Objective To develop a rapid expansion assay and determine the predominant epitope for HBV-specific CD8 T cells from peripheral blood mononuclear cells(PBMC) of patients with acute hepatitis B(AHB).Methods PBMC were isolated from a patient with HLA-A2 positive AHB,and the frequency of HBV-specific CD8 T-cells was determined using 3 kinds of HLA-A2 antigenic peptide florescence-labeled pentamer agents,i.e.S183-191 and S335-343 derived from HLA-A2 restricted HBsAg epitope, and C18-27 derived from HBcAg epitope.Cytokines and corresponding epitopic peptides were used for induction of positive CD8 T-cell expansion and enrichment in PBMC for 1-2 weeks.T epitope-specific CD8 T-cells were quantified and sorted by flow cytometry. Results After cultivation with antigenic peptide stimulation and proper cultural condition, the frequency increased significantly in the pentamer positive HBV-specific CD8 T cells in total CD8 T cells. The peak value was observed in 2-week cultivation. At the time the HBV-specific CD8 T cell frequencies increased from 0.21% to 15.17% for S183-191,and from 0.44% to 1.79% for S335-343,representing 73.8-folds and 3.1-folds increase for each epitope respectively. However, the frequency of C18-27-specific CD8 T-cells was negative and without obvious proliferation after stimulation. Conclusions Under stimulation of HBV epitopic peptide in proper culture condition, the specific CD8 T lymphocytes may be induced and expanded in quantity via sort-term conditioned culture.CD8 T cells specific for S183-191 may play an important role in the control of acute HBV infection.

Objective:To Explore the correlation between silent cerebral infarcti on(SCI) and aged depression. Methods:SCI was analyzed in 90 aged patients w ith or without depression. Results:Incidence rate of SCI was 71.1% in 45 aged p ersons with depression(depression group), but incidence rate of SCI was 13.56% i n 45 aged persons without depression(no depression group). Compared no depressio n group the SCI incidence rate of depression group was obviously increase(P< 0.01).Conclusion: There is significantly correlation between SCI and aged depression

Insulin Resistance(IR) is the core of metabolic syndrome(MR) which is once regarded as deadly quartet.Early intervals can prevent or delay the occurrence and progress of diabetes mellitus, cardiovascular disease.Health education, diet tyerapy, exercises and durgs are of great importance during nursing care.

Objective To explore the surface electromyography(EMG)characteristics of biceps brachii during isometric contraction in children with hemiplegic cerebral palsy. Methods The surface electromyograms signals of biceps brachii of 10 children with hemiplegic cerebral palsy during passive isometric contraction were recorded. Results The intact side registered a stronger EMG than that of the affected side, the integrated electromyography(iEMG)and root mean square(RMS)levels from the intact side were higher than those of affected side significantly(P<0.05).Conclusion The results of electromyographic analysis are consistent with Modified Ashworth Scale.Electromyographic analysis can provide a satisfactory evaluation of the muscle tone in children with hemiplegic cerebral palsy.

Chinese Journal of Rehabilitation Theory and Practice 2008;14(10):958-959
Objective To observe the effect of static balance test and training equipment on the equilibrium function in children with spastic diplegia. Methods 24 children with spastic diplegia were treated with visual feedback balance training besides routine rehabilitation training. All cases were assessed with Berg Balance Scale(BBS) and the static balance test and training equipment when eyes opened and closed, before and after the 8-week training. Results After the 8 week-training,the BBS scores of all children were higher than that before training(P<0.05).In the segment of the static balance test, fall index, weight distribution index and stability index decreased significantly(P<0.05~0.01).Conclusion The equilibrium function of the children with cerebral palsy can be assessed and pertinent training can be performed with balance equipment.

Objective To evaluate the antitumor effect of corosolic acid and its impact on CAM and YSM angiogenesis. Methods The effects of CRA on A549 proliferation was studied by MTT method in vitro.Tumor-bruden nude mice model were established by injecting A549 lung cancer cell marked with bioluminescent to nude mice, and the growth of tumor in mice were detected by IVIS small animal in vivo imaging system.Experiment of chicken embryos eggs are used to observe the role of drugs on CAMand YSMblood vessels. Results The value of IC50 of CRA for A549 in vitro was 26.8μg/mL.A tumor-burdened animal experiment results showed that the CRA to A549 solid tumor has a certain therapeutic effect.CAM and YSM blood vessels of chicken embryos eggs treated with CRA were decreased significantly than negative control group. Conclusion CRA has certain therapeutic effect for A549,which may be related to the inhibition of angiogenesis in tumor tissues.

Objective Within human hepatoma cell lines,we aimed to investigate the effects of the down-regulation by RNAi on different fragments of osteopontin (OPN) in order to discover more effective and accurate sites for OPN.Methods Specific small interfering RNA of OPN (OPNi-1) were synthesized and transfected into human hepatoma cell line (HEP-G2).Fluorescent quantitative PCR and immunohistochemical methods were used to test*the OPN expression levels of mRNA and protein before and after RNAi.Results After transfection, the △CT value of the A fragment was greater than B and C fragments of OPN mRNA in HEP-G2.Before RNAi was added to HEP-G2 cells, the three fragments A,B,C had OPN mRNA CT values of 8.31±1.58,8.78±1.49,8.25±1.51 respectively.Once the RNAi were added, the CT values were measured 48h after for the fragments A,B,and C which were 12.14±1.43,10.22±1.97,10.48±1.88 (P<0.05) respectively. The immunohis tochemical values of A,B,C were down from 6.44±1.67,5.43±2.05,5.45±2.52 to 2.84±1.52,4.43± 1.65,3.95± 1.43 respectively after interference. Conclusions RNAi can inhibit the expression of OPN gene selectively.siRNA targets different segments of OPN,which may have more effects on invasion and metastasis of liver cancer for a more important significance in science and health economics.

Methods A retrospective study was conducted to 297 cases of using Shuxuening Injection in our hospital from January 2016 to June 2016. Results In the 297 cases, 58.9% patients were over 60 years old, and indication coincidence rate of Shuxuening Injection was 52.2%. 91.2% single dose met the instructions and 6.7%solvent use met instructions. Shuxuening Injection in the joint medication also had irrational usage phenomenon. Conclusion There is irrational usage of Shuxuening Injection in our hospital, especially in indication and solvent.
