1.Treatment extracranial carotid artey stenosis with endovascular stent

Objective To discuss the indications, methods and potential complications of endovascular stent placement for stenosis of internal carotid artery (ICA). Methods Symptomatic with ICA stenosis greater than 50%, and asymptomaticwere with ICA stenosis greater than 70% were selected. Self-expanding stent were used in extracranial ICA. Results 40 patients were inplaced 48 stents. All patients had no complications during and after operation. Each case was succssful at instant agiograph. Follow-up X-ray and sonography showed no distortion, movement of stents and restenosis. Conclusions Endovascular stent angioplasty was a safe, effective for stenosis of extracranial ICA. How to prevent complications occur including the embolic particles and restenosis must be studied deeply.
3.Study on association of HLA-DRB1 alleles with gastric carcinoma and Helicobacter pylori infection

Objective To investigate the association of HLA DRB1 alleles with gastric carcinoma and H. pylori infection. Methods HLA DRB1 alleles polymorphism in 63 patients with gastric adenocarcinoma and 136 normal controls were tested with the polymerase chain reaction/sequence specific primer(PCR/SSP) technique. The status of H. pylori infection was determined by histology of gastric biopsy specimens and /or by ELISA. Results Both HLA DRB1 0901 and HLA DRB1 12 were positively associated, and HLA DRB1 15 was negatively associated with gastric adenocarcinoma, but no significant associations were found between patients with and without above HLA DRB1 alleles with regard to patients' age at presentation, sex ratio, tumor site (distal vs proximal), TNM staging, histological grading of tumor or status of H. pylori infection. Conclusions Our results suggest that HLA DRB1 0901, DRB1 12 and DRB1 15 are associated with gastric adenocarcinoma, but are not associated with the clinical features of gastric adenocarcinoma and the status of H. pylori infection.
4.The association between intraplaque hemorrhage of cerebral arteries and acute cerebral infarction

Objective To evaluate the relationship between intraplaque hemorrhage of cerebral arteries and acute cerebral infarcts. Methods 35 patients with severe stenosis in M1 segment of middle cerebral arteries (MCA)were included in this study.Intracranial TOF MRA (time of flight MR angiography)was performed to detect the stenosis in MCA,and DWI (diffusion weighted imaging) was performed to detect cerebral acute infarcts.T1 MPRAGE sequence was positioned on the stenosis in M1 segment of MCA,and intraplaque hemorrhage was determined according to high signal on T1 MRRAGE images.35 patients were divided into two groups:one group with intraplaque hemorrhage and the other group without intraplaque hemorrhage.Whether there was significant differ-ence in the incidences of acute cerebral infarcts between the two groups were determined byχ2 test.Results Intraplaque hemorrhage was detected in 1 5 patients,in which 12 patients had acute cerebral infarcts.There were no intraplaque hemorrhage in 20 patients,in which 9 patients had acute cerebral infarcts.There was significant difference in the incidences of acute cerebral infarcts between the two groups (P =0.046 <0.05).Conclusion There is a higher incidence of acute cerebral infarcts in patients with intraplaque hem-orrhage than those without intraplaque hemorrhage.Intraplaque hemorrhage in severe stenosis of cerebral arteries is a high-risk indi-cation for acute cerebral infarction.
5.Determination of cocaine in rat's serum by high performance liquid chromatography

Objective To develop an assay method for quantitative analysis of cocaine in rat's serum by high-performance liquid chromatography (HPLC). Method Serum samples (30?l) were extracted with 2% isoamyl alcohol and hexane. The organic phase was extracted with 0.1 mol/L HC1. The quantitative analysis was achieved by using a 5?m reversed-phase Kromasil CIS column with mobile phase of 0.04mol/ L phosphate buffer solution (containing 0.26 mmol/L tetrabutylammonium chloride , pH3.2) - acetonitrile (83:17). The wave length of the ultraviolet detection was performed at 235nm. Results The lowest limit of the detection limit was 25ng/ml. The recovery of the cocaine was 71 %. Conclusion The method is sensitive, simple and easy to perform.
6.Video-assisted Thoracoscopic Surgery for Spontaneous Pneumothorax:Report of 61 Cases

Objective To evaluate the efficacy of video-assisted thoracoscopic surgery(VATS)for spontaneous pneumothorax.Methods From January 2003 to June 2006,VATS was carried out in 61 cases of spontaneous pneumothorax in our hospital.Resection or ligatation of pulmonary bullae,and pleurodesis were performed by using Endo-GIA.Results Among the cases,unilateral VATS was performed on 58 patients(an axillary small incision was made in one case),and bilateral VATS was done in 3.No conversion to open surgery or severe postoperative complications occurred in this series.Pulmonary bullae was detected in 56 of all the patients.The operation time was 35-55 min(mean,46 min)for unilateral VATS,and was 85,175,and 190 min respectively for the 3 cases of bilateral VATS.The mean blood loss for unilateral operation was 40 ml(range,30-45 ml),and the blood loss in the 3 cases of bilateral VATS was 55,60,and 200 ml.The thoracic-drainage volume was 230-500 ml(mean,390 ml)in the unilateral cases,and was 350,1030,and 1200 respectively in the 3 bilateral cases.The drainage tube was withdrawn 3-6 d(mean 4 d)later in the unilateral cases,and 3,4,and 5 d later in the 3 bilateral cases.The 61 patients were followed up for 4-24 mon(mean,8 mon),during which one patient experienced recurrent pneumothorax in 2 months and was cured by closed thoracic drainage.Conclusions VATS is safe and effective for patients with spontaneous pneumothorax.
7.Effect of natural killer cells treated by serum of severe preeclampsia patient on apoptosis and endothelin-1 secretion of endothelial cells

Objective To investigate the effect of natural killer (NK) cells treated by serum of severe preeclampsia patient on the function of endothelial cells.Methods Fifteen patients with severe preeclampsia and 15 normal pregnant women from the Obstetrics department,Shengjing Hospital,China Medical University were admitted into this case-control study from January 1,2006 to December 31,2008.NK cells from healthy non-pregnant woman were incubated with 20% serum from severe preeclampsia patients or normal pregnant women for 20 hours.Then,human umbilical vein endothelial cells ( HUVEC) and serum-treated NK cells were co-incubated for 24 hours.Apoptosis of HUVEC was checked by flow cytometry and electronic microscope.Endothelin-1 (ET-1) levels in the supernatants of HUVEC and NK cells were examined by radioimmunologic method.Results In severe preeclampsia group,the percentage of early apoptosis cell (Annexin V-FITC+ /PI+ ) was (23.81±4.79)%,that of late apoptosis cell (AnnexinV-FITC+/PI+ ) was (3.29±1.04) %,while those were (16.59±5.13)% and (2.24±0.72)% respectively in normal pregnant group (P<0.01).There was no significant difference in dead cells (Annexin V-FITC- /PI+ ).Under electronic microscope,typical morphologic changes of apoptosis were shown in severe preeclampsia group.Level of ET-1 in
8.Protective role of tea polyphenols in oxidative stress damage of the rat articular cartilage tissue caused by brick-tea fluorosis

Objective To explore the protective mechanism of tea polyphenols (TPs) ion oxidative stress damage of the rat articular cartilage tissue caused by brick-tea fluorosis. Methods One hundred and twenty wistar male rats were randomly divided into 6 groups according to body mass: fluoride group with drinking water containing 100.00 mg/L F-, fluoride plus TPs group treated with 100.00 mg/L F- and 10.0 g/L TPs, fluoride plus aluminum group fed with 100.00 mg/L F- and 200.00 mg/L Al3+, fluoride plus aluminium and TPs group treated with 100.00 mg/L F-,200.O0 mg/L Al3+ and 10.0 g/L TPs;brick-tea group treated with drinking water containing 100.00 mg/L F-,215.00 mg/L Al3+ and 9.2 g/L TPs, which was steeped by the brick-tea;control group treated with tap water. The animals were bred for three months and then sacrificed. The level of SOD,T-AOC and MDA in blood serum were detected,also the level of NO and cytokine IL-1β and IL-6, the expression of iNOS mRNA and protein in articular cartilage were respectively analyzed by RT-PCR and immunohistochemistry. Results Blood serum SOD level in the fluoride plus aluminum and TPs group[(664.009 ± 29.589)kU/L] was higher compared with that in the fluoride group[(625.328 ± 27.199)kU/L], fluoride plus aluminum group[(652.282±13.926)kU/L], although no statistically significant differences was found(P > 0.05) ;blood serum T-AOC level of the fluoride plus TPs, fluoride plus aluminum and TPs group, brick tea group[(10.874 ± 0.721), (11.871 ± 0.941), (10.380 ± 2.747)kU/L] was higher compared with fluoride group, fluoride plus aluminum group [(8.849 ± 1.887), (8.210 ± 1.740)kU/L], the differences all being statistically significant(P < 0.05) ;blood serum MDA level in the fluoride plus aluminum and TPs group[(3.235 ± 0.446)μmol/L] had significances compared with fluoride group, fluoride plus aluminum group [(3.889 ± 0.387), (4.580 ± 0.474)μmol/L, all P < 0.05)];blood serum NO level in fluoride plus Tps group, fluoride plus aluminum and TPs group, brick-tea group[(23.278 ± 2.386), (20.643 ± 2.623), (24.367 ± 6.072) μmol/L] had tatistical differences compared with fluoride group, fluoride plus aluminum group[(32.962 ± 8.268), (34.909 ± 6.288)μmol/L, all P < 0.05];blood serum IL-1β level of fluoride group, fluoride plus aluminum, fluoride plus Tps, fluoride plus aluminum and TPs group and brick-tea group [(4.728 ± 0.297), (4.412 ± 0.229), (4.432 ± 0.285), (4.516 ± 0.351), (4.614 ±0.2270)n/L] did not have inter-group differences (F = 2.314,P > 0.05);the blood serum IL-6 level of fluoride plus aluminum and TPs group, brick-tea group[(7.231 ± 0.596), (7.325 ± 0.290)ng/L] had statistical differences compared with fluoride plus aluminum[(8.256 ± 0.635)ng/L, P < 0.05]. The iNOS mRNA correspondent expression content of fluoride plus Tps group, fluoride plus aluminum and TPs group, brick-tea group(0.482 ± 0.021,0.447±0.021,0.491 ± 0.022) had statistical differences compared with fluoride group, fluoride plus aluminum group (0.562 ± 0.025,0.591 ± 0.020, all P < 0.05). Cells with positive iNOS protein expression of control group were mainly distributed at the surface layer of joint, while the cells of experiment groups were distributed both at the surface layer and the intermediate layer. Conclusions Tea polyphenols could alleviate oxidative stress damage on the articular cartilage, exerting protection against brick-tea fluorosis on rats through cleaning up free radicals, elevating total anti-oxidation capability, diminishing the generation of lipid peroxide.
9.Role of perforin in severe preeclampsia

Objectives To investigate the possible role of perforin (PFN) in the pathogenesis of severe preeclampsia.Methods Thirty-two cases of severe preeclampsia were included in the study.Thirtytwo cases of normal pregnancy were selected as control group in random.The expression of PFN mRNA in the peripheral blood mononuclear cells (PBMC) was detected by reverse transcription (RT)-PCR, and its correlation with mean arterial pressure was analyzed in severe preeclamptic patients.The expression of PFN protein in the decidua was detected by immunohistochemistry.Results ( 1 ) The expression of PFN mRNA in PBMC:the PFN mRNA level in severe preeclamptic group was 1.19 ± 0.31, and that in normal pregnancy group is 0.82 ± 0.28.The PFN mRNA level in severe preeclamptic group was significantly higher than that of control group (P < 0.0l ).(2)Correlation analysis:the mean blood pressure in severe preeclampsia group was (133 ±5) mm Hg( 1 mm Hg =0.133 kPa).There was significant positive correlation between level of PFN mRNA in PBMC and mean blood pressure in severe preeclamptic patients ( r = 0.701, P = 0.000).(3)Decidual PFN protein expression:PFN protein was mainly expressed in lymphocytes and the cytoplasm of decidual stromal cells.The positive ratio of PFN in the decidua of severe preeclamptic patients was 84% ( 27/32), significantly higher than that of control group (53%, 17/32, P < 0.01 ).Conclusions Expression of PFN was significantly increased in severe preeclampsia, and it was of significant positive correlation with mean blood pressure.PFN may participate in the pathogenesis of severe preeclampsia.
