Tr-treated macrophage foam cells(CD25~+) was significantly reduced. Total cellular cholesterol and cellular cholesteryl ester was siginifi-cantly reduced in CD25~+ cultures relative to no T[TC(total cholesterol): 57.46 ± 17.92 vs 159.48 ± 16.38, P<0.01 ; CE(esterified cholesterol): 26.68 ± 8.88 vs 102.54 ± 16.67, P<0.001] or CD25~- (TC: 58.50±7.00 vs 150.55±25.11, P<0.01; CE: 26.68±8.88 vs96.90 ± 11.95, P<0.001) cul-tures. Moreover, PCR and Western blot analysis showed that the expression of both CD36 and SRA in Tr-treated macrophage foam cells was significantly down-regulated. Conclusion Results collectively suggest that CD4~+ CD25~+ Tr cells may inhibit macrophage foam-cell formation, which is largely attributed to a down-regulated expression in scavenger receptor in Tr-treated macrophage foam cells.
